RESUMO

Cyathostomins are the most prevalent parasitic nematodes of grazing horses. They are responsible for colic and diarrhea in their hosts. After several decades of exposure to synthetic anthelmintics, they have evolved to become resistant to most compounds. In addition, the drug-associated environmental side-effects question their use in the field. Alternative control strategies, like bioactive forages, are needed to face these challenges. Among these, chicory (Cichorium intybus, Puna II cultivar (cv.)) is known to convey anthelmintic compounds and may control cyathostomins in grazing horses. To challenge this hypothesis, we measured fecal egg counts and the rate of larval development in 20 naturally infected young saddle horses (2-year-old) grazing either (i) a pasture sown with chicory (n = 10) or (ii) a mesophile grassland (n = 10) at the same stocking rate (2.4 livestock unit (LU)/ha). The grazing period lasted 45 days to prevent horse reinfection. Horses in the chicory group mostly grazed chicory (89% of the bites), while those of the control group grazed mainly grasses (73%). Cyathostomins egg excretion decreased in both groups throughout the experiment. Accounting for this trajectory, the fecal egg count reduction (FECR) measured in individuals grazing chicory relative to control individuals increased from 72.9% at day 16 to 85.5% at the end of the study. In addition, larval development in feces from horses grazed on chicory was reduced by more than 60% from d31 compared to control individuals. Using a metabarcoding approach, we also evidenced a significant decrease in cyathostomin species abundance in horses grazing chicory. Chicory extract enriched in sesquiterpenes lactones was tested on two cyathostomins isolates. The estimated IC50 was high (1 and 3.4 mg/ml) and varied according to the pyrantel sensitivity status of the worm isolate. We conclude that the grazing of chicory (cv. Puna II) by horses is a promising strategy for reducing cyathostomin egg excretion and larval development that may contribute to lower the reliance on synthetic anthelmintics. The underpinning modes of action remain to be explored further.

In the equine industry, monitoring of the reproduction cycle is key to be able to produce one foal per mare and per year. Ovulation detection is difficult partly due to the variability of the estrus length. Currently, the most reliable method for ovulation detection is transrectal ultrasonography. This technique, however, implies handling of the mare as well as veterinary costs. The aim of this experimentation is to study body temperature variations around ovulation. Nine reproduction cycles were monitored around ovulation. Transrectal ultrasonographies were performed each day as well as blood sampling to dose estradiol-17ß and progesterone to confirm ultrasonographic results. Body temperature was automatically recorded every 10 minutes using an identification chip equipped with a temperature sensor implanted in the mares' neckline. Data were analyzed using linear mixed models. Daily body temperature pattern did not vary between the phases of the reproductive cycle (follicular, ovulatory and luteal). Temperature differences between phases, however, were identified and appeared hourly-specific. There was an increase of temperature at ovulation compared to the end of the follicular phase ranging from 0.51°C ± 0.21°C to 0.92°C ± 0.26°C and occurring between 04:30 and 08:00. Moreover, a significant increase of body temperature was measured during the first days of luteal phase, ranging from 0.29°C ± 0.17°C to 0.60°C ± 0.16°C, between 10:30 and 16:00. Body temperature varied around ovulation and it might be a promising tool for mare reproduction monitoring. A more complete study, however, focusing on the whole cycle is required.

Nulliparity is associated with intra-uterine growth retardation and foal delayed catch-up growth. Older mares produce larger/taller foals than the precedents. Nursing at conception on foal growth had not been investigated yet. In any case, milk production conditions the foal's growth. This study aimed to determine effects of mare parity, age and nursing on subsequent lactation quantity and quality. Saddlebred mares and their foals (N = 43) run as a single herd over the same year were: young (6-7-year-old) primiparous, young multiparous, old (10-16-year-old) multiparous nursing at insemination time or old multiparous barren the previous year. No young nursing nor old multiparous mares were available. Colostrum was collected. Milk production and foal weight were monitored at 3-, 30-, 60-, 90- and 180-days postfoaling. The foal average daily weight gain (ADG) was calculated for each period between two measurements. Milk fatty acid (FA), sodium, potassium, total protein and lactose contents were determined. The primiparous versus multiparous colostrum was richer in immunoglobulin G, with lower production but greater FA contents in milk. The primiparous foals had a lower ADG for 3 to 30 days postpartum period. Old mares' colostrum contained more SFA and less polyunsaturated FA (PUFA) whereas their milk was richer in proteins and sodium and poorer in short-chain-SFA with a reduced PUFA/SFA ratio at 90 days. Nursing mares' colostrum was richer in MUFA and PUFA and late-lactation milk production was reduced. In conclusion, parity, age and nursing at conception affect mare's colostrum and milk production and foal growth and should be considered for broodmares' management.

