RESUMO

Ubiquitin-specific peptidase 15 (USP15), a critical deubiquitinating enzyme, has been demonstrated to improve substrate stabilization by hydrolyzing the bond between the substrate and ubiquitin, and is implicated in multiple carcinogenic processes. Prompted by the information cited from The Cancer Genome Atlas (TCGA) database and the Cancer Proteogenomic Data Analysis Site (cProSite), USP15 is selectively overexpressed in clear cell renal cell carcinoma (ccRCC) samples. We aimed to investigate the function of USP15 on ccRCC malignant features, which was emphasized in its deubiquitination of SHC adaptor protein 1 (SHC1). The overexpression of USP15 promoted the capacity of proliferation, migration, and invasion in ccRCC CAKI1 and 769-P cells, and these malignant biological properties were diminished by USP15 deletion in 786-O cells. USP15 accelerated tumor growth and lung metastasis in vivo. In addition, deubiquitinase USP15 was further identified as a new protector for SHC1 from degradation by the ubiquitination pathway, the post-translational modification. In sequence, transcription factor activating enhancer binding protein 4 (TFAP4) was shown to be partly responsible for USP15 expression at the level of transcription, as manifested by the chromatin immunoprecipitation and pull-down assay. Based on the in vitro and in vivo data, we postulate that USP15 regulated by TFAP4 transcriptionally deteriorates ccRCC malignant biological properties via stabilizing SHC1 by deubiquitination.

KEY MESSAGE: The novel ZmR1CQ01 allele for maize anthocyanin synthesis was identified, and the biological function and regulatory molecular mechanisms of three ZmR1 alleles were unveiled. Anthocyanins in maize are valuable to human health. The R1 gene family is one of the important regulatory genes for the tissue-specific distribution of anthocyanins. R1 gene allelic variations are abundant and its biological function and regulatory molecular mechanisms are not fully understood. By exploiting genetic mapping and transgenic verification, we found that anthocyanin pigmentation in maize leaf midrib was controlled by ZmR1 on chromosome 10. Allelism test of maize zmr1 EMS mutants confirmed that anthocyanin pigmentation in leaf sheath was also controlled by ZmR1. ZmR1CQ01 was a novel ZmR1 allelic variation obtained from purple maize. Its overexpression caused the whole maize plant to turn purple. ZmR1B73 allele confers anthocyanin accumulation in near ground leaf sheath rather than in leaf midribs. The mRNA expression level of ZmR1B73 was low in leaf midribs, resulting in no anthocyanin accumulation. ZmR1B73 overexpression promoted anthocyanin accumulation in leaf midribs. Loss of exon 5 resulted in ZmR1ZN3 allele function destruction and no anthocyanin accumulation in leaf midrib and leaf sheath. DNA affinity purification sequencing revealed 1010 genes targeted by ZmR1CQ01, including the bz2 in anthocyanin synthesis pathway. RNA-seq analysis showed 55 genes targeted by ZmR1CQ01 changed the expression level significantly, and the expression of genes encoding key enzymes in flavonoid and phenylpropanoid biosynthesis pathways were significantly up-regulated. ZmR1 functional molecular marker was developed. These results revealed the effects of transcriptional regulation and sequence variation on ZmR1 function and identified the genes targeted by ZmR1CQ01 at the genome-wide level.

In recent years, morbidity and mortality of prostate cancer (PCa) have increased dramatically, while mechanistic understanding of its onset and progression remains unmet. LncRNA SNHG3 has been proved to stimulate malignant progression of multiple cancers, whereas its functional mechanism in PCa needs to be deciphered. In this study, our analysis in the TCGA database revealed high SNHG3 expression in PCa tissue. Further analysis in starBase, TargetScan, and mirDIP databases identified the SNHG3/miR-152-3p/SLC7A11 regulatory axis. FISH was conducted to assess the distribution of SNHG3 in PCa tissue. Dual-luciferase reporter gene and RIP assays confirmed the relationship among the three objects. Next, qRT-PCR and western blot were conducted to measure expression levels of SNHG3, miR-152-3p, and SLC7A11. CCK-8, colony formation, Transwell, and flow cytometry were carried out to assess proliferation, migration, invasion, methionine dependence, apoptosis, and the cell cycle. It was noted that SNHG3 as a molecular sponge of miR-152-3p stimulated proliferation, migration, and invasion, restrained methionine dependence and apoptosis, and affected the cell cycle of PCa cells via targeting SLC7A11. Additionally, we constructed xenograft tumor models in nude mice and confirmed that knockdown of SNHG3 could restrain PCa tumor growth and elevate methionine dependence in vivo. In conclusion, our investigation improved understanding of the molecular mechanism of SNHG3 modulating PCa progression, thereby generating novel insights into clinical therapy for PCa.

