This study evaluates the use of mercury (Hg) concentrations in fish muscle tissue to determine a species' trophic position (TP) in its environment. A campaign conducted in 2019 along 375 km in the middle Araguaia River basin, Brazil, resulted in 239 organisms from 20 species collected. The highest total mercury (THg) concentrations were found in Pellonacastelnaeana (6.93 µg·g-1, wet weight) and in Triportheus elongatus (3.18 µg·g-1, wet weight), whose TPs were different according to the FishBase database. However, they occupied the same trophic level in this study. The intra-specific comparison showed a difference in Hg concentrations between individuals captured in distinct sites. The study of the biota-sediment accumulation factor (BSAF) showed that spatiality interferes with a species' TP. Statistical analyses revealed that when we used a predicted species' TP based on each individual's size, it explained 72% of the variability in THg concentration across all fish species. Multiple regression analysis confirmed that standard length and FishBase values are positively associated with THg (R2 = 0.943). These results point to Hg as a viable indicator of a fish species' TP since it reflects regional, biological, and environmental factors, as demonstrated here for the middle Araguaia River.

Socio-economic constructs and urban topology are crucial drivers of human mobility patterns. During the coronavirus disease 2019 pandemic, these patterns were reshaped in their components: the spatial dimension represented by the daily travelled distance, and the temporal dimension expressed as the synchronization time of commuting routines. Here, leveraging location-based data from de-identified mobile phone users, we observed that, during lockdowns restrictions, the decrease of spatial mobility is interwoven with the emergence of asynchronous mobility dynamics. The lifting of restriction in urban mobility allowed a faster recovery of the spatial dimension compared with the temporal one. Moreover, the recovery in mobility was different depending on urbanization levels and economic stratification. In rural and low-income areas, the spatial mobility dimension suffered a more considerable disruption when compared with urbanized and high-income areas. In contrast, the temporal dimension was more affected in urbanized and high-income areas than in rural and low-income areas.

Many of our routines and activities are linked to our ability to move; be it commuting to work, shopping for groceries, or meeting friends. Yet, factors that limit the individuals' ability to fully realise their mobility needs will ultimately affect the opportunities they can have access to (e.g. cultural activities, professional interactions). One important aspect frequently overlooked in human mobility studies is how gender-centred issues can amplify other sources of mobility disadvantages (e.g. socioeconomic inequalities), unevenly affecting the pool of opportunities men and women have access to. In this work, we leverage on a combination of computational, statistical, and information-theoretical approaches to investigate the existence of systematic discrepancies in the mobility diversity (i.e. the diversity of travel destinations) of (1) men and women from different socioeconomic backgrounds, and (2) work and non-work travels. Our analysis is based on datasets containing multiple instances of large-scale, official, travel surveys carried out in three major metropolitan areas in South America: Medellín and Bogotá in Colombia, and São Paulo in Brazil. Our results indicate the presence of general discrepancies in the urban mobility diversities related to the gender and socioeconomic characteristics of the individuals. Lastly, this paper sheds new light on the possible origins of gender-level human mobility inequalities, contributing to the general understanding of disaggregated patterns in human mobility.

While significant effort has been devoted to understand the role of intraurban characteristics on sustainability and growth, much remains to be understood about the effect of interurban interactions and the role cities have in determining each other's urban welfare. Here we consider a global mobility network of population flows between cities as a proxy for the communication between these regions, and analyze how it correlates with socioeconomic indicators. We use several measures of centrality to rank cities according to their importance in the mobility network, finding PageRank to be the most effective measure for reflecting these prosperity indicators. Our analysis reveals that the characterization of the welfare of cities based on mobility information hinges on their corresponding development stage. Namely, while network-based predictions of welfare correlate well with economic indicators in mature cities, for developing urban areas additional information about the prosperity of their mobility neighborhood is needed. We develop a simple generative model for the allocation of population flows out of a city that balances the costs and benefits of interaction with other cities that are successful, finding that it provides a strong fit to the flows observed in the global mobility network and highlights the differences in flow patterns between developed and developing urban regions. Our results hint towards the importance of leveraging interurban connections in service of urban development and welfare.

