Although metacommunity dynamics of lentic phytoplankton are relatively well-documented, studies on the role of environmental and spatial processes in shaping phytoplankton communities of large rivers are still scarce. Here, we examined six phytoplankton data sets, which were collected in 1978-2017 from large river-scale segments (mean spatial extent 1117 km) in the Danube River. Our aim was to elucidate role of climatic, spatial and temporal predictors in variation of phytoplankton beta diversity using variance partitioning for compositions of species and functional groups sensu Reynolds. We hypothesised that phytoplankton beta diversity (measured as average distance to group centroid) would be positively related to both climatic heterogeneity and spatial extent used as a proxy for dispersal limitation. Additionally, we tested alternative dispersal models to evaluate different spatial processes structuring phytoplankton community. Our results revealed that spatial variables were more important than climatic factors in controlling both species and functional group composition. Climatic heterogeneity showed significant positive relationship with beta diversity. In contrast, there was no significant relationship between beta diversity and spatial extent, suggesting that spatial effect on beta-diversity was attenuated by anthropogenic disturbance. The better performance of non-directional model compared to model of water directionality suggested that spatial dynamics of phytoplankton metacommunity was in large part regulated by differences in the regional species pools. Spatial and temporal variables outperformed environmental (including climatic) factors in explaining phytoplankton metacommunity structure, indicating that phytoplankton exhibited strong biogeographical patterns. Thus, dispersal limitation interfered with species-sorting processes in determining phytoplankton community structure. In conclusion, our findings revealed that the development of a more reliable bioassessment program of the Danube River should be based on separation into basin regions.

The chemical ecology of rotifers has been little studied. A yet unknown property is presented within some monogonant rotifers, namely the ability to produce an exogenic filamentous biopolymer, named 'Rotimer'. This rotifer-specific viscoelastic fiber was observed in six different freshwater monogonants (Euchlanis dilatata, Lecane bulla, Lepadella patella, Itura aurita, Colurella adriatica and Trichocerca iernis) in exception of four species. Induction of Rotimer secretion can only be achieved by mechanically irritating rotifer ciliate with administering different types (yeast cell skeleton, denatured BSA, epoxy, Carmine or urea crystals and micro-cellulose) and sizes (approx. from 2.5 to 50 µm diameter) of inert particles, as inductors or visualization by adhering particles. The thickness of this Rotimer is 33 ± 3 nm, detected by scanning electron microscope. This material has two structural formations (fiber or gluelike) in nano dimension. The existence of the novel adherent natural product becomes visible by forming a 'Rotimer-Inductor Conglomerate' (RIC) web structure within a few minutes. The RIC-producing capacity of animals, depends on viability, is significantly modified according to physiological- (depletion), drug- (toxin or stimulator) and environmental (temperature, salt content and pH) effects. The E. dilatata-produced RIC is affected by protein disruptors but is resistant to several chemical influences and its Rotimer component has an overwhelming cell (algae, yeast and human neuroblastoma) motility inhibitory effect, associated with low toxicity. This biopolymer-secretion-capacity is protective of rotifers against human-type beta-amyloid aggregates.

Attachment of microorganisms to natural or artificial surfaces and the development of biofilms are complex processes which can be influenced by several factors. Nevertheless, our knowledge on biofilm formation in karstic environment is quite incomplete. The present study aimed to examine biofilm development for a year under controlled conditions in quasi-stagnant water of a hydrothermal spring cave located in the Buda Thermal Karst System (Hungary). Using a model system, we investigated how the structure of the biofilm is formed from the water and also how the growth rate of biofilm development takes place in this environment. Besides scanning electron microscopy, next-generation DNA sequencing was used to reveal the characteristic taxa and major shifts in the composition of the bacterial communities. Dynamic temporal changes were observed in the structure of bacterial communities. Bacterial richness and diversity increased during the biofilm formation, and 9-12 weeks were needed for the maturation. Increasing EPS production was also observed from the 9-12 weeks. The biofilm was different from the water that filled the cave pool, in terms of the taxonomic composition and metabolic potential of microorganisms. In these karstic environments, the formation of mature biofilm appears to take place relatively quickly, in a few months.