The concentration, sources and environmental risks of polycyclic aromatic hydrocarbons (PAHs) in surface water in urban areas of Nanjing were investigated. The range of ∑16PAHs concentration is between 4,076 and 29,455 ng/L, with a mean of 17,212 ng/L. The composition of PAHs indicated that 2- and 3-ring PAHs have the highest proportion in all PAHs, while the 5- and 6-ring PAHs were the least in proportion. By diagnostic ratio analysis, combustion and petroleum were a mixture input that contributed to the water PAH in urban areas of Nanjing. Positive matrix factorization quantitatively identified four factors, including coke oven, coal combustion, oil source, and vehicle emission, as the main sources. Toxic equivalency factors of BaP (BaPeq) evaluate the environmental risks of PAHs and indicate the PAH concentration in surface water in urban areas of Nanjing had been polluted and might cause potential environmental risks. Therefore, the PAH contamination in surface water in urban areas of Nanjing should draw considerable attention.

Background: Immune checkpoint genes (ICGs), which are the cornerstone of immunotherapy, influence the incidence and progression of clear cell renal cell carcinoma (ccRCC). It is important to note that there is not much data in the literature to determine how cuproptosis and antitumor immunity are related. Methods: On the basis of The Cancer Genome Atlas ccRCC dataset (n=526), cuproptosis-related ICGs (CICGs) were used to identify distinct molecular subtypes. Using the Cox regression method, a risk signature was constructed and externally validated using the ICGC (n=91) and primary ccRCC subgroups of GSE22541 (n=24). The molecular and immune characteristics and efficacy of immunotherapy in the subgroups defined by the risk score were investigated. Four risk CICGs were verified through in vitro experiment. Results: We identified two unique molecular subgroups with substantial prognostic differences based on 17 CICGs. The two subtypes clearly differ in terms of the tumor immune microenvironment (TME). A predictive risk signature (CD276, HLA-E, LGALS9, and TNFRSF18) was created and externally confirmed, and their expressions were validated by realtime PCR. The multivariate Cox regression analysis demonstrated that this signature could independently predict survival. Thus, a credible nomogram incorporating the signature, age, stage, and grade was constructed, and discrimination was confirmed using the C-index, calibration curve, and decision curve analyses. The underlying implications for immune checkpoint inhibitors, the landscape of the TME, and single-cell level localization are depicted. In addition, its accuracy in forecasting actual immunotherapeutic results has been proven (CheckMate025 and TCGA-SKCM cohorts). The sensitivity of the two risk groups to various drug-targeted therapy methods was analyzed. Conclusions: The data provided here provide the groundwork for creating customized therapeutic options for individuals with ccRCC. The findings also suggested that researching the cuproptosis-based pathway might improve ccRCC patient better prognosis, development of anti-tumor immunity, and therapeutic strategies for immunotherapy.