Given the rapid recent trend of urbanization, a better understanding of how urban infrastructure mediates socioeconomic interactions and economic systems is of vital importance. While the accessibility of location-enabled devices as well as large-scale datasets of human activities, has fueled significant advances in our understanding, there is little agreement on the linkage between socioeconomic status and its influence on movement patterns, in particular, the role of inequality. Here, we analyze a heavily aggregated and anonymized summary of global mobility and investigate the relationships between socioeconomic status and mobility across a hundred cities in the US and Brazil. We uncover two types of relationships, finding either a clear connection or little-to-no interdependencies. The former tend to be characterized by low levels of public transportation usage, inequitable access to basic amenities and services, and segregated clusters of communities in terms of income, with the latter class showing the opposite trends. Our findings provide useful lessons in designing urban habitats that serve the larger interests of all inhabitants irrespective of their economic status.

The recent availability of digital traces from information and communications technologies has facilitated the study of both individual- and population-level movement with unprecedented spatio-temporal resolution, enabling us to better understand a plethora of socio-economic processes such as urbanization, transportation, impact on the environment and epidemic spreading to name a few. Using empirical spatio-temporal trends, several mobility models have been proposed to explain the observed regularities in human movement. With the advent of the World Wide Web, a new type of virtual mobility has emerged that has begun to supplant many traditional facets of human activity. Here, we conduct a systematic analysis of physical and virtual movement, uncovering both similarities and differences in their statistical patterns. The differences manifest themselves primarily in the temporal regime, as a signature of the spatial and economic constraints inherent in physical movement, features that are predominantly absent in the virtual space. We demonstrate that once one moves to the time-independent space of events, i.e. the sequences of visited locations, these differences vanish, and the statistical patterns of physical and virtual mobility are identical. The observed similarity in navigating these markedly different domains points towards a common mechanism governing the movement patterns, a feature we describe through a Metropolis-Hastings type optimization model, where individuals navigate locations through decision-making processes resembling a cost-benefit analysis of the utility of locations. In contrast to existing phenomenological models of mobility, we show that our model can reproduce the commonalities in the empirically observed statistics with minimal input.

HIV infection may be considered a chronic condition for people living with HIV with access to antiretrovirals and this has effectively increased survival. Moreover, this has also facilitated the emergence of other comorbidities increasing the risk for drug-drug interactions and polypharmacy. The profile of these interactions as well as their consequences for people living with HIV are still not completely elucidated. The objectives of this study were to describe the profile of these interactions, their prevalence and their classification according to the potential for significant or non-significant drug-drug interactions. From June 2015 to July 2016, people living with HIV on follow-up at an Infectious Diseases Referral Center in Belo Horizonte, Brazil have been investigated for the presence of drug-drug interactions. A total of 304 patients were included and the majority (75%) had less than 50 years of age, male (66.4%), and 37.8% self-defined as brown skinned. Approximately 24% were on five or more medications and half of them presented with drug-drug interactions. Patients older than 50 years had a higher frequency of antiretrovirals drug-drug interactions with other drugs compared to younger patients (p=0.002). No relationship was found between the number of drug-drug interactions and the effectiveness of antiretrovirals. As expected, the higher the number of non-HIV medications used (OR=1.129; 95%CI 1.004-1.209; p=0.04) was associated with an increase in drug-drug interactions. The high prevalence of drug-drug interactions found and the data collected should be useful to establish measures of quaternary prevention and to increase the medication security for people living with HIV.

