BACKGROUND: Excessive rates of nitrogen (N) fertilizers may result in elevated concentrations of nitrate (NO3- ) in plants. Considering that many programs of biofortification with trace elements are being performed, it has become important to study how the application of these elements affects plant physiology and, particularly, N utilization in leaf crops. The main objective of the present study was to determine whether the NO3- accumulation and the nitrogen use efficiency was affected by the application of different doses of Zn in Lactuca sativa plants. RESULTS: Zn doses in the range 80-100 µmol L-1 produced an increase in Zn concentration provoking a decrease of NO3- concentration and increase of the nitrate reductase, glutamine synthetase and aspartate aminotransferase activities, as well as the photorespiration processes. As result, we observed an increase in reduced N, total N concentration and N utilization efficiency. Consequently, at a dose of 80 µmol L-1 of Zn, the amino acid concentration increased significantly. CONCLUSION: Adequate Zn fertilization is an important critical player in lettuce, especially at a dose of 80 µmol L-1 of Zn, because it could result in an increase in the Zn concentration, a reduction of NO3- levels and an increase the concentration of essential amino acids, with all of them having beneficial properties for the human diet. © 2016 Society of Chemical Industry.

Zinc (Zn) plays a crucial role in metabolism in both plant and animal life. Zn deficiency is a worldwide problem that has recently gotten worse. This micronutrient shortage can be largely attributed to eating foods that are poor in zinc. If biofortification methods were widely used, Zn enrichment of the organ or tissue of interest would increase dramatically. However, Zn absorption mechanisms in rice plants must be understood on a fundamental level before these methods can be used effectively. Plant systems' Zn transporters and metal chelators play a major role in regulating this intricate physiological characteristic. The Zn efficiency of specific species is affected by a variety of factors, including the plant's growth stage, edaphic conditions, the time of year, and more. Both old and new ways of breeding plants can be used for biofortification. We have highlighted the significance of recombinant and genetic approaches to biofortifying in rice. In this review, we have the metabolic role of zinc in rice, and the different transporter families involved in the transportation of zinc in rice. We have also discussed the combined approaches of agronomic and genetic in zinc biofortification in rice and potential outcomes and future predictions.

Introduction: One-third of the human population consumes insufficient zinc (Zn) to sustain a healthy life. Zn deficiency can be relieved by increasing the Zn concentration ([Zn]) in staple food crops through biofortification breeding. Rice is a poor source of Zn, and in countries predominantly relying on rice without sufficient dietary diversification, such as Madagascar, Zn biofortification is a priority. Methods: Multi-environmental trials were performed in Madagascar over two years, 2019 and 2020, to screen a total of 28 genotypes including local and imported germplasm. The trials were conducted in the highlands of Ankazomiriotra, Anjiro, and Behenji and in Morovoay, a location representative of the coastal ecosystem. Contributions of genotype (G), environment (E), and G by E interactions (GEIs) were investigated. Result: The grain [Zn] of local Malagasy rice varieties was similar to the internationally established grain [Zn] baseline of 18-20 µg/g for brown rice. While several imported breeding lines reached 50% of our breeding target set at +12 µg/g, only few met farmers' appreciation criteria. Levels of grain [Zn] were stable across E. The G effects accounted for a main fraction of the variation, 76% to 83% of the variation for year 1 and year 2 trials, respectively, while GEI effects were comparatively small, contributing 23% to 9%. This contrasted with dominant E and GEI effects for grain yield. Our results indicate that local varieties tested contained insufficient Zn to alleviate Zn malnutrition, and developing new Zn-biofortified varieties should therefore be a priority. GGE analysis did not distinguish mega-environments for grain [Zn], whereas at least three mega-environments existed for grain yield, differentiated by the presence of limiting environmental conditions and responsiveness to improved soil fertility. Discussion: Our main conclusion reveals that grain [Zn] seems to be under strong genetic control in the agro-climatic conditions of Madagascar. We could identify several interesting genotypes as potential donors for the breeding program, among those BF156, with a relatively stable grain [Zn] (AMMI stability value (ASV) = 0.89) reaching our target (>26 µg/g). While selection for grain yield, general adaptation, and farmers' appreciation would have to rely on multi-environment testing, selection for grain [Zn] could be centralized in earlier generations.

