Two studies were conducted to determine the effect of the supplementation of sugar, starch or both on feed, nutrient and energy intake and total tract digestibility in four adult female addax (Addax nasomaculatus) and four adult male Reeves's muntjac (Muntiacus reevesi) - representatives of grazing and browsing ruminants, respectively. Studies in both species were conducted according to 4 × 4 Latin Square Design. Animals had free access to meadow hay (addax) or dehydrated chopped lucerne (muntjac), and were fed a restricted amount of a 'basal diet' consisting of: (1) wheat bran; (2) wheat bran and sucrose (source of sugar); (3) wheat bran and wheat (source of starch); or (4) wheat bran, sucrose and wheat. The amounts of supplemental sucrose and wheat were set to account for 2% and 15%, respectively, of dry matter (DM) consumed. There was no effect of the ~2% sugar supplementation on DM intake of hay by addax, while the ~10% starch supplementation reduced DM intake of hay by 13% (p < 0.01); total DM intake (of hay and the basal diet) was not affected neither by sugar nor starch supplementation. When the diet for addax included wheat, this resulted in a greater intake of crude protein by 15%, lower intake of ADF by 9%, and greater crude protein digestibility by 10% (p ≤ 0.05). The ~2% sugar supplementation did not affect intake of lucerne and total DM intake by muntjac, but the ~10% starch supplementation decreased DM intake of lucerne by 25% (p < 0.01), total DM intake by 7% (p = 0.02) and intake of all nutrients (p ≤ 0.10). In summary, if high intake of roughages by captive ruminants is fundamental for their gastrointestinal functions and health, then starchy feeds supplementation should be limited, as they have an especially negative impact on roughage intake.

The aim of this study was to determine the effect of an increased mono- and disaccharide (MD) intake on selected functions and structure of the gastrointestinal tract (GIT), and selected blood parameters in Reeves's muntjac (Muntiacus reevesi), a small browsing ruminant. Eighteen male muntjacs were fed diets consisting of lucerne (ad libitum), a high fibre pellet (100 g/day) and wheat bran (30 g/day) without (MD0) or with addition of 10 or 20 g of glucose, fructose and sucrose mixture/day (MD10 and MD20, respectively) for 14 days. MD dosages were set to increase intake of these saccharides by 25% and 50% relative to MD0, which resulted in a range of water-soluble carbohydrate content in the consumed dry matter from 7% to 12%. Compared to MD0 animals, MD20 animals had a lower dry matter intake, a higher MD concentrations in the reticulorumen (RR), abomasal and small intestinal digesta, higher ruminal butyrate concentration, higher SGLT1 expression in the epithelium of proximal jejunum, higher plasma glucose, lower RR tissue weight but greater caecal tissue weight (p ≤ 0.05), and had or tended to have shorter papillae and lower mucosa surface area in the Atrium ruminis (by 44%; p = 0.02 and p = 0.10, respectively); MD10 animals tended to have higher MD concentrations in the abomasal and small intestinal digesta (p ≤ 0.10), and a higher amylolytic activity (p = 0.02) as well as a tendency to lower xylanolytic activity in the RR digesta (p = 0.06). MD supplementation did not affect ruminal pH. In conclusion, low to moderate increase of MD intake increased MD concentrations in the RR, abomasal and intestinal digesta, and SGLT1 expression in intestinal epithelium, suggesting incomplete fermentation of those saccharides in the RR. MD supplementation dose-dependently affects structure of GIT in Reeves's muntjac.

Due to the presence of toxic pollutants, soils in former military areas need remedial actions with environmentally friendly methods. Greenhouse experiments were conducted to investigate the aided phytostabilization of multi-heavy metals (HMs), i.e. Cd, Cr, Cu, Ni, Pb, Zn, in post-military soil by Festuca rubra and three mineral amendments (diatomite, dolomite and halloysite). The amendments were applied at 0 and 3.0% to each pot filled with 5 kg of polluted soil. After seven weeks of the phytostabilization, selected soil properties, biomass yield of F. rubra and immobilization of HMs by their accumulation in plant and redistribution among individual fractions in soil were determined. In addition, ecotoxicology parameters of non-amended and amended soil were established using Phytotoxkit (Sinapsis alba) and Ostracodtoxkit (Heterocypris incongruens) tests. The addition of halloysite significantly increased F. rubra biomass. Diatomite significantly increased both the Cd, Cu, Pb and Cr concentrations in the roots and the pH of the soil. The application of halloysite significantly decreased the Cd and Zn contents of the soil after the completion of the experiment. Dolomite and halloysite were more effective in HM immobilization in soil by decreasing their content in an exchangeable fraction than diatomite. These soil amendments significantly differentiated the length of S. alba roots and had a positive effect on the development of H. incongruens.