Equid milk is arousing increasing interest in consumers and researchers because of its similarity in composition to human milk. The low and different protein content makes equid milk it suitable for children with cow milk protein allergy. Both horse and milk production, in many farms, still follow a characteristic and traditional method of separating the foal from the mother to allow milking procedures. This separation lasts at least 2 to 3 h before milking, a time in which the foal remains fasting. This operation is repeated several times a day, as the equid udder has little collection capacity, and milking frequency is one of the most important parameters to increase milk production; it must be emptied often. New partial artificially suckling techniques have been developed that allow the foal to be separated from its mother for many hours without starving. Furthermore, mechanical milking has been introduced in equid milk production, although in-depth knowledge is lacking on milking parameters and how these aspects affect milk production and udder health. Moreover, in some farms, new milking parlors for Equidae have been developed, ensuring that stress is minimized and production and animal welfare are ensured. It is important to develop and apply technologies for equid milk production, evaluating potential effects on welfare, health, and milk production. This represents the broadest perspective and the greatest challenge because of the need to understand management best practices, thinking to the possibility to introduce as soon as possible automatic milking systems that could ensure a good milking frequency.

BACKGROUND: Breeding a mare until she is not fertile or even until her death is common in equine industry but the fertility decreases as the mare age increases. Embryo loss due to reduced embryo quality is partly accountable for this observation. Here, the effect of mare's age on blastocysts' gene expression was explored. Day 8 post-ovulation embryos were collected from multiparous young (YM, 6-year-old, N = 5) and older (OM, > 10-year-old, N = 6) non-nursing Saddlebred mares, inseminated with the semen of one stallion. Pure or inner cell mass (ICM) enriched trophoblast, obtained by embryo bisection, were RNA sequenced. Deconvolution algorithm was used to discriminate gene expression in the ICM from that in the trophoblast. Differential expression was analyzed with embryo sex and diameter as cofactors. Functional annotation and classification of differentially expressed genes and gene set enrichment analysis were also performed. RESULTS: Maternal aging did not affect embryo recovery rate, embryo diameter nor total RNA quantity. In both compartments, the expression of genes involved in mitochondria and protein metabolism were disturbed by maternal age, although more genes were affected in the ICM. Mitosis, signaling and adhesion pathways and embryo development were decreased in the ICM of embryos from old mares. In trophoblast, ion movement pathways were affected. CONCLUSIONS: This is the first study showing that maternal age affects gene expression in the equine blastocyst, demonstrating significant effects as early as 10 years of age. These perturbations may affect further embryo development and contribute to decreased fertility due to aging.

Although puberty can occur as early as 14-15months of age, depending on breed and use, the reproductive career of mares may continue to advanced ages. Once mares are used as broodmares, they will usually produce foals once a year until they become unfertile, and their productivity can be enhanced and/or prolonged through embryo technologies. There is a general consensus that old mares are less fertile, but maternal age and parity are confounding factors because nulliparous mares are usually younger and older mares are multiparous in most studies. This review shows that age critically affects cyclicity, folliculogenesis, oocyte and embryo quality as well as presence of oviductal masses and uterine tract function. Maternal parity has a non-linear effect. Primiparity has a major influence on placental and foal development, with smaller foals at the first gestation that remain smaller postnatally. After the first gestation, endometrial quality and uterine clearance capacities decline progressively with increasing parity and age, whilst placental and foal birthweight and milk production increase. These combined effects should be carefully balanced when breeding mares, in particular when choosing and caring for recipients and their foals.

