BACKGROUND: Although the incidence of postoperative delirium (POD) in the elderly after surgery are rising as individuals are living longer, the pathogenesis of POD remains poorly understood. It has been suggested that miRNA-320 may play a role in POD based on animal study and human study. METHODS: We first carried out an animal study, and designed and conducted a human study based on the result of animal study. The aged rats were randomly assigned to five groups: the control (C), anesthesia and surgery (AS), saline (NS), agomir-320 (AG), and antagomir-320 (AT) groups. Postoperative spatial learning and memory in rats were analyzed by the Morris water maze and the open field tests. The plasma levels of insulin-like growth factor-1 (IGF-1), amyloid precursor protein (APP) proteins, miRNA320 and IGF-1mRNA were measured by ELISA and qRT-PCR, respectively. A total of 240 Chinese Han patients over 65 years who underwent tibial fracture internal fixation were included in the PNDABLE study. POD cases and non-POD controls (1:1 matched) were selected by an anesthesiologist using Confusion Assessment Method. RESULTS: For Group AS, the escape latency was significantly longer and the ratio of time spent in the target quadrant was significantly reduced, APP and miR-320 were upregulated and IGF-1mRNA was downregulated compared with Group C. For Group AG, the escape latency was significantly longer and the ratio of time spent in the target quadrant was significantly reduced, APP and miR-320 were upregulated and IGF-1mRNA was downregulated compared with Group AS. For Group AT, the escape latency was significantly reduced and the ratio of time spent in the target quadrant was significantly longer, APP and miR-320 were downregulated and IGF-1mRNAwas upregulated compared with Group AS. Compared with NPOD patients, the expressions of plasma miR-320 and APP protein were increased and the expression of plasma IGF-1 mRNA was decreased in POD patients after surgery. CONCLUSIONS: MiRNA-320 might play a role in up-regulating the levels of IGF-1mRNA and APP protein, which offered a new target for POD treatment. OBJECTIVE: To investigate the effects of stellate ganglion block on AMP-activated protein kinase and astrocyte in hippocampal neurons in postoperative aged rats. METHODS: A total of 96 male Sprague-Dawley rats aged 18-20 months, weighing 450-550 g, were randomly divided into 4 groups (n = 24): Control group (Group I); Surgery group (Group II); Normal saline + surgery group (Group III); SGB+ surgery group (Group IV). SGB with 0.25% bupivacaine 0.15 ml and operation was conducted after 15 min in groups II, III and IV. The eight rats were randomly sacrificed in each group postoperative 1, 3, and 5 d. The escape latency and swimming distance were recorded by morris water maze. The expression of AMPKmRNA,AMPK and phosphorylated AMPK(p-AMPK) was measured by RT-PCR and Western blot. The expression of GFAP was measured by immunohistochemisty. RESULTS: Compared with I group, the escape latency and swimming distance were significantly prolonged and the expression of AMPK mRNA, AMPK and p-AMPK was up-regulated and the expression of GFAP in astrocyte was significatively increased in groups II, III and IV. Compared with II and III group, the escape latency and swimming distance were significantly shorten and the expression of AMPK mRNA, AMPK and p-AMPK was down-regulated and the expression of GFAP in astrocyte was significatively decreased in group IV. CONCLUSION: SGB can improve the postoperative cognitive function, and the mechanism may be associated with down-regulating the expression of AMPK and restraining the activation of the astrocyte.

The population structure of Protosalanx chinensis is affected by a variety of factors, including water environment, intraspecific differentiation, interspecific competition and commercial fishing. To investigate the growth performance and the current status of resource utilization of P. chinensis in two important fish farming grounds (Dalong Lake and Amuta Lake) in Daqing, Northeast China, we collected 1513 P. chinensis samples from April to December 2018 for measurement and recording the basic biological characteristics. We estimated the parameters including population growth, mortality and exploitation using Von Bertalanffy growth formula, Beverton-Holt model, Pauly's empirical formula and length-based Bayesian biomass estimation method. The results showed that the asymptotic body lengths of Dalong Lake (n=660) and Amuta Lake (n=853) populations were 183.75 and 169.10 mm, respectively. The growth performance indices were both 4.85, and the body mass growth equations were Wt=29.29×(1-e-2.1(t+0.07))3.2977 and Wt=28.72×(1-e-2.5(t+0.07))3.4168, respectively. The ages at inflection were 0.49 a and 0.43 a, and the condition factors were 5.30×10-4±1.38×10-4 and 4.00×10-4±1.61×10-4. The exploitation rates were 0.60 and 0.61, and the relative catch per unit of supplementation were 0.069 and 0.075, respectively. The current differences in population resources between Dalong Lake and Amuta Lake in Daqing were mainly manifested in the parameters related to growth characteristics. The main reason for such differences was changes in the survival conditions of P. chinensis populations after the period of dietary transition induced by the density differences.

