RESUMO

Increasing evidence indicates that disturbance of the clock genes, which leads to systemic endocrine perturbation, plays a crucial role in the pathogenesis of metabolic and liver diseases. Fluorene-9-bisphenol (BHPF) is utilized in the manufacturing of plastic materials but its biological effects on liver homeostasis remain unknown. The impacts and involved mechanisms of BHPF on the liver diseases, metabolism, and circadian clock were comprehensively studied by zebrafish and mouse models. The therapeutic effect of melatonin (MT) was also verified. Zebrafish and mouse models with either acute exposure (0.5 and 1 µmol/L, 1-4 days post-fertilization) or chronic oral exposure (0.5 and 50 mg/(kg·2 days), 30 days) were established with various BHPF concentrations. Herein, we identified a crucial role for estrogenic regulation in liver development and circadian locomotor rhythms damaged by BHPF in a zebrafish model. BHPF mice showed chaos in circadian activity through the imbalance of circadian clock component Brain and Muscle Aryl hydrocarbon receptor nuclear translocator-like 1 in the liver and brain. The liver sexual dimorphic alteration along with reduced growth hormone and estrogens played a critical role in damaged glucose metabolism, hepatic inflammation, and fibrosis induced by BHPF. Besides, sleep improvement by exogenous MT alleviated BHPF-related glucose metabolism and liver injury in mice. We proposed the pathogenesis of metabolic and liver disease resulting from BHPF and promising targeted therapy for liver metabolism disorders associated with endocrine perturbation chemicals. These results might play a warning role in the administration of endocrine-disrupting chemicals in everyday life and various industry applications.

Fluorene-9-bisphenol (BHPF) is widely used in the manufacture of plastic products and potentially disrupts several physiological processes, but its biological effects on social behavior remain unknown. In this study, we investigated the effects of BHPF exposure on anxiety-like and social behavior in female mice and the potential mechanisms, thereby proposing a potential therapy strategy. We exposed female Balb/c mice to BHPF by oral gavage at different doses (0.5, 50 mg/kg bw/2-day) for 28 days, which were found BHPF (50 mg/kg) exposure affected motor activity in the open field test (OFT) and elevated cross maze (EPM), resulting in anxiety-like behaviors, as well as abnormal social behavioral deficits in the Social Interaction Test (SIT). Analysis of histopathological staining results showed that BHPF exposure caused damage to hippocampal neurons in the CA1/CA3/DG region and decreased Nissl pyramidal neurons in the CA1/CA3 regions of the hippocampus, as well as a decrease in parvalbumin neuron expression. In addition, BHPF exposure upregulated the expression of excitatory and inhibitory (E/I) vesicle transporter genes (Vglut1, Vglut2, VGAT, GAD67, Gabra) and axon growth gene (Dcc) in the mouse hippocampus. Interestingly, behavioral disturbances and E/I balance could be alleviated by exogenous melatonin (15 mg/kg bw/2-day) therapy. Our findings suggest that exogenous melatonin may be a potential therapy with protective potential for ameliorating or preventing BHPF-induced hippocampal neuronal damage and behavioral disturbances. This study provided new insight into the neurotoxicological effects on organisms exposed to endocrine-disrupting chemicals and aroused our vigilance in current environmental safety about chemical use.

The increasing side effects of traditional medications used to treat type II diabetes have made research into the development of safer and more effective natural medications necessary. ACT001, a derivative of parthenolide, has been shown to have good anti-inflammatory and antitumor effects; however, its role in diabetes is unclear. The short-chain fatty acid propionate is a common food preservative that has been found to cause disturbances in glucose metabolism in mice and humans. This study aimed to investigate whether sodium propionate could aggravate insulin resistance in obese mice and cause diabetes and to study the alleviative effects and potential mechanisms of action of ACT001 on insulin resistance in diabetic mice. Type II diabetic mice were adminietered sodium propionate combined with a high-fat diet (HFD + propionate) by gavage daily for four weeks. Biochemical analysis showed that ACT001 significantly affected blood glucose concentration in diabetic mice, mainly by downregulating the expression of phosphoenolpyruvate carboxykinase 2 and glucose-6-phosphatase. Meanwhile, the level of fatty acid-binding protein 4 in the liver was significantly decreased. ACT001 has a protective effect on the liver and adipose tissue of mice. In addition, the results of the running wheel experiment indicated that ACT001 alleviated the circadian rhythm disorder caused by insulin resistance to a certain extent. This study revealed the potential mechanism by which ACT001 alleviates insulin resistance and provides ideas for developing natural antidiabetic drugs.