BACKGROUND: As early as pregnancy, maternal mental stress impinges on the child's development and health. Thus, this may cause enhanced risk for premature birth, lowered fetal growth, and lower fetal birth weight as well as enhanced levels of the stress hormone cortisol and lowered levels of the bonding hormone oxytocin. Maternal stress further reduces maternal sensitivity for the child's needs which impairs the mother-child-interaction and bonding. Therefore, prevention and intervention studies on mental stress are necessary, beginning prenatally and applying rigorous research methodology, such as randomized controlled trials, to ensure high validity. METHODS: A randomized controlled trial is used to assess the impact of psychotherapy and telemedicine on maternal mental stress and the child's mental and physical health. Mentally stressed pregnant women are randomized to an intervention (IG) and a not intervened control group. The IG receives an individualized psychotherapy starting prenatal and lasting for 10 months. Afterwards, a second randomization is used to investigate whether the use of telemedicine can stabilize the therapeutic effects. Using ecological momentary assessments and video recordings, the transfer into daily life, maternal sensitivity and mother-child-bonding are assessed. Psycho-biologically, the synchronicity of cortisol and oxytocin levels between mother and child are assessed as well as the peptidome of the colostrum and breast milk, which are assumed to be essential for the adaptation to the extra-uterine environment. All assessments are compared to an additional control group of healthy women. Finally, the results of the study will lead to the development of a qualification measure for health professionals to detect mental stress, to treat it with low-level interventions and to refer those women with high stress levels to mental health professionals. DISCUSSION: The study aims to prevent the transgenerational transfer of psychiatric and somatic disorders from the mother to her child. The effects of the psychotherapy will be stabilized through telemedicine and long-term impacts on the child's and mothers' mental health are enhanced. The combination of psychotherapy, telemedicine and methodologies of ecological momentary assessment, video recording and bio banking are new in content-related and methodological manner. This research paper addresses the hypothesis that calcium salts combined with whole linseed and heat-treated rapeseed cake in one feed additive may efficiently stimulate the productivity of dairy cows and have a positive effect on the functional (health-promoting) properties of milk fat. The article proposes the composition of such an additive (EFA) and evaluates its nutritional effect in the diet of mid-lactation dairy cows. Forty multiparous Polish Holstein-Friesian (PHF) dairy cows were allocated to one of four treatments (10 cows/treatment) and fed a TMR diet without EFA or with EFA in the amount of 1, 2 or 3 kg/d per head for a 63-d-period. Individual intake of dry matter (DMI) and nutrients was determined, as was milk yield and composition, including fatty acid profile, fat soluble vitamins, cholesterol and phospholipids (PLs). Irrespective of the treatment group, cows fed diets with EFA had higher (P < 0.05) DMI, milk yield and milk vitamin D3 and K2 concentration but lower (P < 0.01) milk protein, fat and cholesterol contents. The additive did not affect the milk concentrations of ß-carotene or vitamin A or E. The PLs content was correlated with fat concentration in the milk and decreased as the level of EFA in the diet increased. An increase in phosphatidylcholine in total PLs was accompanied by a reduction in the proportion of sphingomyelin (P < 0.05). The use of EFA increased the proportion of polyunsaturated fatty acids (PUFA) in the total fatty acids in the milk. The addition of EFA in the amount of 3 kg increased the proportion of PUFA by 77% (P < 0.05). In conclusion, the use of an energy-protein feed additive (EFA) increases feed intake and milk yield in cows and alters milk fat composition, improving its functional properties. Higher milk production compensates for the decrease in solids concentration in the milk, which has no effect on their daily yield.