As doenças inflamatórias intestinais são desordens de etiologia idiopática cujo diagnóstico histopatológico é dado por meio de marcadores imunohistoquímicos. Portanto, o objetivo do presente estudo foi revisar os mais recentes ensaios clínicos publicados, que se utilizaram da imunohistoquímica para a avaliação de marcadores úteis no diagnóstico, no tratamento e no acompanhamento de tais doenças. Para isso, realizou-se uma revisão bibliográfica dos trabalhos publicados entre 2014 e 2017, presentes nas bases de dados Pubmed/medline e Cochrane Library, a partir da busca pelos descritores "Inflammatory bowel disease" AND "immunohistochemistry". Os resultados evidenciaram que MMP-9 e calprotectina fecal são marcadores úteis para o diagnóstico das doenças inflamatórias intestinais. Além disso, as quantificações de TNF, IL-6 e Anti-TNF se mostraram úteis para o acompanhamento desses pacientes, principalmente na verificação de melhora ou piora do quadro clínico após a instituição da terapia farmacológica. Concluiu-se, então, que a imunohistoquímica é uma metodologia bastante promissora para a avaliação das DII e, por isso, mais pesquisas que visem a descobertas de outros marcadores e que contribuam para o cuidado desses pacientes devem ser estimuladas.

OBJECTIVE: To examine the relationship between unit-wide Clostridium difficile infection (CDI) susceptibility and inpatient mobility and to create contagion centrality as a new predictive measure of CDI. DESIGN: Retrospective cohort study. METHODS: A mobility network was constructed using 2 years of patient electronic health record data for a 739-bed hospital (n = 72,636 admissions). Network centrality measures were calculated for each hospital unit (node) providing clinical context for each in terms of patient transfers between units (ie, edges). Daily unit-wide CDI susceptibility scores were calculated using logistic regression and were compared to network centrality measures to determine the relationship between unit CDI susceptibility and patient mobility. RESULTS: Closeness centrality was a statistically significant measure associated with unit susceptibility (P < .05), highlighting the importance of incoming patient mobility in CDI prevention at the unit level. Contagion centrality (CC) was calculated using inpatient transfer rates, unit-wide susceptibility of CDI, and current hospital CDI infections. The contagion centrality measure was statistically significant (P < .05) with our outcome of hospital-onset CDI cases, and it captured the additional opportunities for transmission associated with inpatient transfers. We have used this analysis to create easily interpretable clinical tools showing this relationship as well as the risk of hospital-onset CDI in real time, and these tools can be implemented in hospital EHR systems. CONCLUSIONS: Quantifying and visualizing the combination of inpatient transfers, unit-wide risk, and current infections help identify hospital units at risk of developing a CDI outbreak and, thus, provide clinicians and infection prevention staff with advanced warning and specific location data to inform prevention efforts.

The betweenness centrality, a path-based global measure of flow, is a static predictor of congestion and load on networks. Here we demonstrate that its statistical distribution is invariant for planar networks, that are used to model many infrastructural and biological systems. Empirical analysis of street networks from 97 cities worldwide, along with simulations of random planar graph models, indicates the observed invariance to be a consequence of a bimodal regime consisting of an underlying tree structure for high betweenness nodes, and a low betweenness regime corresponding to loops providing local path alternatives. Furthermore, the high betweenness nodes display a non-trivial spatial clustering with increasing spatial correlation as a function of the edge-density. Our results suggest that the spatial distribution of betweenness is a more accurate discriminator than its statistics for comparing  static congestion patterns and  its evolution across cities as demonstrated by analyzing 200 years of street data for Paris.

The city is a complex system that evolves through its inherent social and economic interactions. Mediating the movements of people and resources, urban street networks offer a spatial footprint of these activities. Of particular interest is the interplay between street structure and its functional usage. Here, we study the shape of 472,040 spatiotemporally optimized travel routes in the 92 most populated cities in the world, finding that their collective morphology exhibits a directional bias influenced by the attractive (or repulsive) forces resulting from congestion, accessibility, and travel demand. To capture this, we develop a simple geometric measure, inness, that maps this force field. In particular, cities with common inness patterns cluster together in groups that are correlated with their putative stage of urban development as measured by a series of socio-economic and infrastructural indicators, suggesting a strong connection between urban development, increasing physical connectivity, and diversity of road hierarchies.

