Psilocybe mushrooms, otherwise known as "magic" mushrooms, owe their psychedelic effect to psilocin, a serotonin subtype 2A (5-HT2A) receptor agonist and metabolite of psilocybin, the primary indole alkaloid found in Psilocybe species. Metabolomics is an advanced fingerprinting tool that can be utilized to identify the differences among fungal life stages that may otherwise be unaccounted for. In this study, by using targeted and untargeted (metabolomic) multivariate analysis, we demonstrate that the chemical composition of Psilocybe differs among mycelia, grain mycelia, and fruiting bodies. The preferential accumulation of psilocybin, baeocystin, tryptophan, ergothioneine, and phenylethylamine in fruiting bodies differentiated them from mycelia; however, the levels of alpha-glycerylphosphorylcholine (α-GPC), N-acetylglucosamine, and trimethylglycine were found to be proportionally higher in mycelia than in fruiting bodies based on Pareto-scaled data. Considering the wealth of compounds with therapeutic potential that have been isolated from various fungal genera, it would be pertinent to study the compounds found in Psilocybe mycelia as potential naturally derived therapeutic targets.

Organic phototransistors can enable many important applications such as nonvolatile memory, artificial synapses, and photodetectors in next-generation optical communication and wearable electronics. However, it is still a challenge to achieve a big memory window (threshold voltage response ∆Vth ) for phototransistors. Here, a nanographene-based heterojunction phototransistor memory with large ∆Vth responses is reported. Exposure to low intensity light (25.7 µW cm-2 ) for 1 s yields a memory window of 35 V, and the threshold voltage shift is found to be larger than 140 V under continuous light illumination. The device exhibits both good photosensitivity (3.6 × 105 ) and memory properties including long retention time (>1.5 × 105  s), large hysteresis (45.35 V), and high endurance for voltage-erasing and light-programming. These findings demonstrate the high application potential of nanographenes in the field of optoelectronics. In addition, the working principle of these hybrid nanographene-organic structured heterojunction phototransistor memory devices is described which provides new insight into the design of high-performance organic phototransistor devices.

PROBLEM: The SARS-CoV-2 (COVID-19) pandemic presented numerous challenges to inpatient care, including overtaxed inpatient medicine services, surges in patient censuses, disrupted patient care and educational activities for trainees, underused providers in certain specialties, and personal protective equipment shortages and new requirements for physical distancing. In March 2020, as the COVID-19 surge began, an interdisciplinary group of administrators, providers, and trainees at Brigham and Women's Hospital created an inpatient virtual staffing model called the Virtual Team Rounding Program (VTRP). APPROACH: The conceptual framework guiding VTRP development was rapid-cycle innovation. The VTRP was designed iteratively using feedback from residents, physician assistants, attendings, and administrators from March to June 2020. The VTRP trained and deployed a diverse set of providers across specialties as "virtual rounders" to support inpatient teams by joining and participating in rounds via videoconference and completing documentation tasks during and after rounds. The program was rapidly scaled up from March to June 2020. OUTCOMES: In a survey of inpatient providers at the end of the pilot phase, 10/10 (100%) respondents reported they were getting either "a lot" or "a little" benefit from the VTRP and did not find the addition of the virtual rounder burdensome. During the scaling phase, the program grew to support 24 teams. In a survey at the end of the contraction phase, 117/187 (62.6%) inpatient providers who worked with a virtual rounder felt the rounder saved them time. VTRP leadership collaboratively and iteratively developed best practices for challenges encountered during implementation. NEXT STEPS: Virtual rounding provides a valuable extension of inpatient teams to manage COVID-19 surges. Future work will quantitatively and qualitatively assess the impact of the VTRP on inpatient provider satisfaction and well-being, virtual rounders' experiences, and patient care outcomes.

A fundamental question in developmental biology is how morphogens, such as bone morphogenetic protein (BMP), form precise signaling gradients to impart positional and functional identity to the cells of the early embryo. We combine rigorous mutant analyses with quantitative immunofluorescence to determine that the proteases Bmp1a and Tolloid spatially restrict the BMP antagonist Chordin in dorsoventral (DV) axial patterning of the early zebrafish gastrula. We show that maternally deposited Bmp1a plays an unexpected and non-redundant role in establishing the BMP signaling gradient, while the Bmp1a/Tolloid antagonist Sizzled is surprisingly dispensable. Combining computational modeling and in vivo analyses with an immobile Chordin construct, we demonstrate that long-range Chordin diffusion is not necessary for BMP gradient formation and DV patterning. Our data do not support a counter-gradient of Chordin and instead favor a Chordin sink, established by Bmp1a and Tolloid, as the primary mechanism that drives BMP gradient formation.

