RESUMO

Viral pathogens pose a substantial threat to public health and necessitate the development of effective remediation and antiviral strategies. This short communication aimed to investigate the antiviral efficacy of disinfectants on the surface proteins of human pathogenic viruses. Using in silico modeling, the ligand-binding energies (LBEs) of selected disinfectants were predicted and combined with their environmental impacts and costs through an eco-pharmaco-economic analysis (EPEA). The results revealed that the binding affinities of chemical disinfectants to viral proteins varied significantly (p < 0.005). Rutin demonstrated promising broad-spectrum antiviral efficacy with an LBE of -8.49 ± 0.92 kcal/mol across all tested proteins. Additionally, rutin showed a superior eco-pharmaco-economic profile compared to the other chemicals, effectively balancing high antiviral effectiveness, moderate environmental impact, and affordability. These findings highlight rutin as a key phytochemical for use in remediating viral contaminants.

A phytochemical study on Spermacoce ocymoides has led to the isolation of a novel bis-indole alkaloid, spermaocymine A (2), together with the known alkaloid 4-methyl-borreverine (1), as well as an anthraquinone, 8-hydroxy-2-(hydroxymethyl)-1-methoxyanthracene-9,10-dione (3). The structures of the isolated compounds were elucidated by analyzing spectroscopic and spectrometric data, including one-dimensional (1D)- and 2D-NMR and high resolution (HR)-MS. Newly isolated alkaloid 2 was a C-3,14-stereoisomer of 1, the first natural stereoisomer of related bis-indoles containing an indeno[1,2-b]indole skeleton with an epiminoethano bridge. When 1-3 were assayed against five tumor cell lines including multi-drug resistant cells, compound 1 exhibited potent antiproliferative activity with IC50 values of 6.2-11.5 µM.

A simple and efficient BF3-OEt2 promoted C3-alkylation of indole has been developed to obtain3-indolylsuccinimidesfrom commercially available indoles and maleimides, with excellent yields under mild reaction conditions. Furthermore, anti-proliferative activity of these conjugates was evaluated against HT-29 (Colorectal), Hepg2 (Liver) and A549 (Lung) human cancer cell lines. One of the compounds, 3w, having N,N-Dimethylatedindolylsuccinimide is a potent congener amongst the series with IC50 value 0.02 µM and 0.8 µM against HT-29 and Hepg2 cell lines, respectively, and compound 3i was most active amongst the series with IC50 value 1.5 µM against A549 cells. Molecular docking study and mechanism of reaction have briefly beendiscussed. This method is better than previous reports in view of yield and substrate scope including electron deficient indoles.

Five new quinoline alkaloids, paliasanines A-E (1-5), and 17 known compounds (6-22) were isolated from a methanol extract of Melochia umbellata var. deglabrata leaves. Their chemical structures were elucidated by analysis of HRMS and 1D and 2D NMR spectroscopic data. Compounds 1-5 are the first naturally occurring 3,4-methylenedioxyquinolines incorporating an oxabicyclo[3.2.1]octane unit. Compounds 6 and 7 displayed selective cytotoxicity (IC50 5.9-8.4 µM) against A549 and MCF-7 cell lines, while compounds 1-5 were not active. Compounds 1-3 did not exhibit an anti-HIV effect in MT4 cells, although the related quinolone derivative waltherione A exhibited significant activity. These preliminary results indicate that the 3-methoxy-4-quinolone skeleton might be preferred for both antiproliferative and anti-HIV activities.

