BACKGROUND: This research evaluated the anti-Candida albicans effect of Mexican propolis from Chihuahua. Chemical composition of the ethanolic extract of propolis was determined by GC-MS, HPLC-DAD, and HPLC-MS. The presence of anthraquinone, aromatic acid, fatty acids, flavonoids, and carbohydrates was revealed. RESULTS: The anti-Candida activity of propolis was determined. The inhibitions halos were between 10.0 to 11.8 mm; 25% minimum inhibitory concentration (0.5 mg/ml) was fungistatic, and 50% minimum inhibitory concentration (1.0 mg/ml) was fungicidal. The effect of propolis on the capability of C. albicans to change its morphology was evaluated. 25% minimum inhibitory concentration inhibited to 50% of germ tube formation. Staining with calcofluor-white and propidium iodide was performed, showing that the propolis affected the integrity of the cell membrane. INT1 gene expression was evaluated by qRT-PCR. Propolis significantly inhibited the expression of the INT1 gene encodes an adhesin (Int1p). Chihuahua propolis extract inhibited the proliferation of Candida albicans, the development of the germ tube, and the synthesis of adhesin INT1. CONCLUSIONS: Given the properties demonstrated for Chihuahua propolis, we propose that it is a candidate to be considered as an ideal antifungal agent to help treat this infection since it would not have the toxic effects of conventional antifungals.

Some of the species of the genus Bacopa have been used in Pharmacopoeia worldwide. However, in Mexico, Bacopa monnieri has neither been extensively cultivated nor studied, nor has their use in traditional medicine been reported. The aim of this work was to assess the taxonomic verification of the four wild populations of B. monnieri, the chemical content of their pigments and phenols and to provide an analysis of their potential bioactivity. B. monnieri wild populations from Mexico were validated using molecular markers. Chromatographic profiling using HPLC-PDA revealed 21 compounds comprising 12 chlorophylls and nine carotenoids; of the latter, the major ones were lutein (0.921 ± 0.031 µg/mg of dry extract) and ß-carotene (0.095 ± 0.003 µg/mg of dry extract). The total phenolic content, determined using the Folin-Ciocalteu assay, ranged from 54.8 ± 5.8 to 70.3 ± 2.2 µg of gallic acid equivalents (GAE)/mg. Plant extracts scavenged from the free radical DPPH in IC50 ranged from 130.6 ± 3.0 to 249.9 ± 12.1 µg dry extract/mL. In terms of the anti-inflammatory potential, the most effective extract was from a soil-based plant from Jalisco (BS), reduced from nitric oxide in a RAW 264.7 culture medium, with an IC50 value of 134 µg of dry extract/mL. The BS extract showed a significant neutral lipid-reducing activity in the zebrafish model, ranging from 3.13 µg/mL p < 0.05 to 100 µg/mL p < 0.0001. Overall, the extracts analyzed here for the first time seem promising for future use because of their antioxidant, anti-inflammatory and anti-obesity potential.

A 32-fold increase in laccase activity production by the thermophilic biomass-degrading fungus T. terrestris Co3Bag1 was achieved when the microorganism was grown on a modified medium containing fructose, sodium nitrate, and copper. A 70 kDa laccase (TtLacA), produced under the above conditions, was purified, immobilized in copper alginate gel beads, and characterized. TtLacA, both free and immobilized enzymes, exhibited optimal activity at pH 3.0, at a temperature of 65 and 70 °C, respectively, although both displayed 70% of activity from 40 to 70 °C. Free and immobilized enzymes retained at least 80% of relative activity in the pH range from 3 to 4.6. Immobilized TtLacA manifested a 2.3-fold higher thermal stability than the free form of the enzyme at 60 and 70 °C. Immobilized TtLacA retained 95% initial activity for six consecutive reuse cycles at 60 °C, and also retained 86% of initial activity after 12 days of storage at 4 °C. Based on the biochemical features, thermophilic TtLacA may be an efficient enzyme for dye decolorization and other industrial applications at high temperatures or acidic conditions. This work represents the first report about the immobilization and biochemical characterization of a thermophilic laccase from a member of the genus Thielavia.

