RESUMO

We are developing a robust and economic electro-optical remote sensing methodology to monitor the state of health and hydration of trees, endemic to subtropical regions. We measured reflectance spectra with Fourier transform infrared (FTIR) of three samples of two different oak trees. We find that spectral bands suitable for monitoring the state of the health and senescence of the oak include intervals around 0.9 µm and 1.8 µm. The easiest and the most cost-effective strategy would be to implement an electro-optical remote sensing radiometric system featuring a commercial camera incorporating a traditional charge-coupled device (CCD) or complementary metal-oxide-semiconductor (CMOS) detectors and a wideband transmission filter, from about 0.8 to 1 µm.

RESUMO

Purpose: To analyse the relation between antioxidant genotypes and Dietary Antioxidant Quality score (DAQs) effect on prostate cancer (PCa) risk and aggressiveness in a Spanish population. Methods: Men (N = 155 patients and 152 controls) with PSA values >4 ng/ml were enrolled in the project. DAQs were used considering the daily recommended intake for Spanish people (DRI). Genotyping of 5 SNPs rs662 (PON1), rs10432782 (SOD1), rs4880 (SOD2), rs17650792 (GPX1) and rs1001179 (CAT) were included for the analysis. Results: rs17650792 was statistically significant between case and controls subjects. When comparing D´Amico risk, we found that rs662 (CC), rs10432782 (G allele) and rs17650792 (GG) confer a protection. When testing SNP-antioxidant nutrients interactions, we found an intake of vitamin A and rs100179 (T carriers) and selenium and rs17650792 (G carriers) confers a protection of being in low risk classification. Conclusions: We reported by the first time a correlation between rs662 (PON1) and PCa aggressiveness.

RESUMO

Nanoscale plasmonic particles represent a crucial transformation on optical and electronic properties exhibited by advanced materials. Herein are reported remarkable interferometric optical effects with dependence on polarization for filtering or modulating electronic signals in multilayer nanostructures. Metallic nanoparticles were incorporated in randomly distributed networks of reduced graphene oxide by an in-situ vapor-phase deposition method. The polarization-selectable nonlinear optical absorption contribution on the photoconductivity of reduced graphene oxide decorated with gold nanoparticles was analyzed. Nanosecond pulses at 532 nm wavelength were employed in a two-wave mixing experiment to study photoconduction and nonlinear optical absorption in this nanohybrid material. The ablation threshold of the sample was measured in 0.4 J/cm2. Electrochemical impedance spectroscopy measurements revealed a capacitive response that can be enhanced by gold decoration in carbon nanostructures. A strong two-photon absorption process characterized by 5 × 10-7 m/W was identified as a physical mechanism responsible for the nonlinear photoconductive behavior of the nanostructures. Experimental shift of 1 MHz for the cutoff frequency associated with an electrical filter function performed by the sample in film form was demonstrated. Moreover, amplitude modulation of electronic signals controlled by the polarization of a two-wave mixing experiment was proposed. All-optical and optoelectronic nanosystems controlled by multi-photonic interactions in carbon-based materials were discussed. The key role of the vectorial nature of light in two-wave mixing experiments is a fascinating tool for the exploration of low-dimensional systems.

RESUMO

Polo-like kinase 1 (PLK1) is a serine/threonine kinase involved in several stages of the cell cycle, including the entry and exit from mitosis, and cytokinesis. Furthermore, it has an essential role in the regulation of DNA replication. Together with cyclin A, PLK1 also promotes CDH1 phosphorylation to trigger its ubiquitination and degradation, allowing cell cycle progression. The PLK1 levels in different type of tumors are very high compared to normal tissues, which is consistent with its role in promoting proliferation. Therefore, several PLK1 inhibitors have been developed and tested for the treatment of cancer. Here, we further analyzed PLK1 degradation and found that cytoplasmic PLK1 is ubiquitinated and subsequently degraded by the SCFßTrCP/proteasome. This procedure is triggered when heat shock protein (HSP) 90 is inhibited with geldanamycin, which results in misfolding of PLK1. We also identified CDK1 as the major kinase involved in this degradation. Our work shows for the first time that HSP90 inhibition arrests cell cycle progression at the G1/S transition. This novel mechanism inhibits CDH1 degradation through CDK1-dependent PLK1 destruction by the SCFßTrCP/proteasome. In these conditions, CDH1 substrates do not accumulate and cell cycle arrests, providing a novel pathway for regulation of the cell cycle at the G1-to-S boundary.-Giráldez, S., Galindo-Moreno, M., Limón-Mortés, M. C., Rivas, A. C., Herrero-Ruiz, J., Mora-Santos, M., Sáez, C., Japón, M. Á., Tortolero, M., Romero, F. G1/S phase progression is regulated by PLK1 degradation through the CDK1/ßTrCP axis.

