RESUMO

Heterozygous variants in the hepatocyte nuclear factor 1a (HNF1a) cause MODY3 (maturity-onset diabetes of the young, type 3). In this study, we found a case of novel HNF1a p.Gln125* (HNF1a-Q125ter) variant clinically. However, the molecular mechanism linking the new HNF1a variant to impaired islet ß-cell function remains unclear. Firstly, a similar HNF1a-Q125ter variant in zebrafish (hnf1a+/-) was generated by CRISPR/Cas9. We further crossed hnf1a+/- with several zebrafish reporter lines to investigate pancreatic ß-cell function. Next, we introduced HNF1a-Q125ter and HNF1a shRNA plasmids into the Ins-1 cell line and elucidated the molecular mechanism. hnf1a+/- zebrafish significantly decreased the ß-cell number, insulin expression, and secretion. Moreover, ß cells in hnf1a+/- dilated ER lumen and increased the levels of ER stress markers. Similar ER-stress phenomena were observed in an HNF1a-Q125ter-transfected Ins-1 cell. Follow-up investigations demonstrated that HNF1a-Q125ter induced ER stress through activating the PERK/eIF2a/ATF4 signaling pathway. Our study found a novel loss-of-function HNF1a-Q125ter variant which induced ß-cell dysfunction by activating ER stress via the PERK/eIF2a/ATF4 signaling pathway.

BACKGROUND: Genomic instability is a hallmark of cancer. The G1 checkpoint allows cells to repair damaged DNA that may lead to genomic instability. The high-risk human papillomavirus (HPV) E7 gene can abrogate the G1 checkpoint, yet the mechanism is still not fully understood. Our recent study showed that WDHD1 (WD repeat and high mobility group [HMG]-box DNA-binding protein 1) plays a role in regulating G1 checkpoint of E7 expressing cells. In this study, we explored the mechanism by which WDHD1 regulates G1 checkpoint in HPV E7 expressing cells. METHODS: NIKS and RPE1 derived cell lines were used. Real-time PCR, Rescue experiment, FACS and BrdU labeling experiments were performed to examine role of GCN5 in G1 checkpoint abrogation in HPV-16 E7 expressing cells. RESULTS: In this study, we observed that WDHD1 facilitates G1 checkpoint abrogation by modulating GCN5 in HPV E7 expressing cells. Notably, depletion of WDHD1 caused G1 arrest while overexpression of GCN5 rescued the inhibitory effects of WDHD1 knockdown on G1/S progression. Furthermore, siWDHD1 significantly decreased cell cycle proliferation and DNA synthesis that was correlated with Akt phosphorylation (p-Akt), which was reversed by GCN5 overexpression in HPV E7 expressing cells. CONCLUSIONS: In summary, our data identified a WDHD1/GCN5/Akt pathway leading to the abrogation of G1 checkpoint in the presence of damaged DNA, which may cause genomic instability and eventually HPV induced tumorigenesis.

Cardiac dysfunction caused by excessive alcohol consumption is a specific disease, alcoholic cardiomyopathy (ACM). High-dose alcohol has been found to induce oxidation stress and apoptosis in cardiomyocytes, but the signaling link between alcohol-induced oxidation stress and apoptosis in cardiomyocytes remains to be elucidated. To address the issue, we exposed primary cardiomyocytes from neonatal mouse hearts to high doses of alcohol (50mM, 100mM, and 200 mM). We found that alcohol induced dose-dependent phosphorylation of p66shc, and reactive oxygen species (ROS) production increased in parallel with phosphorylation levels of p66shc. Exposure to alcohol also led to loss of mitochondrial membrane potential and cytochrome c release. Depletion of p66Shc and inhibition of protein kinase C-ß (PKC-ß) successfully reversed all the effects and suppressed alcohol-induced apoptosis in cardiomyocytes. Collectively, our study provides a molecular basis for signaling transduction of alcohol-induced oxidation stress and apoptosis of cardiomyocytes, which may facilitate the prevention and treatment of ACM.

