RESUMO

Tripartite Motif 67 (TRIM67) is an important member of TRIM family proteins, which participates in different cellular processes including immune response, proliferation, differentiation, carcinogenesis, and apoptosis. In recent years, a high fat diet (HFD) has remained one of the main causes of different metabolic diseases and increases in intestinal permeability as well as inducing intestinal inflammation. The current study investigated the protective effects of TRIM67 in the ileum and colon of obese mice. 4-week-old wild-type (WT) C57BL/6N mice and TRIM67 knockout (KO) C57BL/6N mice were selected and randomly divided into four sub-groups, which were fed with control diet (CTR) or HFD for 14 weeks. Samples were collected at the age of 18 weeks for analysis. To construct an in vitro obesity model, over-expressed IPEC-J2 cells (porcine intestinal cells) with Myc-TRIM67 were stimulated with palmitic acid (PA), and its effects on the expression level of TRM67, inflammatory cytokines, and barrier function were evaluated. The KO mice showed pathological lesions in the ileum and colon and this effect was more obvious in KO mice fed with HFD. In addition, KO mice fed with a HFD or CTR diet had increased intestinal inflammation, intestinal permeability, and oxidative stress compared to that WT mice fed with these diets, respectively. Moreover, IPEC-J2 cells were transfected with TRIM67 plasmid to perform the same experiments after stimulation with PA, and the results were found consistent with the in vivo evaluations. Taken together, our study proved for the first time that HFD and TRIM67 KO mice have synergistic damaging effects on the intestine, while TRIM67 plays an important protective role in HFD-induced intestinal damage.

The study investigates the socio-religious factors in the propagation of genetically inherited disease of Beta thalassemia. The disorder which reportedly has a significant protraction through repeated cousin marriages results in the social maladjustment of the parents of the sick children due to constant depression, anxiety, and weak social interaction and may lead to social isolation as well. This research aims to find out the significant effect of socio-religious trends on psychosocial burden of beta thalassemia major among cousin and non-cousin couples in the province of Punjab in Pakistan. It takes a sample of 932 parents of sick children, among whom 735 were married with cousins and 197 with non-cousins, for data collection. The findings reveal that inadequate knowledge of the disease, insufficient or misdirected social support, stigmatization, and marriage breakups caused by the disease, superstitions, and misinterpretations of religion and the subsequent practices accordingly as significant predictors of psychosocial burden of beta thalassemia major among non-cousins and cousin couples. Additionally, it also finds patriarchy as only significant predictors of outcome variable among cousin couples.

In this work, microgels based on tris(2-aminoethyl) amine (TAEA) and glycerol diglycidyl ether (GDE) via simple microemulsion polymerization was prepared as p(TAEA-co-GDE) microgels were used as adsorbent for removal of dichromate (Cr (VI)) and arsenate (As (V)) ions from different aqueous environments. The p(TAEA-co-GDE) microgels were demonstrated very efficient adsorption capacity for Cr (VI), and As (V) that are 164.98 mg/g, and 123.64 mg/g from distilled (DI) water, respectively. The effect of the medium pH on the adsorption capacity of p(TAEA-co-GDE) microgels for Cr (VI) and As (V) ions were investigated. The maximum adsorption capacity was obtained at pH 4.0 for both ions with maximum adsorbed amounts of 160.62, and 98.72 mg/g, respectively. In addition, the microgels were also shown moderate adsorption capacity for Cr (VI) and As (V) from other water sources; tap water with 115.18 mg/g and 82.86 mg/g, sea water with 64.24 mg/g and 46.88 mg/g and creek water with 73.52 mg/g and 59.33 mg/g, respectively. Moreover, the increase in adsorbent dose from 0.025 to 0.125 g enhanced % adsorption of Cr (VI) from 54.13 to 98.03, and As (V) from % 26.72-98.70, respectively. For the adsorption process Langmuir and Freundlich adsorption isotherms were applied and found that Langmuir adsorption isotherm with R2 value of 0.99 for both the metal ions are suitable. Moreover, the experimental adsorption capacities of Cr (VI) and As (V) were found very close to the theoretical values calculated from Langmuir adsorption isotherm. More importantly, the microgels were made magnetic responsive to recover them easily from adsorption medium for reuse studies by applying external magnetic field with little decrease in adsorption capacity. Additionally, reusability of p(TAEA-co-GDE) microgels was also evaluated for adsorption of Cr (VI) and As (V) from DI water.