To encourage recycling in Malaysian households, a waste separation at source programme was implemented that made it mandatory for households to sort their waste into different categories before leaving it out for collection. Penalties designed to act as a deterrent are imposed on households that fail to sort their waste appropriately. But does this deterrence motivate compliance with the programme directives? This study employs the deterrence theory to investigate if deterrence alone is sufficient to motivate households to actively participate in recycling. A total of 866 questionnaires were administered in person in households in Putrajaya and Melaka. Data were analysed using structural equation modelling. The results of the structural model reveal that just 25% of the variance in compliance is explained by the severity of the sanction, and the perceived certainty of penalty imposed. Perceived severity of sanction (ß = 0.149, p = 0.012) and perceived certainty of sanction (ß = 0.383, p = 0.000) were found to contribute significantly to compliance behaviour to the programme directives. However, deterrence alone cannot motivate household participation in waste separation at source because it only explains 25% of compliance behaviour. The present study provides information that could allow policymakers to understand recycling habits better and implement more effective compliance strategies.

OBJECTIVE: To determine the frequency of impaired renal functions and hypertension in rheumatoid arthritis. METHODS: This study was conducted between May 1st 2018 to February 1st 2019 at Rheumatology Division, Department of Medicine Central Park Medical College Lahore, total 260 study participants were selected, demographic detail were asked in detail, disease duration of RA and hypertension, DMARD's, self-use NSAID's,/hakeem medications, smoking were asked in detail, BMI and blood pressure were measured,5 ml of blood was taken by trained phlebotomist, and sent for the estimation of serum urea and creatinine on (COBAS-III) machine, after availability of results each individuals eGFR (creatinine clearance) was calculated by Cockroft Gualt(CG) and Modification in diet in renal disease method (MDRD). RESULTS: In this study the mean age of study participants was 42.4 (± 9.5) years with disease duration of 7.7(±4.8) years, prevalence of Impaired renal functions of 14.6% (n=38) and hypertension in 53.5% (n=139).Regression analysis shows there is significant association between hypertension, smoking and self/hakeem medications with impaired renal functions (p-0.5). Kappa analysis shows both (MDRD & CG methods) had uniformity in picking up cases of impaired renal functions 75.6% (p-0.05). CONCLUSION: In RA decline in renal functions is seen with self-use NSAID's/hakeem medications along with other modifiable factors like smoking and hypertension, while conventional DMARD's don't show association with decline. There is very high prevalence of hypertension in rheumatoid arthritis.

RESUMO

A heterogeneously integrated InP-on-silicon fast tunable filtered feedback laser is demonstrated. The laser device consists of a main Fabry-Pérot cavity connected to an integrated arrayed waveguide grating of which the outputs form external cavities in which semiconductor optical amplifiers can be switched to provide single-mode operation and tunability. The laser can operate at four different wavelengths whereby switching between each wavelength channel is done within one nanosecond. For each wavelength channel 12.5 Gbit/s NRZ-OOK direct modulation is demonstrated. The combination of fast wavelength switching with straightforward wavelength control and high-speed direct modulation characteristics make the demonstrated laser structure very attractive for use in optical packet or burst switching systems.

Iodine-promoted oxidative C-H/C-H cross-coupling of unprotected anilines and 2-methylquinoline to furnish C4-carbonylated aniline (4-aminophenyl)(quinoline-2-yl) methanones in moderate to good yields has been demonstrated. This work provides the first site-selective approach for the synthesis of free amino groups containing methanones including unprecedented C-H functionalization rather than the N-H functionalization of unprotected anilines via the Kornblum oxidation of 2-methylquinoline. Furthermore, we noticed that the incorporation of KOH under standard conditions provides 2-heteroarylbenzothiazoles from benzothiazoles and 2-methylquinoline in good to excellent yields. These transformations do not require any transition metals or peroxides and tolerate various functional groups such as methoxy, hydroxy, bromo, chloro and nitro groups. Moreover, a plausible mechanistic pathway is proposed.

