OBJECTIVES: As a serious complication of moderately severe acute pancreatitis (MSAP) and severe acute pancreatitis (SAP), infected pancreatic necrosis (IPN) can lead to a prolonged course of interventional therapy. Most predictive models designed to identify such patients are complex or lack validation. The aim of this study was to develop a predictive model for the early detection of IPN in MSAP and SAP. METHODS: A total of 594 patients with MSAP or SAP were included in the study. To reduce dimensionality, least absolute shrinkage and selection operator regression analysis was used to screen potential predictive variables, a nomogram was then constructed using logistic regression analysis. The receiver operating characteristic (ROC) curve, calibration curve, and decision curve analysis (DCA) were used to evaluate the discrimination, accuracy, and clinical efficacy of the model. External data were also obtained to further validate the constructed model. RESULTS: There were 476, 118, and 82 patients in the training, internal validation, and external validation cohorts, respectively. Platelet count, hematocrit, albumin/globulin, severity of acute pancreatitis, and modified computed tomography severity index score were independent factors for predicting IPN in MSAP and SAP. The area under the ROC curves were 0.923, 0.940, and 0.817, respectively, in the three groups. There was a good consistency between the actual probabilities and the predicted probabilities. DCA revealed excellent clinical utility. CONCLUSION: The constructed nomogram is a simple and feasible model that has good clinical predictive value and efficacy in clinical decision-making for IPN in MSAP and SAP.

OBJECTIVE: Fatty acids play a critical role in the proper functioning of the brain. This study investigated the effects of a high-fat (HF) diet on brain fatty acid profiles of offspring exposed to maternal gestational diabetes mellitus (GDM). METHODS: Insulin receptor antagonist (S961) and HF diet were used to establish the GDM animal model. Brain fatty acid profiles of the offspring mice were measured by gas chromatography at weaning and adulthood. Protein expressions of the fatty acid transport pathway Wnt3/ß-catenin and the target protein major facilitator superfamily domain-containing 2a (MFSD2a) were measured in the offspring brain by Western blot. RESULTS: Maternal GDM increased the body weight of male offspring (P < 0.05). In weaning offspring, factorial analysis showed that maternal GDM increased the monounsaturated fatty acid (MUFA) percentage of the weaning offspring's brain (P < 0.05). Maternal GDM decreased offspring brain arachidonic acid (AA), but HF diet increased brain linoleic acid (LA) (P < 0.05). Maternal GDM and HF diet reduced offspring brain docosahexaenoic acid (DHA), and the male offspring had higher DHA than the female offspring (P < 0.05). In adult offspring, factorial analysis showed that HF diet increased brain MUFA in offspring, and male offspring had higher brain MUFA than female offspring (P < 0.05). The HF diet increased brain LA in the offspring. Male offspring had higher level of AA than female offspring (P < 0.05). HF diet reduced DHA in the brains of female offspring. The brain protein expression of ß-catenin and MFSD2a in both weaning and adult female offspring was lower in the HF + GDM group than in the CON group (P < 0.05). CONCLUSIONS: Maternal GDM increased the susceptibility of male offspring to HF diet-induced obesity. HF diet-induced adverse brain fatty acid profiles in both male and female offspring exposed to GDM.

A preliminary study was conducted of the chemical, structural properties and immunomodulatory activities of fucoidan isolated from Sargassum Zhangii (SZ). Sargassum Zhangii fucoidan (SZF) was determined to have a sulfate content of 19.74 ± 0.01% (w/w) and an average molecular weight of 111.28 kDa. SZF possessed a backbone structure of (1,4)-α-d-linked-galactose, (3,4)-α-l-fucose, (1,3)-α-d-linked-xylose, ß-d-linked-mannose and a terminal (1,4)-α-d-linked-glucose. The main monosaccharide composition was determined as (w/w) 36.10% galactose, 20.13% fucose, 8.86% xylose, 7.36% glucose, 5.62% mannose, and 18.07% uronic acids, respectively. An immunostimulatory assay showed that SZF, compared to commercial fucoidans (Undaria pitnnaifida and Fucus vesiculosus sources), significantly elevated nitric oxide production via up-regulation of cyclooxygenase-2 and inducible nitric oxide synthase at both gene and protein levels. These results suggest that SZ has the potential to be a source of fucoidan with enhanced properties that may act as a useful ingredient for functional foods, nutritional supplements, and immune enhancers.