Wheat is an important food crop worldwide, providing substantial calories and nourishment. Genetic variability in wheat germplasm is crucial for the development of cultivars with desirable features. This two years study (2020-21 and 2021-22) was conducted to evaluate 13 diverse wheat genotypes factorially combined with foliar-applied zinc sulphate (0, 0.4, 0.6%) arranged in a triplicate randomized complete block design. Boxplot analysis revealed the significant (P < 0.01) phenotypic variation of wheat germplasm for all the studied traits, but maximum variation was observed for yield and Zn biofortification-related traits. Correlation and path analysis revealed a significant (P < 0.01) association among yield and biofortification-related traits. Zinc uptake showed maximum strength of association (r = 0.96, p < 0.01) with grain Zn concentration. The Biplot analysis showed the graphical representation of wheat accessions based on similar characteristics and then assort into distinct groups. Broadsense heritability (Hbs) was calculated to determine the proportion of variation transmitted to future generations. The high value of Hbs for yield and Zn biofortification-related traits indicates that these traits are governed by the additive type of gene action and can be fixed in early segregating generations. In crux, this study validated the genetic variability in existing wheat genotypes for yield and Zn biofortification-related traits and may be helpful to devise an efficient breeding program for wheat Zn biofortification.

Micronutrient deficiency is a serious health issue in resource-poor human populations worldwide, which is responsible for the death of millions of women and underage children in most developing countries. Zinc (Zn) malnutrition in middle- and lower-class families is rampant when daily calorie intake of staple cereals contains extremely low concentrations of micronutrients, especially Zn and Fe. Looking at the importance of the problem, the present investigation aimed to enhance the growth, yield, nutrient status, and biofortification of wheat crop by inoculation of native zinc-solubilizing Bacillus spp. in conjunction with soil-applied fertilizers (NPK) and zinc phosphate in saline soil. In this study, 175 bacterial isolates were recovered from the rhizosphere of wheat grown in the eastern parts of the Indo-Gangetic Plain of India. These isolates were further screened for Zn solubilization potential using sparingly insoluble zinc carbonate (ZnCO3), zinc oxide (ZnO), and zinc phosphate {Zn3(PO4)2} as a source of Zn under in vitro conditions. Of 175 bacterial isolates, 42 were found to solubilize either one or two or all the three insoluble Zn compounds, and subsequently, these isolates were identified based on 16S rRNA gene sequences. Based on zone halo diameter, solubilization efficiency, and amount of solubilized zinc, six potential bacterial strains, i.e., Bacillus altitudinis AJW-3, B. subtilis ABW-30, B. megaterium CHW-22, B. licheniformis MJW-38, Brevibacillus borstelensis CHW-2, and B. xiamenensis BLW-7, were further shortlisted for pot- and field-level evaluation in wheat crop. The results of the present investigation clearly indicated that these inoculants not only increase plant growth but also enhance the yield and yield attributes. Furthermore, bacterial inoculation also enhanced available nutrients and microbial activity in the wheat rhizosphere under pot experiments. It was observed that the application of B. megaterium CHW-22 significantly increased the Zn content in wheat straw and grains along with other nutrients (N, P, K, Fe, Cu, and Mn) followed by B. licheniformis MJW-38 as compared to other inoculants. By and large, similar observations were recorded under field conditions. Interestingly, when comparing the nutrient use efficiency (NUE) of wheat, bacterial inoculants showed their potential in enhancing the NUE in a greater way, which was further confirmed by correlation and principal component analyses. This study apparently provides evidence of Zn biofortification in wheat upon bacterial inoculation in conjunction with chemical fertilizers and zinc phosphate in degraded soil under both nethouse and field conditions.