Vegetable oil is an important part of the human diet and has multiple industrial uses. The rapid increase in vegetable oil consumption has necessitated the development of viable methods for optimizing the oil content of plants. The key genes regulating the biosynthesis of maize grain oil remain mostly uncharacterized. In this study, by analyzing oil contents and performing bulked segregant RNA sequencing and mapping analyses, we determined that su1 and sh2-R mediate the shrinkage of ultra-high-oil maize grains and contribute to the increase in the grain oil content. Functional kompetitive allele-specific PCR (KASP) markers developed for su1 and sh2-R detected su1su1Sh2Sh2, Su1Su1sh2sh2, and su1su1sh2sh2 mutants among 183 sweet maize inbred lines. An RNA sequencing (RNA-seq) analysis indicated that genes differentially expressed between two conventional sweet maize lines and two ultra-high-oil maize lines were significantly associated with linoleic acid metabolism, cyanoamino acid metabolism, glutathione metabolism, alanine, aspartate, and glutamate metabolism, and nitrogen metabolism. A bulk segregant analysis and sequencing (BSA-seq) analysis identified another 88 genomic intervals related to grain oil content, 16 of which overlapped previously reported maize grain oil-related QTLs. The combined analysis of BSA-seq and RNA-seq data enabled the identification of candidate genes. The KASP markers for GRMZM2G176998 (putative WD40-like beta propeller repeat family protein), GRMZM2G021339 (homeobox-transcription factor 115), and GRMZM2G167438 (3-ketoacyl-CoA synthase) were significantly related to maize grain oil content. Another candidate gene, GRMZM2G099802 (GDSL-like lipase/acylhydrolase), catalyzes the final step of the triacylglycerol synthesis pathway and was expressed at significantly higher levels in the two ultra-high-oil maize lines than in the two conventional sweet maize lines. These novel findings will help clarify the genetic basis of the increased oil production in ultra-high-oil maize lines with grain oil contents exceeding 20%. The KASP markers developed in this study may be useful for breeding new high-oil sweet maize varieties.

Background: Ferroptosis is a new type of iron- and reactive oxygen species-dependent cell death, studies on ferroptosis-related long noncoding RNAs (FerLncRNAs) in clear cell renal cell carcinoma (ccRCC) are limited. The purpose of this study was to investigate the potential prognostic value of FerLncRNAs and their relationship with the immune microenvironment and immunotherapy response of ccRCC. Methods: RNA sequencing data of 526 patients with ccRCC were downloaded from The Cancer Genome Atlas (TCGA) database. The patients with ccRCC in TCGA were randomly divided (1:1) into a training and testing cohort. ICGC and GEO databases were used for validation. Screening for FerLncRNAs was performed using Pearson's correlation analysis with the reported ferroptosis-related genes. A FerLncRNA signature was constructed using univariate, LASSO, and multivariate Cox regression analyses in the training cohort. Internal and external datasets were performed to verify the FRlncRNA signature. Four major FRlncRNAs were verified through in vitro experiment. Results: We identified seven FerLncRNAs (LINC00894, DUXAP8, LINC01426, PVT1, PELATON, LINC02609, and MYG1-AS1), and established a risk signature and nomogram for predicting the prognosis of ccRCC. Four major FRlncRNAs were verified with the prognosis of ccRCC in the GEPIA and K-M Plotter databases, and their expressions were validated by realtime PCR. The risk signature can also effectively reflect the immune environment, immunotherapy response and drug sensitivity of ccRCC. These FRlncRNAs have great significance to the implementation of individualized treatment and disease monitoring of ccRCC patients.

