RESUMO

Three distinct microsporidia were identified from parasitic copepods in the northeast Pacific Ocean. Sequencing and phylogenetic analysis of a partial small subunit ribosomal RNA gene (SSU rDNA) sequence identified a genetically distinct variety of Desmozoon lepeophtherii from Lepeophtheirus salmonis on cultured Atlantic salmon Salmo salar, and this was confirmed by transmission electron microscopy. Phylogenetic analysis resolved the SSU rDNA sequence of the second organism in a unique lineage that was most similar to microsporidia from marine and brackish water crustaceans. The second occurred in L. salmonis on Atlantic, sockeye Oncorhynchus nerka, chum O. keta and coho O. kisutch salmon, in Lepeophtheirus cuneifer on Atlantic salmon, and in Lepeophtheirus parviventris on Irish Lord Hemilepidotus hemilepidotus. Replication occurred by binary fission during merogony and sporogony, diplokarya were not present, and all stages were in contact with host cell cytoplasm. This parasite was identified as Facilispora margolisi n. g., n. sp. and accommodated within a new family, the Facilisporidae n. fam. The third, from Lepeophtheirus hospitalis on starry flounder Platichthys stellatus, was recognized only from its unique, but clearly microsporidian SSU rDNA sequence. Phylogenetic analysis placed this organism within the clade of microsporidia from crustaceans.

A new species of sphaerosporid myxosporean, Sphaerospora elwhaiensis sp. n., is described from kidney of non-anadromous sockeye salmon (kokanee) Oncorhynchus nerka (Walbaum) from Lake Sutherland in the northern Olympic Peninsula, Washington, USA. Infection with the parasite was detected in 45% of 177 kokanee examined over 5 years. While conforming to the morphological criteria by which members of the genus are defined, the parasite is distinguished from congeners in salmonids of western North America by a unique combination of valvular sculpting of the myxospore, the relatively large size of the myxospore and monosporous development within the pseudoplasmodium. In addition, nucleotide sequences of the parasite's small and large subunit ribosomal RNA gene are unique. Phylogenetic analyses of these sequences suggested that the parasite is most closely related to freshwater Myxidium spp. and Zschokkella spp. The molecular data have provided further evidence for a polyphyletic association previously recognized among members of the genus and emphasize the need for a taxonomic revision of Sphaerospora Thélohan, 1892 and related genera.

Myxospores consistent with species of Parvicapsula were observed in kidney of 15 of 95 (15.8%) adult pink salmon, Oncorhynchus gorbuscha, collected from the Quinsam River, British Columbia, Canada. The spores were elongate and curved with unequal valves, and 2 spherical-to-subspherical polar capsules within a highly refractile capsular region. The spores were unlike those of P. minibicornis found in nearby populations of Pacific salmon, Oncorhynchus spp. The spore dimensions were similar to those of Parvicapsula pseudobranchicola from Atlantic salmon, Salmo salar, in Norway, and the spores seemed similar to an undescribed Parvicapsula sp. from several Oncorhynchus spp. in Puget Sound, Washington. A sequence of 1,480 base pairs (bp) of the small subunit ribosomal RNA gene (SSU rDNA) of the parasite from pink salmon was most similar to, but distinct from, that of other Parvicapsula spp. The parasite is described as a new species, Parvicapsula kabatai n. sp. Polymerase chain reactions amplified a 158-bp sequence, unique to P. kabatai n. sp., from 22 of 93 (23.7%) adult pink salmon kidney samples, from 3 of 3 juvenile pink salmon collected in the ocean 125 km north of the Quinsam River, and from 2 of 5 archival coho salmon, Oncorhynchus kisutch, samples from Puget Sound. The parasite occurs within the lumen and epithelium of renal tubules and ducts, and within the renal interstitium. Concurrent infections with extrasporogonic stages of the myxosporean CKX, the microsporidian Loma salmonae, and a Myxidium sp. also were observed in the adult pink salmon.

Infections with sea lice species belonging to Lepeophtheirus and Caligus are reported from examinations of 1,309 three-spine sticklebacks collected in coastal British Columbia. Over 97% of the 19,960 Lepeophtheirus specimens and nearly 96% of the 2,340 Caligus specimens were in the copepodid and chalimus developmental stages. The parasites were identified as Lepeophtheirus salmonis and Caligus clemensi based on morphology of adult stages. Between 1,763 and 1,766 base pairs (bp) of 18S rDNA from adult specimens collected from sticklebacks and salmon differed from the GenBank L. salmonis reference sequence by a single bp and were distinct from those of 2 other Lepeophtheirus species. A 530-bp region of 18S rDNA from chalimus stages of Lepeophtheirus obtained from sticklebacks and salmon was identical to that of the L. salmonis reference sequence. The three-spine stickleback is a new host record for L. salmonis. The prevalence of L. salmonis was 83.6% and that of C. clemensi was 42.8%. The intensities of these infections were 18.3 and 4.2, respectively. There was no significant relationship between sea lice abundance and stickleback condition factor. Significant spatial and temporal variations both in abundance of sea lice and surface seawater salinities were measured. The abundance of both sea lice species was lowest in zones in which surface seawater salinity was also lowest. Sticklebacks appear to serve as temporary hosts, suggesting a role of this host in the epizootiology of L. salmonis. The stickleback may be a useful sentinel species with which to monitor spatial and temporal changes in the abundance of L. salmonis and C. clemensi.