Recent decades have been marked by unprecendented environmental changes which threaten the integrity of freshwater systems and their ecological value. Although most of these changes can be attributed to human activities, disentagling natural and anthropogenic drivers remains a challenge. In this study, surface sediments from Lake Ighiel, a mid-altitude site in the Carpathian Mts (Romania) were investigated following high-resolution sedimentological, geochemical, environmental magnetic and diatom analyses supported by historical cartographic and documentary evidence. Our results suggest that between 1920 and 1960 the study area experienced no significant anthropogenic impact. An excellent correspondence is observed between lake proxy responses (e.g., growth of submerged macrophytes, high detrital input, shifts in diatom assemblages) and parameters tracking natural hydroclimate variability (e.g., temperature, NAO). This highlights a dominant natural hydroclimatic control on the lacustrine system. From 1960 however, the depositional regime shifted markedly from laminated to homogenous clays; since then geochemical and magnetic data document a trend of significant (and on-going) subsurface erosion across the catchment. This is paralleled by a shift in lake ecosystem conditions denoting a strong response to an intensified anthropogenic impact, mainly through forestry. An increase in detrital input and marked changes in the diatom community are observed over the last three decades, alongside accelerated sedimentation rates following enhanced grazing and deforestation in the catchment. Recent shifts in diatom assemblages may also reflect forcing from atmospheric nitrogen (N) deposition, a key recent drive of diatom community turnover in mountain lakes. In general, enhanced human pressure alongside intermittent hydroclimate forcing drastically altered the landscape around Lake Ighiel and thus, the sedimentation regime and the ecosystem's health. However, paleoenvironmental signals tracking natural hydroclimate variability are also clearly discernible in the proxy data. Our work illustrates the complex link between the drivers of catchment-scale impacts on one hand, and lake proxy responses on the other, highlighting the importance of an integrated historical and palaeolimnological approach to better assess lake system changes.

Reduced body size is among the universal ecological responses to global warming. Our knowledge on how altered body size affects ecosystem functioning in ectothermic aquatic organisms is still limited. We analysed trends in the cell size structure of phytoplankton in the middle Danube River over a 34-year period at multiple levels: (1) average cell size of assemblages (ACS), (2) within the centric diatom community and (3) in the dominant centric diatom taxon: Stephanodiscus. We asked whether global warming and human impacts affected the average cell size of phytoplankton. Also, whether the altered size structure affected how chlorophyll-a, as an ecosystem functioning measure, relates to the ACS of phytoplankton. The cell size of phytoplankton decreased significantly at all organisation levels, and the assemblages became more dispersed in cell size over time. Environmental variables related to global warming and human impacts affected the ACS of phytoplankton significantly. The relationship between chlorophyll-a and the ACS of phytoplankton shifted from negative linear to broad and then narrow hump shape over time. Longer water residence time, warming and decline in nutrients and suspended solids decrease the ACS of phytoplankton in the middle Danube and expectedly in other large rivers. Our results suggest that cell size decrease in phytoplankton, especially of centric diatoms, constrains planktic algal biomass production in large rivers, independently of algal density. Such cell size decrease may also affect higher trophic levels and enhance the more frequent occurrence of "clear-water" plankton in large, human-impacted rivers under global change.

Lake snow, caused by the freshwater centric diatom Lindavia intermedia, has become problematic in several large, oligotrophic New Zealand lakes over the past decade. Macroaggregates produced by L. intermedia foul fishing lines, intake screens, and water filters, and have a negative impact on recreational values. It was confirmed that the fibers constituting lake snow are composed of chitin, two chitin synthase genes (chs1 and 2) from L. intermedia were characterized, new qPCR-based tools to quantify the abundance of the species and measure expression of chs2 relative to the reference gene act1 (the product of which has cytoskeletal functions) were developed. The strong heterogeneity and mucilaginous nature of lake snow samples create particular difficulties for calibrations of gene or transcript copy numbers with cell densities and obtaining high yields of mRNA. However, data collected from four lakes during November 2018 and February and May 2019 show that abundance of L. intermedia is always high when lake snow is also abundant, but that a full range of L. intermedia abundance can occur when lake snow is absent, suggesting that chitin production is not obligate in L. intermedia. This result is consistent with the available data for chs2 expression, which suggest higher transcription when lake snow is abundant. Lake snow production by L. intermedia therefore requires an as yet undetermined stimulus independent of cell abundance.

The stress dominance hypothesis (SDH) postulates that strong environmental gradients drive trait convergence in communities over limiting similarity. Previous studies, conducted mostly with terrestrial plant communities, found controversial evidence for this prediction. We provide here the first test for SDH for epiphytic diatoms. We studied community assembly in diatom communities of astatic ponds. These water bodies serve as a good model system for testing SDH because they exhibit stress gradients of various environmental factors. Functional diversity of diatom communities was assessed based on four traits: (1) combined trait reflecting the trade-off between stress tolerance and competitive dominance, (2) cell size, (3) oxygen requirement and (4) N-uptake strategy. According to our results, salinity, pH and the width of the macrophyte belt appeared as significant predictors of the trait convergence/divergence patterns presumably acting through influencing the availability of carbon dioxide and turbidity. Lower trait diversity was found in turbid, more saline and more alkaline ponds and functional diversity was higher in transparent, less saline and less alkaline ponds. Overall, our results supported the stress dominance hypothesis. In habitats representing increased environmental stress, environmental filtering was the most important community assembly rule, while limiting similarity became dominant under more favourable conditions.

