RESUMO

BACKGROUND: ΦC31 integrase, a site-specific recombinase, can efficiently target attB-bearing transgenes to endogenous pseudo attP sites within mammalian genomes. The sequence features of endogenous binding sites will help us to fully understand the site-specific recognition function by ΦC31 integrase. The present study was aimed to uncover the global map of ΦC31 integrase binding sites in bovine cells and analysis the features of these binding sites by comprehensive bioinformatics methods. RESULTS: In this study, we constructed a ChIP-seq method that can be used to uncover the global binding sites by phiC31 integrase. 6740 potential ΦC31 integrase binding sites were identified. A sequence motif was found that contains inverted repeats and has similarities to wild-type attP site. Using REPEATMASKER, we identified a total of 20,183 repeat-regions distributed in 50 repeat types for the 6740 binding sites. These sites enriched in "regulation of GTPase activity" of in the GO category of biological process and KEGG pathway of signal transmembrane transporter activity. CONCLUSION: This study is the first time to uncover the global map of binding sites for ΦC31 integrase using ChIP-sequencing method and analysis the features of these binding sites. This method will help us to fully understand the mechanism of the site-specific integration function by phiC31 integrase and will potentially boost its genetic manipulations in both gene therapy and generation of transgenic animals.

RESUMO

Current marine oil spill detection and monitoring methods using high-resolution remote sensing imagery are quite limited. This study presented a new bottom-up and top-down visual saliency model. We used Landsat 8, GF-1, MAMS, HJ-1 oil spill imagery as dataset. A simplified, graph-based visual saliency model was used to extract bottom-up saliency. It could identify the regions with high visual saliency object in the ocean. A spectral similarity match model was used to obtain top-down saliency. It could distinguish oil regions and exclude the other salient interference by spectrums. The regions of interest containing oil spills were integrated using these complementary saliency detection steps. Then, the genetic neural network was used to complete the image classification. These steps increased the speed of analysis. For the test dataset, the average running time of the entire process to detect regions of interest was 204.56 s. During image segmentation, the oil spill was extracted using a genetic neural network. The classification results showed that the method had a low false-alarm rate (high accuracy of 91.42%) and was able to increase the speed of the detection process (fast runtime of 19.88 s). The test image dataset was composed of different types of features over large areas in complicated imaging conditions. The proposed model was proved to be robust in complex sea conditions.

RESUMO

Oil spills generate a large cost in environmental and economic terms. Their identification plays an important role in oil-spill response. We propose an oil spill detection method with improved adaptive enhancement on X-band marine radar systems. The radar images used in this paper were acquired on 21 July 2010, from the teaching-training ship "YUKUN" of the Dalian Maritime University. According to the shape characteristic of co-channel interference, two convolutional filters are used to detect the location of the interference, followed by a mean filter to erase the interference. Small objects, such as bright speckles, are taken as a mask in the radar image and improved by the Fields-of-Experts model. The region marked by strong reflected signals from the sea's surface is selected to identify oil spills. The selected region is subject to improved adaptive enhancement designed based on features of radar images. With the proposed adaptive enhancement technique, calculated oil spill detection is comparable to visual interpretation in accuracy.

RESUMO

OBJECTIVE: To provide references for effective implementing cost management for public hospitals through establishing time-driven activity-based management (TDABM) system. The TDABM system was established from hospital cost accounting, budget, control, and performance. RESULTS: The established TDABM system could improve the precision of hospital cost accounting, improve medical staff's working efficiency, realize the whole process of cost management, and enhance the competitiveness of the hospital. CONCLUSION: The activity of implementing TDABM in public hospitals had practical significance.

RESUMO

Pressure enhanced coagulation and sedimentation technique is an effective way for blue algae treatment. It is not clear whether Cyanobacteria balloon rupture will cause Cyanobacteria cells rupture, resulting in high intracellular concentrations of microcystin LR leak into the water, affecting drinking water safety. Therefore, in this study experimental comparative study of pressure and pre-oxidation of water containing Cyanobacteria was carried out to examine the microcystin LR concentration changes and Cyanobacteria removal efficiency. The results showed that microcystin concentration increase was not significant by the pre-treatment with Cyanobacteria water pressure, while the pre-oxidation process caused a significant increase in the concentration of microcystin. After 0.5-0.8 MPa pressure coagulation and sedimentation, removal of Cyanobacteria basically was over 90%, up to 93.5%, while the removal rate by pre-oxidation was low and unstable. Effluent turbidity is also significantly better in the pre-pressure method than the pre-oxidation. The results indicated that pressure enhanced coagulation is a safe and reliable method for Cyanobacteria removal.
