RESUMO

Cyathophorella adiantum (Griff.) M. Fleisch. (Division-Bryophyta, Family-Daltoniaceae), an Eastern Himalayan moss was studied for the first time to identify the volatiles derived from cellular and membrane bound fatty acids. A high capacity sample enrichment probe (SEP) was used for extraction of headspace volatile (HSV) molecules followed by GC-MS analysis. Different short-chain oxylipins like alkenes, alkanes, saturated and unsaturated alcohols, saturated and unsaturated aldehydes, ketones were identified along with free and esterified fatty acids, cyclo compounds and some by-products of secondary metabolites. Fatty acid analysis of neutral lipids (NL) and phospholipids (PL) of this plant exhibits the predominance of C16 and C18 fatty acids. It also reveals some interesting information that might indicate the possible fatty acid precursors for volatile generation and their sources in this plant.

In lizards, the epidermal glands of the femoral and precloacal regions are involved in the production of semiochemicals. In addition to its femoral glands, the giant girdled lizard, or sungazer, Cordylus giganteus, which is endemic to South Africa, has generation glands as an additional potential source of semiochemicals. These epidermal glands are described as glandular scales that overlay the femoral glands and are included in the normal epidermal profile located in the femoral (thigh) and anterior antebrachial (fore-leg) regions of the male sungazer. GC-MS analysis of the generation gland secretions and the trimethylsilyl derivatives of some of the steroidal constituents was employed to identify 59 constituents, including alkenes, carboxylic acids, alcohols, ketones, aldehydes, esters, amides, nitriles, and steroids. The quantitative differences of the volatile constituents of the fore- and hind-leg generation glands were compared between individuals. This is the first report on the chemical composition of generation glandular material of lizards.

Smoke plays an intriguing role in promoting the germination of seeds of many species following a fire. Recently, a bicyclic compound containing a condensed butenolide moiety, 3-methyl-2H-furo[2,3-c]pyran-2-one (1), was reported as a potent germination promoter from plant-derived smoke. In this study, a related butenolide, 3,4,5-trimethylfuran-2(5H)-one (2), which inhibits germination and significantly reduces the effect of 1 when applied simultaneously, was also isolated from plant-derived smoke. The interaction of these compounds with opposing actions on seed germination may have important ecological implications in a post-fire environment and could be useful molecules for understanding the events involved in breaking seed dormancy and promoting seed germination.

The giant girdled lizard or sungazer, Cordylus giganteus, is endemic to South Africa. It has been suggested that in this species, as in other lizard species, epidermal glands in the femoral, pre-cloacal regions, and cloacal glands are the main sources of semiochemicals and that these secretions could play an important role at different levels of the social biology of the animals. To gain a better understanding of the nature of the femoral gland secretions of the sungazer, characterization of the constituents of the secretions was carried out. By using GC-MS analysis, in conjunction with auxiliary techniques, such as solventless sample introduction and trimethylsilyl derivatization, 53 relatively involatile compounds, including carboxylic acids, alcohols, ketones, esters, and steroids, were identified in the secretions of both sexes. The study showed that the secretions of male and female sungazers contain only semi-volatile chemicals.

The development of a sensitive and solvent-free method for the measurement of estrone (E(1)) and 17beta-estradiol (17beta-E(2)) in human urine samples is described. The deconjugated estrogens were derivatized in situ with acetic acid anhydride and the derivatives were extracted directly from the aqueous samples using stir bar sorptive extraction (SBSE). The compounds containing a secondary alcohol function are further derivatized by headspace acylation prior to thermal desorption and gas chromatography/mass spectrometry (GC/MS). A number of experimental parameters, including salt addition, temperature and time, were optimized to increase the recovery of E(1) and 17beta-E(2) by SBSE. The derivatization reactions were also optimized to obtain the highest yields of the acylated estrogens. Detection limits of 0.02 and 0.03 ng mL(-1) were obtained for E(1) and 17beta-E(2), respectively. The method was applied to determine the effect of conjugated equine estrogen intake on the excretion of E(1) and 17beta-E(2) in human urine samples. Increased levels of the endogenous estrogens were detected after administering a standard dose of Premarin to a female volunteer. Routine monitoring of estrogen levels is recommended to avoid a high urinary excretion of E(1) and 17beta-E(2), nowadays enlisted as endocrine disrupting chemicals (EDCs), during hormone replacement therapy.

A sample enrichment probe (SEP) consisting of a thin rod of an inert material and provided at one end with a short sleeve of polydimethylsilicone rubber was used for the high-capacity sample enrichment of analytes from gaseous and aqueous samples for analysis by gas chromatography (GC) and its hyphenated techniques. The silicone rubber was exposed to the analytical sample, after which the end of the rod carrying the silicone rubber was introduced into the injector and the analytes thermally desorbed and analysed by GC. This technique is similar to, but differs from, solid-phase microextraction (SPME) in that a much larger volume of the sorptive phase is employed, the sorptive phase is not introduced into the inlet of the GC via a needle and the injector is opened to the atmosphere for the introduction and removal of the SEP. In the determination of volatile and semi-volatile organic compounds in gaseous and aqueous media, the SEP technique gave results comparable with those obtained by the stir-bar-sorptive extraction (SBSE) and high-capacity sorption probe (HCSP) techniques. Implementation of the SEP technique requires only minor adaptations to the gas chromatograph and does not require any auxiliary thermal desorption and cryotrapping equipment.

