RESUMO

After spinal cord injury, astrocytes undergo a reactive process and form an astroglial scar, which impedes the regeneration of axons. The role of Runx2 in promoting the transformation of astrocytes in the central nervous system is well-established. However, it remains unclear whether Runx2 also plays a role in the development of astroglial scar, and the precise underlying mechanism has yet to be identified. Recently, our study using cell culture and animal models has demonstrated that Runx2 actually suppresses astrocyte activation and the formation of astroglial scar following injury. The initial results demonstrated an increase in the expression of Runx2 in astrocytes following in vivo injury. Subsequently, the overexpression of Runx2 resulted in the inhibition of astrocyte activation, reduction in the total area of astroglial scar, and restoration of neural function after 14 days of injury. However, these effects were reversed by CADD522. These findings indicate that Runx2 could potentially serve as a therapeutic intervention for spinal cord injury (SCI). Furthermore, our findings suggest that the Nuclear-matrix-targeting signal (NMTS) of Runx2 is associated with its effect. In summary, the study's results propose that targeting Runx2 may be a promising treatment approach for reactive astrocytes and astroglial scar in the recovery of SCI.

RESUMO

Photothermal techniques are infrared remote sensing techniques that have been used for biomedical applications, as well as industrial non-destructive testing (NDT). Machine learning is a branch of artificial intelligence, which includes a set of algorithms for learning from past data and analyzing new data, without being explicitly programmed to do so. In this paper, we first review the latest development of machine learning and its applications in photothermal techniques. Next, we present our latest work on machine learning for data analysis in opto-thermal transient emission radiometry (OTTER), which is a type of photothermal technique that has been extensively used in skin hydration, skin hydration depth profiles, skin pigments, as well as topically applied substances and skin penetration measurements. We have investigated different algorithms, such as random forest regression, gradient boosting regression, support vector machine (SVM) regression, and partial least squares regression, as well as deep learning neural network regression. We first introduce the theoretical background, then illustrate its applications with experimental results.

Objectives: This study aimed to explore the relationship between red blood cell distribution width (RDW) and all-cause mortality in critically ill patients undergoing intra-aortic balloon pumping (IABP) in the intensive care unit (ICU). Methods: This study retrospectively analyzed data from the Medical Information Mart for Intensive Care IV (MIMIC-IV) database. The primary endpoint was the 30-day mortality rate, while the secondary endpoint was the in-hospital mortality rate. Restricted cubic splines were used to assess the dose-response relationship. The receiver operating characteristic (ROC) curve and Kaplan-Meier curve analysis were carried out to evaluate the predictive performance of RDW. Moreover, multiple logistic regression analyses and subgroup analyses were conducted to investigate the relationship between RDW and 30-day mortality. Finally, propensity score matching (PSM) was performed to adjust for the imbalance of covariates. Results: In total, 732 patients were finally identified from the MIMIC-IV database in this study. The RDW of patients in the non-survivor group was significantly higher compared with those in the survivor group (P < 0.01). Multiple logistic regression analyses corroborated RDW was an independent predictor of all-cause 30-day mortality in critically ill patients post-IABP. Meanwhile, ROC analysis identified an RDW cutoff of 14.2%. High RDW patients exhibited a 131% (OR = 2.31, 95% CI: 1.49-3.61) elevated risk of 30-day mortality after adjusting for confounders in multivariable logistic regression. After PSM, 412 patients were included in the matched cohort. In the original and matched cohorts, the high RDW group had higher 30-day and in-hospital mortality rates, as well as longer ICU stays. Lastly, the area under the ROC curve for 30-day mortality was 0.686, with an optimal cutoff point of 14.2 for RDW (sensitivity: 69.09 % and specificity: 63.32%). Conclusion: RDW could be a simple and valuable prognostic tool to predict mortality in critically ill patients after IABP.

The content of gallic acid (GA) is positively correlated with the quality grade of tea. Here, we developed a colorimetric method based on raspberry-like N-doped Mn3O4 nanospheres (N-Mn3O4 NSs) with oxidase-like activity for GA assay. Modulating the electronic structure of Mn3O4 by N doping could promote the catalysis ability, and the produced oxygen vacancies (OVs) can provide high surface energy and abundant active sites. The N-Mn3O4 NSs presented low Michaelis-Menten constant (Km) of 0.142 mM and maximum initial velocity (Vmax) of 9.8 × 10-6 M s-1. The sensor exhibited excellent analytical performance towards GA detection, including low LOD (0.028 µM) and promising linear range (5 âˆ¼ 30 µM). It is attributed that OVs and O2- participated in TMB oxidation. Based on the reaction color changes, a visualized semi-quantitative GA detection could be realized via a smartphone-based system. It could be applied for evaluating GA quality in market-purchased black tea and green tea.

We reported the anti-cervical cancer effect of proprietary saponin content from seeds of Impatiens balsamina L., Hosenkoside A. Our study found that Hosenkoside A significantly promotes cell apoptosis and cell cycle arrest after administration, exhibiting anti-tumor effects. Then the transcriptome sequencing results after administration showed that Hosenkoside A had a significant inhibitory effect on Histone deacetylase 3 (HDAC3). After sufficient administration time, the inhibition of HDAC3 expression level leads to a significant decrease in lysine acetylation at histone 3 sites 4 and 9, blocking the activation of Signal transducer and activator of transcription 3 (STAT3) and achieving anti-tumor effects. In addition, we encapsulated Hosenkoside A into polypeptide metal complexes (PMC) to form slow-release spheres. This material breaks down in the tumor environment, not only does it solve the problem of low drug solubility, but it also achieves targeted sustained-release drug delivery. Under the same concentration of stimulation, the PMC complex group showed better anti-tumor effects in both in vitro and in vivo experiments.
