RESUMO

This study was designed to describe normal axial skeletal structure in common roach Rutilus rutilus from putative unaffected environmental conditions, and the occurrence of skeletal malformations in the fish from an area under radiation contamination. Specimens were collected from water bodies of the Techa Cascade Reservoirs located near the Mayak atomic industry plant in the River Ob' drainage, Chelyabinsk Province, Russia. One sample was collected from Lake Irtyash, a reservoir of drinkable water, supplying the town of Ozersk, and the other one from a technical reservoir which is a storage of liquid radioactive waste from Mayak and characterized by high radioactive contamination (mostly (90)Sr and (137)Cs). A comparison was made with historical material collected from the River Ob' before the middle of the 20th century, i.e. before the environment became affected by radioactive contamination. A high number of abnormalities of the axial skeleton were detected in both Mayak samples, in 94 and 97% of examined specimens, in contrast to about 20% in the historical specimens. The abnormalities were in both the unpaired fins and the vertebral column, including the caudal complex and included supernumerary elements, fusions, deformities and displacement of the elements. Most axial skeleton abnormalities, however, were minor, such as splitting, shortening or deformation of spines. Severe defects, such as extensive scolioses, lordoses and kyphoses, were not found. The causes of the abnormalities were not identified in this study, but the high incidence of malformations may be attributed to genetically determined imbalance during development. The almost equal distribution of abnormalities among the fish from non-contaminated and radioactive contaminated reservoirs may be explained by either recent gene flow within the population of R. rutilus in the River Techa system or the effect of unknown unfavourable environmental factors such as chemical pollution.

Genetic variation and geographical structuring of vimba Vimba vimba were analysed across 26 sites (80 individuals) by means of mtDNA sequences (cyt b gene, mitochondrial control region) to localize hypothesized glacial refugia and to reconstruct postglacial recoloniation routes. Although genetic diversity among sequenced individuals was low, a combined analysis of the two sequenced fragments revealed a western (central and northern Europe: Danube, Elbe and lakes of Sweden) and an eastern clade (eastern Europe: Dnieper-South Bug, Don, Neman). Furthermore, a number of divergent ancestral haplotypes distributed around the Black and Caspian Seas became apparent. Mismatch analyses supported a sudden expansion model for the populations of the western clade between 50 and 10 000 bp. Overall, the study provides strong evidence for a northward and westward expansion of V. vimba from two refugial regions located in the Danubian drainage and the northern Pontic regions respectively.

We used DNA sequence variation at the mitochondrial cytochrome b gene (1141 bp) to assess the phylogeography of Barbus fishes in the Black Sea region. Our aim was to test whether the recent ( approximately 22 000-7500 years ago) freshwater phase of the Black Sea was a conduit for gene flow among freshwater fishes that today are found in streams entering the saltwater Black Sea. Deep phylogeographical breaks suggestive of allopatric divergence were observed between four regional groups of populations. Coalescent simulation used to distinguish between this and an alternative scenario that the phylogeographical structure was due to random lineage sorting showed that the contemporary populations were unlikely (P < 0.001) to have been founded by a single ancestral population. Divergences between the lineages (0.86-2.54%) were dated to the Middle to Late Pleistocene using distances and a molecular clock corrected for superimposed substitutions. Taken together, this evidence suggests that multiple refugial populations survived over several later glaciations in the vicinity of the Black Sea. This Pontic refugium served as the primary source for the postglacial expansion throughout Europe as far as the Atlantic basin. However, only one of the phylogeographical lineages contributed to this dispersion, whereas the others remain restricted to the Black Sea region and followed independent evolutionary trajectories.