Zinc (Zn) deficiency in soil is a serious constraint affecting the yield and nutritional quality of wheat and, in turn, human health. Zn fertilization for enhancing its density in grains is a prominent technological solution for the problem. Accordingly, the present study (pot experiment) was undertaken to (i) assess the impacts of different Zn fertilization technologies on yield, concentrations of Zn, phytic acid (PA), iron (Fe) and also the bioavailability of Zn in grains and (ii) determine the optimised Zn fertilization technology that balances all the above attributes. To achieve this, six Zn fertilization technologies, namely, soil fertilization alone, combined soil and foliar fertilization at maximum tillering, jointing, flowering, dough stages and also foliar fertilization alone were tested and compared with control (no Zn) in forty different soil series representing two distinct soil orders, Inceptisols and Alfisols. Results showed that relative effectiveness of different Zn fertilization technologies varied for the crop attributes studied. Soil + foliar fertilization was superior in increasing grain yield (10-13% over the control). Moreover, for an optimum balance among all the tested attributes including bioavailability of Zn to human, foliar Zn fertilization at later crop growth stage (i.e. dough) combined with soil fertilization was the best. It was found that biofortified wheat grains obtained through Zn fertilization, on an average, could supply about 1.5 times more bioavailable Zn than the normal grains. Therefore, the outcomes of this study can provide a guideline for sustainable and quality wheat production, which will help address the malnutrition challenge.

Improving the concentration and bioavailability of zinc (Zn) in cereal grains is an important way to solve the problem of Zn deficiency in human body. The bioavailability of Zn is related to both its distribution and speciation in grains. In the current study, we examined the differences of Zn concentration, distribution, and speciation within grains among wheat cultivars with similar high grain yield but contrasting grain Zn concentration using synchrotron micro X-ray fluorescence (µ-XRF) and X-ray absorption near-edge structure (XANES). Results showed that compared to the low-Zn cultivar, the Zn concentration was 103, 50, 76, 33, and 64% higher in the crease region, aleurone layer, scutellum, embryonic axis, and endosperm of the high-Zn cultivar, respectively. Zinc mainly colocalized with phosphorus (P) in the aleurone layer and the scutellum, but less colocalization of Zn with P and a much lower concentration ratio of P/Zn were found in the high-Zn cultivar. Sulfur (S) is present in the form of scattered spots in the endosperm in accord with Zn, but the colocalization of Zn with S was predominant in the modified aleurone layer and the nucellar projection of the high-Zn cultivar. XANES results showed the lower proportion of Zn-phytate in the high-Zn cultivar. Findings indicated that it is possible to select the high-yield wheat cultivar with both high grain Zn concentration and high bioavailability, which provide a new perspective for genetic Zn biofortification.

An effective solution to global human zinc (Zn) deficiency is Zn biofortification of staple food crops, which has been hindered by the low available Zn in calcareous soils worldwide. Many culturable soil microbes have been reported to increase Zn availability in the laboratory, while the status of these microbes in fields and whether there are unculturable Zn-mobilizing microbes remain unexplored. Here, we use the culture-independent metagenomic sequencing to investigate the rhizosphere microbiome of three high-Zn (HZn) and three low-Zn (LZn) wheat cultivars in a field experiment with calcareous soils. The average grain Zn concentration of HZn was higher than the Zn biofortification target 40 mg kg-1, while that of LZn was lower than 40 mg kg-1. Metagenomic sequencing and analysis showed large microbiome difference between wheat rhizosphere and bulk soil but small difference between HZn and LZn. Most of the rhizosphere-enriched microbes in HZn and LZn were in common, including many of the previously reported soil Zn-mobilizing microbes. Notably, 30 of the 32 rhizosphere-enriched species exhibiting different abundances between HZn and LZn possess the functional genes involved in soil Zn mobilization, especially the synthesis and exudation of organic acids and siderophores. Most of the abundant potential Zn-mobilizing species were positively correlated with grain Zn concentration and formed a module with strong interspecies relations in the co-occurrence network of abundant rhizosphere-enriched microbes. The potential Zn-mobilizing species, especially Massilia and Pseudomonas, may contribute to the cultivars' variation in grain Zn concentration, and they deserve further investigation in future studies on Zn biofortification.