RESUMO

A novel cycloartane triterpenoid alkaloid, kleinhospitine E (1), six new cycloartane triterpenoids (2-7), three known cycloartane triterpenoids (8-10), and taraxerone (11) were isolated from a methanol extract of Kleinhovia hospita. Their structures were elucidated by 1D- and 2D-NMR spectroscopy as well as HRMS analysis. The absolute configurations of all isolated compounds were determined from their ECD spectra by comparison with theoretical values. Kleinhospitine E (1) is the first cycloartane alkaloid possessing an unusual γ-lactam with an oxopropylidene side chain. Compounds 2, 3, and 6 were assigned as cycloartane triterpenoids with a 9α,10α-cyclopropyl ring, which is found rarely among naturally occurring compounds, while 4 and 5 were established as isomers of compound 3 containing a 21,23-diacetal side chain. Biological evaluation revealed that compounds 4 and 9 exhibited more potent antiproliferative activities against a multidrug-resistant tumor cell line compared with its parent chemosensitive cell line. Furthermore, compound 6 exhibited submicromolar anti-HIV activity.

Although vascular occlusion has long been noted in peri-burn tissue, the literature is inconsistent regarding the nature of the occlusion, with articles in the 1940s claiming that erythrocytes were the culprit and in the 1980s-1990s that microthrombi were responsible. To better define the nature of vessel occlusion, we studied two porcine burn models, a hot comb horizontal injury model and a vertical injury progression model. In both cases, tissue from the first two days after burn were stained with hemotoxylin and eosin, or probed for platelets or for fibrinogen/fibrin. Erythrocytes, identified as nonstained, clumped, anuclear, 5 µm cells, occluded most blood vessels (BVs) in both burn models. In contrast, platelet or fibrinogen/fibrin antibodies stained BV occlusions minimally at early time points, and only up to 16% of deep dermal BVs at 48 hours in the hot comb model and up to 7% at 24 hours in the vertical injury progression model. Treatment of animals with a fibronectin-derived peptide (P12), which limits burn injury progression and can dilate peripheral microvasculature, reduced erythrocyte occlusion by at least 50%, speeded healing and reduced scarring. Early erythrocyte aggregation, rather than thrombosis, explains the ineffectiveness of anticoagulants to prevent burn injury progression.

2-Styryl quinolines (9a-l) have been synthesized regioselectively from 2-methyl-quinoline by using NaOAc in water acetic acid binary solvents and evaluated for their antibacterial activity against both Gram-positive and Gram-negative strains. Among these, the compounds 12 and 8 were found to be active against both bacterial strains. Compounds 9b, 9f, 9g, 9i, 9j and 9k were the most active among the series exhibiting MIC values ranging between 1.9 and 31.2 µg ml(-1) against different bacterial strains. Compounds 9j and 9k were found to be as potent as the standard drug ciprofloxacin against Micrococcus luteus, Klebsiella planticola and Staphylococcus aureus. In addition, the compounds showed bactericidal activity; compound 9j was found to be better than ciprofloxacin, with an MBC value of 0.9 µg ml(-1) against both M. luteus and K. planticola. The compounds also inhibited biofilm formation, and compound 9j was found to be equipotent to erythromycin against M. luteus and S. aureus MLS16. Further, theoretical studies such as those on druggable properties and PMI plot have been carried out.

A series of compounds consisting of 25 novel oxadiazole-benzohydrazone hybrids (6-30) were synthesized through a five-step reaction sequence and evaluated for their ß-glucuronidase inhibitory potential. The IC50 values of compounds 6-30 were found to be in the range of 7.14-44.16µM. Compounds 6, 7, 8, 9, 11, 13, 18, and 25 were found to be more potent than d-saccharic acid 1,4-lactone (48.4±1.25µM). These compounds were further subjected for molecular docking studies to confirm the binding mode towards human ß-d-glucuronidase active site. Docking study for compound 13 (IC50=7.14±0.30µM) revealed that it adopts a binding mode that fits within the entire pocket of the binding site of ß-d-glucuronidase. Compound 13 has the maximum number of hydrogens bonded to the residues of the active site as compared to the other compounds, that is, the ortho-hydroxyl group forms hydrogen bond with carboxyl side chain of Asp207 (2.1Å) and with hydroxyl group of Tyr508 (2.6Å). The other hydroxyl group forms hydrogen bond with His385 side chain (2.8Å), side chain carboxyl oxygen of Glu540 (2.2Å) and Asn450 side-chain's carboxamide NH (2.1Å).