Double emulsions may be able to protect and release in a controlled manner bioactive compounds during digestion of food matrices. It was hypothesized that the physical state and solid content in the inner phases of water-in-oil-in-water (W1/O/W2) emulsions may affect the overall stability and the release behavior of bioactives during in vitro digestion. Therefore, hydrophobic (phytosterols or Vitamin D3) and hydrophilic (Vitamin B12) molecules were incorporated in double emulsions prepared either with a liquid (soybean oil - SO) or oil-fat gel (soybean oil+trimyristin - STO) lipid phase and liquid internal aqueous phase. In addition, the impact of a gelled inner aqueous phase was studied, using high methoxyl pectin. W1/O/W2 emulsions were prepared with polyglycerol polyricinoleate (PGPR) and sodium caseinate as emulsifiers. After the 30min in vitro gastric stage, all double emulsions showed no significant change in size. Lipid crystals were visible in the STO emulsions. Fat crystallization, and the formation of an oil fat gel, led to coalescence of the inner aqueous droplets. The inner aqueous droplets were no longer visible by confocal microscopy after the initial stages of 2h in vitro duodenal digestion. Fat crystals and droplets of non-spherical shape were also noted in the STO double emulsions up to 25min of in vitro duodenal stage. Overall, the STO emulsions had a higher extent of free fatty acid release and consequent bioactive transfer compared to the SO emulsions. The presence of the medium chain fatty acids (from trimyristin), in addition to the surface-to-core distribution of the hydrophobic bioactives within the oil droplet were key factors in lipid digestibility and bioactive release. The STO and SO samples did not differ in terms of the release of the hydrophilic molecule, vitamin B12, over time. On the other hand, there was a significant increase in the stability of the inner water phase, after gastric digestion, when this phase was gelled with high methoxyl pectin. This work demonstrated that the physical properties of the different internal phases of W1/O/W2 influenced lipid digestion and bioactive transfer kinetics during in vitro digestion.

Cassava (Manihot esculenta Crantz) is a staple food and an important source of starch, and the attributes of its tuberous root largely depend on the variety. The proteome of cassava has been investigated; however, to date, no study has focused on varieties that reveal the molecular basis of phenotypical characteristics. Therefore, we aimed to compare the proteome of the tuberous roots of six cassava varieties that differed in carbohydrates, carotenoids, and resistance to diseases, among other attributes. Two-dimensional gels showed 146 differential spots between the varieties, and the functional roles of some differential proteins were correlated to phenotypic characteristics of the varieties, such as the amount of carbohydrates or carotenoids and the resistance to biotic or abiotic stresses. The results obtained here highlight elements that might help to direct the improvement of new cultivars of cassava, which is an economically and socially relevant crop worldwide.

A comparative proteomic investigation between the pre-climacteric and climacteric mango fruits (cv. Keitt) was performed to identify protein species with variable abundance during ripening. Proteins were phenol-extracted from fruits, cyanine-dye-labeled, and separated on 2D gels at pH 4-7. Total spot count of about 373 proteins spots was detected in each gel and forty-seven were consistently different between pre-climacteric and climacteric fruits and were subjected to LC-MS/MS analysis. Functional classification revealed that protein species involved in carbon fixation and hormone biosynthesis decreased during ripening, whereas those related to catabolism and the stress-response, including oxidative stress and abiotic and pathogen defense factors, accumulated. In relation to fruit quality, protein species putatively involved in color development and pulp softening were also identified. This study on mango proteomics provides an overview of the biological processes that occur during ripening.

This study aimed to identify risk factors for voice disorders (hoarseness in the previous six months as an initial manifestation, and vocal cord nodules as a more severe manifestation of overt disease) among teachers. The cross-sectional study included 747 women teachers from elementary and middle schools in the public school system in Vitória da Conquista, Bahia State, Brazil. Data were collected using a self-applied, standardized questionnaire. Hoarseness in the previous six months was reported by 59.2% of teachers and vocal cord nodules by 12.9%. Logistic regression techniques showed that hoarseness was statistically associated with > 24 classroom hours per week, work in more than one school, and having to make an effort to speak. Vocal cord nodules were associated with working > 5 years as a teacher, work in more than one school, working at another job besides teaching, and having to make an effort to speak. In conclusion, voice disorders are frequent among schoolteachers and are associated with multiple occupational risk factors, besides purely biological ones.