To enhance the utilization of summer-autumn tea, a water-soluble polysaccharide (D1N1) was isolated through a series of techniques including hot water extraction, ethanol precipitation, and column chromatography. The structure of D1N1 was determined through the utilization of ultraviolet, Fourier-transform infrared, high-performance anion-exchange chromatography, gas chromatography-tandem mass spectrometry, and nuclear magnetic resonance. The results revealed that glucose was the predominant component of D1N1, accounting for 95% of its composition. Additionally, D1N1 also contained galactose, arabinose, and rhamnose. The molecular weight (Mw) of D1N1 was determined to be 224.71 kDa. The backbone of D1N1 consisted of →4)-α-D -Glcp (1→, →3,4)-α-D-Galp-(1→, →4,6)-α-D -Glcp (1→ at a molar ratio of 35:1:1, and branching at the O-3 position of →3,4)-α-D-Galp-(1→ and O-6 position of →4,6)-α-D-Glcp (1→ with α-D -Glcp (1→. In addition, the antioxidant activity of D1N1 was also evaluated. D1N1 exhibited excellent antioxidant bioactivity against the DPPH, superoxide anion radical, and ABTS+ radical. These findings provide a theoretical basis for the application of summer-autumn tea polysaccharide as a potential functional food.

With the rapid development of the times, people have entered the era of intelligence, and the application of big data algorithms is becoming increasingly widespread. The satisfaction of consumers in online shopping is closely related to communication and interaction during shopping. Based on this, this article studies the impact of online e-commerce interaction on consumer satisfaction based on big data algorithms. This article introduces the mediating variable of consumer satisfaction from the perspective of interaction, constructs a model between interaction and trust, and studies the internal impact mechanism of online interaction on consumer satisfaction in online shopping. This article takes the JD interactive shopping platform as the research object, and analyzes and explores the target consumer satisfaction of the women's clothing interactive shopping platform. Analyzed the impact of interaction on merchant qualifications and service satisfaction evaluations, store size, and logistics of purchased goods. The research results indicate that the normalization coefficients of the H1a and H1b pathways are 0.131 and 0.118, respectively, which are slightly smaller, indicating that the impact of perceived risk on consumer satisfaction is not significant. Meanwhile, the CR in H1 is a positive number and the direction of influence is positive, which is contrary to the assumption. Therefore, it is necessary to calibrate the initial model. After correction, the GFI value is 0.816, AGFI value is 0.825, RMSEA value is 0.042, TFI value is 0.930, CFI value is 0.955, PGFI value is 0.718, and PNFI value is 0.810. The degree is within an acceptable range. Therefore, when implementing interactive shopping, e-commerce companies need to create a good shopping environment for the implementation of interactive activities between sellers and customers. The impact of online e-commerce interaction based on big data algorithms on consumer satisfaction is a hot topic. Personalized recommendations can improve consumer satisfaction and loyalty, but data privacy and security issues are also receiving increasing attention. In addition, it is also necessary to consider the fairness and bias issues of the algorithm, as well as the transparency issues of data analysis and decision-making. On the premise of ensuring data privacy and security, it is necessary to improve the fairness and transparency of algorithms to improve consumer satisfaction and trust, and achieve sustainable development.

The exopolysaccharide (EPS) form Paecilomyces cicadae TJJ1213 possessed immunomodulatory activity in vitro, but whether it could regulate the immune system and intestinal microbiota in vivo remained unknown. In this study, the cyclophosphamide (CTX)-induced immunosuppressive mouse model was established to explore the immunomodulatory activity of EPS. Results showed that EPS could increase the immune organ indices, promote the secretion of serum immunoglobulins and up-regulate the expression of cytokines. Additionally, EPS could repair CTX-induced intestinal injury by increasing the expression of tight junction proteins and promoting the production of short-chain fatty acids. Moreover, EPS could remarkably enhance immunity through TLR4/MyD88/NF-κB and mitogen-activated protein kinase (MAPK) signaling pathways. Furthermore, EPS regulated intestinal microbiota by increasing the abundance of beneficial bacteria (Muribaculaceae, Lachnospiraceae NK4A136, Bacteroides, Odoribacter) and reducing the level of harmful bacteria (Alistipes, Helicobacter). In conclusion, our study suggested that EPS had the abilities to enhance immunity, restore intestinal mucosal injury and modulate intestinal microbiota, and may serve as a potential prebiotic to maintain health in the future.

