RESUMO

The excessive and uncontrolled application of antibiotics in the fish farming industry, coupled with a lack of health monitoring and medication practices, is a driving force behind the escalating development of antimicrobial resistance. The present study assessed and compared qualitative field diffusion (QFD) and disk diffusion (DD) assays for the detection of antimicrobial residues (ARs) in diverse freshwater aquaculture fish. A total of 380 freshwater aquaculture fish (160 fresh and 180 frozen) samples were systematically collected between January and June 2021 from various retail stores located in Erbil Governorate, Iraq. Based on QFDA results, overall, ARs were detected (52; 15.3%) at a relatively lower frequency with comparatively higher frequency (21; 31.1%) in fresh than (31; 17.2%) frozen fish samples. On the other hand, DDA also revealed a comparable (45; 13.2%) prevalence rate of ARs. However, a low detection was observed more in fresh (17; 10.6%) than frozen (28; 15.6%) fish samples. Moreover, no statistically significant disparity (χ2 = 0.069; p = 0.79) between two assays and types of fish was recorded. In conclusion, the results of the present study showed that detecting a considerable frequency of ARs in these fish samples raises concerns about potential threats to public health. This underscores the necessity for understanding antibiotic application in aquaculture and its potential connection to antibiotic resistance in bacterial pathogens. Such comprehension is pivotal for formulating and implementing effective control and farm management strategies to address this pressing issue.

Vibrio parahaemolyticus is a zoonotic disease transmitted to humans when handling or consuming improperly cooked fish meat. This study aimed to evaluate the effect of thermal treatment on V. parahaemolyticus isolates. Different heat treatment methods are used to determine the best methods for controlling V. parahaemolyticus, isolated from fish meat, which include microwave, low-temperature long-time, and high-temperature short-time methods. The V. parahaemolyticus isolates significantly declined in bacteria count when they were kept at 4°C, and 25°C for a long time, and the V. parahaemolyticus isolates significantly declined in bacteria count manner when they were kept at -20°C for a long time. The high temperature and long-time exposure at 75°C/25 minutes by moist heat, 87°C/5 minutes by dry heat, and 70°C/20 minutes by frying heat were enough to kill V. parahaemolyticus isolates. This work can be useful to decrease the hazards of infections related to V. parahaemolyticus and reduce the causes of fishborne pathogens.

Helicobacter pylori is an important and common bacterial pathogen in humans. The accumulated evidence of the existence of H. pylori in water from different environmental sources suggests a water-borne transmission route. This study aimed to investigate the occurrence of H. pylori in different water sources used by human populations in Dhamar Governorate, Yemen. 250 samples were randomly collected from the municipal water supply network, wells, and springs. The samples were processed, plated onto modified campy-blood agar, and incubated under microaerobic conditions for 4-10 days. Bacterial identification was based on morphological properties and biochemical tests. Bacteriological analysis showed that 9.6% and 13.2% of tap and surface water samples were contaminated with H. pylori, respectively. Despite a higher frequency in samples from rural areas, these were not significantly (p=0.068) more contaminated than the samples from urban areas. Regarding the seasonal variations of H. pylori detection, 85.71% of positive samples were detected in the late winter and spring seasons (February to May). To conclude, H. pylori transmission through water is likely to occur in Dhamar Governorate. Further prospective studies are highly recommended to provide further evidence and a clearer picture of H. pylori transmission.

Although Helicobacter pylori (H. pylori) is one of the most common bacterial pathogens of human, its natural reservoirs are still unclear. There is an increasing number of reports that document the occurrence of H. pylori in various foods. This study aimed at isolation of H. pylori from chicken meat sampled. Two hundred and sixty samples were collected randomly from slaughterhouses and markets in Dhamar Governorate, Yemen. Samples were enriched in Brain-Heart Infusion broth in microaerophilic conditions before inoculating the Camp-Blood agar and EYE agar plates. Results showed that 13.8% of samples were contaminated evidenced by H. pylori growth via traditional culture method on agar media. No significant differences between sample types (thighs and breast muscles) (p=0.353) or the sampling source (p=0.816) were observed. Autumn season was associated with increased occurrence of H. pylori. The source of H. pylori in food is still not identified. Proper cooking and good sanitation practices are highly recommended to avoid the infection. Further studies addressing the potential sources of H. pylori are highly suggested.