The question of whether one large, continuous area or many smaller habitats maintain more species is one of the most relevant questions in conservation ecology, and it is referred to as the SLOSS (Single Large Or Several Small) dilemma in the literature. This question has not yet been raised in the case of microscopic organisms, therefore we investigated whether or not the SLOSS dilemma could apply to phytoplankton and benthic diatom metacommunities. Benthic diatom and phytoplankton diversity in pools and ponds of different sizes (ranging between 10-2-107 m2) was studied. Species richness of water bodies belonging to neighbouring size categories was compared step by step across the whole size gradient. With the exception of the 104-105 m2 and 105-106 m2 size categories, where phytoplankton and benthic diatom richness values of the SL water bodies were higher than that of the SS ones, findings showed that the diversity of several smaller (SS) sized waters was higher than that in single large water bodies (SL) throughout the whole studied size range. The proportion of the various functional groups of algae, including both the benthic diatoms and phytoplankton, showed remarkable changes from the smaller water bodies to large sized ones.

The spatial response of epiphytic diatom communities to environmental stress was studied in a moderately saline wetland area located in the plain of Danube-Tisza Interfluve, Hungary. The area is characterised by World War II bomb crater ponds and can be regarded as an excellent ecological model system where the dispersion of species is slightly limited by distance. To study the effect of environmental variables on the communities, canonical correspondence analysis was applied. Salinity, pH, total suspended solids, total phosphorous and depth proved to be significant environmental drivers in this analysis. The ecological status of the ponds was assessed with Ziemann's halobity index, as the trophity-depending metric cannot be applied to these habitats (due to the naturally high phosphorus content). Ponds in "good" ecological status significantly differed from those appertaining to water quality category of "not-good" ecological status considering characteristic of natural astatic soda pans (e.g. salinity, pH, ammonium, total phosphorous concentration, nitrogen:phosphorous ratio and turbidity). The differences between epiphytic diatom communities inhabiting the ponds were detected using non-parametric multidimensional scaling. The samples formed three groups according to the types of ponds ("transparent", "transitional" and "turbid") based on the width of the macrophyte belt around them. Indicator species related to the ecological status of the ponds and diatom communities contributing to the separation of groups of ponds were identified. One of the indicator species differed from species already described. Light and scanning electron microscopy features and phylogenetic analyses based on three genes (18S and 28S rRNA genes, rbcL) proved that it was a new species of Nitzschia genus, closely related to Nitzschia frustulum and Nitzschia inconspicua. Therefore, description of a new species, Nitzschia reskoi Ács, Duleba, C.E.Wetzel & Ector is proposed. We concluded that the increasing abundance of Nitzschia reskoi was a signal of the degradation of the intermittent saline wetlands.

Neurodegenerative diseases are linked to a systemic enzyme resistance of toxic aggregated molecules and their pathological consequences. This paper presents a unique phenomenon that Philodina acuticornis, a bdelloid rotifer, is able to catabolize different types of neurotoxic peptide and protein aggregates (such as beta-amyloids /Aß/, alpha-synuclein, and prion) without suffering any damage. P. acuticornis is capable of using these aggregates as an exclusive energy source (i.e., as 'food', identified in the digestive system and body) in a hermetically isolated microdrop environment, increasing their survival. As regards Aß1-42, five other bdelloid rotifer species were also found to be able to perform this phenomenon. Based on our experiments, the Aß1-42-treated bdelloid rotifers demonstrate significantly increased survival (e.g. mean lifespan = 51 ± 2.71 days) compared to their untreated controls (e.g. mean lifespan = 14 ± 2.29 days), with similar improvements in a variety of phenotypic characteristics. To our knowledge, no other animal species have so far been reported to have a similar capability. For all other microscopic species tested, including monogonant rotifers and non-rotifers, the treatment with Aß1-42 aggregates proved to be either toxic or simply ineffective. This paper describes and proves the existence of an unprecedented in vivo catabolic capability of neurotoxic aggregates by bdelloid rotifers, with special focus on P. acuticornis. Our results may provide the basis for a new preclinical perspective on therapeutic research in human neurodegenerative diseases.