The aim of the study was to determine the effect of fructose supplementation in the diet on feed intake, nutrient digestibility and digesta retention time in Reeves's muntjac (Muntiacus reevesi), a browsing cervid. In Experiment 1, six adult males of Reeves's muntjac were used in a replicated 3 × 3 Latin square design and fed a diet consisting of dehydrated chopped lucerne (ad libitum), high-fibre pellet (120 g/day) and wheat bran (30 g/day) without (F0) or with addition of 12 and 24 g fructose/day (F12 and F24, respectively). In Experiment 2, the same six adult muntjacs were used in crossover design and fed F0 or F12. Doses of supplemental fructose were set to increase intake of water-soluble carbohydrates (WSC; ≈40 g/day; ≈8% of WSC in consumed dry matter [DM]) by 25 and 50% relative to F0. Feed intake was controlled daily (Experiment 1 and 2) and total tract digestibility and digesta retention time were determined (Experiment 2). In Experiment 1, DM intake of chopped dehydrated lucerne decreased with fructose supplementation (F0 vs. F12 and F24; p = .01) but was not different between F12 and F24 (p = .76). Total DM intake was also not different between treatments (p ≥ .13). In Experiment 2, DM intake of lucerne, total DM intake and nutrient digestibility was not affected by fructose supplementation (p ≥ .17), but mean retention time of long particles in the whole GIT tended to be longer for F12 compared to F0 (p = .09). Under conditions of the current study, additional fructose intake (resulting in a range of WSC content in consumed DM from 8.6% to 13%) had only minor impact on feed intake and investigated functions of the gastrointestinal tract of Reeves's muntjac.

AIMS: Catheter-associated urinary tract infections (CAUTI) caused by Proteus mirabilis are very difficult to treat due to the ability of biofilm formation and drug resistance of these bacteria. The aim of this study was to assess the antibiofilm activity of phages and develop phage cocktail to combat biofilm of P. mirabilis strains. METHODS AND RESULTS: Planktonic forms and biofilms of 50 tested uropathogenic P. mirabilis strains showed different sensitivity to 13 phages used. Phages 39APmC32, 65APm2833 and 72APm5211 presenting strong antibiofilm activity were selected as cocktail components. The antibiofilm activity of phage cocktails was similar or slightly higher than that of the most active phage. A three-phage cocktail inhibited biofilm formation and destroyed biofilms of the same number of strains or 2-3 strains more compared to a single phage. The components of the three-phage cocktail did not block each other's activity. CONCLUSIONS: The potential of developed anti-P. mirabilis phage cocktail as an antibiofilm agent was proved. SIGNIFICANCE AND IMPACT OF THE STUDY: In this study, three broad host range phages presenting strong anti-P. mirabilis biofilm activity were selected. Additionally, high stability of these viruses makes them a useful tool for controlling the biofilms.

The aim of this study was to characterize sewage sludge from mechanical-biological wastewater treatment plants (WWTPs) in terms of total concentrations of heavy metals (HMs) and their distribution. Moreover, HM mobility, stability and ecological risk were assessed by calculating the mobility factor (MF), the reduced partition index (Ir) and the ecological risk index (ERI). Hierarchical cluster analysis (CA) was used to group the HMs in the sludges on the basis of their MF, Ir and ERI. The concentration of HMs decreased in the following order: Zn > Cu > Ni > Pb > Cd. HM mobility, as indicated by the MF values, was as follows: 18.7%-30.6% (Zn), 8.1%-36.4% (Ni), 6.8%-11.1% (Cu), 3.2%-32.2% (Pb) and 0% (Cd). Based on the average Ir values, the stability of the metals in the sludge increased in this order: Zn (0.23)=Pb (0.23)

