RESUMO

The antibacterial and antiviral functions of human defensin 5 lay the foundation for its role as a core host protective component. In addition, HD5 also has the function of inhibiting tumor proliferation and immune regulation. However, everything has two sides; cytotoxic and proinflammatory properties may exist, while HD5 performs physiological functions. Accordingly, the modification and engineering of HD5 are particularly important. Therefore, this review summarizes the role of HD5 in various aspects of host defense, as well as modification of HD5 to ameliorate the biological activity, with a view to promoting the clinical use of HD5.

Diflubenzuron is widely used as a benzoylurea insecticide, and its impact on human health should not be underestimated. Therefore, the detection of its residues in food and the environment is crucial. In this paper, octahedral Cu-BTB was fabricated using a simple hydrothermal method. It served as a precursor for synthesizing Cu/Cu2O/CuO@C with a core-shell structure through annealing, creating an electrochemical sensor for the detection of diflubenzuron. The response of Cu/Cu2O/CuO@C/GCE, expressed as ΔI/I0 exhibited a linear correlation with the logarithm of the diflubenzuron concentration ranging from 1.0 × 10-4 to 1.0 × 10-12 mol·L-1. The limit of detection (LOD) was determined to be 130 fM using differential pulse voltammetry (DPV). The electrochemical sensor demonstrated excellent stability, reproducibility, and anti-interference properties. Moreover, Cu/Cu2O/CuO@C/GCE was successfully employed to quantitatively determine diflubenzuron in actual food samples (tomato and cucumber) and environmental samples (Songhua River water, tap water, and local soil) with good recoveries. Finally, the possible mechanism of Cu/Cu2O/CuO@C/GCE for monitoring diflubenzuron was thoroughly investigated.

Vibrio mimicus (V. mimicus) is a pathogen causing serious vibriosis in aquatic animals. Hepcidin and ß-Defensin1 are two important antibacterial peptides (AMPs) with broad-spectrum antibacterial activity in fish. In mammals, some evidences demonstrated that interleukin-1ß (IL-1ß) primarily promote AMPs expression via activating classical NF-κB pathway, but it still remains unclear in fish. Here, the temporal and spatial expression patterns of grass carp IL-1ß (gcIL-1ß) gene and two AMPs genes (gchepcidin and gcß-defensin1) in tissues post-V. mimicus infection and anti-V. mimicus activity of these two AMPs in vitro were detected, showing that V. mimicus infection significantly elevated the mRNA levels of these three genes in the immune-related tissues although their expression patterns were not entirely consistent, and both gcHepcidin and gcß-Defensin1 possessed anti-V. mimicus activity in vitro. Subsequently, the recombinant gcIL-1ß (rgcIL-1ß) was expressed prokaryotically in an inclusion body, which could promote proliferation of grass carp head kidney leukocytes (gcHKLs) and enhance respiratory burst activity and phagocytic activity of head kidney macrophages. Stimulation with rgcIL-1ß was able to significantly regulate the mRNA expression of key regulatory genes (il-1RI, traf6, tak1, ikkß, iκBα and p65) involved in the activation of classical NF-κB pathway, and then induce gcTAK1 phosphorylation, promote gcp65 nuclear translocation and enhance endogenous gcIL-1ß expression at both mRNA and protein levels, implying NF-κB pathway was activated. More importantly, exogenous rgcIL-1ß stimulation also significantly up-regulated both gcHepcidin and gcß-Defensin1 mRNA levels against V. mimicus, and the regulatory effect was blocked or inhibited by NF-κB inhibitor PDTC. Taken together, our results demonstrated for the first time that grass carp IL-1ß stimulation could significantly enhance the expression of these two anti-V.mimicus AMPs via activating classical NF-κB pathway.

BACKGROUND: Intestinal microbiota is a novel drug target of metabolic diseases, especially for those with poor oral bioavailability. Nuciferine, with poor bioavailability, has an anti-hyperlipidemic effect at low dosages. PURPOSE: In the present study, we aimed to explore the role of intestinal microbiota in the anti-hyperlipidemic function of nuciferine and identify the key bacterial targets that might confer the therapeutic actions. METHODS: The contribution of gut microbes in the anti-hyperlipidemic effect of nuciferine was evaluated by conventional and antibiotic-established pseudo-sterile mice. Whole-metagenome shotgun sequencing was used to characterize the changes in microbial communities by various agents. RESULTS: Nuciferine exhibited potent anti-hyperlipidemic and liver steatosis-alleviating effects at the doses of 7.5-30 mg/kg. The beneficial effects of nuciferine were substantially abolished when combined with antibiotics. Metagenomic analysis showed that nuciferine significantly shifted the microbial structure, and the enrichment of Akkermansia muciniphila was closely related to the therapeutic effect of nuciferine. CONCLUSIONS: Our results revealed that gut microbiota played an essential role in the anti-hyperlipidemic effect of nuciferine, and enrichment of Akkermansia muciniphila represented a key mechanism through which nuciferine exerted its therapeutic effects.