Network models of healthcare systems can be used to examine how providers collaborate, communicate, refer patients to each other, and to map how patients traverse the network of providers. Most healthcare service network models have been constructed from patient claims data, using billing claims to link a patient with a specific provider in time. The data sets can be quite large (106-108 individual claims per year), making standard methods for network construction computationally challenging and thus requiring the use of alternate construction algorithms. While these alternate methods have seen increasing use in generating healthcare networks, there is little to no literature comparing the differences in the structural properties of the generated networks, which as we demonstrate, can be dramatically different. To address this issue, we compared the properties of healthcare networks constructed using different algorithms from 2013 Medicare Part B outpatient claims data. Three different algorithms were compared: binning, sliding frame, and trace-route. Unipartite networks linking either providers or healthcare organizations by shared patients were built using each method. We find that each algorithm produced networks with substantially different topological properties, as reflected by numbers of edges, network density, assortativity, clustering coefficients and other structural measures. Provider networks adhered to a power law, while organization networks were best fit by a power law with exponential cutoff. Censoring networks to exclude edges with less than 11 shared patients, a common de-identification practice for healthcare network data, markedly reduced edge numbers and network density, and greatly altered measures of vertex prominence such as the betweenness centrality. Data analysis identified patterns in the distance patients travel between network providers, and a striking set of teaming relationships between providers in the Northeast United States and Florida, likely due to seasonal residence patterns of Medicare beneficiaries. We conclude that the choice of network construction algorithm is critical for healthcare network analysis, and discuss the implications of our findings for selecting the algorithm best suited to the type of analysis to be performed.

ABSTRACT Riffles are unique habitats regarding to assemblage structure. However, understanding how these assemblages respond to environmental variations in small spatial extents, as within a stream, is a challenge. We assess whether the quantitative structure and the trophic composition of fish assemblages vary predictably among stream riffles. We predict that the variation of environmental conditions will explain species abundance and trophic composition, with the latter presenting higher predictive power, since species would be filtered according to their traits (e.g. diet). Moreover, we expect that the low among-riffle dispersal limitation within a stream and the strong habitat filtering would result in lower importance of spatial variables in the structure of riffle fish assemblages. We tested these predictions by studying 18 riffles of a stream in the Central Brazil. Environmental variables, but not spatial ones, were the most important in explaining the variation in assemblages structure. Environmental variables explained a greater portion of the trophic structure variation (R2=0.62) than of abundance (R2=0.37), indicating that the variation on the trophic traits at community level are more predictable. These results also indicate that these assemblages are subject to environmental control, highlighting the importance of riffle characteristics in driving ecological processes within streams.


RESUMO Corredeiras são habitats singulares no que se refere à estrutura das suas assembleias. Entretanto, a compreensão sobre como essas assembleias respondem às variações ambientais em pequenas extensões espaciais, como dentro de um riacho, ainda é um desafio. Avaliamos se a estrutura quantitativa das assembleias de peixes, assim como a sua composição trófica variam previsivelmente entre corredeiras de riachos. Predizemos que a variação nas condições ambientais explicará a abundância das espécies, assim como a sua composição trófica, entretanto, com uma maior proporção da variação explicada para a segunda, já que as espécies seriam filtradas de acordo com seus atributos (e.g. dieta). Além disso, esperamos que a pequena limitação para a dispersão entre as corredeiras de um mesmo riacho, associada à forte filtragem ambiental, resultaria em uma menor importância de variáveis espaciais na estruturação das assembleias de corredeiras. Para testar essas predições, estudamos 18 corredeiras de um rio do Brasil Central. As variáveis ambientais, e não as espaciais, foram mais importantes para explicar a variação na estrutura das assembleias. As variáveis ambientais explicaram uma maior proporção da variação da composição trófica (R2=0,62), em comparação com a abundância (R2=0,37), indicando maior previsibilidade na variação dos atributos relacionados a dieta em nível de comunidade. Esses resultados também indicam que essas assembleias são sujeitas a forte controle ambiental, destacando a importância das características desses habitats nos processos ecológicos dentro dos riachos.