RESUMO

In 2015, a predictive model for invasive bacterial infection (IBI) in febrile young infants with altered urine dipstick was published. The aim of this study was to externally validate a previously published set of low risk criteria for invasive bacterial infection in febrile young infants with altered urine dipstick. Retrospective multicenter study including nine Spanish hospitals. Febrile infants ≤90 days old with altered urinalysis (presence of leukocyturia and/or nitrituria) were included. According to our predictive model, an infant is classified as low-risk for IBI when meeting all the following: appearing well at arrival to the emergency department, being >21 days old, having a procalcitonin value <0.5 ng/mL and a C-reactive protein value <20 mg/L. IBI was considered as secondary to urinary tract infection if the same pathogen was isolated in the urine culture and in the blood or cerebrospinal fluid culture. A total of 391 patients with altered urine dipstick were included. Thirty (7.7 %) of them developed an IBI, with 26 (86.7 %) of them secondary to UTI. Prevalence of IBI was 2/104 (1.9 %; CI 95% 0.5-6.7) among low-risk patients vs 28/287 (9.7 %; CI 95% 6.8-13.7) among high-risk patients (p < 0.05). Sensitivity of the model was 93.3 % (CI 95% 78.7-98.2) and negative predictive value was 98.1 % (93.3-99.4). Although our predictive model was shown to be less accurate in the validation cohort, it still showed a good discriminatory ability to detect IBI. Larger prospective external validation studies, taking into account fever duration as well as the role of ED observation, should be undertaken before its implementation into clinical practice.

RESUMO

The overall incidence of spinal tuberculosis (TB) appears to be stable or declining in most European countries, but with an increasing proportion of cases in the foreign-born populations. We performed a retrospective observational study (1993-2014), including all cases of spinal TB diagnosed at a Barcelona hospital to assess the epidemiological changes. Fifty-four episodes (48·1% males, median age 52 years) of spinal TB were diagnosed. The percentage of foreign-born residents with spinal TB increased from 14% to 45·2% in the last 10 years (P = 0·017). Positive Mycobacterium tuberculosis testing in vertebral specimens was 88·2% (15/17) for GeneXpert MTB/RIF. Compared with natives, foreign-born patients were younger (P < 0·01) and required surgery more often (P = 0·003) because of higher percentages of paravertebral abscess (P = 0·038), cord compression (P = 0·05), and persistent neurological sequelae (P = 0·05). In our setting, one-third of spinal TB cases occurred in non-native residents. Compared with natives, foreign-born patients were younger and had greater severity of the disease. The GeneXpert MTB/RIF test may be of value for diagnosing spinal TB.