INTRODUCTION: Human brucellosis is one of the most common zoonosis infections, with an important impact on the health and economy worldwide. This study aimed to update and provide epidemiological information on this infection and evaluate Rose Bengal Test, which is used as an essential diagnostic test for brucellosis in Erbil. METHODOLOGY: A total of 325 participants seeking care and reporting fever at Rizgary Teaching Hospital were enrolled. Blood samples were tested for Brucella spp. antibodies using Rose Bengal Test and blood culture followed by species identification. A questionnaire was administered to detect the risk factors. RESULTS: The prevalence of probable and confirmed brucellosis was 12.3% (95% CI 9.2-16.3) and 9.5% (95% CI 6.8-13.2) respectively. The majority of cases were in the age group of 18-39 years. Brucellosis was significantly associated with raw milk consumption (OR = 10.3 95% CI 5-22.4) and contact with livestock (OR = 11.5 95% CI 5.6-23.9). Brucella melitensis (58.1%) and Brucella abortus (41.9%) are the dominant species in the area. The sensitivity, specificity, positive predictive value, and negative predictive value of the Rose Bengal Test in comparison to the blood culture were 100%, 96.9%, 77.5 %, and 100% respectively. CONCLUSIONS: Brucellosis is a significant cause of fever in Erbil and could be diagnosed by the Rose Bengal Test taking into account the compatibility of clinical features with the positive result. The vaccination of livestock and boiling or pasteurization of milk are essential procedures to reduce the frequency of human brucellosis.

INTRODUCTION: Besides stomach, the oral cavity is the second reservoir of Helicobacter pylori (H. pylori) that plays an important role in oral diseases and recurrent gastric infection. This study aimed to determine the risk factors of oral H. pylori infection for better human health. METHODOLOGY: Saliva samples from 280 subjects who visited the dental clinics were collected for the H. pylori antigen test. The data regarding age, gender, residence, frequency of tooth brushing, presence of dental caries and/or periodontitis were reported for each participant. Stool antigen Helicobacter pylori test was used to detect gastric infection. RESULTS: The overall prevalence of H. pylori in oral cavity and stool were 40.4%; 95% CI [34.8-46.2], and 36.4%; 95% CI [31-42.2], respectively and were not statistically significant (p = 0.546). The differences of positive rates of H. pylori infection according to the presence of periodontitis (54.7% vs 30.1%, p < 0.001, OR; 95% CI: 2.8 (1.7-4.6)) and dental caries (47.1% vs 32%, p = 0.001, OR; 95% CI: 2.2 (1.3-3.5)) were statistically significant. Living in rural areas increases the risk of acquiring H. pylori infection compared with urban areas (46.9% vs 34.7%, p < 0.001, OR; 95% CI: 0.6 (0.4-1)). CONCLUSIONS: The H. pylori antigen test on saliva could be used as evidence of gastric infection. Oral diseases including periodontal diseases and caries are important risk factors for H. pylori colonization, so the professional treatment of these diseases may reduce the rate of new and recurrent gastric infection by H. pylori.

Although antibiotics are valuable drugs for treatment of certain infections, their presence in foodstuff derived from animals is a potential public health hazard. They pose a serious threat as they are implicated in direct toxicity; allergic reactions; disturbance of the normal gut microbiota, carcinogenesis, and emergence of antibioticresistant bacteria. This study investigated the occurrence of antibiotic residues in raw milk samples derived from cows and buffaloes. Samples were collected randomly from different retail outlets in Erbil city (Iraq) from January 1st to June 30th, 2019. The residues were detected by two diffusion assays against Bacillus subtilis bacteria on agar plates. The total occurrence of residues ranged from 11.9% to 13.4% of screened milk samples. No significant differences were found between milk type or location of animal rearing (urban or suburban). Regarding the seasonal variations, spring was found to be associated with gradual decrease in antibiotic residues levels in milk. Such occurrence rate of residues is alarming and require authorities to observe the quality of raw milk introduced to markets for consumption. Further evaluation of antibiotic stability period in raw milk is also necessary.