The biomass-degrading thermophilic ascomycete fungus Thielavia terrestris Co3Bag1 produces TtCel7A, a native bifunctional cellulase/xylanase GH7 family. The purified TtCel7A, with an estimated molecular weight of 71 kDa, was biochemically characterized. TtCel7A displayed an optimal pH of 5.5 for both activities and an optimal temperature of 60 and 50 °C for cellulolytic and xylanolytic activities, respectively. The half-lives determined for cellulase activity were 140, 106, and 41 min at 50, 60, and 70 °C, respectively, whereas the half-lives observed for xylanase activity were 24, 10, and 1.4 h at 50, 60, and 70 °C, respectively. The KM and Vmax values were 3.12 mg/mL and 50 U/mg for cellulase activity and 0.17 mg/mL and 42.75 U/mg for xylanase activity. Circular dichroism analysis suggests changes in the secondary structure of TtCel7A in the presence of CMC as the substrate, whereas no modifications were observed with beechwood xylan. TtCel7A displayed the excellent capability to hydrolyze CMC, beechwood xylan, and complex substrates such as oat bran, wheat bran, and sugarcane bagasse, with glucose and cellobiose being the main products released; also, slightly less endo cellulase and xylanase activities were observed. Thus, suggesting TtCel7A has an exo- and endomode of action. Based on the characteristics of the enzyme, it might be considered a good candidate for industrial applications.

Temperature is the main factor that impacts germination and therefore the success of annual crops, such as chia (Salvia hispanica L.), whose seeds are known for their high nutritional value related to its oil. The effect of temperature on germination is related to cardinal-temperature concepts that describe the range of temperature over which seeds of a particular species can germinate. Therefore, in this study, in addition to calculated germinative parameters such as total germination and germination rate of S. hispanica seeds, the effectiveness of non-linear models for estimating the cardinal temperatures of chia seeds was also determined. We observed that germination of S. hispanica occurred in cold to moderate-high temperatures (10-35 °C), having an optimal range between 25 and 35 °C, with the highest GR and t50 at 30 °C. Temperatures higher than 35 °C significantly reduced germination. Output parameters of the different non-linear models showed that the response of chia germination to temperature was best explained by beta models (B). Cardinal temperatures calculated by the B1 model for chia germination were: 2.52 ± 6.82 °C for the base, 30.45 ± 0.32 °C for the optimum, and 48.58 ± 2.93 °C for the ceiling temperature.

Genetic studies in rare species are important to determine their patterns of genetic diversity among populations and implement conservation plans aimed to reduce extinction risks. Pachyphytum caesium is an endemic species with extremely small populations in central Mexico. This work analyzes the diversity and genetic structure of Pachyphytum cesium (Crassulaceae) to change the conservation status and propose conservation strategies. Twelve dominant ISSR markers were used to describe the genetic diversity of all known populations. Additionally, we carried out two simulations to explore how the loss of individuals or the local populations extinction affect the genetics parameters of the species. The average results indicated moderate levels of genetic diversity (number of alleles = 89.7 ± 3.9, expected heterozygosity = 0.212 ± 0.0, and percentage of polymorphic loci = 56.1 ± 3.9), parameters that decreased significantly when simulations were performed in P. caesium. Additionally, a genetic structure of the populations was found with low gene flow (Nm = 0.92). Genetic parameters are negatively affected by the loss of individuals and the local extinction of populations. Based on our results, we propose to reclassify the conservation status of the species in danger of extinction, hence a series of conservation strategies are provided to prevent its disappearance.

Cellulomonas uda produces Xyn11A, moderately thermostable xylanase, with optimal activity at 50 °C and pH 6.5. An improvement in the biochemical properties of Xyn11A was achieved by site-directed mutagenesis approach. Wild-type xylanase, Xyn11A-WT, and its mutant Xyn11A-N9Y were expressed in Escherichia coli, and then both enzymes were purified and characterized. Xyn11A-N9Y displayed optimal activity at 60 °C and pH 7.5, an upward shift of 10 °C in the optimum temperature and an upward shift of 1 unit in optimum pH; also, it manifested an 11-fold increase in thermal stability at 60 °C, compared to that displayed by Xyn11A-WT. Molecular dynamics simulations of Xyn11A-WT and Xyn11A-N9Y suggest that the substitution N9Y leads to an array of secondary structure changes at the N-terminal end and an increase in the number of hydrogen bonds in Xyn11A-N9Y. Based on the significant improvements, Xyn11A-N9Y may be considered as a candidate for several biotechnological applications.