This study investigated the interaction among Kluyveromyces marxianus G-Y4 (G-Y4), Lacticaseibacillus paracasei GL1 (GL1) and Lactobacillus helveticus SNA12 (SNA12) that isolated from Tibetan kefir grains. Additionally, the effects of G-Y4 on the growth and biofilm formation of GL1 and SNA12 were determined. The results indicated that G-Y4 promoted the growth of GL1 and SNA12 and improved their biofilm-forming ability. Furthermore, the dead cells of G-Y4 were found that could enhance bacterial biofilm formation, and the cell wall polysaccharide (CWPS) produced by G-Y4 was performed to be key substances that promote the formation of bacterial biofilms. Moreover, the structure of soluble cell wall polysaccharides (SCWP) and insoluble cell wall polysaccharide (NCWP) of G-Y4 were studied to determine their contribution to biofilm formation. Results showed that G-Y4-SCWP was α-mannan with the main chain of a →6)-α-d-Manp-(1→ unit and the branch structure of →2)-α-d-Manp-(1. At the same time, G-Y4-NCWP was a glucan rich in ß-(1→3), ß-(1→2), or ß-(1→4) linkages.

The prebiotic properties of two purified fractions (GL1-E1 and GL1-E2) of exopolysaccharides (EPSs) from Lacticaseibacillus paracasei GL1 were investigated through in vitro fermentation of pure and human fecal cultures. The results indicated that the simulated digestion under saliva, gastric, and small intestinal conditions had no effect on GL1-E1 and GL1-E2. Additionally, GL1-E1 and GL1-E2 can be used as substrates for Lactobacillus and Lactococcus growth. It was also found that both were gradually degraded and utilized by the gut microbiota. As fermentation proceeded, the pH continued to decrease. Additionally, the total short-chain fatty acid (SCFA) production significantly increased, especially the major SCFA of formic, lactic, and acetic acid. Furthermore, GL1-E1 and GL1-E2 could significantly regulate the composition of the gut microbiota, by increasing the relative abundances of Bacteroides and Phascolarctobacterium, and decreasing the pathogenic bacteria Escherichia-Shigella, Klebsiella, and Fusobacterium. These results suggest that GL1-E1 and GL1-E2 have the potential to be developed as a prebiotic.

Oxidative stress is a key process in the development of neurodegenerative diseases. More attention is needed to screen natural antioxidants and explore pharmacological mechanisms. Natural product polysaccharides with no toxic side effects have powerful antioxidant activity. Two purified intracellular polysaccharide fractions (IPS1 and IPS2) from Paecilomyces cicadae TJJ1213 was isolated. Then, a model of H2 O2 -induced oxidative stress in PC12 cells was established to investigate the neuroprotective role of IPS and elucidate the potential protection mechanism. Results showed that IPS1 and IPS2 reduced reactive oxygen species (ROS) production, inhibited the leakage of lactate dehydrogenase (LDH) and Ca2+ and attenuated the expression of apoptotic proteins. In addition, western blots displayed that IPS1 and IPS2 significantly inhibited mitophagy induced by H2 O2 in PC12 cells via PINK/Parkin pathway. Therefore, IPS1 and IPS2 deserved further investigation as protective agents against neurodegenerative diseases.

The rapid development and wide application of artificial intelligence is deeply affecting all aspects of human society. Combine artificial intelligence with the accounting industry, use computers to efficiently and automatically process accounting information, and let the accounting industry move towards the intelligent era. This can help people reduce the workload and speed up work efficiency. In recent years, with the rapid development of economy and technology, the use of financial instrument vouchers has exploded, but the processing requirements of financial instrument vouchers have become more and more efficient. Traditional accounting information processing methods, due to the staff's energy and ability, it is often difficult to quickly and accurately handle accounting information. This makes the processing of accounting information lack of timeliness, the degree of utilization of accounting information by enterprises is relatively low, and the demand for intelligent processing of accounting information is constantly pressing. In view of the above problems, this paper uses image processing technology to intelligently identify the content of accounting information to achieve automatic ticket input, improve work efficiency, reduce error rate and reduce labor costs. By simulating the actual 230 invoice images, the results show that the recognition accuracy rate is as high as 98.7%. The results show that the method is effective and has great application value, which is of great significance to the artificial intelligence of accounting information processing.