BACKGROUND: Congenital toxoplasmosis is associated with severe complications. German state health insurance covers rubella, but not toxoplasmosis, immunity screening. We analysed the effect of socioeconomic factors on the efficiency of private toxoplasmosis screening during pregnancy. METHODS: Toxoplasmosis and rubella screening data (n = 5402 mothers) were collected within the population-based Survey of Neonates in Pomerania (SNiP). RESULTS: At the first-trimester screening, 34.4 % (88.1 %) of expecting mothers were immune to toxoplasmosis (rubella). Susceptibility for toxoplasmosis (rubella) was observed in 39.6 % (8.9 %) and 25.8 % (2.95 %) were not tested. Data on a 2(nd) screening were available in a subgroup of women with negative immunity showing less than 45 % participation rate. Active toxoplasmosis (no rubella) infection was observed in 0.3 % (n = 17) of pregnant women. A multiple logistic regression model (AIC = 719.67; AUC = 0.725) revealed that the likelihood of participating in a second toxoplasmosis screening increased among women with a good level of education and a steady partnership and decreased with paternal unemployment and the absence of breastfeeding. The highest probability of non-participation in toxoplasmosis screening was found among women with temporal burden and family responsibilities. A cost-benefit analysis showed that covering general screening for toxoplasmosis with health insurance saved costs. CONCLUSION: Toxoplasmosis carried a substantial risk of infection during pregnancy. Although increased socioeconomic status was positively associated with the participation in toxoplasmosis screening, this was not the case when pregnant women had strong temporal burden and family responsibilities. This data supports the need for toxoplasmosis screening among pregnant women as a general healthcare benefit covered by insurance.

Ovarian cancer still represents a challenge in gynecological oncology. Most patients are diagnosed in an advanced tumor stage. No specific screening or prevention strategies for ovarian cancer exist as of yet. Interleukin 8 (IL-8) is a pro-inflammatory chemokine known for its angiogenetic activity, and is supposedly responsible for tumor-associated angiogenesis in several malignant tumors. The aim of the study was to investigate the susceptibility of patients with an IL-8 gene polymorphism to developing ovarian cancer. Four single nucleotide polymorphisms (SNPs) (IL-8 -251, IL-8 +781, IL-8 +1633 and IL-8 +2767) of the IL-8 gene were screened, using the PCR method in 268 patients with ovarian cancer and 426 healthy women as a control group. Significant associations were noted in patients with the IL-8 +781 (T/T) genotype (p=0.0048) with increased frequencies of ovarian cancer, while women with the IL-8 +781 (C/C) allele suffer from ovarian cancer significantly less frequently (p=0.0003). Furthermore, the IL-8 +2767 (T/T) genotype is also associated with a higher risk of ovarian cancer (p=0.0177). Our results indicate, for the first time, that IL-8 polymorphism is associated with ovarian cancer.

In the luteal phase, human endometrial stromal cells (HESCs) undergo proliferation, migration and differentiation during the decidualization process under the control of the ovarian steroids progesterone and estrogen. Proper decidualization of stromal cells is required for blastocyst implantation and the development of pregnancy. The proliferation, migration and differentiation of HESCs in decidualization do not require the presence of a blastocyst but are greatly accelerated during implantation. Lysophosphatidic acid (LPA) and sphingosine-1-phosphate (S1P) are potent bioactive lysophospholipids that have critical roles in various physiological and pathophysiological processes, including inflammation, angiogenesis and cancer. The expression of the enzymes involved in LPA and S1P turnover and their receptors in HESCs during decidualization has not been characterized yet. We found that the LPAR1 and LPAR6 and S1PR3 receptors are highly expressed in HESCs. LPAR1, autotaxin (ATX), an LPA producing enzyme and lipid phosphate phosphatase 3 were up-regulated during decidualization. Interestingly, the expression of all S1P receptor subtypes and LPA receptors (LPAR2-6) mRNA was down-regulated after decidualization. We found that SPHK1 is highly expressed in HESCs, and is up-regulated during decidualization. S1P phosphatase SGPP1 and S1P lyase SGPL1 are highly expressed in HESCs. SGPP1 mRNA expression was significantly up-regulated in decidualized HESCs. In conclusion, this study shows the first time that specific LPA and S1P receptors and their metabolizing enzymes are highly regulated in HESCs during decidualization. Furthermore, we suggest that LPAR1 receptor-mediated signaling in HESCs may be crucial in decidualization process. SPHK1 activity and high turnover of S1P and LPA might be essential for precise regulation of their signaling during decidualization of human endometrium and implantation.