A novel ferric-phosphate induced shape memory (SM) hydrogel is prepared by the one-step copolymerization of isopropenyl phosphonic acid (IPPA) and acrylamide (AM) in the presence of a crosslinker polyethylene glycol diacrylate (PEGDA). Different from the traditional SM hydrogels, our SM hydrogel can be processed into various shapes as needed and recovers to its original form in 'multiconditions' such as in the presence of a reducing agent or in the presence of a competitive complexing agent. This unique feature is attributed to the fact that the oxidized ferric ions show a high complexation ability with phosphate groups of IPPA, which acts as a physical crosslinker to form the secondary networks within the hydrogels to induce the shape memory effect. The memory behavior was totally reversible, owing to Fe3+ that can be reduced to Fe2+ and extracted by the complexing agent. Particularly, the SM hydrogels exhibit controllable and good mechanical characteristics by introduction of the ferric ions, i.e., the elastic modulus can increase from 2 kPa to 70 kPa dramatically. Learning from biological systems, phosphate-metal ion based hydrogels could become an attractive candidate for various biomedical and environmental applications.

Given the steady increase in breast cancer rates in both the developed and developing world, there has been a concerted research effort undertaken worldwide to understand the molecular mechanisms underpinning the disease. The data generated from numerous clinical trials and experimental studies shed light on different aspects of the disease. We present a new version of the ROCK database (rock.icr.ac.uk), which integrates such diverse data types allowing unique analyses of published breast cancer experimental data. We have added several new data types and analysis modules to ROCK, which allow the user to interactively query and research the huge amounts of available experimental data and perform complex correlations across studies and data types such as gene expression, genomic copy number aberrations, micro RNA expression, RNA interference, survival analysis, clinical annotation and signalling protein networks. We present the recent and major functional updates and enhancements to the ROCK resource, including new analysis modules and microRNA and NGS data integration, and illustrate how ROCK can be used to confirm known experimental results as well as generate novel leads and new experimental hypotheses using the Wnt signalling cell surface receptor FZD7 and the Myc oncogene. ROCK provides a unique breast cancer analysis platform of integrated experimental datasets at the genomic, transcriptomic and proteomic level. This paper presents how ROCK has transitioned from being simply a database to an interactive resource useful to the broader breast cancer research community in our effort to facilitate research into the underlying molecular mechanisms of breast cancer.

This study was conducted to evaluate the prevalence of bovine gammaherpesvirus 4 (BoHV-4) among healthy cattle and buffaloes as well as those associated with different diseases (respiratory tract infection, mastitis and reproductive tract infection) in District Chakwal, Pakistan. Blood, swab and milk samples of cattle and buffaloes were randomly collected from different areas of Chakwal. DNA was isolated from the samples and subjected to nested PCR using thymidine kinase gene primers. Out of 300 samples (200 blood, 50 swab and 50 milk samples) from both species (cattle and buffalo), an overall prevalence of BoHV-4 of 3.33% was obtained. Samples from cattle showed a higher species-specific prevalence (4.16%) than samples from buffalo (2.78%). One sample out of 50 swab samples and 1 out of 50 milk samples were also positive for BoHV-4. DNA sequencing of a positive PCR product from cattle confirmed that the sequence was from the thymidine kinase gene of BoHV-4. Phylogenetic analysis also revealed close similarities with other BOHV-4 thymidine kinase sequences. To detect BoHV-4 antibodies, an indirect ELISA was also performed. Two hundred blood samples were also collected from the same animals in nonanticoagulant-containing tubes for the isolation of serum and were subjected to indirect ELISA. Sixteen samples (8%) were positive for BoHV-4 antibodies. This study will be useful in further diagnoses of BoHV-4 in Pakistan and in devising measures to control the spread of BoHV-4.

Requirement elicitation stands as a pivotal activity within requirement engineering, gaining even greater significance in the context of global software development. Effective communication among stakeholders assumes paramount importance in this arena. Factors such as time zone disparities, cultural variations, and language differences exert a formidable impact on communication within the sphere of global software development. These dynamics inevitably impinge upon timely coordination, potentially compromising the software's quality. In response, researchers have proffered communication models tailored for requirement elicitation within the ambit of global software development. The purpose of this study is to conduct an in-depth critical review of existing communication models for demand elicitation in global software development. Through this comprehensive review, we aim to discern prevailing publication trends, provide an introductory overview, and illuminate the strengths and limitations inherent in the existing communication models. By identifying these limitations, we seek to advance a novel, low-cost communication approach designed primarily for demand elicitation in global software development. To culminate our endeavor, we will undertake a case study-based experiment, meticulously designed to assess the efficacy and practical utility of the proposed techniques.