BACKGROUND: The distant metastasis is the primary cause of cancer morbidity and mortality for bladder cancer (BLCA) paitents. All the recommended therapy for it largely depends on how far the cancer has invaded. It has been confirmed that epithelial to mesenchymal transition (EMT) is the leading reason for the BLCA metastasis which makes BLCA difficult to cure. The aim of the present study is to identify the BLCA-related genes that can be used as the new prognostic biomarker and treatment target, and to investigate the functional mechanisms of TEAD4 in EMT dysregulation. METHODS: The "limma" R package was used to identify the differentially expressed genes (DEGs) between the normal and the tumor samples from TCGA BLCA and GTEx databases. Kaplan-Meier and UniCox analysis were used to filter DEGs with prognostic value in BLCA. Step muti-Cox analysis was used to construct a prognostic risk score model based on clinical phenotype characters. Gene set enrichment analysis (GSEA) was performed to explore the possible molecular mechanisms affecting the prognosis in BLCA. Unsupervised hierarchical clustering analysis was performed to evaluate the effects of EMT process on the prognosis. Single-sample GSEA (ssGSEA) was used to calculate the correlation betweeen the expression of DEGs and EMT enrichment scores. TEAD4 expression and its association with pathological grading and survival were appraised in samples from TCGA dataset and BLCA tissue microarray. Colony formation assays and CCK8 assays were performed to study the changes in BLCA cell proliferation when the TEAD4 levels was down- or up-regulated in BLCA cells. Transwell and wound healing assays were utilized to analyze the impact of TEAD4 on the invasion and metastasis of the BLCA cells. Western Blot was carried out to detect the changes of EMT-related markers and the active molecules involved in PI3K/AKT signaling in BLCA cells. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis was conducted on the genes related to TEAD4 expression. 740Y-P (activator of PI3K/AKT pathway) and LY294002 (inhibitor of PI3K/AKT pathway) were applied to evaluate the contribution of PI3K/AKT signaling pathway in the EMT of BLCA cells. To examine the in vivo effect of TEAD4 on tumor metastasis, we designed a metastatic nude-mouse model by tail vein injection of TEAD4-knockdown BLCA cells. And PET/CT imaging was used to assess the extent of lung metastases. RESULTS: A total of 1592 DEGs were recognized, among which 4 DEGs have been identified as independent prognostic factors for BLCA, such as FASN, IGFL2, PLOD1 and TEAD4. TCGA BLCA samples were divided into significantly different low- and high-risk groups according to the median risk score; GSEA analysis showed that HALLMARK EMT pathway was the top enriched gene signature when compared high-risk and low-risk groups, which was also verified by unsupervised cluster analysis. EMT signature-derived ssGSEA scores demonstrated that TEAD4 had the most positive correlation with EMT process. In addition, TEAD4 expression was upregulated in TCGA BLCA samples and correlated with pT stage, tumor stage and tumor grade. Functional studies showed that TEAD4 knockdown via lentiviral TEAD4 shRNA inhibited cell migration and invasion in vitro and in vivo, with the reduced expression of EMT related markers in BLCA cell lines; the migration and invasion of TEAD4 knockdown cells could be restored by ectopic expression of TEAD4. Meanwhile, KEGG enrichment analysis of genes related to TEAD4 expression showed that enrichment was significantly related to PI3K/AKT pathway. The pathway inhibitor LY294002 blocked the TEAD4-induced enhancement of migration and invasion as well as the expression EMT-related markers, whereas the agonist 740Y-P rescued the decreased migration, invasion and EMT induced by TEAD4 knockdown. CONCLUSIONS: TEAD4 is closely correlated with poor prognosis in BLCA and mediates its metastasis through regulating EMT via PI3K/AKT pathway, proving that TEAD4 is not only an effective biomarker for predicting the prognosis but also a great potential target for treatment of metastatic BLCA.

Circular RNA (circRNA) is a long non­coding RNA molecule with a closed loop structure lacking a 5'cap and 3'tail. circRNA is stable, difficult to cleave and resistant to RNA exonuclease or RNase R degradation. circRNA molecules have several clinical applications, especially in tumors. For instance, circRNA may be used for non­invasive diagnosis, therapy and prognosis. Exosomes play a crucial role in the development of tumors. Exosomal circRNA in particular has led to increased research interest into tumorigenesis and tumor progression. Additionally, exosomal circRNA plays a role in cell­cell communication. Exosomal circRNA facilitates tumor metastasis by altering the tumor microenvironment and the pre­metastatic niche. Additionally, studies have revealed the mechanism by which exosomal circRNA affects malignant progression through signal transduction. Moreover, exosomal circRNA promotes tumor metastasis by regulating gene expression, RNA transcription and protein translation. In this review, the biological features and clinical application of exosomal circRNA are described, highlighting the underlying mechanisms through which they regulate tumor metastasis. The application of circRNA as clinical diagnostic biomarkers and in the development of novel therapeutic strategies is also discussed.

