RESUMO

The AT-hook motif nuclear localized (AHL) gene family is a highly conserved transcription factors involved in plant growth, development, and stress responses. However, AHLs have not been systematically analyzed in radish (Raphanus sativus). Therefore, we performed genome-wide identification and expression pattern, gene structure, and function verifications of radish AHLs. We identified 52 radish AHLs (RsAHL1-RsAHL52), which were unevenly distributed across nine chromosomes. Phylogenetic analysis showed that the RsAHLs were divided into two clades (A and B) and subdivided into three types (I, II, and III). Collinearity analysis revealed that the 52 RsAHLs produced 49 repeat events. Tissue expression profiles revealed differential expression of RsAHLs across different tissues, with higher expression observed in flower organs, particularly petals and anthers. qRT-PCR results indicated that RsAHLs responded to abscisic acid, methyl jasmonate, and abiotic stress (low and high temperatures and drought). Additionally, RsAHL14 induced a dwarf phenotype in tomato plants, and RsAHL14-overexpression tomato plants presented significantly decreased expression levels of the gibberellin (GA) synthetic genes ent-Copalyl diphosphatase, GA3ox-3/-4/-5, and GA20ox-1/-2/-3, but significantly increased expression of the degradation gene GA2ox-1/-3. Thus, RsAHL14 might affect plant growth by regulating GA content. Collectively, our study comprehensively identified RsAHLs in radish and provided a reference for further research on these genes.

RESUMO

Rapid social-economic development introduces modern lifestyles into rural areas, not only bringing numerous modern products but also new pollutants, such as chlorinated paraffins (CPs). The rural Tibetan Plateau has limited industrial activities and is a unique place to investigate this issue. Herein we collected 90 free-range chicken egg pool samples across the rural Tibetan Plateau to evaluate the pollution status of CPs. Meanwhile, CPs in related soils, free-range chicken eggs from Jiangxi, and farmed eggs from markets were also analyzed. The median concentrations of SCCPs (159 ng g-1 wet weight (ww)) and MCCPs (1390 ng g-1 ww) in Tibetan free-range chicken eggs were comparable to those from Jiangxi (259 and 938 ng g-1 ww) and significantly higher than those in farmed eggs (22.0 and 81.7 ng g-1 ww). In the rural Tibetan Plateau, the median EDI of CPs via egg consumption by adults and children were estimated to be 81.6 and 220.2 ng kg-1 bw day-1 for SCCPs and 483.4 and 1291 ng kg-1 bw day-1 for MCCPs, respectively. MCCPs might pose potential health risks for both adults and children in the worst scenario. Our study demonstrates that new pollutants should not be ignored and need further attention in remote rural areas.

RESUMO

The issue of hypersonic boundary layer transition prediction is a critical aerodynamic concern that must be addressed during the aerodynamic design process of high-speed vehicles. In this context, we propose an advanced mesoscopic method that couples the gas kinetic scheme (GKS) with the Langtry-Menter transition model, including its three high-speed modification methods, tailored for accurate predictions of high-speed transition flows. The new method incorporates the turbulent kinetic energy term into the Maxwellian velocity distribution function, and it couples the effects of high-speed modifications on turbulent kinetic energy within the computational framework of the GKS solver. This integration elevates both the transition model and its high-speed enhancements to the mesoscopic level, enhancing the method's predictive capability. The GKS-coupled mesoscopic method is validated through a series of test cases, including supersonic flat plate simulation, multiple hypersonic cone cases, the Hypersonic International Flight Research Experimentation (HIFiRE)-1 flight test, and the HIFiRE-5 case. The computational results obtained from these cases exhibit favorable agreement with experimental data. In comparison with the conventional Godunov method, the new approach encompasses a broader range of physical mechanisms, yielding computational results that closely align with the true physical phenomena and marking a notable elevation in computational fidelity and accuracy. This innovative method potentially satisfies the compelling demand for developing a precise and rapid method for predicting hypersonic boundary layer transition, which can be readily used in engineering applications.

RESUMO

Oceanic submesoscale processes are ubiquitous in the North Pacific Subtropical Gyre (NPSG), where the biological carbon pump is generally ineffective. Due to difficulties in collecting continuous observations, however, it remains uncertain whether episodic submesoscale processes can drive significant changes in particulate organic carbon (POC) export into the mesopelagic ocean. Here we present observations from high-frequency Biogeochemical-Argo floats in the NPSG, which captured the enhanced POC export fluxes during the intensifying stages of a submesoscale front and a cyclonic eddy compared to their other life stages. A higher percentage of POC export flux was found to be transferred to the base of mesopelagic layer at the front compared to that at the intensifying eddy and the mean of previous studies (37% vs. ~10%), suggesting that the POC export efficiency was significantly strengthened by submesoscale dynamics. Such findings highlight the importance of submesoscale fronts for carbon export and sequestration in subtropical gyres.