Thermal treatment of sewage sludge produces fly ash, also known as the air pollution control residue (APCR), which may be recycled as a component of artificial lightweight aggregates (ALWA). Properties of APCR are typical: high content of Ca, Mg, P2O5, as well as potential to induce alkaline reactions. These properties indicate that ALWA prepared with a high content of APCR may remove heavy metals, phosphorus, and ammonium nitrogen from wastewater with high efficiency. The aim of this preliminary study was to determine the optimal composition of ALWA for potential use as a filter media in wastewater treatment systems. Five kinds of ALWA were produced, with different proportions of ash (shown as percentages in subscripts) in mixture with bentonite: ALWA0 (reference), ALWA12.5, ALWA25, ALWA50, and ALWA100. The following parameters of ALWA were determined: density, bulk density, compressive strength, hydraulic conductivity, and removal efficiency of ions Zn(2+), NH4 (+), and PO4 (3-). Tests showed that ALWA had good mechanical and hydraulic properties, and might be used in wastewater filtering systems. Phosphates and zinc ions were removed with high efficiency (80-96%) by ALWA25-100 in static (batch) conditions. The efficiency of ammonium nitrogen removal was low, <18%. Artificial wastewater treatment performance in dynamic conditions (through-flow), showed increasing removal efficiency of Zn(2+), PO4 (3-) with a decrease in flow rate.

This study aimed to determine the effect of supplementing liquid feeds with a mixture of ethyl esters of polyunsaturated fatty acid of linseed oil (EEPUFA; α-linolenic acid-64.5%, linoleic acid-16.1%, and oleic acid-19.4%) on feed intake, body weight gain, feed efficiency, and health of dairy calves. Thirty-six healthy female Holstein-Friesian calves (7 d of age, 41.2 ± 4.0 kg) were assigned to one of two treatment groups (18 calves per group), i.e., control or EEPUFA, and fed liquid feed (whole milk (WM) or milk replacer (MR)) either without or with 10 mL/d of EEPUFA supplementation, respectively, for 56 days (till 63 d of age). Average daily intake of WM and MR was similar between treatments (p = 0.94). Average daily total DM intake and average daily starter feed DM intake were higher for the EEPUFA group (p = 0.05 and p = 0.01, respectively). The average daily body weight gain was also higher for the EEPUFA group (55 g/d; p = 0.03), although final body weight turned out not to be significantly different between groups (75.6 kg vs. 79.0 kg, control vs. EEPUFA, respectively; p = 0.20). Supplementation of liquid feeds with EEPUFA did not affect feed efficiency (p = 0.37) and most of investigated health parameters. However, the percentage of days with diarrhea relative to the number of days receiving treatment was higher in the control group than the EEPUFA group (76 vs. 42, respectively; p = 0.04). Although the results of this preliminary study are promising, further research is needed to establish the dose effect of EEPUFA on the performance and health of calves.

Brucella spp. and Sinorhizobium meliloti are alphaproteobacteria that share not only an intracellular lifestyle in their respective hosts, but also a crucial requirement for cell envelope components and their timely regulation for a successful infectious cycle. Here, we report the characterization of Brucella melitensis mucR, which encodes a zinc finger transcriptional regulator that has previously been shown to be involved in cellular and mouse infections at early time points. MucR modulates the surface properties of the bacteria and their resistance to environmental stresses (i.e., oxidative stress, cationic peptide, and detergents). We show that B. melitensis mucR is a functional orthologue of S. meliloti mucR, because it was able to restore the production of succinoglycan in an S. meliloti mucR mutant, as detected by calcofluor staining. Similar to S. meliloti MucR, B. melitensis MucR also represses its own transcription and flagellar gene expression via the flagellar master regulator ftcR. More surprisingly, we demonstrate that MucR regulates a lipid A core modification in B. melitensis. These changes could account for the attenuated virulence of a mucR mutant. These data reinforce the idea that there is a common conserved circuitry between plant symbionts and animal pathogens that regulates the relationship they have with their hosts.