Waxy maize has many excellent characteristics in terms of its nutritional and economic value. In recent decades, the waxy maize germplasm has increased dramatically as a result of different selection methods. We collected 200 waxy maize inbred accessions from different origins to study their genetic diversity and phylogenetic relationships, and to identify new waxy mutations. A simple sequence repeat (SSR) analysis revealed wide genetic diversity among the 200 waxy maize accessions. The maize accessions were clustered into three groups. We sequenced the waxy gene from the first to the 14th exon. Nucleotide variation analysis of 167 waxy maize and 14 flint maize lines revealed some nucleotide differences in the waxy gene among different waxy maize groups, and much narrower nucleotide diversity in waxy maize than in flint maize. In a phylogenetic analysis, waxy maize carrying the same mutation allele clustered together, and waxy maize carrying different mutation alleles distributed in different groups; waxy maize was intermixed with flint maize in each branch, and wx-D7 waxy maize separated significantly from waxy maize lines carrying wx-D10, wx-124 and wx-hAT mutant alleles. The wx-hAT was a new waxy mutation identified in this study. It consisted of a 2286-bp transposon inserted into the middle of exon three of the waxy gene. A PCR marker specific for the wx-hAT allele was developed. These results will be useful for the utilization and preservation of the waxy maize germplasm, and the PCR marker has potential uses in waxy maize breeding programs.

In recent years, autophagy has become a research hotspot in the field of pancreatic adenocarcinoma (PAAD) due to its ambiguous roles in pancreatic tumor progression. Hence, it is necessary to assess its clinical significance in a larger cohort of patients with PAAD. Here, we identified autophagy-related genes with prognostic value in PAAD and constructed a risk model based on these genes. We found that patients in high-risk group were significantly associated with poor prognosis. Genome mutation analysis suggested that KRAS and TP53 mutations were significantly higher in high-risk groups. In addition, functional enrichment analysis showed that high-risk groups were associated with immune cell infiltration and tumor-associated signaling pathways. We further performed CIBERSORT analysis and observed increased macrophage infiltration in high-risk group, but decreased B and T cell counts compared to that in low-risk group. Gene set enrichment analysis indicated that the Hippo pathway was enriched in the high-risk group. Further, using weighted gene co-expression network analysis, Yes-associated protein 1 (YAP1) was identified as a critical hub gene. Interestingly, we found that the autophagy status and YAP1 expression status could influence each other, thus creating a positive feedback loop. In conclusion, in this study, we highlighted the clinical significance of autophagy in pancreatic cancer, constructed an autophagy-related prognostic predictive system, and identified a promising target for autophagy regulation in pancreatic cancer.

To explore the effects of different types of forest environments for forest therapy, this study focused on forest resting environments. Seven representative forest resting environments found in field research in Beijing were used as independent variables and were shown to subjects by a virtual reality (VR) video. Stress level was used as the dependent variable, and blood pressure, heart rate, salivary amylase, and the Brief Profile of Mood States (BPOMS) were used as physiological and psychological indicators. A between-subjects design was used in the experiment. A total of 96 subjects were randomly assigned to each environment type, and only one type of forest resting environment was observed. Through the relevant sample t-test and one-way analysis of variance, the pre- and post-test data of the indicators were analyzed. This study found that all the seven different types of forest resting environments can produce stress relief effects to some extent. Different types of forest resting environments have different effects on relieving stress. The most natural environment does not have the most significant effect on stress relief. A water landscape has a positive effect on the relief of stress. The conclusions of this study are conducive to the better use of the forest environment for forest therapy services.

Environmental pollution by heavy metals resulting from rapid economic development is a major concern. Soil, water, wheat, and rice samples were collected from the Lihe River Watershed in the Taihu Region (east China). In this study area, many types of industrial plants, including ceramics factories, plants working with refractory materials, and chemical plants are densely distributed and cause serious heavy metal pollution. In addition, well-developed transportation and agricultural activities are also important sources of heavy metals. Thus, the concentrations of selected heavy metals including cadmium (Cd), chromium (Cr), copper (Cu), nickel (Ni), lead (Pb), and zinc (Zn) in the samples were analyzed to evaluate their potential integral risk (IR) to the health of the local population. Accordingly, the spatial distribution pattern of the IR values was determined in the study. The soil in the study area showed heavy Cd pollution, whereas the pollution by other elements was relatively slight. When the proportions of grain samples in which the concentrations exceeded the tolerance limits were examined, the grains were primarily contaminated with Pb, Ni, Cd, and Zn; and less contaminated with Cu and Cr. The drinking water of the local inhabitants was safe. The average IR value was 3.53 for adults and 3.91 for children, indicating that both adults and children may experience adverse health effects. The spatial distribution pattern of the IR values among the exposed populations in the study area showed high values in the eastern and middle parts, with maximum values >5, and low values in the western part, with minimum values <2. This is consistent with the distributions of the industries and the population. The study may provide a basis for comparison to other regions both in China and worldwide.