A plant used in an Indonesian traditional herbal medicine as a diabetes treatment and known locally as "Jampu Salo" was collected on Sulawesi Island, Indonesia. It was identified as Syzygium oblanceolatum (C. B. Rob.) Merr. (Myrtaceae) and found for the first time in Sulawesi; it was previously reported only in the eastern Philippines and Borneo. A phytochemical study of S. oblanceolatum led to the isolation of three unprecedented meroterpenoids, syzygioblanes A-C (1-3, respectively). These compounds might be biosynthesized through [4+2] cycloaddition of various germacrane-based cyclic sesquiterpenoids with the flavone desmethoxymatteucinol to form a spiro skeleton. The unique and complex structures were elucidated by microcrystal electron diffraction analysis in addition to general analytical techniques such as high-resolution mass spectrometry, various nuclear magnetic resonance methods, and infrared spectroscopy. Synchrotron X-ray diffraction and calculations of electronic circular dichroism spectra helped to determine the absolute configurations. The newly isolated compounds exhibited collateral sensitivity to more strongly inhibit the growth of a multidrug resistant tumor cell line compared to a chemosensitive tumor cell line.

There are many advantages of the hydropower industry, as an environmentally friendly resource, and also some challenges that need to be overcome to fully exploit this sustainable and renewable natural resource. The primary objective of this study is to find out the impact of hydropower factors and economic growth on the agriculture industry output among the EU27 nations within the time factor 1990 to 2021. Adopting the autoregressive distributed lag (ARDL), the findings show a significant positive effect could occur in agriculture industry growth of the European Union (EU13)-emerging economies using hydropower factors than in EU14-emerged economies. On one hand, among additional factors, economic growth and institutional quality contribute more positively to agriculture growth in EU13 economies than in EU14 economies. On the other hand, population density contributes more negatively to agriculture growth in EU13 economies than in EU14 economies. The findings show there can be a positive significant growth increase in the EU13 agriculture industry using fossil fuel output than in EU14 economies. The results show that growth could be sustained in the agricultural industry of the European nations by increasing the level of hydropower production as this will help in attaining sustainable development by the year 2030. This will therefore help in mitigating the effect of climatic changes due to environmental pollution. The projected calculations were seen to be reliable and valid and this was attested to by the three estimators used in the study (pooled mean group, mean group, and dynamic fixed effect). This study recommended that European nations could leverage hydroelectricity to achieve sustainable growth and development. The legislative arms of the government of these European nations should as well show more interest in green energy to achieve security and sustainable development in hydroelectricity production. Decision-makers in the EU nations should buttress more emphasis on sustainable means through which hydropower could be used to attain sustainable irrigation systems for the agriculture industry and thus minimize the demand for fossil fuels and reduce CO2-related emissions in the future tine ahead.

There are many advantages of geothermal energy as an environmentally friendly resource; however, there are quite a several challenges that need to be overcome to completely harness sustainable and renewable energy that is also natural. The primary aim of this study is to examine what influence geothermal energy will have on land use changes among the considered 27 states in the European Union from the time being 1990 to 2021. The study adopts the auto-regressive distributed lag (ARDL); the findings show that geothermal energy growth could be leveraged to achieve remarkable growth in land use change among the 13 European developing economies than among the 14 EU developed economies. On the other hand, results from analysis further show that a remarkable decrease in land use change could be better attained among the 14 EU developed economies that among the 13 EU developing economies as a result of institutional quality. Furthermore, the result suggests that through economic growth, there could be a remarkable increase in land use change among the 14 EU developed economies than among the 13 EU developing economies. It was further revealed by the study that the level of land use change among the 27 EU nations could be remarkably increased, boosting the level of geothermal energy production that will assist in attaining the aims behind the 2030 energy union. This will eventually help in curbing the incidence of climate change and pollution in the environment; the projected calculations are observed to be valid, as confirmed through the chosen three estimators for this research. The chosen estimators are the pooled mean group, mean group, and dynamic fixed effect. The regulations and governors in 27 European Union countries should give priority to using geothermal in their renewable energy mix to reduce the incidence of changes in land structures. Also, an increased level of efficiency and effectiveness should be made to the generation of geothermal energy by state actors and investors to prompt sustainability and attainability with no further depreciation in agricultural and forest natural states.

