RESUMO

A method using label-free surface enhanced Raman spectroscopy (SERS) based on substrate design is provided for an early detection and differentiation of spike glycoprotein mutation sites in live SARS-CoV-2 variants. Two SERS-active substrates, Au nanocavities (Au NCs) and Au NPs on porous ZrO2 (Au NPs/pZrO2), were used to identify specific peaks of A.3, Alpha, and Delta variants at different concentrations and demonstrated the ability to provide their SERS spectra with detection limits of 0.1-1.0% (or 104-5 copies/mL). Variant identification can be achieved by cross-examining reference spectra and analyzing the substrate-analyte relationship between the suitability of the analyte upon the hotspot(s) formed at high concentrations and the effective detection distance at low concentrations. Mutation sites on the S1 chain of the spike glycoprotein for each variant may be related and distinguishable. This method does not require sample preprocessing and therefore allows for fast screening, which is of high value for more comprehensive and specific studies to distinguish upcoming variants.

Smartphone equipment has promoted the widespread use of new media communication, and users have changed from being passive to actively receiving information. This has changed people's lifestyles, and has enriched the convenience and entertainment value of knowledge acquisition. However, the new media communication systems can be too complex, and the interface design for users to interact with the new media may not be sufficiently intuitive, which causes interface usability problems. Therefore, this research study focuses on the concept of affordance and the impact of user perception pertinent to smartphone applications on frequency modulation (FM) new media interface design. The experiment is a between-subjects design using one-way ANOVA to examine three different operation modes, namely the Litchi, Himalayas and Archimedes types. The experimental data were obtained through task performance and subjective evaluation. The results indicate that: (1) Visual information presentation methods, such as viewing and deleting, affect user perception. The three operating modes revealed significant differences, with the Himalayas type taking the least amount of task performance time. (2) There were significant differences among the different operation modes, with the Himalayas type the best in terms of the users' subjective evaluation. (3) The overall analysis of task performance and satisfaction consistently showed that the Himalayas type was better than the Litchi and Archimedes types in all aspects. (4) Smartphone user interface applications provide users with cognitive support, objects, functions, and sensory affordance, which enhance the user's interactive experience of FM new media.

This study examined how users acquire spatial knowledge in an onscreen three-dimensional virtual environment when using overview maps. This experiment adopted a three (the size of overview maps) x two (the transparency of overview maps) between-subjects design. Three levels of the size of overview maps were evaluated, i.e., 1/2, 1/8, and 1/16 screen size. Comparisons between 20% transparent and 80% transparent were made. We asked 108 participants to complete spatial perception tasks and fill out questionnaires regarding their feelings. The results indicate the following: (1) The effects of the transparency of overview maps on users' spatial perception vary with the size of overview maps. The 80% transparent overview map is significantly more efficient than the 20% transparent overview map in the condition of 1/2 screen size. However, the result is opposite in the condition of 1/8 screen size. (2) Users like the 80% transparent overview map significantly better than the 20% transparent overview map in the condition of 1/2 screen size. (3) Concerning subjective evaluations of satisfaction, preference, and system usability, overview maps in the condition of 1/8 screen size are significantly better than those in the condition of 1/2 screen size.

UNLABELLED: Understanding the underlying molecular mechanisms in burn wound progression is crucial to providing appropriate diagnoses and designing therapeutic regimens for burn patients. When inflammation becomes unregulated, recurrent, or excessive, it interferes with burn wound healing. Autophagy, which is a homeostatic and catabolic degradation process, was found to protect against ischemic injury, inflammatory diseases, and apoptosis in some cases. In the present study, we investigated whether far-infrared (FIR) could ameliorate burn wound progression and promote wound healing both in vitro and in a rat model of deep second-degree burn. We found that FIR induced autophagy in differentiated THP-1 cells (human monocytic cells differentiated to macrophages). Furthermore, FIR inhibited both the NLRP3 inflammasome and the production of IL-1ß in lipopolysaccharide-activated THP-1 macrophages. In addition, FIR induced the ubiquitination of ASC, which is the adaptor protein of the inflammasome, by increasing tumor necrosis factor receptor-associated factor 6 (TRAF6), which is a ubiquitin E3 ligase. Furthermore, the exposure to FIR then promoted the delivery of inflammasome to autophagosomes for degradation. In a rat burn model, FIR ameliorated burn-induced epidermal thickening, inflammatory cell infiltration, and loss of distinct collagen fibers. Moreover, FIR enhanced autophagy and suppressed the activity of the NLRP3 inflammasome in the rat skin tissue of the burn model. Based on these results, we suggest that FIR-regulated autophagy and inflammasomes will be important for the discovery of novel therapeutics to promote the healing of burn wounds. KEY MESSAGES: Far-infrared (FIR) induced autophagy in THP-1 macrophages. FIR suppressed the NLRP3 inflammasome through the activation of autophagy. FIR induced the ubiquitination of ASC by increasing TRAF6. FIR ameliorated burn wound progression and promoted wound healing in a rat burn model.

This study investigates the effects of picture amount on subjective evaluation. The experiment herein adopted two variables to define picture amount: column ratio and picture size. Six column ratios were employed: 7:93,15:85, 24:76, 33:67, 41:59, and 50:50. Five picture sizes were examined: 140 x 81, 220 x 127, 300 x 173, 380 x 219, and 460 x 266 pixels. The experiment implemented a within-subject design; 104 participants were asked to evaluate 30 web page layouts. Repeated measurements revealed that the column ratio and picture size have significant effects on preference, balance, and dynamic feel. The results indicated the most appropriate picture amount for display: column ratios of 15:85 and 24:76, and picture sizes of 220 x 127, 300 x 173, and 380 x 219. The research findings can serve as the basis for the application of design guidelines for future web page interface design.

Automatic dynamic displays, e.g., scrolling displays, are frequently used to present text information on small screens. This study examined the effects of three dynamic displays [leading, scrolling, and rapid serial visual presentation (RSVP)] and three presentations speeds [171, 250, and 305 wpm (words per minute)] on subjects' reading comprehension for different types of small screens for laptops, personal digital assistants (PDAs), and mobile phones. 12 college students who were native speakers of Chinese (4 men and 8 women between the ages of 19 and 36 years) participated. Scores for reading comprehension indicated (1) for laptops, both leading displays and RSVP, at speeds of about 250 wpm, comprehension was highest; (2) for PDAs and mobile phones, scrolling displays and RSVP, speeds of about 305 wpm were more suitable for presenting information in Chinese text; (3) consequently, RSVP appeared to be the most viable dynamic display on small screens and led to the highest reading comprehension for these Chinese readers.