The threat posed by the degradation of the soil environment by metal (-oid)s has been lead to the improvement of existing or search for new remediation methods; in this case, the application of environmentally friendly nanomaterials falls into this trend. The study applied a technique of aided phytostabilization for the immobilization of metal (-oid)s in soil with the application of nanosized halloysite and biochar (nBH), along with Lolium perenne L. Its effectiveness was assessed in terms of changing temperature conditions (16 cycles of freeze and thaw cycles, (FTC)) on the content of As, Cu, Pb and Zn in the soil, roots, and above-ground parts of the tested plant, chemical fraction distributions of metal (-oid)s and their stability (based on reduced partition index, Ir). The biomass yield in nBH-amended soil was 2-fold higher compared to control soil, but it decreased by 1.6-fold after FTC. nBH facilitated more bioaccumulation of As, Pb and Zn than Cu in plant roots, before than after FTC. nBH increased pH in phytostabilized soil, but it was not affected by changing FTC. In soil nBH-phytostabilized total concentration of metal (-oid)s significantly decreased compared to control soil, for As and Cu below permissible value, regardless of FTC. Soil amendment and changing temperature conditions affected metal (-oid)s redistribution in soil. As a result, the stability of As increased from 0.50 to 0.66, Cu from 0.49 to 0.52, Pb from 0.36 to 0.48 and Zn from 0.39 to 0.47. These findings suggest that nBH can immobilize metal (-oid)s in phytostabilized soil under changing temperature conditions.

Removal of heavy metals (HMs) from soil is a priority in soil washing/soil flushing. However, for further management of remediated soil, it should be characterized in detail. This study presents, for the first time, an evaluation of soil quality after column flushing with new-generation washing agents (WAs) recovered from municipal sewage sludge (dissolved organic matter, DOM; soluble humic-like substances, HLS; soluble humic substances, SHS) and Na2EDTA as a standard benchmark. Sandy loam soil was spiked with industrial levels of Cu, Pb and Zn, then flushed in a column reactor at two WA flow rates (0.5 and 1.0 ml/min). Soil quality was assessed by determining both physico-chemical (pH, total HMs and their mobility, soil organic matter, OM, humic substances, HS and their fractions, macroelements) and biological indicators (dehydrogenase activity, DHA; germination rate, GR; and inhibition factors for roots and shoots of Triticum aestivum). Total residual HMs contents and HMs contents in the mobile fraction were significantly lower in soil flushed at 1.0 ml/min than in soil flushed at 0.5 ml/min. With all WAs, the decrease in Cu content was larger than that of the other HMs, however this HM most effectively was removed with DOM. In contrast, Pb most effectively was removed by HLS and Na2EDTA, and DOM should not be used to remediate Pb-contaminated soil, due to its very low effectiveness. Flow rate did not appear to affect the fertilizing properties of the soil, DHA activity or soil toxicity indicators. Soil flushing with all SS_WAs increased OM, HS, and exchangeable P, K and Na content in remediated soils, but decreased exchangeable Ca content, and in most cases, exchangeable Mg content. Soil flushing substantially improved DHA activity and GR, but only slightly improved the shoot and root inhibition factors.

This paper presents the first tests of Cu (7875 mg/kg), Pb (1414 mg/kg) and Zn (566 mg/kg) removal from contaminated soil with sewage-sludge-derived washing agents (SS_WAs) (dissolved organic matter, DOM; soluble humic-like substances, HLS; soluble humic substances, SHS) and Na2EDTA (as a standard benchmark) in column experiments. Flow rates of 0.5 ml/min and 1 ml/min were used. Using a 1. order kinetic model, the kinetic constant (k), the maximum concentrations of each metal removed (Cmax), and the initial rates of metal removal (r) were established. At both flow rates, stable flow velocity was maintained for approximately eight pore volumes, for flushing times of 8 h (1.0 ml/min) and 16 h (0.5 ml/min). Although the flow rate did not influence k, it influenced Cmax: at 1 ml/min, Cmax values were higher than at 0.5 ml/min. For Cu and Zn, but not Pb, k was about twofold higher with Na2EDTA than with SS_WAs. Although Na2EDTA gave the highest kCu, Cmax,Cu was highest with DOM (Na2EDTA, 66%; DOM 73%). For Pb removal, HLS was the most effective SS_WA (77%; Na2EDTA was 80% effective). kZn was about twofold higher with Na2EDTA than with SS_WAs. Cmax,Zn was highest with HLS. The quick mobilization of Cu, Pb and Zn with most of the WAs corresponded to efficient metal removal from the exchangeable (F1) fraction.