Backgrounds: AngioVac is used for the percutaneous removal of vegetations and for debulking of large vegetations in patients who are not surgical candidates.This study aims to identify the demographics, echocardiographic features, indications, improvement of the tricuspid valve regurgitation, and survival outcomes of patients who have undergone AngioVac vegetectomy reported in the literature. Methods: A systematic review was performed to identify articles reporting suction thromectomy or vegetation removal using the AngioVac system for RSIE (right sided infective endocarditis). Survival on discharge was our primary outcome. Additionally, we evaluated indications for suction thrombectomy and TR improvement. Categorical variables were expressed as percentages and ratios. Results: A total of 49 studies were identified. The most common risk factor was intravenous drug abuse seen in 45% (20/49) and cardiovascular implantable electronic device (CIED) in 45% (20/49). Circulatory shock was seen in 35% of patients. The causative organism was gram positive cocci (86%). Moderate to severe TR was present in 74% of cases with documented echocardiograms. Indications for AngioVac were poor surgical candidacy (81%) or to reduce septic emboli risk (19%). Survival at discharge was 93%. TR improvement was reported only in 16% cases and remained unchanged/worsened in 84%. Conclusion: AngioVac procedure is an alternative treatment for critically ill patients who cannot undergo surgery. To understand the survival, safety and candidacy of patients undergoing this procedure, further randomized control studies and literature reviews are needed. The improvement or worsening of tricuspid regurgitation in patients with TR valve involvement is another factor to be investigated.

Soil-borne diseases cause serious economic losses in agriculture. Managing diseases with microbial preparations is an excellent approach to soil-borne disease prevention. However, microbial preparations often exhibit unstable effects, limiting their large-scale application. This review introduces and summarizes disease-suppressive soils, the relationship between carbon sources and the microbial community, and the application of human microbial preparation concepts to plant microbial preparations. We also propose an innovative synthetic microbial community assembly strategy with synergistic prebiotics to promote healthy plant growth and resistance to disease. In this review, a new approach is proposed to improve traditional microbial preparations; provide a better understanding of the relationships among carbon sources, beneficial microorganisms, and plants; and lay a theoretical foundation for developing new microbial preparations.

Au-doped ZnO-samarium nitrate (Sm) nanoparticles with fixed concentrations of Sm (1 wt %) and various concentrations of Au (0.0, 0.5, 1.0 and 1.5 wt %) were prepared and used as photoelectrodes to enhance the photovoltaic efficiency of dye-sensitized solar cells (DSSCs). The cell fabricated with 1.5 wt % of Au-doped ZnO-Sm nanoparticles film achieved an optimal efficiency of 4.35%, which is about 76% higher than that of 0.0 wt % of Au-doped ZnO-Sm-based cell (2.47%). This increase might be due to the formation of a blocking layer at the ZnO-Sm/Au interface, which inhibits the recombination of electrons. This increase may also be attributed to the addition of rare-earth ions in ZnO to enhance the non-absorbable wavelength region of light via up/down-conversion of near-infrared and ultraviolet radiations to visible emission and reduce the recombination loss of electron in the cell. The efficiency of cells may be increased by the blocking layer and up/down-conversion process and thus promote the overall performance of the cells. This work indicates that Au-doped ZnO-Sm nanoparticle films have the potential to increase the performance of DSSCs.

Low-cost and washable resistive switching (RS) memory devices with stable retention and low operational voltage are important for higher speed and denser non-volatile memories. In the case of green electronics, pectin has emerged as a suitable alternative to toxic metal oxides for resistive switching applications. Herein, a pectin-based thin film was fabricated on a fluorine-doped tin oxide glass substrate for RS mechanism. The presence of sp3-C groups with low binding energy corresponds to tunable charged defects and the oxygen vacancies confirmed by the O 1s spectra that plays a decisive role in the resistive switching mechanism, as revealed by X-ray photoemission spectroscopy (XPS). The surface morphology of the pectin film shows homogeneous growth and negligible surface roughness (38.98 ± 9.09). The pectin film can dissolve in DI water (10 minutes) owing to its ionization of carboxylic groups, that meet the trends of transient electronics. The developed Ag/pectin/FTO-based memory cell exhibits stable and reproducible bipolar resistive switching behavior along with an excellent ON/OFF ratio (104) and negligible electrical degradation was observed over 30 repeated cycles. Hence, it appears to be a valuable application for green electronics. Indeed, biocompatible storage devices derived from natural pectin are promising for high-density safe applications for information storage systems, flexible electronics, and green electronics.