RESUMO

Principal component analysis (PCA) is a variable reduction method used on over-parameterized data sets with a vast number of variables and a limited number of observations, such as Dairy Herd Improvement (DHI) data, to select subsets of variables that describe the largest amount of variance. Cluster analysis (CA) segregates objects, in this case dairy herds, into groups based upon similarity in multiple characteristics simultaneously. This project aimed to apply PCA to discover the subset of most meaningful DHI variables and to discover groupings of dairy herds with similar performance characteristics. Year 2011 DHI data was obtained for 557 Upper Midwest herds with test-day mean ≥200 cows (assumed mostly freestall housed), that remained on test for the entire year. The PCA reduced an initial list of 22 variables to 16. The average distance method of CA grouped farms based on best goodness of fit determined by the minimum cophenetic distance. Six groupings provided the optimal fitting number of clusters. Descriptive statistics for the 16 variables were computed per group. On observations of means, groups 1, 2, and 6 demonstrated the best performances in most variables, including energy-corrected milk, linear somatic cell score (log of somatic cell count), dry period intramammary infection cure rate, new intramammary infection risk, risk of subclinical intramammary infection at first test, age at first calving, days in milk, and Transition Cow Index. Groups 3, 4, and 5 demonstrated the worst mean performances in most the PCA-selected variables, including DIM, age at first calving, risk of subclinical intramammary infection at first test, and dry period intramammary infection cure rate. Groups 4 and 5 also had the worst mean herd performances in energy-corrected milk, Transition Cow Index, linear somatic cell score, and new intramammary infection risk. Further investigation will be conducted to reveal patterns of management associated with herd categorization. The PCA and CA should be used when describing the multivariate performance of dairy herds and whenever working with over-parameterized data sets, such as DHI databases.

RESUMO

We construct a topological invariant that classifies density matrices of symmetry-protected topological orders in two-dimensional fermionic systems. As it is constructed out of the previously introduced Uhlmann phase, we refer to it as the topological Uhlmann number n_{U}. With it, we study thermal topological phases in several two-dimensional models of topological insulators and superconductors, computing phase diagrams where the temperature T is on an equal footing with the coupling constants in the Hamiltonian. Moreover, we find novel thermal-topological transitions between two nontrivial phases in a model with high Chern numbers. At small temperatures we recover the standard topological phases as the Uhlmann number approaches to the Chern number.

RESUMO

We introduce the Uhlmann geometric phase as a tool to characterize symmetry-protected topological phases in one-dimensional fermion systems, such as topological insulators and superconductors. Since this phase is formulated for general mixed quantum states, it provides a way to extend topological properties to finite temperature situations. We illustrate these ideas with some paradigmatic models and find that there exists a critical temperature Tc at which the Uhlmann phase goes discontinuously and abruptly to zero. This stands as a borderline between two different topological phases as a function of the temperature. Furthermore, at small temperatures we recover the usual notion of topological phase in fermion systems.

RESUMO

Information Retrieval focuses on finding documents whose content matches with a user query from a large document collection. As formulating well-designed queries is difficult for most users, it is necessary to use query expansion to retrieve relevant information. Query expansion techniques are widely applied for improving the efficiency of the textual information retrieval systems. These techniques help to overcome vocabulary mismatch issues by expanding the original query with additional relevant terms and reweighting the terms in the expanded query. In this paper, different text preprocessing and query expansion approaches are combined to improve the documents initially retrieved by a query in a scientific documental database. A corpus belonging to MEDLINE, called Cystic Fibrosis, is used as a knowledge source. Experimental results show that the proposed combinations of techniques greatly enhance the efficiency obtained by traditional queries.

RESUMO

Lymphocytes play a significant role in the immunological processes of the bovine mammary gland and were found to be the dominant cell population in the milk of healthy udder quarters. The objective of this study was to investigate the quantitative relationship between CD2(+) T and CD21(+) B lymphocytes using flow cytometry. In a first study, quarter foremilk samples from apparently healthy udder quarters [somatic cell counts (SCC) ≤100,000 cells/mL; n=65] were analyzed and compared with diseased quarters (SCC >100,000 cells/mL; n=15). Percentages of CD2(+) T cells were significantly higher in milk samples with SCC ≤100,000 cells/mL than in those with SCC >100,000 cells/mL, whereas percentages of CD21(+) B cells developed in the opposite direction. As a result of this opposing trend, a new variable, the CD2/CD21 index-representing the percentages of CD2(+) cells per CD21(+) cells-was defined. Although diseased quarters with SCC >100,000 cells/mL and the detection of major pathogens revealed generally CD2/CD21 indices <10, values >10 were observed in apparently healthy quarters. Hence, a CD2/CD21 index cutoff value of 10 may be suitable to aid differentiation between unsuspicious and microbiologically suspicious or diseased udder quarters. To test whether CD2/CD21 indices <10 were primarily related to pathogens, quarters with SCC ≤100,000 cells/mL and >100,000 cells/mL with different bacteriological status (culture negative, or minor or major pathogens) were selectively examined in a second biphasic study. In the first trial, 63 udder quarters were analyzed and 55 of these quarters were able to be sampled again in the second trial carried out 14 d later. In both trials, results of the first study were confirmed. Indeed, CD2/CD21 indices <10 were also found in quarters showing SCC ≤100,000 cells/mL and containing minor or major pathogens at the time of the current or previous bacteriological analysis. The results of our examinations indicated a clear relationship between the CD2/CD21 index and the bacteriological status of the mammary gland. In combination with SCC, it offers a new marker for quick differentiation of unsuspicious and microbiologically suspicious or diseased udder quarters.