Currently, the quest for bamboo materials with high color fastness, rich colors and environmental friendliness is rapidly rising due to its potential applications in construction, furniture and decoration. However, finding an easy-to-operate and environmentally friendly dye for bamboo is a necessary task because of the difficulty in treating the dyeing waste liquid of acid dyes and the complexity of the production process of reactive dyes.Five formulations involving metal polyphenol complexes were employed to straightforwardly produce eco-friendly dyed bamboo and the impact of various formulations on the light aging resistance of the dyed veneers was examined. The results indicated that the light resistance of bamboo veneer dyed with the solution containing only FeSO4·7H2O and tannic acid reached level 4, surpassing the undyed bamboo veneer by three levels. The mechanism of enhanced lightfastness of dyed bamboo veneer was elucidated by XPS analysis. The polyphenol iron complex serves a dual purpose: it absorbs ultraviolet rays and scavenges free radicals within the system. Additionally, it reduced the oxidation of phenolics in the substrate, transforming them into dark-colored quinone structures. This process enhanced the light-aging resistance of the finishing materials. Therefore, this work provides a simple and environmentally friendly method for changing the color of bamboo and provides a new idea for the selection of dyes for bamboo dyeing in actual production.

Because of its long running time, complex working environment, and for other reasons, a gear is prone to failure, and early failure is difficult to detect by direct observation; therefore, fault diagnosis of gears is very necessary. Neural network algorithms have been widely used to realize gear fault diagnosis, but the structure of the neural network model is complicated, the training time is long and the model is not easy to converge. To solve the above problems and combine the advantages of the ResNeXt50 model in the extraction of image features, this paper proposes a gearbox fault detection method that integrates the convolutional block attention module (CBAM). Firstly, the CBAM is embedded in the ResNeXt50 network to enhance the extraction of image channels and spatial features. Secondly, the different time-frequency analysis method was compared and analyzed, and the method with the better effect was selected to convert the one-dimensional vibration signal in the open data set of the gearbox into a two-dimensional image, eliminating the influence of the redundant background noise, and took it as the input of the model for training. Finally, the accuracy and the average training time of the model were obtained by entering the test set into the model, and the results were compared with four other classical convolutional neural network models. The results show that the proposed method performs well both in fault identification accuracy and average training time under two working conditions, and it also provides some references for existing gear failure diagnosis research.

Food enriched with organic selenium is considered as a good source for selenium supplementation. In the current research, we cultivated Pleurotus citrinopileatus with medium containing different levels of sodium selenate, evaluated the antioxidant activity, and sequenced the transcriptome of the Se-enriched mushroom. Selenium content in Se-enriched mushroom is dependent on selenium level in the surroundings. The ABTS total radical scavenging ability was not significantly different between selenium enriched mushroom and the control, though the total phenol content was increased in Se-enriched mushroom. Transcriptome sequencing showed a total of 1036 differentially expressed genes, including 987 upregulated genes and 49 downregulated genes. These differentially expressed genes are involved in 20 metabolic pathways, most of which are involved in carbon metabolism and amino acid biosynthesis, while many differentially expressed genes are in growth, plasma membrane, and protein binding. It needs to be noted that the sulfur metabolism and ABS transporters, which are closely related with selenium metabolism and transportation, are particularly enriched. The mushroom P. citrinopileatus has strong ability to uptake selenium from the surroundings, which modifies many biological pathways. This paper provides a theoretical reference for the development of Se-enriched fungal foods.

Alloying 3d transition metals with Pt has been discovered as an effective strategy to boost the catalytic activity in oxygen reduction reaction (ORR), which, however, often raises the insufficient catalyst durability issue due to rapid leaching of the 3d metal elements. To overcome this issue and realize enhancements in both the activity and the durability properties, here we report a new catalytic structure based on PtGa ultrathin alloy nanowires (NWs), which feature an unconventional strong p-d hybridization interaction. Relative to commercial Pt catalyst, the optimum Pt4.31Ga NWs catalyst exhibited 10.5- and 12.1-fold enhancement in the ORR mass activity and specific activity, respectively. Particularly, the Pt4.31Ga NWs catalyst showed only 15.8% loss in the mass activity after 30 000 cycles of durability test, as compared to a big decrease of 79.6% for the commercial Pt catalyst. Our mechanistic studies find a strong p-d hybridization interaction between Ga and Pt that accounts for the improved ORR performance via synergistically optimizing the surface electronic structure, enhancing the oxidation resistance of Pt, and suppressing the leaching of lattice Ga. We believe this work provides new perspectives to design active and durable electrocatalysts toward ORR.

Proton-exchange membranes (PEMs), characterized by selectively permitting the transfer of protons and acting as a separator in electrochemical devices, have attracted immense attention. The composite membrane, fabricated from organic polymer matrix and high proton-conducting metal-organic framework (MOF), integrates the excellent physical and chemical performances of the polymer and MOF, achieving collective properties for good-performance PEMs. In this study, we demonstrate that MOF-801 shows remarkable proton conductance with σ = 1.88 × 10-3 S cm-1 at 298 K and 98% relative humidity (RH), specifically, together with extra stability to hydrochloric acid or diluting sodium hydroxide aqueous solutions and boiling water. Furthermore, the composite membranes (denoted MOF-801@PP- X, where X represents the mass percentage of MOF-801 in the membrane) have been fabricated using the sub-micrometer-scale crystalline particles of MOF-801 and blending the poly(vinylidene fluoride)-poly(vinylpyrrolidone) matrix, and these PEMs display high proton conductivity, with σ = 1.84 × 10-3 S cm-1 at 325 K 98% RH. A composite membrane as PEM was assembled into H2/O2 fuel cell for tests, indicating that these membrane materials have vast potential for PEM application on electrochemical devices.