BACKGROUND: Bone marrow trephine biopsy is a well established minor surgical procedure for the inspection of bone marrow usually done along with bone marrow aspiration. The objective of this study was to evaluate the length of trephine biopsies and the rate of positivity for diagnosis as well as unfit biopsies in various length ranges. METHODS: This retrospective study was conducted at Fauji Foundation Hospital and Foundation University Medical College Rawalpindi from Jan 2007 to Dec 2009. A total of 394 trephine biopsy reports were collected and reviewed. The criterion for adequate trephine biopsy was > or = 1.5 Cm. The biopsies were divided into four groups according to length, i.e., group-1: > or = 1.5 Cm, group-2: 1-1.4 Cm, group-3: 0.5-0.9 Cm, and group-4: < 0.5 Cm. The adequacy of trephine biopsy length and rate of positive diagnosis as well as unfit biopsies were compared. RESULTS: Total 394 trephine biopsies were reviewed. Group-1 included 88 biopsies and 87 (98.9%) had positive diagnosis. Group-2 included 137 biopsies and 133 (97.1%) had positive diagnosis. Group-3 included 99 biopsies and 91 (92%) had positive diagnosis. Group-4 included 70 biopsies and 57 (81.4%) had positive diagnosis. There was no significant difference between group-1 and group-2 for the rate of positivity of diagnosis (p = 0.65). In group-1, 1 (1.1%) was unfit for evaluation, in group-2, 4 (2.9%) were unfit, in group-3, 8 (8%) were unfit, and in group-4, 13 (18.5%) were unfit for evaluation. Total 26 trephine biopsies were unfit for evaluation, out of which 13 (50%) belonged to group-4. Trephine biopsies that were unfit for evaluation were 4 (4.9%) in 2007, 17 (10.5%) in 2008, and 5 (3.3%) in 2009. CONCLUSION: Although 22.3% biopsies were of recommended length there was no significant difference in rate of positive diagnosis between biopsies of > or = 1.5 Cm and 1-1.4 Cm.

Societal determinants of health are of recognized importance for understanding the causal association of society and health of an individual. Iron deficiency anemia (IDA) is a challenging public health problem across the globe instigating from a broader sociocultural background. It is more prevalent among pregnant women, children under the age of five years, and adolescent girls. Adolescent girls are vulnerable to develop IDA because of additional nutritional demand of the body needed for growth spurt, blood loss due to onset of menarche, malnourishment, and poor dietary iron intake. In this study, we explore the societal determinants of anemia among adolescent girls in Azad Jammu and Kashmir (AJK), Pakistan. A cross-sectional study was conducted in the Muzaffarabad division of AJK on randomly selected 626 adolescent girls. The data were collected using a pretested self-administered interview schedule comprising mainly closed-ended questions with a few open-ended questions. Descriptive statistics was computed for describing the data, and bivariate regression and logistic regression were used to determine the association of anemia with its societal determinants. Multiple linear regression is used to determine the relationship of different determinants (independent variables) with the hemoglobin level (dependent variable) of the respondents. The prevalence of anemia among adolescent girls is 47.9%, of which 47.7% have mild anemia, 51.7% have moderate anemia, and 5.7% have severe anemia, which reveals that anemia is a severe public health problem among adolescent girls in the study area. The findings aver that anemia occurrence was significantly associated with the respondent's and her parental education, economic well-being, prevalence of communicable diseases, menstrual disorder, exercise habits, meals regularity, and type of sewerage system.

Psychosocial hazards present in workplaces are being actively investigated by researchers from multiple domains. More research and resources are required to investigate the debilitating consequences of these hazards in the developing and underdeveloped countries where this issue remains one of grave concern. This study aims at investigating the psychometric properties of Malaysian version of Copenhagen Psychosocial Questionnaire for reliability and validity purpose. The Malaysian version of COPSOQ is a multidimensional questionnaire; it comprises of 7 major formative constructs and 28 variables with an additional inclusion of two variables which are organizational loyalty and physiological health biomarkers (blood pressure and body mass index) that explicate a reflective construct which has 93 items all catering to assess psychosocial determinants present in workplace environments. Each formative second-order construct is further categorized into different reflective first-order constructs. The focus of this study was only on first-order reflective constructs. Probability sampling was used for data collection from 300 respondents working in industries with a response rate of 100%; structural equation modeling technique was applied for data analysis. All psychometric analysis performed on reflective constructs gave reliable results which demonstrate the validity of Bahasa Melayu (BM-COPSOQ) and its comprehensiveness of including relevant dimensions particularly in context to Asian region. The BM-COPSOQ will fill up the knowledge gap and provide a bridge between researchers, work professionals and practitioners, and many other workplaces for the best understanding of psychosocial work environment.