RESUMO

BACKGROUND: Osteoid osteoma is a benign osteoblastic tumor with a nidus of <20 mm in maximum diameter. There are several treatment options, all of them aiming either to resect or to eliminate the nidus. PURPOSE: To report and to describe the benefits of treating non-spinal osteoid osteoma by percutaneous computed tomography-guided resection, according to our experience. STUDY DESIGN: Retrospective case series METHODS: Between 1992 and 2008, 54 patients with non-spinal osteoid osteoma underwent primary treatment with percutaneous CT-guided resection. In all cases, the materials obtained were processed for pathology and microbiology. RESULTS: Fifty-four patients with a mean age of 22.7 years (range 10-47), of whom 46 were males (85.2 %) and 8 were females (14.8 %). The lesion size ranged between 5 and 15 mm with an average size of 6.9 mm. The resection was considered complete by the CT study in all 54 cases. Of all the specimens sent to pathology, the histological diagnosis was achieved in 41 (75.9 %). Cure was obtained in 50 patients (92.6 %) and the other four patients required a second surgery using the same technique, after which all of them achieved clinical and radiological improvement (100 %). CONCLUSION: Percutaneous computed tomography-guided resection of non-spinal osteoid osteomas provides good results, similar to other surgical techniques, with the advantages of being a simple, mini invasive, safe and economic procedure without the need for specific materials. Level of evidence, IV.

RESUMO

Introduction: Physical and non-physical processes that occur in nature may influence biological processes, such as dissemination of infectious diseases. However, such processes may be hard to detect when they are complex systems. Because complexity is a dynamic and non-linear interaction among numerous elements and structural levels in which specific effects are not necessarily linked to any one specific element, cause-effect connections are rarely or poorly observed. Methods: To test this hypothesis, the complex and dynamic properties of geo-biological data were explored with high-resolution epidemiological data collected in the 2001 Uruguayan foot-and-mouth disease (FMD) epizootic that mainly affected cattle. County-level data on cases, farm density, road density, river density, and the ratio of road (or river) length/county perimeter were analyzed with an open-ended procedure that identified geographical clustering in the first 11 epidemic weeks. Two questions were asked: (i) do geo-referenced epidemiologic data display complex properties? and (ii) can such properties facilitate or prevent disease dissemination? Results: Emergent patterns were detected when complex data structures were analyzed, which were not observed when variables were assessed individually. Complex properties-including data circularity-were demonstrated. The emergent patterns helped identify 11 counties as 'disseminators' or 'facilitators' (F) and 264 counties as 'barriers' (B) of epidemic spread. In the early epidemic phase, F and B counties differed in terms of road density and FMD case density. Focusing on non-biological, geographical data, a second analysis indicated that complex relationships may identify B-like counties even before epidemics occur. Discussion: Geographical barriers and/or promoters of disease dispersal may precede the introduction of emerging pathogens. If corroborated, the analysis of geo-referenced complexity may support anticipatory epidemiological policies.