Previous research has shown that individuals suffering from depression and generalized anxiety disorder (GAD) seem to have inhibitory control deficits compared with healthy controls. However, few studies have been conducted in Spanish-speaking countries. Thus, this study aims to analyze the performance on the Stroop Color and Word Test (SCWT) between groups of Colombian participants with clinical levels of depression and GAD symptoms and a nonclinical control group. According to previous research, we expected to find significant differences in inhibitory control among groups. An ex post facto design was implemented. The SCWT was administered to a total sample of 105 individuals (64.8% women, M = 22.94 years, SD = 4.62), including 27 depressed and 15 anxious participants according to their scores on the Personal Health Questionnaire-9 and the Generalized Anxiety Disorder-7, respectively. Bayesian t-tests showed that depressed participants showed the same processing speed but lower scores on inhibitory control than healthy controls, BF = 13.70, δ = 0.50, 95% CI [0.08, 0.94]. Conversely, anxious participants showed deficits in processing speed, SCWT-Word: BF = 16.19, δ = 0.68, 95% CI [0.15, 1.24]; SCWT-Color: BF = 5.98, δ = 0.50, 95% CI [-0.01, 1.04], but not in inhibitory control compared with the nonanxious counterparts. This study provides preliminary evidence concerning the inhibitory control deficits in Colombian depressed individuals and processing speed deficits in those experiencing clinical levels of GAD symptoms.

Using the rabbit corneal epithelial cell line RCE1(5T5) as a model, we analyzed three differentiation stages, distinguished on basis to the growth state of cultured cells and after studying the expression of transcription factors such as Oct4, Pax6 and ΔNp63α, selected differentiation markers, and signaling or epigenetic markers such as Notch receptors and Prdm3. Namely, proliferative non-differentiated cells, committed cells, and cells that constitute a stratified epithelium with a limbal epithelial-like structure. RNAseq based transcriptome analysis showed that 4891 genes were differentially expressed among these stages displaying distinctive gene signatures: proliferative cells had 1278 genes as gene signature, and seem to be early epithelial progenitors with an Oct4+, KLF4+, Myc+, ΔNp63α+, ABCG2+, Vimentin+, Zeb1+, VANGL1+, Krt3-, Krt12- phenotype. Committed cells had a gene signature with 417 genes and displayed markers indicative of the beginning of corneal differentiation, and genes characteristic of proliferative cells; we found the possible participation of Six3 and Six4 transcription factors along this stage. The third stage matches with a stratified corneal epithelium (gene signature comprising 979 genes) and is typified by an increase in the expression of WNT10A and NOTCH 2 and 3 signaling and Cux1 transcription factor, besides Pax6, KLF4 or Sox9. The differentiated cells express about 50% of the genes that belong to the Epidermal Differentiation Complex (EDC). Analysis of the differences between corneal epithelium and epidermis could be crucial to understand the regulatory mechanisms that lead to the expression of the differentiated phenotype.

The concept of rule-governed behavior (RGB) has been used in the behavior-analytic literature as a way to analyze complex human behavior, including thinking and problem-solving. Relational frame theory suggests the existence of two main functional types of RGB termed pliance and tracking. In this paper, we describe the development of the Generalized Tracking Questionnaire (GTQ) and the preliminary evaluation of its psychometric properties and validity through three studies, with a total of 1155 participants. In Study 1, a pool of items describing the main characteristics of generalized tracking was designed and evaluated by experts on the RFT account of RGB. The resulting 11 items were administered to 460 undergraduates to examine the understandability and psychometric quality of the items. The exploratory factor analysis indicated that the GTQ can be seen as a unidimensional scale, with all items exhibiting high factor loadings and corrected item-total correlations. In Study 2, the GTQ was administered online to a sample of 464 non-clinical participants and a clinical sample of 125 participants. The one-factor model of the GTQ obtained a good fit in the conducted confirmatory factor analysis. The GTQ showed measurement invariance across gender and clinical and nonclinical participants. It also obtained excellent internal consistency and correlated in theoretically coherent ways with other constructs. In Study 3, the GTQ and a neuropsychological battery of executive functions were administered to 105 participants. The GTQ showed statistically significant, medium-size correlations with working memory tests, verbal fluency, planning, and behavioral inhibition. In conclusion, the GTQ seems to be a promising measure to advance in the empirical analysis of functional classes of RGB.