Bran is the main by-product of wheat milling and the part of the grain with the highest Zn content. We investigated the effects of the particle sizes (coarse, D50 = 375.4 ± 12.3 µm; medium, D50 = 122.3 ± 7.1 µm; and fine, D50 = 60.5 ± 4.2 µm) and addition level (5-20%) of Zn-biofortified bran on the quality of flour and Chinese steamed bread. It was studied to determine if the Zn content of steamed bread could be enhanced without deleterious effects on quality. Dough pasting properties, such as peak viscosity, trough viscosity, final viscosity, breakdown, and setback, decreased significantly as the bran addition level was increased from 5 to 20% but did not significantly differ as a result of different bran particle sizes. Bran incorporation significantly increased hardness, gumminess, chewiness, and adhesiveness, whereas the springiness, cohesiveness, and specific volume of steamed bread decreased with the increase in bran addition. The optimal sensory score of steamed bread samples in the control and Zn fertilizer groups were obtained under 5% bran addition resulting in comparable flavor, and texture relative to control. Meanwhile, the Zn content of the steamed bread in the Zn fertilizer group was 40.2 mg/kg, which was 55.8% higher than that in the control group. Results indicated that adding the appropriate particle size and amount of bran would be an effective and practical way to solve the problem of the insufficient Zn content of steamed bread.

Zn is an essential micronutrient for living organisms and, in that capacity, it is added to animal feed in intensive livestock production to promote growth and eliminate diseases. Alfalfa (Medicago sativa L.) may have the potential to compensate and substitute the need for chemical Zn additives in feeds as a Zn-biofortified feed crop when grown on Zn-enriched soils. Thus, this possibility was investigated with a greenhouse experiment using three soils with Zn concentrations (mg kg-1) of 189 (soil A), 265 (soil B) and 1496 (soil C). Ethylenediamine-N,N'-disuccinate acid (EDDS) and Nitrilotriacetic acid (NTA) at different rates (0 as control, 0.5, 2 and 5 mmol kg-1) were applied as soil additives to enhance the phytoextraction efficiency of alfalfa. The results showed that Zn was highly transferable in alfalfa tissues in the three soils even without additives. EDDS was more effective than NTA in enhancing Zn phytoextraction by alfalfa. The maximum Zn accumulation in the third cutting shoots was obtained with the EDDS concentration of 5 mmol kg-1 in soil A and of 2 mmol kg-1 in soil B, with a 462% and 162% increase compared with controls, respectively. However, the higher EDDS concentration resulted in a significant reduction in biomass production. In soil C, all EDDS concentrations resulted in similar Zn accumulations in the third shoot. To improve the phytoextraction efficacy of Zn while minimizing its phytotoxicity on alfalfa, the rate of 2 mmol kg-1 EDDS proved to be optimal for soil B, and 0.5 mmol kg-1 EDDS for soils A and C. Findings suggest that phytoextraction of Zn-enriched soil can be combined with Zn biofortification, thus allowing to recycle Zn into biomass that can, to an extent, substitute Zn feed additives. This study provided a primary data set for the combination of Zn-biofortification and Zn-phytoextraction.