RESUMO

Woodchip bioreactors have gained popularity in many countries as a conservation practice for reducing nitrate load to freshwater. However, current methods for assessing their performance may be inadequate when nitrate removal rates (RR) are determined from low-frequency (e.g., weekly) concurrent sampling at the inlet and outlet. We hypothesised that high-frequency monitoring data at multiple locations can help improve the accuracy of quantifying nitrate removal performance, enhance the understanding of processes occurring within a bioreactor, and therefore improve the design practice for bioreactors. Accordingly, the objectives of this study were to compare RRs calculated using high- and low-frequency sampling and assess the spatiotemporal variability of the nitrate removal within a bioreactor to unravel the processes occurring within a bioreactor. For two drainage seasons, we monitored nitrate concentrations at 21 locations on an hourly or two-hourly basis within a pilot-scale woodchip bioreactor in Tatuanui, New Zealand. A novel method was developed to account for the variable lag time between entry and exit of a parcel of sampled drainage water. Our results showed that this method not only enabled lag time to be accounted for but also helped quantify volumetric inefficiencies (e.g., dead zone) within the bioreactor. The average RR calculated using this method was significantly higher than the average RR calculated using conventional low-frequency methods. The average RRs of each of the quarter sections within the bioreactor were found to be different. 1-D transport modelling confirmed the effect of nitrate loading on the removal process as nitrate reduction followed Michaelis-Menten (MM) kinetics. These results demonstrate that high-frequency temporal and spatial monitoring of nitrate concentrations in the field allows improved description of bioreactor performance and better understanding of processes occurring within woodchip bioreactors. Thus, insights gained from this study can be used to optimise the design of future field bioreactors.

RESUMO

This paper reports on a methodology to elaborate interdigitated nanoelectrode devices (INDs) at the wafer scale, relying on a mix-and-match process which combines proximity optical lithography and electron beam lithography. An optimum exposure dose allowed fabricating nanodevices, at the wafer level, with a successful yield of 97%. The final devices are bonded onto conventional TO-8 packages. Electrical characterization in a short-circuited nanoelectrode is performed, revealing a 230 µΩ cm resistivity value at 23 °C. A MEMS-based spotter made of cantilevers (called Bioplume) has been used to obtain precise functionalization of the INDs with sub-picoliter volume solutions. These INDs are the basis of multiple tunnel junction nanodevices, intended to serve as novel highly sensitive nanobiosensors.

RESUMO

The increase in children obesity worldwide has been of particular concern in recent decades. Environmental factors have been proposed as contributors to obesity, and there is a growing concern over obesogens, environmental chemicals with potential obesity-related endocrine-disrupting properties. In this regard, bisphenol A (BPA) and its analogues are suspected to have obesogenic properties. Current document report on the activities of the fellow, undertaken during the fourth, 2020-2021 cycle of the EU-FORA programme at the University of Granada, Institute of Nutrition and Food Science, in Spain. The work programme offered by the hosting site was related to the extrapolation of bisphenols exposure following the determination of these compounds in food frequently consumed by children and in their biological samples. The fellow has participated in the recruitment of the study population in the health centres. In addition, she has participated in the collection of the children biological samples, anthropometric measurements and dietary surveys and in the optimisation of the laboratory methodology for the extraction of bisphenols in biological samples. All these activities also provided the fellow an opportunity to develop her data science related skills, which will benefit her professional development. In addition, the fellow gained an overview of various topics related to food safety risk assessment by attending the EU-FORA dedicated training modules.

RESUMO

Excessive use of insecticides has led to resistance of some pathogenic organisms (nematodes, bacteria and fungi), environmental contamination, and the presence of hazardous residues. Therefore, the aim of the present study was to evaluate synthetic metabolites derived from previous studies with edible mushrooms against the soybean weevil Rhyssomatus nigerrimus Fahraeus (Curculonidae) because of the relevance of pest control in an economically important crop. Furthermore, this is one of the first studies where edible fungal molecules are evaluated for the control of these insects. Initially, two in vitro tests (toxic effect and immersion) were evaluated against R. nigerrimus. In these tests, sensitivity and viability were determined in the 2% Tween control in water. For these two tests, the synthetic metabolites pentadecanoic acid (PNA), palmitic acid (PMA), stearic acid (STA), linoleic acid (LNA), ß-sitosterol (ßT) were evaluated individually as well as in combinations, "the fraction of standards (E1)". Based on the results obtained, the dip test was selected to evaluate the mixtures of two standards (1. PMA + ßT, 2. PMA + PNA, 3. PMA + LNA, 4. PMA + STA, 5. STA + ßT, 6. STA + PNA, 7. STA + LNA, 8. PNA + ßT, 9. PNA + LNA, 10. LNA + ßT), three (1. PNA + ßT + LNA, 2. PNA + ßT + STA, 3. STA + LNA + PNA and 4. STA + LNA + ßT) and four (PNA, ßT, LNA and STA). The results showed that the mixture of three standards caused a higher percentage of mortality relative to the control group: l. PNA + ßT + LNA and 2. PNA + ßT + STA with 54.44 and 48% mortality of R. nigerrimus insects exposed for 15 days. These results show the importance of evaluating mixtures of molecules against R. nigerrimus.