Polymerase chain reaction (PCR) and microscopic examination of stained kidney sections were used to diagnose infections with the myxozoan parasite Parvicapsula minibicornis in maturing Fraser River salmon. In 2 series of collections, the parasite was detected in 109 of 406 migrating sockeye salmon Oncorhynchus nerka belonging to Early Stuart, Early Summer and Summer run-timing groups, mainly upper Fraser River stocks. However, the parasite was detected neither in fish at sea nor once they had migrated several 100 km upstream. Prevalence then increased to 95% or greater at the spawning grounds. Histological examination of kidney was less sensitive than PCR in detecting the parasite in salmon collected from the earliest sites in both collections found positive by PCR. Severity of infection was greatest at the spawning grounds. Development of infection in sockeye, measured by prevalence, severity or by the rate of false-negative histological diagnoses, appeared to be a useful estimate of in-river residence time. Prevalence and severity of infections in sequential samples of Harrison River and Weaver Creek sockeye stocks collected from the Harrison River indicated that more time had elapsed since parasite transmission than would be predicted based on migration distance alone. Pink salmon Oncorhynchus gorbuscha, coho salmon O. kisutch and chinook salmon O. tshawytscha were found to be infected with the parasite. Development of P. minibicornis in pink salmon was most similar to that in sockeye. Pink and coho salmon may be at risk to the pathological consequences of P. minibicornis infection.

The myxosporean parasite Parvicapsula minibicornis is described from adult sockeye and coho salmon during spawning migrations in tributaries of the Columbia River in Canada and the United States. These observations extend the known distribution of this parasite from the Fraser River drainage basin. The parasite was identified in Columbia River salmonids using polymerase chain reaction (PCR) and by in situ hybridization, but unlike in Fraser River salmon, it was not observed in conventional histological preparations of the kidney. Prevalence of the parasite determined by PCR was higher in spawning sockeye from the Fraser River than in those from the Okanagan River. Our ability to explain the relatively low prevalence and absence of clinical P. minibicornis infections in Columbia River salmon is hampered by our poor understanding of the life cycle of this parasite.

An unidentified myxosporean parasite (CKX) is described from the kidney of approximately 80% of spawning coho salmon Oncorhynchus kisutch (Walbaum) in British Columbia, Canada and Washington, United States of America. Morphological features were described using light and electron microscopy. Sequencing of polymerase chain reaction (PCR) amplified 18S ribosomal RNA gene and in situ hybridisation were used to further characterise CKX. The parasite occurred with a focal distribution within tubule epithelial cells, the tubule lumen and the interstitium as primary cells containing from one to at least 16 secondary cells. Luminal stages were degenerate and sporogony was not observed. In situ hybridisation using a digoxygenin-labelled DNA probe confirmed CKX to be the source of DNA used in PCR analyses. CKX 18S rDNA sequences were most similar (97%) to those of Sphaerospora oncorhynchi. Phylogenetic analysis revealed similarities among the 18S rDNA sequences of CKX, S. oncorhynchi and Myxidium lieberkuehni. CKX is hypothesised to be the abortive extrasporogonic development of a Sphaerospora sp. or Myxidium sp. occurring in immune-incompetent spawning and senescent salmon.

The prevalence, intensity, and abundance of sea lice belonging to Lepeophtheirus or Caligus clemensi are reported from threespine stickleback (Gasterosteus aculeatus) collected from the Broughton Archipelago region of coastal British Columbia, Canada, between 2005 and 2008. In total, 25,130 sea lice were collected from 7,684 sticklebacks. The prevalence of Lepeophtheirus ranged from 51% in 2005 to 11% in 2008 and that of C. clemensi from 56% in 2007 to 24% in 2008. Chalimus stages accounted for approximately 69% of all Lepeophtheirus and 88% of Caligus specimens. Cytochrome c oxidase subunit 1 (COI) gene sequences, useful in distinguishing reference specimens belonging to 8 species of Lepeophtheirus, Caligus, and Bomolochus, were used to identify the Lepeophtheirus specimens from stickleback as L. salmonis (71%) and L. cuneifer (29%). A COI phylogenetic analysis confirmed a monophylogenetic origin of Lepeophtheirus but not of Caligus. Two genotypes were resolved in L. cuneifer, i.e., genotype A occurred twice as often as genotype B. Virtually all immature Lepeophtheirus specimens from juvenile salmon were L. salmonis. The results emphasized the need to accurately identify immature sea lice as a prerequisite to understanding sea lice ecology. The threespine stickleback may be a useful sentinel species for the abundance and diversity of the sea lice that are also parasites of wild and farmed salmon in coastal ecosystems in British Columbia.

Ciliated protists were isolated from the ovarian fluid of apparently healthy adult rainbow trout (Oncorhynchus mykiss) maintained in freshwater. The organism was identified as Pseudocohnilembus persalinus based on morphometric and morphological analysis of silver-stained specimens obtained from culture and on analysis of ribosomal RNA gene sequences. The cytochrome c oxidase subunit 1 gene sequence of this organism also was characterized. This ciliate has been reported previously as free living only in saline environments and as an endosymbiont in a marine teleost, the olive flounder (Paralichthys olivaceus). A cyst-like stage may have facilitated the novel occurrence of this organism as an endosymbiont in rainbow trout.