A wide range of root-associated mutualistic microorganisms have been successfully applied and documented in the past for growth promotion, biofertilization, biofortification and biotic and abiotic stress amelioration in major crops. These microorganisms include nitrogen fixers, nutrient mobilizers, bio-remediators and bio-control agents. The present study aimed to demonstrate the impact of salt-tolerant compatible microbial inoculants on plant growth; Zn biofortification and yield of wheat (Triticum aestivum L.) crops grown in saline-sodic soil and insight of the mechanisms involved therein are being shared through this paper. Field experiments were conducted to evaluate the effects of Trichoderma harzianum UBSTH-501 and Bacillus amyloliquefaciens B-16 on wheat grown in saline-sodic soil at Research Farm, ICAR-Indian Institute of Seed Sciences, Kushmaur, India. The population of rhizosphere-associated microorganisms changed dramatically upon inoculation of the test microbes in the wheat rhizosphere. The co-inoculation induced a significant accumulation of proline and total soluble sugar in wheat at 30, 60, 90 and 120 days after sowing as compared to the uninoculated control. Upon quantitative estimation of organic solutes and antioxidant enzymes, these were found to have increased significantly in co-inoculated plants under salt-stressed conditions. The application of microbial inoculants enhanced the salt tolerance level significantly in wheat plants grown in saline-sodic soil. A significant increase in the uptake and translocation of potassium (K+) and calcium (Ca2+) was observed in wheat co-inoculated with the microbial inoculants, while a significant reduction in sodium (Na+) content was recorded in plants treated with both the bio-agents when compared with the respective uninoculated control plants. Results clearly indicated that significantly higher expression of TaHKT-1 and TaNHX1 in the roots enhances salt tolerance effectively by maintaining the Na+/K+ balance in the plant tissue. It was also observed that co-inoculation of the test inoculants increased the expression of ZIP transporters (2-3.5-folds) which ultimately led to increased biofortification of Zn in wheat grown in saline-sodic soil. Results suggested that co-inoculation of T. harzianum UBSTH-501 and B. amyloliquefaciens B-16 not only increased plant growth but also improved total grain yield along with a reduction in seedling mortality in the early stages of crop growth. In general, the present investigation demonstrated the feasibility of using salt-tolerant rhizosphere microbes for plant growth promotion and provides insights into plant-microbe interactions to ameliorate salt stress and increase Zn bio-fortification in wheat.

Soybean is recognized as one of the most important prospective protein sources for human nutrition under conditions of climate change and population growth. Occurrence of Se and Zn deficiency in vast areas over the globe inhabited by up to 2 billion people, induced search for a comprehensive solution to these problems through the efficient Se/Zn biofortification of soybean seeds (beans). To assess the Se/Zn accumulation efficiency and the physiological status of soybean plants, a pot experiment on Se and Zn enrichment in beans was conducted. It consisted of applying 15 different Se-deficient soil treatments with these elements during the flowering phase, alone or in dose combinations. Application of Se alone, besides Se accumulation in soybean, reduced Zn uptake from soil, but caused alterations in Zn translocation, and its multiple enrichment in beans. Addition of Zn alone promoted both Zn and Se enrichment in beans. Joint Se/Zn application in increasing doses appeared to have a strong synergistic effect on accumulation of these elements in beans and enhanced the physiological functions of the soybean. This manifested itself in the growth of photosynthetic production and soybean biomass, and in the improvement of lipid peroxidation status (REC, MDA and proline content indices). Toxicity symptoms indicated the maximum Se/Zn doses. Several-fold higher contents of Se and Zn in soybean straw compared to spiked soil suggest its possible use as Se/Zn-rich soil amendment in accordance with the circular economy goals. These novel findings may significantly contribute to human health improvement in Se and Zn deficient regions.

Zinc (Zn) deficiency is an important problem worldwide, adversely impacting human health. Using a field trial in China, we compared the foliar application of both ZnO nanoparticles (ZnO-NPs) and ZnSO4 on winter wheat (Triticum aestivum L.) for increasing the Zn concentration within the grain. We also used synchrotron-based X-ray fluorescence microscopy and laser ablation inductively coupled plasma mass spectrometry to examine the distribution of Zn within the grain. We found that ZnO-NPs increase the Zn concentration in the wheat grain, increasing from 18 mg·kg-1 in the control up to 40 mg·kg-1 when the ZnO-NPs were applied four times. These grain Zn concentrations in the ZnO-NP-treated grains are similar to those recommended for human consumption. However, the ZnO-NPs were similar in their effectiveness to ZnSO4. When examining trace element distribution in the grain, the trace elements were found to accumulate primarily in the aleurone layer and the crease region across all treatments. Importantly, Zn concentrations in the grain endosperm increased by nearly 30-fold relative to the control, with markedly increasing Zn concentrations within the edible portion. These results demonstrate that ZnO-NPs are a suitable fertilizer for increasing Zn within wheat grain and can potentially be used to improve human nutrition.