Orphan nuclear receptor Nur77 has been reported to be implicated in a diverse range of metabolic processes, including carbohydrate metabolism and lipid metabolism. However, the detailed mechanism of Nur77 in the regulation of metabolic pathway still needs to be further investigated. In this study, we created a global nur77 knockout zebrafish model by CRISPR/Cas9 technique, and then performed whole-organism RNA sequencing analysis in wildtype and nur77-deficient zebrafish to dissect the genetic changes in metabolic-related pathways. We found that many genes involved in amino acid, lipid, and carbohydrate metabolism changed by more than twofold. Furthermore, we revealed that nur77-/- mutant displayed increased total cholesterol (TC) and triglyceride (TG), alteration in total amino acids, as well as elevated glucose. We also demonstrated that the elevated glucose was not due to the change of glucose uptake but was likely caused by the disorder of glycolysis/gluconeogenesis and the impaired ß-cell function, including downregulated insb expression, reduced ß-cell mass, and suppressed insulin secretion. Importantly, we also verified that targeted expression of Nur77 in the ß cells is sufficient to rescue the ß-cell defects in global nur77-/- larvae zebrafish. These results provide new information about the global metabolic network that Nur77 signaling regulates, as well as the role of Nur77 in ß-cell function.

In this study, three yeast α-mannans (LZ-MPS, MC-MPS, and G-MPS) were extracted from different sources of Kluyveromyces marxianus. The total sugar content of the three α-mannans ranged from 91.13-97.10%, whereas no proteins were detected. A structural arrangement was proposed using ultraviolet spectroscopy, Fourier-transform infrared spectroscopy, and one-dimensional and two-dimensional nuclear magnetic resonance. The main chain of the three yeast α-mannans was formed by a →6)-α-D-Manp-(1→ unit, which was slightly different from the repeating unit of the branch structure. The prebiotic potential of LZ-MPS, MC-MPS, and G-MPS was assessed using in vitro fermentation with pure and faecal cultures. The three yeast α-mannans could be utilised as substrates for the growth of Lactobacillus and Lactococcus strains. In addition, the three yeast α-mannans markedly regulated the intestinal microbiota composition by increasing the relative abundances of Bacteroides, Parabacteroides, and Phascolarctobacterium and decreasing the abundance of pathogenic bacteria.

This study investigated the supramolecular structure features and immunomodulatory effects of two exopolysaccharide fractions (EPS1 and EPS2) from Paecilomyces cicada TJJ1213 in vitro. AFM images revealed that EPS1 and EPS2 displayed different morphological features at different concentrations. Congo red and XRD assay further proved that EPS1 and EPS2 mainly exhibited amorphous structure with random coil conformation in solution. Furthermore, the immunomodulatory effect of EPSs was investigated on RAW264.7 cells. Results showed that EPS1 and EPS2 could enhance the phagocytic activity and induce the NO production and could also significantly up-regulate the mRNA expression of iNOS, TNF-α, IL-6, IL-1ß, IFN-γ and IL-4. Western blot assay analysis demonstrated that EPSs increased protein expression of TLR4 and the nuclear translocation of NF-κB p50/p65. Additionally, the phosphorylation levels of MAPKs proteins (p38, ERK and JNK) were also remarkably increased. Thus, EPSs could active TLR4-NF-κB/MAPKs signaling pathways to exert the immunomodulatory effect on macrophages.