Metastasis/recurrence has been the most fundamental characteristic of hepatocellular cancer (HCC) and the ultimate cause of most HCC-related deaths. However, there are still a limited number of reliable tumor markers that can be used to predict the possibility of metastasis/recurrence in an HCC patient after operation. Recently, much evidence has shown that glypican-3 (GPC3) can be a useful tool to identify the early development of HCC, but little research has been done to test its usefulness as a prognostic marker related to post-operative metastasis/recurrence in HCC patients. In this study, the expression of GPC3 and its relationship with clinicopathological factors were determined by immunohistochemical analysis in 61 primary HCC patients. The potential prognostic value of GPC3 was investigated by comparing the survival time between HCC patients with high and low GPC3 expression. The results demonstrated that GPC3 expression was closely related with metastasis/recurrence in an HCC patient who can receive the operation. The risk of metastasis/recurrence after surgery in an HCC patient with high GPC3 expression was increased to 3.214 as compared to that of an HCC patient with low GPC3 expression. Survival analysis showed that HCC patients with high GPC3 expression had a significantly shorter overall survival time than HCC patients with low GPC3 expression (P=0.003). Further, multivariate analysis showed that GPC3 expression was a significant, independent prognostic parameter (P=0.030) for HCC patients. Overall, the study indicates that GPC3 might be a valuable marker closely related with prognosis and post-operative metastasis/recurrence in HCC patients.

RESUMO

Four styles of spectral line shape function and their transformation relation were investigated theoretically. The photon number and energy distribution as functions of frequency and wavelength resulting from Doppler broadening were investigated, and the maximum intensity and FWHM (full width at half maximum intensity) were obtained. The results show that the photon number distribution as function of frequency is almost the same as the energy distribution as function of frequency, while the photon number distribution as function of wavelength is almost the same as the energy distribution as function of wavelength. The method has been presented by analyzing the Doppler spectral line shape function, with which the ratio of density between two spectral lines can be obtained according to the ratio of maximum value of two spectral lines.

Electroacupuncture preconditioning at acupoint Baihui (GV20) can reduce focal cerebral ischemia/reperfusion injury. However, the precise protective mechanism remains unknown. Mitochondrial fission mediated by dynamin-related protein 1 (Drp1) can trigger neuronal apoptosis following cerebral ischemia/reperfusion injury. Herein, we examined the hypothesis that electroacupuncture pretreatment can regulate Drp1, and thus inhibit mitochondrial fission to provide cerebral protection. Rat models of focal cerebral ischemia/reperfusion injury were established by middle cerebral artery occlusion at 24 hours after 5 consecutive days of preconditioning with electroacupuncture at GV20 (depth 2 mm, intensity 1 mA, frequency 2/15 Hz, for 30 minutes, once a day). Neurological function was assessed using the Longa neurological deficit score. Pathological changes in the ischemic penumbra on the injury side were assessed by hematoxylin-eosin staining. Cellular apoptosis in the ischemic penumbra on the injury side was assessed by terminal deoxyribonucleotidyl transferase-mediated dUTP-digoxigenin nick end labeling staining. Mitochondrial ultrastructure in the ischemic penumbra on the injury side was assessed by transmission electron microscopy. Drp1 and cytochrome c expression in the ischemic penumbra on the injury side were assessed by western blot assay. Results showed that electroacupuncture preconditioning decreased expression of total and mitochondrial Drp1, decreased expression of total and cytosolic cytochrome c, maintained mitochondrial morphology and reduced the proportion of apoptotic cells in the ischemic penumbra on the injury side, with associated improvements in neurological function. These data suggest that electroacupuncture preconditioning-induced neuronal protection involves inhibition of the expression and translocation of Drp1.

Dielectric barrier discharge has become a hot issue in low temperature plasma research field because of wide prospect for its application in industry. In the present paper, an uniform glow discharge was realized in atmospheric pressure helium with a parallel planar dielectric barrier discharge device and an uniform plasma was generated in helium at atmospheric pressure. Electrical method was used to separate the discharge current from the total current signal and the waveform of discharge current could be obtained. The relations among the discharge current, applied voltage, gap voltage and wall charge during the uniform glow discharge were used to analyze the dynamical behavior of wall charges during their accumulation. The results show that the wall charges were mainly accumulated during the existence of discharge current pulse, while the quantity of wall charges continued to increase until the gap voltage changed its polarity after the disappearance of discharge current pulse, and finally the quantity of wall charges decreased until the next breakdown. These experimental results are important for the further study of wall charges' effect in the dielectric barrier glow discharge and its application in industry.