Wheat is a staple food crop and a major source of both the essential micronutrient zinc (Zn) and the toxic heavy metal cadmium (Cd) for humans. Since Zn and Cd are chemically similar, increasing Zn concentrations in wheat grains (biofortification), while preventing Cd accumulation, is an agronomic challenge. We used two Swiss agricultural long-term field trials, the "Dynamic-Organic-Conventional System Comparison Trial" (DOK) and the "Zurich Organic Fertilization Experiment" (ZOFE), to investigate the impact of long-term organic, mineral and combined fertilizer inputs on total and phytoavailable concentrations of soil Zn and Cd and their accumulation in winter wheat (Triticum aestivum L.). "Diffusive gradients in thin films" (DGT) and diethylene-triamine-pentaacetic acid (DTPA) extraction were used as proxies for plant available soil metals. Compared to unfertilized controls, long-term organic fertilization with composted manure or green waste compost led to higher soil organic carbon, cation exchange capacity and pH, while DGT-available Zn and Cd concentrations were reduced. The DGT method was a strong predictor of shoot and grain Cd, but not Zn concentrations. Shoot and grain Zn concentrations correlated with DTPA-extractable and total soil Zn concentrations in the ZOFE, but not the DOK trial. Long-term compost fertilization led to lower accumulation of Cd in wheat grains, but did not affect grain Zn. Therefore, Zn/Cd ratios in the grains increased. High Zn and Cd inputs with organic fertilizers and high Cd inputs with phosphate fertilizers led to positive Zn and Cd mass balances when taking into account atmospheric deposition and fertilizer inputs. On the other hand, mineral fertilization led to the depletion of soil Zn due to higher yields and thus higher Zn exports than under organic management. The study supports the use of organic fertilizers for reducing Cd concentrations of wheat grains in the long-term, given that the quality of the fertilizers is guaranteed.

The amount of Zn absorbed from Zn-biofortified wheat material has been determined using an in vivo model of Zn absorption. The erythrocyte linoleic:dihomo -γ-linolenic acid (LA:DGLA) ratio was used as a biomarker of Zn status. Two groups of chickens (n = 15) were fed different diets: a high-Zn (46.5 µg Zn g-1) and a low-Zn wheat-based diet (32.8 µg Zn g-1). Dietary Zn intakes, body weight, serum Zn, and the erythrocyte fatty acid profile were measured, and tissues were taken for gene expression analysis. Serum Zn concentrations were greater in the high Zn group (p < 0.05). Duodenal mRNA expression of various Zn transporters demonstrated expression upregulation in the birds fed a low Zn diet (n = 15, p < 0.05). The LA:DGLA ratio was higher in the birds fed the low Zn diet (p < 0.05). The higher amount of Zn in the biofortified wheat resulted in a greater Zn uptake.

The effects of foliar-applied ZnO nanoparticles (ZnO NPs) and ZnSO4 on the winter wheat ( Triticum aestivum L.) grain yield and grain quality were studied under field conditions, with the distribution and speciation of Zn within the grain examined using synchrotron-based X-ray fluorescence microscopy and X-ray absorption spectroscopy. Although neither of the two Zn compounds improved the grain yield or quality, both increased the grain Zn concentration (average increments were 5 and 10 mg/kg for ZnSO4 and ZnO NP treatments, respectively). Across all treatments, this Zn was mainly located within the aleurone layer and crease of the grain, although the application of ZnO NPs also slightly increased Zn within the endosperm. This Zn within the grain was found to be present as Zn phosphate, regardless of the form in which Zn was applied. These results indicate that the foliar application of ZnO NPs appears to be a promising approach for Zn biofortification, as required to improve human health.

Zn deficiency is currently listed as a major risk factor for human health. Recently, a complimentary solution to mineral malnutrition termed 'biofortification' has been proposed. The aim of this study was to investigate the possible effects of a Zn-biofortification program on Zn levels, amino acidic profile and the phytochemicals content in an edible leafy vegetable, such as Brassica oleracea cv. Bronco. Our results indicate that supplementation of 80-100µM Zn is optimal for maintaining the normal growth of plants and to promote the major Zn concentration in the edible part of B. oleracea. Any further increase of Zn supply induced an accumulation of total amino acids, and increased the enzymatic activities involved in sulfur assimilation and synthesis of phenols, finally resulting in a foliar accumulation of glucosinolates and phenolic compounds. Thus, it could be proposed that the growth of B. oleracea under 80-100µM Zn may increase the intake of this micronutrient and other beneficial compunds for the human health.