Considering the ongoing loss of aquatic habitats, anthropogenic ponds are gaining importance as substitute habitats. It is therefore important to assess their functioning in comparison to their natural precursors. Here we assess the biodiversity value of sodic bomb crater ponds by comparing their gamma diversity to that of natural reference habitats, astatic soda pans, and assess their importance on the landscape level by studying alpha and beta diversity. We studied aquatic organisms ranging from algae to vertebrates in a dense cluster of 54 sodic bomb crater ponds in Central Europe. Despite the overall small area of the pond cluster, gamma diversity was comparable to that found in surveys of natural habitats that encompassed much wider spatial and temporal scales. We also found a considerable number of species shared with reference habitats, indicating that these anthropogenic habitats function as important refuge sites for several species that are associated with the endangered soda pans. Moreover, we found a number of regionally or worldwide rare species. Among the components of beta diversity, species replacement dominated community assembly. Individual ponds contributed similarly to beta diversity in terms of replacement, being equally important for maintaining high gamma diversity and emphasising the role of the pond network rather than individual ponds. This pattern was seen in all studied groups. Bomb crater ponds therefore acted as important contributors to aquatic biodiversity. Considering the tremendous losses of ponds throughout Europe, anthropogenic ponds should be taken into consideration in nature conservation, especially when occurring in pond networks.

Skeletonema potamos is a poorly known freshwater species in the ancestrally and predominantly marine genus Skeletonema. With phylogenetic analysis of two nuclear {partial SSU (18S) and partial LSU (28S) rDNA)} and two chloroplast (rbcL and psbC) genes, we verified its placement within the genus Skeletonema and identified the mostly brackish species, Skeletonema subsalsum, as its closest known relative. Comparisons of SSU and LSU rRNA genes from S. potamos populations from Europe and North America revealed no intraspecific variation. Skeletonema potamos can be a dominant element of the phytoplankton community in various ecosystems, including the River Danube. We tracked phytoplankton composition in the River Danube weekly from 1979 to 2 012, and throughout this period, S. potamos exhibited a strong increase in proportion of total phytoplankton abundance and biomass - an increase that was correlated with increasing water temperature over the same time period. Current records indicate a temperate distribution of S. potamos, but ecological data predict possible expansion of its geographic range and increase in seasonal duration within existing habitats in response to the warming of surface waters.

Seasonal dynamics of all major protozoan groups were investigated in the plankton of the River Danube, upstream of Budapest (Hungary), by bi-weekly sampling over a 1-year long period. Sixty-one heterotrophic flagellate, 14 naked amoeba, 50 testate amoeba, 4 heliozoan and 83 ciliate morphospecies were identified. The estimated abundance ranges of major groups throughout the year were as follows: heterotrophic flagellates, 0.27-7.8 x 10(6)ind.l(-1); naked amoebae, max. 3300ind.l(-1); testaceans, max. 1600ind.l(-1); heliozoans, max. 8500ind.l(-1); ciliates, 132-34,000ind.l(-1). In terms of biovolume, heterotrophic flagellates dominated throughout the year (max. 0.58mm(3)l(-1)), and ciliates only exceeded their biovolume in summer (max. 0.76mm(3)l(-1)). Naked amoeba and heliozoan biovolume was about one, and testacean biovolume 1-3, orders of magnitude lower than that of ciliates. In winter, flagellates, mainly chrysomonads, had the highest biomass, whilst ciliates were dominated by peritrichs. In 2005 from April to July a long spring/summer peak occurred for all protozoan groups. Beside chrysomonads typical flagellates were choanoflagellates, bicosoecids and abundant microflagellates (large chrysomonads and Collodictyon). Most abundant ciliates were oligotrichs, while Phascolodon, Urotricha, Vorticella, haptorids, Suctoria, Climacostomum and Stokesia also contributed significantly to biovolume during rapid succession processes. In October and November a second high protozoan peak occurred, with flagellate dominance, and slightly different taxonomic composition.

Cyclotella meneghiniana Kützing is one of the most commonly found and intensively studied freshwater diatom species. However, it is considered taxonomically problematic because of its unusually wide ecological range and large frustule ultrastructural variation. As part of a study of morphological and genetic variation in this morphospecies, we surveyed nucleotide variation in the hypervariable D1/D2 regions of the 28S rDNA, in the ribosomal internal transcribed spacer region (containing ITS1, the 5.8S rDNA and ITS2) and in the 18S rDNA in a collection of 20 sympatric strains. High genetic variability and strong indications of genetic structure among the Cyclotella meneghiniana strains were found. Representatives of four genetically distinct--apparently reproductively isolated--groups were revealed among them. The random distribution of ITS variation within these four groups indicated that the genetic structure in Cyclotella meneghiniana can probably be explained by the presence of cryptic sexual species rather than by the lack of allogamous sexual reproduction. The morphological features traditionally used for species identification in this group cannot distinguish these putative cryptic species.