RESUMO

A multi-stage municipal wastewater treatment system is proposed to comply with Mexican standards for discharge into receiving water bodies. The system is located in Santa Fe de la Laguna, Mexico, an area with a temperate climate. It was designed for 2,700 people equivalent (259.2 m3/d) and consists of a preliminary treatment, a septic tank as well as two modules operating in parallel, each consisting of a horizontal subsurface-flow wetland, a maturation pond and a vertical flow polishing wetland. After two years of operation, on-site research was performed. An efficient biochemical oxygen demand (BOD5) (94-98%), chemical oxygen demand (91-93%), total suspended solids (93-97%), total Kjeldahl nitrogen (56-88%) and fecal coliform (4-5 logs) removal was obtained. Significant phosphorus removal was not accomplished in this study (25-52%). Evapotranspiration was measured in different treatment units. This study demonstrates that during the dry season wastewater treatment by this multi-stage system cannot comply with the limits established by Mexican standards for receiving water bodies type 'C'. However, it has demonstrated the system's potential for less restrictive uses such as agricultural irrigation, recreation and provides the opportunity for wastewater treatment in rural areas without electric energy.

RESUMO

We study in this work the dynamics of a collection of identical hollow spheres (ping-pong balls) that rest on a horizontal metallic grid. Fluidization is achieved by means of a turbulent air current coming from below. The upflow is adjusted so that the particles do not levitate over the grid, resulting in quasi-two-dimensional dynamics. We show that the behavior of diffusion and correlations in this system is particularly rich. Noticeably as well (and related to the complex dynamical behavior), a variety of phases appear, with important peculiarities with respect to analogous setups. We observe gas, liquid, glass, and hexagonal crystal phases. Most notably, we show that the melting of the hexagonal crystal occurs in coexistence with a liquid phase. This strikingly differs from the corresponding transition in a purely two-dimensional systems of air-fluidized disks, for which no phase coexistence has been reported in the literature.

RESUMO

BACKGROUND: Immune checkpoint inhibitors (ICI) have been associated with several immune-related adverse events, including sarcoidosis-like reactions (SLR). SLR, which has a low prevalence but an increasing incidence, is similar to sarcoidosis in terms of histology, and clinical and radiological manifestations. The most commonly affected organs are hilar and mediastinal lymph nodes and skin. SLR is an exclusion diagnosis, so a lymph node biopsy can be useful to distinguish between tumor progression and SLR, particularly in tumors in which nodal involvement is very common. PATIENTS AND METHODS: We performed a retrospective analysis of SLR in all cancer patients receiving ICIs in our institution between January 2016 and June 2020. RESULTS: Among the 1063 treated patients, seven experienced SLR, four of whom were symptomatic (cough, skin lesions, arthralgia), with time to onset ranging from 1.5 to 6.7 months after ICI initiation. All seven patients had bilateral hilar lymphadenopathy, and granulomatous reactions in five of the six patients with lymph node biopsies. SLR improved in all patients, including four patients who continued with ICI. Three patients received corticosteroids and/or stopped ICI therapy. Four of these patients had partial responses at the time SLR was identified. CONCLUSION: Management of SLR lacks a consensus recommendation, although corticosteroids and/or stopping the ICI are generally implemented. The potential consequences of stopping anticancer treatment should be taken into consideration, particularly in the absence of clear management recommendations.