Given that plant uptake and transport systems for metals have some similarities, zinc (Zn)-biofortified cultivars may concurrently accumulate non-essential toxic heavy metals in grains. However, Zn-biofortified cultivars have never been tested for heavy metal accumulation in grains. In a pot experiment, we compared Zn-biofortified wheat (Zincol-2016) with a standard wheat (Faisalabad-2008) cultivar on heavy-metal-contaminated soils for yield response and grain accumulation of Zn, lead (Pb) and cadmium (Cd), without or with Zn fertilisation (8mgZnkg-1). The soils, collected from agricultural fields in (i) industrial zone and (ii) peri-urban area, had been receiving industrial and city effluents for >20years. In the two soils, Zn fertilisation significantly (P≤0.05) increased grain yield of both cultivars. Zinc fertilisation increased grain Zn concentration of Zincol-2016 and Faisalabad-2008 by respectively 32 and 18% in industrial-zone soil, and by 15 and 2% in peri-urban soil. Averaged across Zn rates, Zincol-2016 accumulated in grains more than double the Zn amount than Faisalabad-2008 in industrial-zone soil. At 0mgZnkg-1, grain Pb and Cd concentrations were respectively 26 and 33% greater in Zincol-2016 than Faisalabad-2008 in industrial-zone soil, and 86 and 50% greater in Zincol-2016 than Faisalabad-2008 in peri-urban soil. Zinc fertilisation significantly (P≤0.05) decreased concentration of Pb and Cd in grains of both cultivars. In industrial-zone soil, a toxic level of Pb in grains (0.24mgkg-1) was attained at control rate of Zn by Zincol-2016, and was decreased to a safe level (0.07mgkg-1) by application of 8mgZnkg-1. Therefore, biofortified cultivars should not be grown in contaminated soils, and/or sufficient Zn must be applied, to decrease accumulation of non-essential toxic heavy metals in grains. Moreover, future breeding efforts should be directed toward selection of biofortified cultivars that would selectively accumulate Zn in grains, but not the contaminants.

One of the important factors that influences Zn deficiency tolerance and grain Zn loading in crops is the within-plant allocation of Zn. Three independent experiments were carried out to understand the internal Zn distribution patterns in rice genotypes grown in Zn-sufficient and Zn-deficient agar nutrient solution (ANS). In one of the experiments, two rice genotypes (IR55179 and KP) contrasting in Zn deficiency tolerance were leaf-labeled with (65)Zn. In the other two experiments, two Zn biofortification breeding lines (IR69428 and SWHOO) were either root- or leaf-labeled with (65)Zn. Rice genotype IR55179 showed significantly higher Zn deficiency tolerance than KP at 21 and 42 days after planting. When KP was Zn-deficient, it failed to translocate (65)Zn from the labeled leaf to newly emerging leaves. Similarly, the root-to-shoot translocation of unlabeled Zn was lower in KP than in IR55179. These results suggest that some Zn-efficient rice genotypes have greater ability to translocate Zn from older to actively growing tissues than genotypes sensitive to Zn deficiency. Among the two Zn biofortication breeding lines that were leaf-labeled with (65)Zn at 10 days before panicle initiation stage, (65)Zn distribution in the grains at maturity was similar between both genotypes in Zn-sufficient conditions. However, under Zn-deficient conditions, SWHOO accumulated significantly higher (65)Zn in grains than IR69428, indicating that SWHOO is a better remobilizer than IR69428. When the roots of these two Zn biofortication breeding lines were exposed to (65)Zn solution at 10 days after flowering, IR69428 showed higher root uptake of (65)Zn than SWHOO in Zn-sufficient conditions, but (65)Zn allocation in the aerial parts of the plant was similar between both genotypes.