The globalization of agricultural trade has dramatically altered global nitrogen flows by changing the spatial pattern of nitrogen utilization and emissions at a global scale. As a major trading country, China uses a large amount of nitrogen, which has a profound impact on global nitrogen flows. Using data on food production and trade between China and 26 other countries and regions, we calculated nitrogen inputs and outputs in food production ecosystem in each country. We estimated nitrogen flows in international food trade and analyzed their impact on nitrogen pollution in China. We divided nitrogen flows into embodied and virtual nitrogen flows. Embodied nitrogen is taken up by the plant and incorporated into the final food product, whereas virtual nitrogen is lost to the environment throughout the food production process and is not contained in the final food product. Our results show that China mainly imports food products from America and Asia, accounting for 95 % of all imported food. Asia (mainly Japan) and Europe are the main exporters of food from China, with Japan and the EU accounting for 17 and 10 % of all exported food, respectively. Total nitrogen inputs and outputs in food production in China were 55,400 and 61,000 Gg respectively, which were much higher than in other countries. About 1440 and 950 Gg of embodied and virtual nitrogen respectively flow into China through the food trade, mainly from food-exporting countries such as the USA, Argentina, and Brazil. Meanwhile, 177 and 160 Gg of embodied and virtual nitrogen respectively flow out of China from the export of food products, mainly to Japan. China's net food imports have reduced 720 and 458 Gg for nitrogen utilization and outputs, respectively, which accounted for 1.3 and 0.78 % of total nitrogen inputs and outputs in China. These results suggest that food trade in China has a profound effect on nitrogen flows and has greatly reduced environmental impacts on nitrogen pollution in China.

The assessment of critical loads of metals in soil can be used as an important tool for evaluation and for risk precaution of future inputs of metal in order to avoid the occurrence of heavy metal pollution and its long-term risks for people. In this study, critical loads of Cd, Cu, and Pb in farming and non-farming areas of Kunshan were calculated based on three main effects. Two of these effects, limit value of daily metals dose and different environmental water quality criteria are new ways to calculate the critical content of heavy metals. The mean value of critical loads decreased in the order Cu>Pb>Cd when calculated using mass balance effects, child health risk effects, and adult health risk effects. Critical loads were highest in the areas near construction land, areas of low critical load were scattered throughout the city. The areal proportion of critical load exceedance is greatest for Pb based on mass balance effects, followed by Cu based on water quality effects, and Cd based on mass balance effects. Exceedances only occurred in 6% and 3% of farming areas for water quality effects for Cd and Pb when compared critical load values to the input fluxes in the Yangtze River delta. However, for these metals, values were up to 83% and 100%, respectively, based on mass balance effects. Exceedances completely covered non-farming areas for each effect for Pb. Most exceedances occurred in the north and south of the city in non-farming areas. Spatially explicit critical loads of heavy metals based on the different effects can serve as a reference for controlling the emissions of heavy metals effectively and meeting the demands of different management objectives.

Soil-crop system is an important way that heavy metals harm the ecological environment and human health. To research and understand the process of heavy metal absorption, migration and accumulation in soil-crop system is important for the prevention and control of heavy metal pollution and the health of human beings. In this paper, we established a model for crop uptake of heavy metals to calculate the heavy metals contents of wheat roots, stems, leaves and grains, and we analyzed the accumulation process of heavy metals in roots, stems, leaves and grains in the growth cycle of wheat. The predicted values were compared with the measured values to test the accuracy of the model. The results demonstrated that different parts of wheat had different heavy metal absorption capacity, the absorption of the roots was the strongest, followed by leaves, and the absorption capacity of stems and grains was weak. In addition, the contents of different heavy metals in each part of wheat were also significantly different. The content of Cu was the highest, followed by Ni, while the contents of Pb and Cd were small. In the process of wheat growth, the heavy metal accumulation rate of stem, leaf and grain began to slow down at 90, 60 and 100 days respectively, and the concentration of heavy metals reached the maximum gradually, while the accumulation rate of heavy metals in roots showed a growing trend. We studied the process of absorption, migration and accumulation of heavy metals in soil-crop system by using numerical simulation technology, which can provide scientific basis for preventing the ecological and health risks of heavy metal pollution.