BACKGROUND: Infection in orthopedic surgery is a disaster both for the patient and surgeon. Although its incidence has been reduced due to modern theatre facilities and aseptic measures but in developing countries its prevalence is still high. It is better to prevent infection rather than to treat it. The objective of this study is to know the frequency of infection in orthopedic implant surgery in a public hospital and to evaluate the risk factors, causative organism, complications and treatment. METHODS: This prospective study was conducted in orthopedic unit-B Ayub Teaching Hospital, Abbottabad, from 1st April 2007 to 30th October 2007. Close fracture cases admitted for internal fixation devices were included. The exclusion criteria were soft tissue surgery, wounds and open fractures needing external fixation devices. The follow up was done for six months. RESULTS: Infection developed in 6 patients (5.76%), out of which superficial and deep infections were 2 (1.92%) and 4 (3.84%) respectively. There were 2 infection cases in each stage of the infection i.e. early, delayed and late. The staphylococcus aureus was the commonest organism, i.e., 3 (50%) out of 6. The age of the patients was more than 60 years in 3 (2.88%) patients, 30 to 60 years in 2 (1.92%) patients and below 30 year in 1 (0.96%) patient. The smoking history was in 2 (1.92%) patients. CONCLUSION: Infection rate in our study was quite high and needs proper measures to control it because it had great financial burden on patient and on hospital resources and could lead to morbidity and mortality in patients. We could not find significant risk factors in our small sample size study although the infection was relatively more common in patients with advanced age, prolonged surgery time, smoking and skin abrasion at fracture site. Commonest organism was Staphylococcus aureus.

RESUMO

Gastric cancer is the 2nd most common cause of death among all cancers and is the 4th most common cancer in the world. The number of deaths due to gastric cancer is about 800,000 annually. Gastric cancer is more common in men as compared to women and is 3rd most common cancer after colorectal and breast cancers in women. A progressive rise in the incidence rate has been observed in females over the last 5 years. The highest incidence of stomach cancer is in China, South America and Eastern Europe. The incidence of gastric cancer has 20 fold variation worldwide. Global variation is linked by two factors which play important role in developing gastric cancer. One is infection with Helicobacter pylori and the 2nd is diet. South Asia is a region with low risk, despite a high prevalence of H.pylori. Gastric carcinoma is common in southern region of India. Gastric cancer is more readily treated if diagnosed early. This study aims to provide awareness about gastric cancer as well as an updated knowledge about risk factors and epidemiology of gastric cancer in Pakistan.

RESUMO

We are presenting an interesting case of skin metastasis from adenocarcinoma prostate. Interestingly bone metastasis were absent as documented on radioisotope bone scan and PSA levels were normal. As it was a hormone refractory disease, he was started on mitoxantrone and prednisolone chemotherapy. Lung metastatis appeared after 2 cycles of chemotherapy. He died of progressive disease 4 months later.

Foot and mouth disease (FMD) is a highly contagious disease of ruminants that causes huge economic losses around the globe. However, the prevalence of FMDV in small ruminants has been overlooked in Pakistan. A seroepidemiological study was conducted in Chakwal, Faisalabad and Khanewal districts of Punjab, Pakistan to determine the prevalence of FMD in sheep and goats. A total 1200 serum samples were collected from sheep (n = 180) and goats (n = 920) and were subjected to 3ABC non-structural protein enzyme-linked immunosorbent assay for detection of antibodies against non-structural proteins of FMD virus. Samples collected from clinical cases were also confirmed for FMDV using RT-PCR. The overall seroprevalence of FMD in sheep and goats was 21% (n = 252) while 19.44% (n = 35) in sheep and 21.27% (n = 217) prevalence was recorded in goats. Highest seroprevalence (32.5%) was observed in southern Punjab (Khanewal), followed by (25.75%) central Punjab (Faisalabad) and the lowest seroprevalence (4.75%) was detected in northern Punjab (Chakwal). There was no statistically significant difference in seroprevalence between sheep and goats. Among different risk factors tested, age and sex were found to be significantly associated with the prevalence of disease while pregnancy and herd type had no association with the prevalence of the disease. This study illustrate that FMD is highly prevalent in sheep and goats in Punjab. Therefore, a broader study is needed to ascertain the countrywide prevalence of FMD in small ruminants.