BACKGROUND: CD4+CD25high regulatory T (TReg) cells modulate antigen-specific T cell responses, and can suppress anti-viral immunity. In HTLV-1 infection, a selective decrease in the function of TReg cell mediated HTLV-1-tax inhibition of FOXP3 expression has been described. The purpose of this study was to assess the frequency and phenotype of TReg cells in HTLV-1 asymptomatic carriers and in HTLV-1-associated neurological disease (HAM/TSP) patients, and to correlate with measures of T cell activation. RESULTS: We were able to confirm that HTLV-I drives activation, spontaneous IFNgamma production, and proliferation of CD4+ T cells. We also observed a significantly lower proportion of CTLA-4+ TReg cells (CD4+CD25high T cells) in subjects with HAM/TSP patients compared to healthy controls. Ki-67 expression was negatively correlated to the frequency of CTLA-4+ TReg cells in HAM/TSP only, although Ki-67 expression was inversely correlated with the percentage of CD127low TReg cells in healthy control subjects. Finally, the proportion of CD127low TReg cells correlated inversely with HTLV-1 proviral load. CONCLUSION: Taken together, the results suggest that TReg cells may be subverted in HAM/TSP patients, which could explain the marked cellular activation, spontaneous cytokine production, and proliferation of CD4+ T cells, in particular those expressing the CD25highCD127low phenotype. TReg cells represent a potential target for therapeutic intervention for patients with HTLV-1-related neurological diseases.

Metallothionein (MT), a low-molecular-weight protein with high cysteine content, seems to be related to neoplastic resistance to oncologic treatment and therefore has been studied as a prognostic factor for a variety of human malignant tumors. MT overexpression in neoplasms of ectodermal origin is usually associated with a poor prognosis. MT expression was evaluated in 60 samples of oral squamous cell carcinoma by immunohistochemistry to study its prognostic influence on oral cancer. Possible associations of MT immunoexpression were also investigated with respect to clinical stage (TNM), histological grading, and proliferation index (Ki-67) of the lesions. No significant statistical correlation was observed among these variables. The impact on overall survival was assessed by uni and multivariate statistical tests. Mean MT labeling index was 60%. High MT labeling indexes (over 76%) predicted shorter survival in univariate statistical analysis. In multivariate analysis, MT labeling index and clinical stage were independent prognostic factors. MT overexpression in oral squamous cell carcinoma seems to be related to a worse prognosis for patients.

RESUMO

Pesquisou-se a ocorrência, na parede das veias do plexo pampiniforme de chagásicos crônicos, de acúmulos intracelulares de Trypanosoma cruzi e sinais inflamatórios (flebife). Para tal fim, colheram-se, à necropsia, 23 pares de funículos espermáticos, epidídimos e testículos, sendo 17 de chagásicos crônicos e 6 de controles (näo chagásicos). Em cada caso, foram feitos múltiplos cortes das gônadas e dos vasos; fez-se pesquisa de T. cruzi por imuno-histoquímica nos funículos espermáticos de todos os casos. Näo se observaram parasitos nas paredes vasculares. Notou-se flebite crônica inespecífica, focal e discreta, em cinco chagásicos (bilateral em três pacientes) e dois controles; havia infiltraçäo mononuclear discreta do interstício funicular em treze chagásicos e cinco controles. A análise estatística dos resultados (X²) näo revelou diferenças significativas. Conclui-se que o ambiente hormonal devido à testosterona näo parece favorecer a infecçäo da parede dos vasos gonadais por Trypanosoma cruzi, embora, segundo a literatura, o referido hormônio pareça ter açöes imunodepressoras

RESUMO

Infection of non-adherent TG180 murine sarcoma cells with Toxoplasma gondii was compared, at the ultrastructural level, in both in vivo and in vitro conditions. Suspensions of 3.0 x 10(6) TG180 cells infected in vitro with 1.0 x 10(6) parasites of the RH strain were harvested between the first and 6th day post-infection and processed for transmission electron microscopy. In vivo infection was made by intraperitoneal inoculation in mice of 1.0 x 10(6) TG180 cells, that were co-inoculated with a parasite suspension at the same cell concentration. Cells were harvested 10, 20, 30 min and 24, 48 h post-inoculation and processed for transmission electron microscopy at the same conditions of the in vitro culture. It was observed TG180 murine sarcoma cells with intense and equivalent intracellular parasitism in both conditions. Host cells with parasitophorous vacuoles containing up to 16 parasites, as well as parasites undergoing mitoses or presenting a bradyzoite-like morphology, were frequently seen in both culture methods.