There are many advantages of geothermal energy, as an environmental friend resource. This heat radiation emanating from beneath the earth's surface presents man with good opportunities to harness it and makes a good level of agricultural food production and its processing in the EU region. The primary objective of this research is to examine the impact of geothermal energy on agri-food supply among the 27 European countries (EU27), within the time frame 1990 to 2021. The study adopted the autoregressive distributed lag (ARDL), and the findings from this study revealed that agri-food supply can increase significantly among the 13 European countries (EU13 emerging economies), leveraging on geothermal energy and economic growth variables than in the EU14 emerged economies. Furthermore, the outcome of this study showed that there could be a significant decrease in the food products coming from agricultural practices among the 13 European countries (EU13 emerging economies), due to an ineffective population density than in EU14 emerged economies. Furthermore, fossil fuel and institutional quality contribute more positively to the agri-food supply in the EU14 emerged economies than in the EU13 emerging economies. This results in an outcome that means that the agri-food supply among the EU13 emerging economies could be greatly boosted by replacing fossil fuel consumption with geothermal energy, and this facilitates the attainment of the European energy goals by the year 2030. Substituting fossil fuels with geothermal will also assist in minimizing the risks of environmental pollution and climate change. All projected calculations were seen as valid in this study, and this was confirmed by the three estimators adopted which are the pooled mean group, the mean group, and the dynamic fixed effect. This study, therefore, recommends that the 27 European countries should lay more emphasis on geothermal energy production as this will help in ensuring food security in the region. Policymakers and other government authorities as well as local and foreign investors should make more investments in geothermal energy resources as this study has proven that this will lead to agri-food security and sustainability. OBJECTIVE: The main goals of this investigation were to i) assess the population structure and genetic diversity and ii) determine the efficiency of the ongoing breeding program in a closed flock of Angora rabbits through pedigree analysis. METHODS: The pedigree records of 6,145 animals, born between 1996 to 2020 at NTRS, ICAR-CSWRI, Garsa were analyzed using ENDOG version 4.8 software package. The genealogical information, genetic conservation index and parameters based on gene origin probabilities were estimated. RESULTS: Analysis revealed that, 99.09% of the kits had both parents recorded in the whole dataset. The completeness levels for the whole pedigree were 99.12%, 97.12%, 90.66%, 82.49%, and 74.11% for the 1st, 2nd, 3rd, 4th, and 5th generations, respectively, reflecting well-maintained pedigree records. The maximum inbreeding, average inbreeding and relatedness were 36.96%, 8.07%, and 15.82%, respectively. The mean maximum, mean equivalent and mean completed generations were 10.28, 7.91, and 5.51 with 0.85%, 1.19%, and 1.85% increase in inbreeding, respectively. The effective population size estimated from maximum, equivalent and complete generations were 58.50, 27.05, and 42.08, respectively. Only 1.51% of total mating was highly inbred. The effective population size computed via the individual increase in inbreeding was 42.83. The effective numbers of founders (fe), ancestors (fa), founder genomes (fg) and non-founder genomes (fng) were 18, 16, 6.22, and 9.50, respectively. The fe/fa ratio was 1.12, indicating occasional bottlenecks had occurred in the population. The six most influential ancestors explained 50% of genes contributed to the gene pool. The average generation interval was 1.51 years and was longer for the sireoffspring pathway. The population lost 8% genetic diversity over time, however, considerable genetic variability still existed in the closed Angora population. CONCLUSION: This study provides important and practical insights to manage and maintain the genetic variability within the individual flock and the entire population.