Nicotiana alata Link et Otto, widely used in landscaping, is not only of great ornamental value but also of high commercial and medical value. The global potential habitat of N. alata and the environmental factors affecting its distribution are not that clear at present. To provide a reference for the reasonable and extensive planting of N. alata now and in the future, the MaxEnt model was used to predict its global suitable habitats under current and future climate conditions, respectively, based on global geographic distribution data of N. alata and the current and future world bioclimatic variables. The results showed that mean temperature of the driest quarter (bio9), precipitation of driest month (bio14), precipitation seasonality (bio15) and max temperature of warmest month (bio5), were the key bioclimatic variables governing the distribution of N. alata. The global suitable habitats of N. alata were mainly distributed in Europe, the United States, southeastern South America, and China under current climate conditions. Compared with current climate conditions, the future climate decreased suitable habitats of N. alata under SSP1-2.6, and SSP2-4.5 scenario and increased suitable habitats of N. alata under SSP3-7.0, and SSP5-8.5 climatic scenarios. The results provided valuable information and theoretical reference for the reasonable planting of N. alata.

Fucoidan is a sulfated algal polyanionic polysaccharide that possesses many biological activities. In this paper, a fucoidan (SHF) polysaccharide was extracted from Sargassum hemiphyllum collected in the South China Sea. The SHF, with a molecular weight of 1166.48 kDa (44.06%, w/w), consisted of glucose (32.68%, w/w), galactose (24.81%, w/w), fucose (20.75%, w/w), xylose (6.98%, w/w), mannose (2.76%, w/w), other neutral monosaccharides, and three uronic acids, including glucuronic acid (5.39%, w/w), mannuronic acid (1.76%, w/w), and guronuronic acid (1.76%, w/w). The SHF exhibited excellent immunostimulatory activity. An immunostimulating assay showed that SHF could significantly increase NO secretion in macrophage RAW 264.7 cells via upregulation of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) levels based on both gene expression and protein abundance. These results suggest that SHF isolated from Sargassum hemiphyllum has great potential to act as a health-boosting ingredient in the pharmaceutical and functional-food fields.

The sediment microenvironment has an important effect on the release of endogenous phosphorus. In this study, the influence of two different in-situ covering materials on the sediment microenvironment were compared, and the controlling effect of endogenous phosphorus release were studied. The sediment microenvironment was represented by the concentration of NH4+-N, Fe2+ in interstitial water, and microbial activity. The results showed that the concentrations of NH4+-N and Fe2+ were lower under ACPM coverage than those of the Phoslock® group, and the microbial activity was higher, indicating that the redox potential of ACPM coverage was higher than that of Phoslock®. Furthermore, the bottom sediment microenvironment was in an aerobic state. Compared with the Phoslock® group, the DIP concentrations in both the overcover water and porewater in the ACPM group were higher, indicating that Phoslock® was superior to ACPM with respect to the phosphate adsorption capacity, suggesting that the sediment microenvironment was not the only criterion for phosphorus adsorption. In the fixation process of endogenous phosphorus, both of the covering materials were conducive to the fixation of inter phosphorus, and Ca-P increased significantly. On the other hand, oxidizing ACPM led to an increase in NH4Cl-P and a significant decrease in Fe/Al-P. This promotes the release of active phosphorus, which is conducive to the cleaning of the sediment and phosphorus release.

Adding biochar from harvested reed to sediments is a new method to control the release of sedimentary phosphorus. Three types of Mg-modified biochars were prepared by pyrolysis of reed modified by magnesium chloride at 300, 450, and 600℃.The phosphate adsorption characteristics of the three types of biochars were analyzed by isothermal adsorption experiments. Biochar MBC-450 with good phosphate adsorption effects was selected as the material for the following experiments. Taking the sediment and overlying water of a campus river as the experimental object, we studied the adsorption of phosphate in overlying water and the control of sedimentary phosphorus by Mg-modified biochar under different dosage modes (mixing and covering). The concentration of DIP in the overlying water could be effectively reduced by biochar mixing and covering, and the accumulative phosphorus adsorption capacity increased by 17.3% (mixing) and 11.7% (covering) compared with that of the control. The control effect of sedimentary phosphorus by biochar mixing was more obvious; the DIP in sedimentary water decreased by 14.7%, 18.9%, and 35.36% from 0-2 cm to 4-6 cm compared with the control. The DIP in sedimentary water decreased by 33.3%, -28.2%, and 12.9% when covered with biochar. Compared with the control, the proportion of NH4 Cl-P in the sediment of 0-2 cm and 2-4 cm increased by 15% and 15% (mixing) and 12% and 2% (covering), respectively, while BD-P in TP decreased by 7% and 9% (mixing) and 6% and 3% (covering), respectively, and the Al-P in TP decreased by 7% and 6% (mixing) and 7% and -1% (covering), respectively. The other forms of phosphorus did not change significantly. Biochar mixing and covering can both improve the microbial activity in surface sediment, and biochar mixing can improve the microbial activity in deeper sediments more significantly.
