In recent years, recurrent droughts have significantly affected spring barley production, reducing the quantity and quality of grain. This study aims to identify genotype-specific traits and the drought resilience of six different Hordeum vulgare L. (spring barley) genotypes, while also examining the potential of potassium application and fungal endophyte Serendipita indica inoculation to mitigate the negative effects of dry periods during the growing season. Field experiments were conducted over a three-year period from 2020 to 2022, measuring physiological, growth, and yield parameters. To get insight into the physiological state of the plants, we measured the soluble sugars content and the ratio of stable carbon isotopes in the flag leaf tissue, which reflects conditions during its formation. The dominant factors that influenced the measured parameters were the genotypes and seasons, as well as their interaction, rather than other experimental factors. The results showed that the Spitfire and Accordine varieties were the best performing in both the 2020 and 2021 seasons, as indicated by their yield. However, in the drier 2022 season, the yield of these two varieties decreased significantly (to 55% for Spitfire and to 69% for Accordine of their yield in 2021), while for the arid-region genotypes, it remained at the same level as the previous year. This study sheds light on the potential of various genotypes to withstand periods of drought and the effectiveness of using potassium application and S. indica inoculation as mitigation approaches.

Wheat and barley genera represent a wide range of genotypes from Triticeae group grown around the globe. The broad plasticity of Triticeae phenotypes mirrors the robustness of their genomes revealing a high level of gene homeology. Publication and annotation of the reference genome sequences for spring barley Morex and Chinese Spring wheat represents an important milestone enabling the researchers to precisely identify and annotate nearly all proteins. Due to the broad range of environments used for wheat and barley cultivation and their economical importance, proteomic studies focused on their responses to environmental stresses including combined stress treatments. Most of the Triticeae stress proteomics studies are comparative ones aimed at determination of differentially abundant proteins (DAPs) between two or more genotypes with contrasting stress tolerance. Studies focused on subcellular fractions and protein posttranslational modifications (PTMs) are still relatively rare although PTMs play a crucial role in modulation of protein biological function. Functional and interactomics studies are needed although gene homeology and the resulting protein functional redundancy practically excludes the utilization of knock-out mutants. The alternatives could represent either gene overexpression in a heterologous system such as A. thaliana or transient posttranscriptional gene silencing using RNAi. Publication of complete reference genome sequences together with novel technological approaches such as pQTL mapping boost the Triticeae proteomics studies not only to provide data but also to contribute to designing novel genotypes with improved adaptations to ever changing environments.

The main problems of crop gene banks comprise heterogeneity of accessions, resulting from mechanical admixtures or out-crossing during their multiplication, and especially the mislabeling of accessions. These discrepancies can adversely affect the results of many expensive research and breeding projects that are based on the use of gene bank resources. To tackle these problems, 860 single-plant progenies (SPPs) of 172 accessions of the Czech winter barley core collection were grown and tested with a set of 53 isolates representing the global virulence/avirulence diversity of powdery mildew. Seventy-one resistance phenotypes encompassed the diversity of known specific resistances and their combinations. Based on testing groups of five SPPs, 94 accessions had one phenotype found in all five SPPs (homogeneous accessions), whereas in 78 accessions (45.3%) more than one phenotype was identified (heterogeneous accessions). In three varieties, specific resistances against the whole set of isolates were detected, but due to high adaptability of the pathogen, they are not recommended for breeding resistant cultivars. Selected SPPs were integrated in the gene bank and are now a reliable source of genotypically pure seed with defined powdery mildew resistance genes that can be used by breeders and researchers. The results obtained can be used to verify authenticity of accession genotype and pedigree, particularly for older varieties for which no other original criteria are available.

A cross-sectional field study on the prevalence of Dicrocoelium dendriticum was performed in the Emmental. The study included 211 bovines, 170 equines, 20 ovines, 46 caprines and 23 rabbits (from 119 farms). In addition, laboratory routine diagnostic data obtained from 2.840 animals--all originating from the same area of investigation--were assessed in the same way. The infection extent concerning the different animal species were the following: bovines 46%, equines 12%, ovines 30%, caprines 48% and rabbits 9%. Univariate analyses of baseline epidemiological data identified no significant risk factors, with the exception of the type of stable used. Bovines kept in a modern free ranging stable had a significantly lower chance of infection with D. dendriticum than cattle in conventional tie stalls. The epidemiological data characterizing the area of investigation suggest the following procedure to reduce the problem of dicrocoeliosis: Pasturing animals of all ages should be regularly dewormed (e.g. every six week during pasture) using a compound effective against D. dendriticum. A treatment is especially indicated at the time after pasture in autumn or before housing the animals in winter. In spring, only animals having pastured the year before need to be treated prior to pasture in the new year. However, it is recommended to perform an economic analysis comparing costs of treatment versus putative costs of damage prior to the initiation of a strategic campaign: animal welfare aspects have to be considered. The laboratory routine diagnostic data showed infection extent similar to those of the cross-sectional study: bovines 60%, equines 24%, ovines 26%, caprines 31%, rabbits 32%. Atypical hosts such as dogs and cats exhibited low infection extent (3% and 1%, respectively), rather reflecting a gastro-intestinal passage of parasite eggs ingested by consumption of infected livers or by coprophagy of ruminant faeces.