Scenedesmus obtusiusculus AT-UAM, isolated from Cuatro Ciénegas wetlands in Mexico was taxonomically, molecularly and biochemically compared to S. obtusiusculus CCAP 276/25 (Culture Collection of Algae and Protozoa, Scotland, UK). Analysis of Internal Transcribed Spacer 2 (ITS2) secondary structures confirmed that the mexican strain belongs to S. obtusiusculus with one change in the ITS2 nucleotide sequence. However, both strains exhibited different biochemical and fatty acid profiles and therefore biotechnological potential, emphasizing the need for deeper studies among strains of the same species. Furthermore, the biochemical variations of S. obtusiusculus AT-UAM under nitrogen starvation and different levels of irradiance were evaluated. The maximum lipid production (1730 mg L-1) was obtained at 613 µmol m-2 s-1 while the highest carbohydrate content (49%) was achieved at 896 µmol m-2 s-1. Additionally, this strain was capable of storing lipids (∼52%) and carbohydrates (∼40%) under outdoor condition depending on the light availability in the cultivation broth.

The candidiasis caused by C. albicans is a public health problem. The abuse of antifungals has contributed to the development of resistance. B. morelensis has demonstrated antibacterial and antifungal activities. In this work the activity of the essential oil of B. morelensis was evaluated and for its two pure compounds with analysis of the different mechanisms of pathogenesis important for C. albicans. The essential oil was obtained by the hydro-distillation method and analyzed using GC-MS. The anti-Candida activity was compared between to essential oil, α-Pinene and γ-Terpinene. GC-MS of the essential oil demonstrated the presence of 13 compounds. The essential oil showed antifungal activity against four C. albicans strains. The most sensitive strain was C. albicans 14065 (MFC 2.0 mg/mL and MIC50 0.125 mg/mL) with α-Pinene and γ-Terpinene having MFCs of 4.0 and 16.0 mg/mL respectively. The essential oil inhibited the growth of the germ tube in 87.94% (8.0 mg/mL). Furthermore, it was observed that the essential oil diminishes the transcription of the gene INT1. This work provides evidence that confirms the anti-Candida activity of the B. morelensis essential oil and its effect on the growth of the germ tube and transcription of the gene INT1.

The zygomycete fungus Lichtheimia ramosa H71D, isolated from sugarcane bagasse compost, was identified by applying phylogenetic analysis based on the DNA sequence of the Internal Transcribed Spacer (ITS), and subsequent secondary structure analysis of ITS2. L. ramosa H71D was able to grow over a wide range of temperatures (25-45 °C), manifesting optimal growth at 37 °C. A 64 kDa xylanase (named LrXynA) was purified from the culture supernatant of L. ramosa H71D grown on 2% carboxymethylcellulose (CMC), as the only carbon source. LrXynA displayed optimal activity at pH 6 and temperature of 65 °C. The enzyme retained more than 50% of its maximal activity over a broad range of pH values (4.5-7.5). Enzyme half-life (t½) times at 55, 65 and 75 °C were 80, 25, and 8 min, respectively. LrXynA showed higher affinity (k M of 2.87 mg/mL) and catalytic efficiency (k cat /k M of 0.651 mg s/mL) towards Beechwood xylan in comparison to other substrates such as Birchwood xylan, Oat-spelt xylan, CMC, Avicel and Solka floc. The predominant final products from LrXynA-mediated hydrolysis of Beechwood xylan were xylobiose and xylotriose, suggesting that the enzyme is an endo-ß-1,4 xylanase. Scanning electron microscopy (SEM) imaging of sugar cane bagasse (SCB) treated with LrXynA, alone or in combination with commercial cellulases, showed a positive effect on the hydrolysis of SCB. To our knowledge, this is the first report focusing on the biochemical and functional characterization of an endo-ß-1,4 xylanase from the thermotolerant and fast-growing fungus Lichtheimia ramosa.

The genus Siderastrea exhibits high levels of morphological variability. Some of its species share similar morphological characteristics with congeners, making their identification difficult. Siderastrea stellata has been reported as an intermediary of S. siderea and S. radians in the Brazilian reef ecosystem. In an earlier study conducted in Mexico, we detected Siderastrea colonies with morphological features that were not consistent with some siderastreid species previously reported in the Gulf of Mexico. Thus, we performed a combined morphological and molecular analysis to identify Siderastrea species boundaries from the Gulf of Mexico. Some colonies presented high morphologic variability, with characteristics that corresponded to Siderastrea stellata. Molecular analysis, using the nuclear ITS and ITS2 region, corroborated the morphological results, revealing low genetic variability between S. radians and S. stellata. Since the ITS sequences did not distinguish between Siderastrea species, we used the ITS2 region to differentiate S. stellata from S. radians. This is the first report of Siderastrea stellata and its variability in the Gulf of Mexico that is supported by morphological and molecular analyses.