OBJECTIVE: This study isolated the chemical compounds and evaluated the cytotoxic activity of the crude hexane extract of Cleome rutidospermae herb (CRH). METHODS: The isolate was purified using silica gel, column chromatography, and preparative thin layer chromatography (PTLC). Furthermore, the structure of the compounds was identified by spectroscopic methods using 1D, 2D NMR, and mass spectrometry. The cytotoxic activity of CRH at a concentration of 20 ug/mL was also tested against MCF-7, A549, KB, KB-VIN, and MDA-MB-231 cancer cells using the sulforhodamine B (SRB) method. RESULTS: The CRH contained compounds of unsaturated fatty acid, saturated fatty acid, lipid, glycerol, ω-3 fatty acid, and cholesterol. Two compounds were obtained from the plant, and their structures were identified as (1) Stigmasta-5,22-dien-3-ol (STML) and (2) 1,2-Benzene dicarboxylic acid, 1,2-bis (2-Ethylhexyl) esters (DEHP). These compounds were reported in this plant for the first time. In comparison, CRH had % growth inhibition in the proliferation of MCF-7 cells up to 28.1%, with cancer cells A549, KB, KB-VIN, and MDA-MB-231 by >50% Compared to the negative DMSO of 0.20%, while the positive control could inhibit the growth of all cancer cells (100%). CONCLUSION: Our findings suggested that crude herb from the plant CRH was the potential for breast cancer treatment.

Safflower (Carthamus tinctorius L.) has been explored as a source of natural antioxidant. However, quercetin 7-O-beta-D-glucopyranoside and luteolin 7-O-beta-D-glucopyranoside, as its bioactive compounds, possessed poor aqueous solubility, limiting its efficacy. Here, we developed solid lipid nanoparticles (SLNs) decorated with hydroxypropyl beta-cyclodextrin (HPßCD) incorporated into dry floating gel in situ systems to control the release of both compounds. Using Geleol® as a lipid matrix, SLNs were <200 nm in size with >80 % of encapsulation efficiency. Importantly, following the decoration using HPßCD, the stability of SLNs in gastric environment was significantly improved. Furthermore, the solubility of both compounds was also enhanced. The incorporation of SLNs into gellan gum-based floating gel in situ provided desired flow and floating properties, with <30 s gelation time. The floating gel in situ system could control the release of bioactive compounds in FaSSGF (Fasted-State Simulated Gastric Fluid). Furthermore, to assess the effect of food intake on release behavior, we found that the formulation could show a sustained release pattern in FeSSGF (Fed-State Simulated Gastric Fluid) for 24 h after being released in FaSGGF for 2 h. This indicated that this combination approach could be a promising oral delivery for bioactive compounds in safflower.

In most carbon dioxide (CO2) capture processes, chemical absorption using an amine solvent is widely used technology; however, the solvent is prone to solvent degradation and solvent loss which leads to the formation of corrosion. This paper investigates the adsorption performance of amine-infused hydrogels (AIFHs) to increase carbon dioxide (CO2) capture by leveraging the potency of amine absorption and adsorption properties of class F fly ash (FA). The solution polymerization method was used to synthesize the FA-grafted acrylic acid/acrylamide hydrogel (FA-AAc/AAm), which was then immersed in monoethanolamine (MEA) to form amine infused hydrogels (AIHs). The prepared FA-AAc/AAm showed dense matrices morphology with no obvious pore at the dry state but capable of capturing up to 0.71 mol/g CO2 at 0.5 wt% FA content, 2 bar pressure, 30 °C reaction temperature, 60 L/min flow rate, and 30 wt% MEA contents. Cumulative adsorption capacity was calculated and Pseudo-first order kinetic model was used to investigate the CO2 adsorption kinetic at different parameters. Remarkably, this FA-AAc/AAm hydrogel is also capable of absorbing liquid activator that was 1000% more than its original weight. FA-AAc/AAm can be used as an alternative AIHs that employ FA waste to capture CO2 and minimize the GHG impact on the environment.