The electron excitation temperature was measured by the intensity ratio of two spectral lines in argon dielectric barrier discharge (DBD) at atmospheric pressure. The spectral range is from 690 to 800 nm. It is shown that all of the spectral lines are attributed to neutral Ar atoms. The spectral line 763.51 nm (2P(6)-->1S(5)) and 772.42 nm (2P(2)-->1S(3)) are chosen to estimate the electron excitation temperature. The experimental results show that the electron excitation temperature is in the range of 0.1-0.5 eV. The electron excitation temperature increases with increasing applied voltage, but decreases with increasing gas flow rate. The electron excitation temperature in flowing Ar gas discharge is much different from that in static Ar gas discharge. The result is of great importance to industrial application of DBD.

Neovascularization induced by vascular endothelial growth factor (VEGF) represents an appealing approach for treating ischemic heart disease. However, VEGF therapy has been associated with transient therapeutic effects and potential risk for hemangioma growth. Adult mesenchymal stem cells (MSCs) derived from bone marrow are a promising source for tissue regeneration and repair. In order to achieve a safe and persistent angiogenic effect, we have explored the potential of autologous MSCs transplantation to enhance angiogenesis and cardiac function of ischemic hearts. One week after myocardial infarction induced by occlusion of left anterior descending artery, autologous MSCs expanded in vitro was administrated intramyocardially into the infarct area of the same donor rats. By 2 months, MSCs implantation significantly elevated VEGF expression levels, accompanied by increased vascular density and regional blood flow in the infarct zone. The neovascularization resulted in a decreased apoptosis of hypertrophied myocytes and markedly improved the left ventricular contractility (ejection fraction: 79.9+/-7.6% vs. 37.2+/-6.9% in control animals). Therefore, mechanisms underlying MSCs improvement of cardiac functions may involve neovascularization induced by differentiation of MSCs to endothelial cells and para-secretion of growth factors, in addition to the apoptosis reduction and previously reported cardiomyocytes regeneration. Two months after cell transplantation, there are significant improvement of left ventricular function. Hence, autologous MSCs transplantation may represent a promising therapeutic strategy free of ethical concerns and immune rejection, for neovascularization in ischemic heart diseases.

A new HPLC method for the determination of paeoniflorin in rat serum with solid-phase extraction (SPE) for preconcentration is introduced. Paeoniflorin and an internal standard (pentoxifylline) were extracted from serum by means of SPE using cartridges with octadecyl chemically bound phase. The HPLC separation was then performed on a reversed-phase C(18) column using acetonitrile-water (18:82, v/v) as eluting solvent system, and UV detection at 230 nm to measure the analyte with a limit of quantitation about 10 ng ml(-1). The calibration curve for paeoniflorin was linear (r=0.9938) in the concentration range of 10-1200 ng ml(-1), both intra- and inter-day precision of the paeoniflorin were determined and their coefficience of variation did not exceed 10%. The validated method has been successfully applied for pharmacokinetic studies of paeoniflorin from rat serum after oral administration of Guan-Xin-Er-Hao decoction.

The experimental setup of dielectric barrier discharge was designed which is propitious to optical measurement. Temporal behavior of light emission of dielectric barrier discharges (filamentary model) in air at atmospheric pressure was measured by using optical method. Temporal behavior of dielectric barrier discharges was obtained. The experimental results show that the discharge burst in each half cycle of applied voltage consists of a series of discharge pulses, the duration of each discharge pulse is about 30-50 ns, and the interval of the neighboring discharge pulses is about a few hundred ns. The result is of great importance to the application of dielectric barrier discharges.

RESUMO

The global light emission of dielectric barrier discharge with pattern mode in air was measured and compared with the global current obtained with a small resistor. The results show that the moments and the amplitudes of the pulses in light emission correspond to those in the global current, respectively. So the discharge current can be measured by optical methods. Further more, the temporal aspect of the microdischarges in dielectric barrier discharge was obtained by using this method. It is believed that the results are of great importance to the study of spatiotemporal dynamics in dielectric barrier discharge, and are valuable to the study of gas discharge as well.