A hyperthermophilic and thermostable xylanase of 82 kDa (TtXynA) was purified from the culture supernatant of T. terrestris Co3Bag1, grown on carboxymethyl cellulose (CMC), and characterized biochemically. TtXynA showed optimal xylanolytic activity at pH 5.5 and at 85 °C, and retained more than 90% of its activity at a broad pH range (4.5-10). The enzyme is highly thermostable with a half-life of 23.1 days at 65 °C, and active in the presence of several metal ions. Circular dichroism spectra strongly suggest the enzyme gains secondary structures when temperature increases. TtXynA displayed higher substrate affinity and higher catalytic efficiency towards beechwood xylan than towards birchwood xylan, oat-spelt xylan, and CMC. According to its final hydrolysis products, TtXynA displays endo-/exo-activity, yielded xylobiose, an unknown oligosaccharide containing about five residues of xylose and a small amount of xylose on beechwood xylan. Finally, this report represents the description of the first fungal hyperthermophilic xylanase which is produced by T. terrestris Co3Bag1. Since TtXynA displays relevant biochemical properties, it may be a suitable candidate for biotechnological applications carried out at high temperatures, like the enzymatic pretreatment of plant biomass for the production of bioethanol.

Water effluents from urban and industrial areas carry large amounts of sediments and pollutants affecting corals populations, inducing bioerosion, increasing diseases and promoting the development of algae that compete for space with corals. In the Veracruz Reef System National Park (VRSNP) coral reefs are strongly affected by human activities carried out in the area. Gallega and Galleguilla reefs are among the most affected by wastewater discharges from the industrial (petrochemical and metallurgical) and urban areas in their vicinity. To assess the potential impact of this contamination on corals in the VRSNP, a chemical composition and morphology study of 76 Pseudodiploria colonies collected in reefs Gallega, Galleguilla, Isla Verde and Isla de Enmedio, was performed. Fragments of ~10 cm2 were collected and boric acid at 0.5 % was used to remove tissue from the skeleton; once clean, the morphology of each sample was determined with a scanning electron microscope (SEM). Subsequently, to test the chemical composition, an energy dispersion spectroscopy of X-ray chemical microanalysis (EDSX) was performed in the SEM. We found that corals from Gallega and Galleguilla reefs, located closer to human populations, presented high levels of tungsten and the skeleton exhibited multiple perforations. In contrast, corals from the farthest offshore reefs (Isla Verde and Isla de Enmedio) exhibited lower levels of tungsten and fewer perforations in their skeleton. These results demonstrated that anthropogenic activities in the NPVRS are affecting corals skeleton, highly damaging and promoting their bioerosion. The presence of traces of tungsten in the skeleton of corals is an evidence of the damage that waste discharges are causing to coral reefs. Discharges of large amounts of contaminants promoted the growth of harmful species that grow and develop into the corals skeleton, causing its bioerosion, and making them susceptible to disease and physical damage. This study is the first evidence of the effects of contamination on these species; therefore, further studies are necessary to determine the impact of pollution on their biology and survival. Rev. ResumenLos arrecifes de coral se encuentran bajo condiciones intensas de estrés causado por las actividades antropogénicas y el incremento de las poblaciones humanas en las zonas costeras. Las descargas de aguas de origen urbano e industrial transportan sedimentos y contaminantes que afectan a las poblaciones de corales, induciendo la bioerosion, el aumento de enfermedades en los corales y promueven el desarrollo de algas que compiten por espacio con los corales. En el Parque Nacional Sistema Arrecifal Veracruzano (NPVRS) los arrecifes de coral son afectados fuertemente por las actividades humanas que se llevan a cabo en la zona. Los arrecifes Gallega y Galleguilla son de los más afectados por las descargas de aguas residuales provenientes de la industria (petroquímica y metalúrgica) y de áreas urbanas que desembocan sus aguas en las proximidades de los arrecifes. Para evaluar el posible impacto de las descargas de aguas en los corales del NPVRS, se realizó un estudio de la composición química y morfología de 76 colonias de Pseudodiploria en los arrecifes Gallega, Galleguilla, Isla Verde e Isla de Enmedio. Se recolectaron fragmentos de ~10 cm2, el tejido del esqueleto fue removido utilizando ácido bórico al 0.5 %. Una vez limpia la muestra, la morfología fue analizada con un microscopio electrónico de barrido (SEM), posteriormente, para analizar la composición química de las muestras, realizamos una espectroscopia de dispersión de energía o micro-análisis químico de rayos X (EDSX) en el SEM. Encontramos que los corales de los arrecifes Gallega y Galleguilla que se encuentran ubicados cerca de poblaciones humanas, presentan altos niveles de tungsteno y el esqueleto exhibe múltiples agujeros. En contraste, los corales de los arrecifes más lejanos (Isla Verde e Isla de En medio) mostraron niveles más bajos de tungsteno y un menor número de agujeros en su esqueleto. Nuestros resultados demuestran que las actividades antropogénicas en el NPVRS, están afectando el esqueleto de los corales y promueven la bioerosión. Las descargas de grandes cantidades de contaminantes hacia las zonas costeras, promueven el crecimiento de especies dañinas que crecen y se desarrollan dentro del esqueleto de los corales, causando bioerosión del esqueleto, haciéndolos susceptibles a enfermedades y daños físicos. Debido a que este estudio es la primera evidencia de los efectos de la contaminación sobre esta especie de corales, son necesarios más estudios para determinar el impacto de la contaminación sobre su biología y la supervivencia de los corales.

