A novel CO releasing material has been prepared via a one-pot synthesis-encapsulation strategy using the hierarchical metal organic framework [Zn2(dhtp)] (dhtp = 2,5-dihydroxyterephthalate) as a host of the photoactivable molybdenum tricarbonyl complex [Mo(CNCMe2CO2H)3(CO)3].

RESUMO

The novel non-conventional metallodrug [(η6-p-cymene)2Ru2mitoxantronato]Cl2 (1) exhibits redox activity, DNA intercalation ability, cathepsin B and D inhibition and in vitro antitumor activity able to circumvent cisplatin resistance. Moreover, 1 can be successfully incorporated into MIL100(Fe) as a proof of concept of the feasibility of metal-organic frameworks as carriers of non-conventional drugs.

RESUMO

Disruption of microRNA (miRNA) expression patterns is now being recognized as a hallmark of human cancer. The causes of these altered profiles are diverse, and, among them, we found the existence of defects in the miRNA processing machinery. However, little is known about how these alterations affect the biology of the underlying tumors. Herein, we show that colorectal cancer cells with an impairment in DICER1, a major miRNA biogenesis gene, undergo enrichment of tumor stemness features and an epithelial-to-mesenchymal transition. These phenotypes are associated with the downregulation of miRNAs, such as miR-34a, miR-126 and those of the miR-200 family, that target critical coding genes in these pathways. Most importantly, DICER1 impairment also induces the acquisition of a greater capacity for tumor initiation and metastasis, two properties associated with cancer stem cells.

OBJECTIVE: To analyse cases of radial head and neck fractures in children and compare them with the literature. METHOD: Retrospective and descriptive study of 21 children with radial head and neck fractures. The following parameters were collected: demographics, comorbidity, classification, treatment, need for rehabilitation, lack of range of motion (ROM), time for recovery and complications. RESULTS: The series included 11 males, and the mean age was 8.3 years. The right side was affected in 14 patients. Twelve cases had an associated ipsilateral elbow injury. According to the Chambers classification, 15 cases belonged to group A, while in the Steele-Graham classification, 12 cases were in group I. Eleven patients were treated with immobilization only, 4 percutaneously, and 6 by open reduction and internal fixation (ORIF). Eleven of them needed rehabilitation and despite this, 8 did not achieve full mobility. The mean time to obtain the greatest ROM was 4.71 months. Eight patients had complications, with the most common being neuroapraxia and valgus deformity of the elbow. DISCUSSION AND CONCLUSIONS: Treatment of paediatric radius head and neck fractures must be step-wise, from immobilization only, manual and/or percutaneous reduction, to ORIF, whichever is less indicated. In this respect, both the transcapital needle and/or removal the radius head should be avoided. The most common complication is lack of supination, especially in cases treated by ORIF. The posterior interosseous neuroapraxia was the most common of the rest of complications.

OBJECTIVE: To determine the bone mineral density (BMD) values in children and adolescents with moderate and severe infantile cerebral palsy (ICP) in our catchment area, and compare these values with a healthy population. MATERIAL AND METHOD: A prognostic study of cases and controls for the assessment of BMD in patients from 2 to 18 years old with infantile cerebral palsy belonging to the Gross Motor Function Classification System (GMFCS) Groups IV and V. The BMD measurements were performed at distal femur level, dividing this region into 3 areas following the forearm protocol. RESULTS: The BMD for each of the three areas studied results in the final sample of 69 patients were much lower than the reference levels. There was a statistically significant difference (P<.05) between the BMD values in the two sub-groups studied. DISCUSSION: The greater the involvement, from a neurological point of view, in patients classified as Group V shows a very low BMD compared to patients of similar sex and age. The acquisition of bone capital in patients with ICP does not follow the normal pattern of the healthy population.