The soil system is a key component of the environment that can serve as a sink of pollutants. Using processed waste for aided phytostabilization of metals (HMs) in contaminated soils is an attractive phytoremediation technique that integrates waste utilization and recycling. In this study, we evaluated the effect of biologically and thermally processed wastes, i.e. sewage sludge compost (CSS), poultry feather ash (AGF) and willow chip biochar (BWC), on phytostabilization of contaminated soil from a steel disposal dump. Greenhouse experiments with Lupinus luteus L. and amendments (dosage: 3.0%, w/w) were conducted for 58 days. Soil toxicity was evaluated with Ostracodtoxkit and Phytotoxkit tests. At the end of the experiment, soil pH, plant biomass yield, and HM accumulation in plant tissues were determined. HM distribution, HM stability (reduced partition index) and potential environmental risk (mRI index) in the soil were assessed. During phytostabilization, changes in the diversity of the rhizospheric bacterial community were monitored. All amendments significantly increased soil pH and biomass yield and decreased soil phytotoxicity. AGF and BWC increased accumulation of individual HMs by L. luteus roots better than CSS (Cu and Cr, and Ni and Zn, respectively). The soil amendments did not improve Pb accumulation by the roots. Improvements in HM stability depended on amendment type: Ni and Pb stability were improved by all amendments; Zn stability, by AGF, and BWC; Cd stability, by AGF; and Cr stability, by BWC. AGF reduced the mRI most effectively. Microbial diversity in amended soils increased with time of phytostabilization and was up to 9% higher in CSS amended soil than in control soil. AGF application favored the abundance of the genera Arenimonas, Brevundimonas, Gemmatimonas and Variovorax, whose metabolic potential could have contributed to the better plant growth and lower mRI in that soil. In conclusion, AGF and BWC have great potential for restoring steel disposal dump areas, and the strategies researched here can contribute to achieving targets for sustainable development.

Biochar has received extensive attention because of its multi-functionality for agricultural and environmental applications. Despite its many benefits, there are concerns related to the long-term safety and implications of its application, mainly because the mechanisms affecting soil and organism health are poorly quantified and understood. This work reviews 259 sources and summarises existing knowledge on biochar's adverse effects on soil from a multiangle perspective, including the physicochemical changes in soil, reduced efficiency of agrochemicals, potentially toxic substances in biochar, and effects on soil biota. Suggestions are made for mitigation measures. Mixed findings are often reported; however, the results suggest that high doses of biochar in clay soils are likely to decrease available water content, and surface application of biochar to sandy soils likely increases erosion and particulate matter emissions. Furthermore, biochar may increase the likelihood of excessive soil salinity and decreased soil fertility because of an increase in the pH of alkaline soils causing nutrient precipitation. Regarding the impact of biochar on (agro)chemicals and the role of biochar-borne toxic substances, these factors cannot be neglected because of their apparent undesirable effects on target and non-target organisms, respectively. Concerning non-target biota, adverse effects on reproduction, growth, and DNA integrity of earthworms have been reported along with effects on soil microbiome such as a shift in the fungi-to-bacteria ratio. Given the diversity of effects that biochar may induce in soil, guidelines for future biochar use should adopt a structured and holistic approach that considers all positive and negative effects of biochar.

In recent years, a lot of attention has been given to searching for new additives which will effectively facilitate the process of immobilizing contaminants in the soil. This work considers the role of the enhanced nano zero valent iron (nZVI) strategy in the phytostabilization of soil contaminated with potentially toxic elements (PTEs). The experiment was carried out on soil that was highly contaminated with PTEs derived from areas in which metal waste had been stored for many years. The plants used comprised a mixture of grasses-Lolium perenne L. and Festuca rubra L. To determine the effect of the nZVI on the content of PTEs in soil and plants, the samples were analyzed using flame atomic absorption spectrometry (FAAS). The addition of nZVI significantly increased average plant biomass (38%), the contents of Cu (above 2-fold), Ni (44%), Cd (29%), Pb (68%), Zn (44%), and Cr (above 2-fold) in the roots as well as the soil pH. The addition of nZVI, on the other hand, was most effective in reducing the Zn content of soil when compared to the control series. Based on the investigations conducted, the application of nZVI to soil highly contaminated with PTEs is potentially beneficial for the restoration of polluted lands.