El fenómeno de la ilusión de la mano falsa (IMF) se ha propuesto como un diseño que permite discriminar procesos perceptuales a partir de entradas sensoriales multimodales. Se han reportado varias modificaciones y variables que podrían afectar la generación del fenómeno. Este estudio se planteó bajo un diseño descriptivo-correlacional del fenómeno. Se realizó el procedimiento en jóvenes (16-48 años M = 20.86 años, DE = 4.4) colombianos para replicar el fenómeno de IMF. Los sujetos se dividieron al azar en dos grupos que diferían en el tipo de estimulación (sincrónica y desincronizada). La identificación de la IMF se realizó mediante la escala reportada en múltiples estudios, mostrando que en algunas variables como la estimulación se presentaba una diferencia en el reporte de IMF (P2. X² = 5.856: p < 0.05; P3. X² = 6.655: p <0.01 y P7(X² = 4.764: p < 0.05). Otras variables como sexo y dominancia manual mostraron algunas diferencias. La escala presenta una consistencia interna aceptable (α = 0.8). Los resultados de la IMF son similares a algunos estudios pero con puntuaciones menores que en los estudios iniciales, la diferencia en cuanto al género en tareas perceptuales podría explicar gran parte de los resultados.


The Rubber Hand Illusion (RHI) has been proposed as a design which allows to discriminate perceptual processes based on multimodal sensory inputs. Several modifications and variables have been reported that could affect the generation of the phenomenon. This study was conducted using a descriptive correlational design of this phenomenon. The procedure was conducted with young Colombian individuals (16-48 years-M = 20.86 years, SD = 4.4), seeking to replicate the RHI. The subjects were randomly divided into two groups that differed in type of stimulation (synchronised and unsynchronised). Identification of the RHI was performed using the scale reported in multiple studies, showing that the RHI report had differences in some variables such as stimulation (P2. X² = 5.856: p <.05; P3. X² = 6.655: p <.01 y P7 (X² = 4.764: p <.05). Other variables, such as gender and handedness, also showed some differences. The scale has an acceptable internal consistency ( X= 0.8). The results of the RHI are similar to some studies, but with lower scores than in the initial studies. The difference in gender in perceptual tasks could explain most of the results.

Two isoforms of a protease inhibitor were isolated by ion-exchange chromatography of tepary bean (Phaseolus acutifolius G.) seed proteins. The main isoform was used to determine the amino acid sequence of the protein. It is an 80 amino acid residue protein with a molecular mass of 8765 Da, showing sequence homology with the Bowman-Birk family of protease inhibitors. Several regions with amino acid microheterogeneity were found, corroborating the possible presence of isoforms. Mass spectrometry analysis was carried out to confirm isoforms. The presence of dimer and trimer forms of the inhibitor was shown through electrophoresis and mass spectrometry. Another unusual characteristic for this inhibitor was its ability to bind metals. The presence of four sequential histidines at the N-terminal end of the protein could account for this binding. Mass spectrometry and atomic absorption spectroscopy support the presence of calcium in the native inhibitor.