The Kruppel-like factor (KLF) proteins are multitasked transcriptional regulators with an expanding tumor suppressor function. KLF2 is one of the prominent members of the family because of its diminished expression in malignancies and its growth-inhibitory, pro-apoptotic and anti-angiogenic roles. In this study, we show that epigenetic silencing of KLF2 occurs in cancer cells through direct transcriptional repression mediated by the Polycomb group protein Enhancer of Zeste Homolog 2 (EZH2). Binding of EZH2 to the 5'-end of KLF2 is also associated with a gain of trimethylated lysine 27 histone H3 and a depletion of phosphorylated serine 2 of RNA polymerase. Upon depletion of EZH2 by RNA interference, short hairpin RNA or use of the small molecule 3-Deazaneplanocin A, the expression of KLF2 was restored. The transfection of KLF2 in cells with EZH2-associated silencing showed a significant anti-tumoral effect, both in culture and in xenografted nude mice. In this last setting, KLF2 transfection was also associated with decreased dissemination and lower mortality rate. In EZH2-depleted cells, which characteristically have lower tumorigenicity, the induction of KLF2 depletion 'rescued' partially the oncogenic phenotype, suggesting that KLF2 repression has an important role in EZH2 oncogenesis. Most importantly, the translation of the described results to human primary samples demonstrated that patients with prostate or breast tumors with low levels of KLF2 and high expression of EZH2 had a shorter overall survival.

Silicon wafers have been silylated with VTMS (vinyltrimethoxysilane) and hydrolyzed. Subsequently, PVP (polyvinyl pyrrolidone) was grafted onto the silylated surface by two different techniques: the grafting-through (GT) and the grafting-onto techniques (GO). The measurement of contact angles along with the topography analysis by atomic force microscopy (AFM) has allowed monitoring the different stages of the process and the temporal evolution of polymer grafting. The results have demonstrated the feasibility of both methods of grafting but have shown that the GT method gives a higher density of polymer-grafted chains. The AFM technique in adequate liquid environments has been proven to permit the surface density of chains to be distinguished by both methods and to estimate the length of the resulting PVP chains.

The shortcomings of the current formulation for calculating the adhesion force for drops and bubbles with noncircular contact lines are discussed. A general formulation to evaluate the adhesion force due to surface forces is presented. Also, a novel methodology, that is, IBAFA, image based adhesion force analysis, was developed to allow implementation of the general formulation. IBAFA is based on the use of multiple profile images of a drop. The images are analyzed (1) to accurately reconstruct the contact line shape, which is analytically represented by a Fourier cosine series, and (2) to measure contact angles at multiple locations along the contact line and determine the contact angle distribution based on a linear piecewise interpolation routine. The contact line shape reconstruction procedure was validated with both actual experiments and simulated experiments. The procedure for the evaluation of the adhesion force was tested using simulated experiments with synthetic drops of known shapes. A comparison with current methods showed that simplifying assumptions (e.g., elliptical contact line or linear contact angle distribution) used in these methods result in errors up to 76% in the estimated adhesion force. However, the drop adhesion force evaluated using IBAFA results in small errors on the order of 1%.

RESUMO

Obturator internus muscle abscess (pyomyositis) is extremely rare in temperate environments, although there has been a recent reported increase in prevalence. There usually is a delay in diagnosis and onset of treatment. In addition to our case, we reviewed all cases of pyomyositis of hip muscles found in a review of the recent medical literature.

PURPOSE: To evaluate the postoperative astigmatism and complications after manual multi-phacofragmentation (MPF) cataract surgery through a 3.2 mm clear corneal incision (CCI). SETTING: Department of Ophthalmology, Hospital General de Segovia, Segovia, Spain. METHODS: In a retrospective study, 50 eyes of 50 patients had cataract surgery by MPF. Patients were divided into 3 groups according to type of cataract. This technique uses a racquet-shaped nucleotome, a spatula, and 2 manipulators, right and left, for nuclear fragments. With these instruments, the nucleus is fragmented into multiple small pieces that are extracted from the anterior chamber. Astigmatism was evaluated before surgery and 7 days and 1 and 3 months after surgery by Gaussian distribution (P <.05). Intraoperative and postoperative complications were recorded. RESULTS: Mean postoperative astigmatism was 0.77 diopters (D) +/- 0. 29 (SD) at 7 days, 0.47 +/- 0.22 D at 1 month, and 0.21 +/- 0.22 D at 3 months. During surgery, 2 cases (4%) of slight transient intracameral bleeding in the anterior chamber occurred. After surgery, 5 eyes (10%) had corneal edema, 2 (4%) had iritis, and 3 (6%) had ocular hypertension. CONCLUSION: Using MPF, soft and hard nuclei can be removed through a 3.2 mm CCI. The astigmatic results were favorable and the complications few.