Objetivou-se identificar fatores associados a alterações vocais (rouquidão nos últimos seis meses, como uma manifestação inicial, e calo nas cordas vocais, como uma manifestação mais severa, de doença já instalada) entre professoras. Um estudo epidemiológico de corte transversal investigou 747 professoras da rede municipal de ensino de Vitória da Conquista, Bahia, Brasil, coletando dados num formulário padronizado, auto-aplicado. A queixa de rouquidão nos últimos seis meses foi referida por 59,2 por cento das professoras e a do diagnóstico médico de calo nas cordas vocais, por 12,9 por cento. Técnicas de regressão logística revelaram que rouquidão nos últimos seis meses estava estatisticamente associada a trabalhar > 24 horas em sala de aula, trabalhar em mais de uma escola e fazer força para falar. A queixa de calo nas cordas vocais estava associada a trabalhar > 5 anos como docente, trabalhar em mais de uma escola, trabalhar em outra atividade além da docência e fazer força para falar. Conclui-se que alterações vocais são freqüentes nesta população e estão associadas a múltiplos fatores ocupacionais, além daqueles de cunho fundamentalmente biológico.

As relações entre as condições de trabalho e a saúde de professores têm merecido vários estudos, recentemente. Contudo, são raros os trabalhos baseados em diagnósticos de doenças ocupacionais. Este trabalho buscou identificar as doenças ocupacionais diagnosticadas mais freqüentemente nos atendimentos realizados a professores pelo Centro de Estudos da Saúde do Trabalhador (CESAT) de Salvador, Bahia. Foi realizado um estudo descritivo de série de casos com base em dados secundários. A população-alvo foi formada por todos os professores atendidos pelo CESAT de 1991 a 2001. Foram calculadas a proporção de professores com doença ocupacional entre os atendidos e as proporções de algumas doenças ocupacionais específicas. Foram analisados os dados referentes a 235 professores. A média de idade foi de 42 anos, com predominância do sexo feminino (97por cento). Do total, 66por cento dos professores atendidos receberam diagnóstico de doença ocupacional. As doenças mais freqüentes foram: doenças da laringe e das cordas vocais, síndrome do túnel do carpo, síndrome do manguito rotatório, epicondilites, bursites do ombro, tendinites, rinites, sinusites e faringites crônicas e alérgicas.

The scientific literature on teachers' health is scarce, recent, and focuses predominantly on stress and burnout. This study describes the labor conditions of private school teachers in Vitória da Conquista, Bahia State, Brazil. Information on 250 teachers from the ten largest schools in the municipality was collected through a self-applied questionnaire. The most relevant characteristics of teachers' work, evaluated by the Job Content Questionnaire were: speed of work, creativity at work, and relations with colleagues. The most frequent complaints related to posture, mental strain, and voice problems. Prevalence of minor psychological disorders according to the Self Reporting Questionnaire-20 was 41.5%, strongly associated with long periods of intense concentration on the same job and excessive work. Results suggest an association between the prevalence of minor psychological disorders and certain characteristics of teaching work, emphasizing teachers' exposure to stress.

A literatura científica sobre a saúde dos professores é escassa e recente, enfocando especialmente o desgaste e estresse. Este trabalho objetivou descrever as condições de trabalho e saúde dos professores da rede particular de ensino da cidade de Vitória da Conquista, Bahia, Brasil. Num questionário auto-aplicado foram coletadas informações de 250 professores de dez escolas. Entre as características do trabalho docente, avaliadas pelo Job Content Questionnaire, destacaram-se ritmo acelerado de trabalho, ser criativo e ter boas relações com as pessoas no trabalho. As queixas de saúde mais freqüentes estavam relacionadas à postura corporal, à saúde mental e a queixas relacionadas à voz. A prevalência de distúrbios psíquicos menores (DPM), medida pelo Self Reporting Questionnaire-20, foi de 41,5 por cento e estava fortemente associada a longos períodos de intensa concentração em uma mesma tarefa e volume excessivo de trabalho. Os resultados sugerem relação entre a prevalência de DPM e algumas características do trabalho docente, evidenciando desgaste psicológico do educador.