The urine of the cheetah, Acinonyx jubatus, is almost odorless, and probably for this reason, it has not attracted much attention from scientists. Using gas chromatography-mass spectrometry, we identified 27 and 37 constituents in the headspace vapor of the urine of male and female cheetah, respectively. These constituents, composed of hydrocarbons, short-chain ethers, aldehydes, saturated and unsaturated cyclic and acyclic ketones, 2-acetylfuran, dimethyl disulfide, dimethyl sulfone, phenol, myristic acid (tetradecanoic acid), urea, and elemental sulfur, are all present in the headspace vapor in very small quantities; dimethyl disulfide is present in such a low concentration that it cannot be detected by the human nose. This is only the second example of elemental sulfur being secreted or excreted by an animal. It is hypothesized that the conversion of sulfur-containing compounds in the cheetah's diet to elemental sulfur and to practically odorless dimethyl sulfone enables this carnivore to operate as if "invisible" to the olfactory world of its predators as well as its prey, which would increase its chances of survival.

A simple and fast method for the measurement of 4-hydroxynonenal (4HNE), a highly toxic end-product of lipid peroxidation, in urine samples is described. The method combines stir bar sorptive extraction (SBSE) with two derivatization steps, followed by thermal desorption and GC/MS. 4HNE is derivatized in situ with O-(2,3,4,5,6-pentafluorobenzyl) hydroxylamine and the oxime is extracted from the aqueous phase with SBSE. The 4HNE-oxime is further acylated by headspace derivatization prior to thermal desorption. Derivatization reactions and extraction were optimized in terms of reagent quantities, temperature and time. The method is linear over a concentration range of 0.5-5 ng mL(-1) with a correlation coefficient of 0.997. The limit of detection and limit of quantitation are 22 and 75 pg mL(-1) urine, respectively. The high sensitivity of the method allows the measurement of physiological concentrations of 4HNE in urine samples.

During its natural life cycle, the yellow dog tick, Haemaphysalis leachi, has three hosts, and it has to spend enough time on each of them to complete a blood meal. When irritated, the females of this tick species produce a cuticular secretion that contains a dog-repelling allomone. This improves the tick's chances of survival by deterring the dog from biting the tick off its body. Employing response-guided isolation techniques in conjunction with gas chromatography-mass spectrometry, the defensive allomone of H. leachi was found to consist of the six homologous aliphatic aldehydes from hexanal to undecanal. A mixture of synthetic versions of these six aldehydes in quantities corresponding to those secreted by one tick elicited strong aversion reactions in the majority of dogs of various breeds.

INTRODUCTION: The inflammation- and infection-seeking properties of (131)I-labeled ornidazole, a 5-nitroimidazole derivative, have recently been reported. Whole-body images in rabbits showed a more rapid uptake in inflamed areas compared to (67)Ga. In the present study, two novel (123)I-labeled 2-methyl-4-nitroimidazole derivatives were synthesized and their infection-seeking properties compared with those of (67)Ga and (123)I-labeled ornidazole. METHODS: Radiolabeling was carried out by means of iodide-for-tosylate, triflate or halogen exchange. Various methods were utilized in order to synthesize the labeling precursors for the (123)I-labeled novel compounds. Serum stability studies on all of the (123)I-labeled tracers were followed by gamma camera imaging studies on rabbits artificially infected with Escherichia coli bacteria. RESULTS AND CONCLUSIONS: The (123)I-labeled tracers were obtained in moderate to good radiochemical yields (34-80%) and acceptable radiochemical purities (93-99%). In contrast to (123)I-labeled ornidazole, 1-[(1-hydroxy-3-[(123)I]iodoprop-2-yloxy)methyl]-2-methyl-4-nitroimidazole (2) and 1-[(1-[(123)I]iodoprop-2-yloxy)methyl]-2-methyl-4-nitroimidazole (3) showed high serum stability. Compared to noninfected controls, all of the (123)I-labeled tracers showed increased uptake at the area of induced infection after 6 and 24 h, but the uptake was significantly lower than in the case of (67)Ga over the same period. Tracer 3 showed a slightly superior uptake after 6 h than the other (123)I-labeled tracers over the same period. The advantage of the initially slightly faster rate at which nitroimidazole tracers appear to accumulate in the infection area in comparison to (67)Ga might not outweigh the advantage of the eventual higher target to nontarget ratio displayed by (67)Ga.