The invasion of the pleural membrane by a malignant pleural tumor can lead to the production of malignant pleural effusion (MPE), resulting in the symptoms of dyspnea, and some patients have cough, sputum and other symptoms, which are easily confused with pneumonia. In this case, the initial diagnosis of the patient is pneumonia, and the final diagnosis is pneumonia combined with pleural malignancy. Therefore, if the patient has unexplained symptoms of bloody pleural effusion, it is necessary to examine for malignant tumors and should actively perform thoracentesis and drainage, look for malignant cells in the pleural effusion cell precipitation, evaluate the nature of pleural effusion, conduct pleural biopsy tissue examination, and determine the type and source of lung malignancy by the combined application of cell block technology and immunohistochemistry. Take the cytological examination results in pleural effusion seriously, and finally, surgical or immunotherapy can be performed.

Soybean whey is a kind of agricultural by-product enriched with nutritional value but with low utilization. The extracellular polysaccharides secreted by lactic acid bacteria during the fermentation possess a variety of structural characteristics and beneficial properties. In this study, an exopolysaccharide (EPS) was isolated from Leuconostoc citreum 1.2461 after fermentation in optimized soybean whey-enriched 10% sucrose at 37 °C for 24 h. The water-soluble EPS-1 was obtained by DEAE-52 anion exchange chromatography, and the structural characterization of EPS-1 was investigated. The EPS-1 was homogeneous with an average molecular weight of 4.712 × 106 Da and consisted mainly of glucose. Nuclear magnetic resonance (NMR) spectrum and flourier transform infrared (FT-IR) spectrum indicated that the EPS-1 contained →3)-α-D-Glcp-(1→ and →6)-α-D-Glcp-(1→ residues. The rheological properties of EPS-1 under the conditions of changing shear rate, concentration, temperature and coexisting ions showed its pseudoplastic fluid behaviors. In addition, the EPS-1 exhibited certain scavenging activity on the ABTS radical and chelating activity on metal ions at relatively high concentrations. Furthermore, EPS-1 with a certain concentration was confirmed to have significant protective effects on yeast cell injury induced by hydrogen peroxide. This study reported the structural characteristics of exopolysaccharide from Lc. citreum 1.2461 and provides a basis for its potential application in the field of functional foods.

Two exopolysaccharide fractions (GL1-E1 and GL1-E2) of Lacticaseibacillus paracasei GL1 were isolated with the molecular weights of 3.9 × 105 Da and 8.2 × 105 Da, respectively. Both fractions possessed mannose, glucose, and galactose in molar ratios of 1.16:1.00:0.1, and 3.81:1.00:0.12, respectively. A structural arrangement of two fractions was proposed by methylation, one-dimensional and two-dimensional nuclear magnetic resonance experiments. The backbone of GL1-E1 consisted of →4)-α-D-Glcp(1→, →3,4)-α-D-Manp(1→, →3,6)-α-D-Manp(1→, →6)-α-D-Manp(1→, and →6)-α-D-Galp(1→ with α-D-Glcp at branching point. The backbone of GL1-E2 consisted of →4)-α-D-Glcp(1→, →3,4)-α-D-Manp(1→, →3,6)-α-D-Manp(1→, →6)-α-D-Manp(1→, →6)-α-D-Galp(1→, and →4)-ß-D-Manp(1→, and the side chain also consisted of α-D-Manp residue. In addition, the differential scanning calorimetry (DSC) analysis indicated that both GL1-E1 and GL1-E2 had good thermal stability. Furthermore, the two fractions could promote the viability of RAW264.7 cells and exert an immunomodulatory role by enhancing phagocytosis, increasing nitric oxide (NO) release and promoting the expression of cytokines.