This study aimed to determine the prevalence and antimicrobial sensitivity of Campylobacter species in raw milk sold at retail vending in Erbil city. Three hundred and fifty (350) samples were aseptically collected from retail raw milk shops between January and June 2019. For isolation of Campylobacter spp., samples were cultured on selective media and tested for biotype and antimicrobials susceptibility by disk diffusion assay. The overall prevalence of Campylobacter spp. was 12.6%. Campylobacter jejuni was significantly prevalent (65.9%) among other Campylobacter species. Antimicrobial susceptibility testing showed complete sensitivity to tetracycline, rifampicin, and neomycin. On the other hand, total resistance to ampicillin and trimethoprim was observed. Strikingly, as low as 56.8% and 72.7% of isolates are still sensitive to the drugs of choice in campylobacteriosis treatment; ciprofloxacin and erythromycin respectively. This resistance pattern of Campylobacter found in this study is critically alarming owing to the insusceptibility to the aforementioned antibiotics commonly used as the drugs of choice for campylobacteriosis treatment. Increase in Campylobacter prevalence in raw milk was associated with warm season. These levels prevalence and resistance worth further investigations and effective countermeasures owing to potential public health hazards.

Heavy metal contamination of poultry meat is a critical issue for human health due to associated risks of cytotoxicity and systemic pathologies after ingestion of such metals. A total of twenty chicken liver samples were collected from markets of Erbil city and analyzed for ten heavy metals contents by Inductively Coupled Plasma Optical Emission Spectrometry. The targeted metals were cadmium (Cd), cobalt (Co), chromium (Cr), copper (Cu), manganese (Mn), nickel (Ni), lead (Pb), mercury (Hg), zinc (Zn) and selenium (Se). The average concentrations (mg/kg) of targeted trace elements were 0.06±0.027, 0.06±0.05, 2.05±0.34, 1.85±0.47, 0.15±0.17, and 33.53±5.24 for Co, Cr, Cu, Mn, Ni, and Zn respectively. Copper (Cu) levels significantly exceeded the maximum permissible limit of WHO. Moreover, the average concentrations of toxic heavy metals and selenium were 0.07±0.037, 0.278±0.10, 0.11±0.083, and 2.01±0.454 mg/kg for Cd, Pb, Hg, and Se respectively. Hg and Pb levels exceeded the permissible limits of WHO. Higher levels of Cu and Hg in poultry may pose a serious threat to consumers which demand countermeasures and precautions to be taken. Iraqi Standards Authority and relevant official institutions are strongly recommended to regulate safe disposal of heavy metal waste in the environment to reduce animal exposure to such metals.

The present study was conducted to evaluate the quality of drinking water in randomly selected schools in Erbil city, Kurdistan Region, Iraq. The water quality indices such as the Heavy metal Pollution Index (HPI) and Heavy metal Evaluation Index (HEI) were applied to characterize water quality. Eighteen schools were incorporated and sampled for their water storage tanks available to students. Water samples and sediment samples from tanks floor were analyzed by Inductively Coupled Plasma Optical Emission Spectrometer for the determination of twenty-two metal elements. In drinking water samples, all detected metals did not exceed the permissible limits of the World Health Organization. The results of this study showed that the average values of HPI and HEI for As, Cd, Cr, Cu, Fe, Pb, Mn, Ni, and Zn were 54.442 and 0.221, respectively. According to data of the water quality indices, the schools drinking water quality are good and suitable for drinking in terms of heavy metals. However, sediments samples contained high concentrations of all elements including the toxic heavy metals (As, Cd, Cr, and Pb). Re-suspension of sediments into water column after refilling storage tanks can pose a serious threat to students drinking water from such vessels. It is therefore recommended that proper storage tanks are provided to the schools accompanied by continuous sanitation and hygiene practice to mitigate the corrosion of tanks to avoid health risks of toxic metals.