Gestational length is highly variable in horses ranging from 320 to 360 days. Thus, determining parturition time is an important challenge for the horse industry. Body temperature can be used in cows and ewes as an indicator of parturition. Thus, the aim of this study is to determine if temperature can also be used as indicator of foaling. Thirty-nine mares were monitored over two foaling seasons (2018 and 2019). They were housed in 16 m2 stalls with access to pasture in group three times a week from 10:00 to 16:00. Night watch as well as video monitoring was ensured during foaling periods. Body temperature was monitored using an identification and temperature sensor microchip implanted in the neckline. Measurement were taken manually every 2 h from 5 days before to 6 h after parturition by moving a microchip reader close to the mares' neck. Mares were equipped with a tail accelerometer recording tail movements and lateral recumbency 24 h before parturition. In addition, behaviour was monitored by video analysis in the hour preceding expulsion of the foal in 8 individuals in 2019. Relationships between behavioural and temperature data were explored throughout principal component analysis (PCA). All foals were born healthy and no human intervention was required during foaling. Mean daily body temperature decreased significantly by 0.3 °C (95%; range: 0.42 to -0.19 °C) between the day of parturition and the mean temperature of the 5 preceding days. A significant temperature decrease was also detected 12 h before and at the onset of parturition. With a 0.5 °C threshold, foaling could be detected 12 h in advance with 96.6% sensitivity and 95.0% specificity, respectively. Tail movements were more frequent and shorter with impending parturition. Body temperature was positively correlated with increased frequency and duration of specific behaviours (flehmen, looking at their flank and rump scratching against the stall wall). In conclusion, as in other species, body temperature was related to signs usually associated with impeding parturition, with a significant temperature drop observed from 12 h before and at the time of foaling. Providing automated measurements become available, temperature monitoring could become an additional tool to predict parturition in mares.

In clinics, temperature is used as an indicator of health. Mostly rectal temperature is recorded, requiring handling and time. Temperature-sensitive identification microchips could be an alternative. Foals (26 males and 17 females), 4-12 months old, were housed in stalls over two winters (December-February). They were equipped with an identification and temperature sensor microchip implanted in the neckline. Temperature was recorded using an antenna located near the drinking trough. Animals were fed concentrated feed and forage twice daily, with free access to water. Rectal temperatures (79 measurements) were recorded simultaneously in 26 animals. Data were analyzed with a linear mixed model, using natural cubic splines for the mean curve and a random horse effect. All animals remained healthy throughout the study. More than 100,000 recordings were obtained. Mean temperature for all individuals at all times was 37.5 ± 0.1°C. Time of the day affected temperature with a daily amplitude of 0.96°C (P < .001). Lowest temperatures were observed before dawn, the acrophase occurring around 18:00, with a smaller increase around midday. Mean temperature was 0.26°C higher in males (P < .05). It was also 0.1°C higher in light (<200 kg) compared with heavier foals (P < .001). Temperature decreased with increasing daylight (-0.35°C over the study period, P < .001). Microchip and rectal temperatures remained within normal limits and were significantly correlated (R2 = 0.16, P < .001). This noninvasive tool does not require extra-handling and will allow a better monitoring of normal body temperature values taking into consideration time of the day, meal time, and sex.

PURPOSE: To compare the effects of 8-wk eccentric (ECC) versus concentric (CON) training using downhill and uphill running in rats on whole body composition, bone mineral density (BMD), and energy expenditure. METHODS: Animals were randomly assigned to one of the following groups: 1) control (CTRL), 2) +15% uphill-running slope (CON), 3) -15% downhill-running slope (ECC15), and 4) -30% downhill-running slope (ECC30). Those programs enabled to achieve conditions of isopower output for CON and ECC15 and of iso-oxygen uptake (V˙O2) for CON and ECC30. Trained rats ran 45 min at 15 m·min five times per week. Total body mass, fat body mass, and lean body mass (LBM) measured through EchoMRI™, and 24-h energy expenditure including basal metabolic rate (BMR) assessed using PhenoMaster/LabMaster™ cage system were obtained before and after training. At sacrifice, the right femur was collected for bone parameters analysis. RESULTS: Although total body mass increased in all groups over the 8-wk period, almost no change occurred for fat body mass in exercised groups (CON, -4.8 ± 6.18 g; ECC15, 0.6 ± 3.32 g; ECC30, 2.6 ± 6.01 g). The gain in LBM was mainly seen for ECC15 (88.9 ± 6.85 g) and ECC30 (101.6 ± 11.07 g). ECC was also seen to positively affect BMD. An increase in BMR from baseline was seen in exercise groups (CON, 13.9 ± 4.13 kJ·d; ECC15, 11.6 ± 5.10 kJ·d; ECC30, 18.3 ± 4.33 kJ·d) but not in CTRL one. This difference disappeared when BMR was normalized for LBM. CONCLUSIONS: Results indicate that for iso-V˙O2 training, the impact on LBM and BMD is enhanced with ECC as compared with CON, and that for isopower but lower V˙O2 ECC, an important stimulus for adaptation is still observed. This provides further insights for the use of ECC in populations with cardiorespiratory exercise limitations.