Traditional Chinese medicine (TCM) has been used in clinical settings for over 2000 years in China. The study of the absorption, distribution, metabolism, and excretion (ADME) of TCM in vivo could be beneficial for the discovery of the active components in TCM. However, the conventional strategies used for ADME research are based on rodent models and have the characteristics of lengthy experimental periods, complex processes, and extensive data processing, which make it difficult to perform rapid analyses and high-throughput ADME screening of the medicinal components of TCM. In this study, an integrated high-throughput research strategy for the in vivo ADME analysis of TCM was established based on a zebrafish model. Accordingly, a combination of ultra-performance liquid chromatography-high-resolution mass spectrometry (UPLC-HRMS), desorption electrospray ionization-mass spectrometry (DESI-MS) imaging, and in-house non-targeted precise-and-thorough background-subtraction (PATBS) data post-processing techniques were successfully applied for the analysis of the metabolism of zebrafish exposed to Xiaoke pills. A total of 49 compounds related to Xiaoke pills (including 13 prototypical components and 36 metabolites) were detected in zebrafish. In total, 32 of them, including puerarin, daidzein, deoxyschizandrin, formononetin, and glibenclamide, which have been identified to have hypoglycemic activity in our previous studies and are phase I and phase II metabolites resulting from the hydroxylation, demethylation, glucuronidation, sulfation, and glycosylation of the prototypical components in vivo, were found in rats treated with Xiaoke pills. Furthermore, the overall distribution of the known compounds in zebrafish exposed to Xiaoke pills was explored using DESI-MS. In summary, this study provides a practical approach for the high-throughput screening of the active components of TCM using a zebrafish model.

Exopolysaccharides (EPSs) from lactic acid bacteria have special functions and complex structures, but the function and structure of EPSs of the important dairy starter, Lactococcus (L.) lactis subsp. lactis, are less known. This study investigated the cytotoxicity, antioxidant capacities, rheological characteristics, chemical structure and expression of biosynthetic genes of EPSs of the L. lactis subsp. lactis IMAU11823. The EPSs showed strong reducing power and no cytotoxicity. EPS-1 comprised glucose and mannose (molar ratio of 7.01: 1.00) and molecular weight was 6.10 × 105 Da, while EPS-2 comprised mannose, glucose and rhamnose (7.45: 1.00: 2.34) and molecular weight was 2.93 × 105 Da. EPS-1 was a linear structure comprised two sugar residues, while EPS-2 was more complex, non-linear, and comprised eight sugar residues. In additions, our study proposed an EPS biosynthesis model for the IMAU11823 strain. The current findings have broadened the understanding of the formation, structure and function of complex EPSs of IMAU11823.

In this study, a high-throughput sequencing technique was used to analyze bacterial and fungal diversity of two traditional Tibetan kefir grains from Linzhi (K1) and Naqu (K2) regions. Comparative bioinformatic analyses indicated that Lactobacillus kefiranofaciens, L. kefiri and Kluyveromyces marxianus were the main dominant strains in K1 and K2. In order to research the relationship of the growth of kefir grains, the biofilm and the extracellular polysaccharides (EPS) produced by microorganisms, the proliferation rate of kefir grains, the yield and chemical structure of EPS and the optimal days for biofilm formation were determined. The results showed that the growth rate, the yield of EPS and the biofilm formation ability of K1 were higher than K2, and the optimal day of their biofilm formation was the same in 10th day. Additionally, the live cells, dead cells and EPS in biofilm formation of K1 and K2 were observed by fluorescence microscope to clarify the formation process of kefir grains. To determine the influence of microbial interactions on biofilm and the formation of kefir grains, the essential role of microbial quorum sensing needs further attention.

Soybean whey is a high-yield but low-utilization agricultural by-product in China. In this study, soybean whey was used as a substrate of fermentation by Lacticaseibacillus plantarum 70810 strains. An exopolysaccharide (LPEPS-1) was isolated from soybean whey fermentation by L. plantarum 70810 and purified by ion-exchange chromatography. Its preliminary structural characteristics and antioxidant activity were investigated. Results show that LPEPS-1 was composed of mannose, glucose, and galactose with molar ratios of 1.49:1.67:1.00. The chemical structure of LPEPS-1 consisted of →4)-α-D-Glcp-(1→, →3)-α-D-Galp-(1→ and →2)-α-D-Manp-(1→. Scanning electron microscopy (SEM) revealed that LPEPS-1 had a relatively rough surface. In addition, LPPES-1 exhibited strong scavenging activity against DPPH and superoxide radicals and chelating ability on ferrous ion. This study demonstrated that soybean whey was a feasible fermentation substrate for the production of polysaccharide from L. plantarum 70810 and that the polysaccharide could be used as a promising ingredient for health-beneficial functional foods.