Waterlogging frequently occurs in Southern China in summer and significantly affects waxy maize growth. This study investigated the physicochemical properties of starch from six waxy maize varieties exposed to waterlogging for 1-7 days after pollination. Waterlogging decreased the starch granule size. Starch maximum absorption wavelength, iodine-binding capacity, crystallinity, and peak intensities in response to waterlogging depended on varieties. Swelling power and solubility in response to waterlogging increased in Wannuo5 and decreased in the other five varieties. Gelatinization and pasting temperatures were only slightly affected by waterlogging. Gelatinization enthalpy was unaffected in Nongkeyu301, increased in Guangbainuo5, and decreased in the other four varieties. Peak and breakdown viscosities decreased and retrogradation percentage increased when plants were subjected to waterlogging after pollination. In conclusion, waterlogging decreased starch granule size, crystallinity, swelling power, and solubility, resulting in deteriorated starch quality (i.e., low swelling, less sticky and easy to retrograde).

China's rapid development of industrialization and urbanization causes the growing problem of heavy metal pollution of soil, threatening environment and human health. Therefore, prevention and management of heavy metal pollution become particularly important. Critical loads of heavy metals are an important management tool that can be utilized to prevent the occurrence of heavy metal pollution. Our study was based on three cases: status balance, water environmental effects and health risks. We used the steady-state mass balance equation to calculate the critical loads of Cd, Cu, Pb, Zn at different effect levels and analyze the values and spatial variation of critical loads. In addition, we used the annual input fluxes of heavy metals of the agro-ecosystem in the Yangtze River delta and China to estimate the proportion of area with exceedance of critical loads. The results demonstrated that the critical load value of Cd was the minimum, and the values of Cu and Zn were lager. There were spatial differences among the critical loads of four elements in the study area, lower critical loads areas mainly occurred in woodland and high value areas distributed in the east and southwest of the study area, while median values and the medium high areas mainly occurred in farmland. Comparing the input fluxes of heavy metals, we found that Pb and Zn in more than 90% of the area exceeded the critical loads under different environmental effects in the study area. The critical load exceedance of Cd mainly occurred under the status balance and the water environmental effect, while Cu under the status balance and water environmental effect with a higher proportion of exceeded areas. Critical loads of heavy metals at different effect levels in this study could serve as a reference from effective control of the emissions of heavy metals and to prevent the occurrence of heavy metal pollution.

Polycyclic aromatic hydrocarbons (PAHs) have become a major type of pollutant in urban areas and their degree of pollution and characteristics of spatial distribution differ between various regions. We conducted a comprehensive study about the concentration, source, spatial distribution, and health risk of 16 PAHs from urban to rural soils in Nanjing. The mean total concentrations of 16 PAHs (∑16PAHs) were 3330 ng g(-1) for urban soils, 1680 ng g(-1) for suburban soils, and 1060 ng g(-1) for rural soils. Five sources in urban, suburban, and rural areas of Nanjing were identified by positive matrix factorization. Their relative contributions of sources to the total soil PAH burden in descending order was coal combustion, vehicle emissions, biomass burning, coke tar, and oil in urban areas; in suburban areas the main sources of soil PAHs were gasoline engine and diesel engine, whereas in rural areas the main sources were creosote and biomass burning. The spatial distribution of soil PAH concentrations shows that old urban districts and commercial centers were the most contaminated of all areas in Nanjing. The distribution pattern of heavier PAHs was in accordance with ∑16PAHs, whereas lighter PAHs show some special characteristics. Health risk assessment based on toxic equivalency factors of benzo[a]pyrene indicated a low concentration of PAHs in most areas in Nanjing, but some sensitive sites should draw considerable attention. We conclude that urbanization has accelerated the accumulation of soil PAHs and increased the environmental risk for urban residents.