RESUMO

Short chain chlorinated paraffins (SCCPs) are ubiquitous persistent organic pollutants. They have been widely detected in plant-based foods and might cause adverse impacts on humans. Nevertheless, uptake and accumulation mechanisms of SCCPs in plants remain unclear. In this study, the soil culture data indicated that SCCPs were strongly absorbed by roots (root concentration factor, RCF>1) yet limited translocated to shoots (translocation factor<1). The uptake mechanism was explored by hydroponic exposure, showing that hydrophobicity and molecular size influenced the root uptake and translocation of SCCPs. RCFs were significantly correlated with logKow values and molecular weights in a parabolic curve relationship. Besides, it was extremely difficult for SCCPs to translocate from shoots back to roots via phloem. An active energy-dependent process was proposed to be involved in the root uptake of SCCPs, which was supported by the uptake inhibition by the low temperature and metabolic inhibitor. Though SCCPs at environmentally relevant concentrations had no negative impacts on root morphology and chlorophyll contents, it caused obvious changes in cellular ultrastructure of root tip cells and induced a significant increase in superoxide dismutase activity. This information may be beneficial to moderate crop contamination by SCCPs, and to remedy soils polluted by SCCPs with plants.

RESUMO

Quantum dots (QDs) are widely applied and inevitably released into the environment. The biotransformation of Se in typical CdSe/ZnS QDs coated with glutathione (CdSe/ZnS-GSH) to volatile alkyl selenides and the fate of alkyl selenides in the hydroponically grown rice system were investigated herein. After a 10-day exposure to CdSe/ZnS-GSH (100 nmol L-1), seven alkyl selenides, dimethyl selenide (DMSe), dimethyl diselenide (DMDSe), methyl selenol (MSeH), ethylmethyl selenide (EMSe), ethylmethyl diselenide (EMDSe), dimethyl selenenyl sulfide (DMSeS), and ethylmethyl selenenyl sulfide (EMSeS), were detected in the exposure system using the suspect screening strategy. CdSe/ZnS-GSH was first biotransformed to DMSe and DMDSe by plant and microorganisms. The generated DMSe was volatilized to the gas phase, adsorbed and absorbed by leaves and stems, downward transported, and released into the hydroponic solution, whereas DMDSe tended to be adsorbed/absorbed by roots and upward transported to stems. The airborne DMSe and DMDSe also partitioned from the gas phase to the hydroponic solution. DMSe and DMDSe in the exposure system were further transformed to DMSeS, EMSeS, EMSe, EMDSe, and MSeH. This study gives a comprehensive understanding on the behaviors of Se in CdSe/ZnS-GSH in a rice plant system and provides new insights into the environmental fate of CdSe/ZnS QDs.

RESUMO

Currently, researchers are looking for efficient and sustainable methods to synthesize biochar for the adsorption of pollutants. In this study, biochar with high specific surface area, tunable pore structure, and abundant functional groups were prepared from waste cotton woven (WCW) using low-dosage Fe(NO3)3 activation at 400-900 °C. The biochar obtained at 800 °C possessed the excellent specific surface area of 1167.37 m2/g with a unique micro-mesoporous structure. XRD analysis showed that the Fe species changed from Fe2O3 to Fe3O4 and then Fe0 with the increase of pyrolysis temperature. TEM images further confirmed the template effect of iron oxides for mesoporous formation. The effect of Fe(NO3)3 on the pyrolysis pathway of waste cotton woven was systematically investigated by TG and XPS analyses to explore the pore development of biochar. The results indicated that Fe(NO3)3 could enhance the dehydration, decarbonylation and dehydroxylation of WCW components, thereby reducing the temperature required for WCW pyrolysis. Moreover, the synergistic effect of Fe and N species improved the development of microporous and mesoporous structure through carbon structure corrosion and reorganization, and volatile release. Additionally, satisfactory adsorption capacity for Eriochrome Black T (456.01 mg/g) of the prepared biochar was obtained at 25 °C. This study demonstrated that low-dosage Fe(NO3)3 activation of waste cotton woven could be used as a facile method to prepare promising inexpensive biochar for contaminants removal.

RESUMO

Radish is an economically important root vegetable worldwide. In this study, the 217 cultivated radish accessions were collected and genotyped. To detect the genotypes of these accessions, a total of 24 structure variation (SV) markers distributed on nine chromosomes were employed to analyze genetic diversity and construct a core germplasm collection of radish. The results of polymorphism information content (PIC) indicated a good polymorphism of these SV markers. Population structure analysis and principal component analysis (PCA) results showed that the 217 radish accessions fell into three main populations (P1, P2, and P3). Genetic diversity analysis showed that these populations were highly associated with geographical distribution. The values of the fixation index (FST) indicated a high genetic diversity between P2 and P3, and a moderate genetic diversity between P1 and P2, and P1 and P3. Furthermore, the 43 core germplasm were exploited for creating cytoplasmic male sterility (CMS) lines and cultivating new radish varieties. The high genetic diversity of 217 radish germplasms will not only provide valuable resources for future genetic mapping and functional genomic research, but also facilitate core germplasm utilization and the molecular breeding of radish.

RESUMO

Background: RAB11A, a member of the GTPase family, acts as a regulator in diverse cancers development. The dysregulation of the FAK/AKT signaling pathway is mainly related to tumorigenesis. This study aimed to investigate the possible effect of RAB11A in prostate cancer and further explore the potential mechanisms. Results: In this study, we illustrated the tumor-promoting effects of RAB11A based on in vivo and in vitro experiments. RAB11A expression was upregulated in prostate cancer cells. RAB11A knockdown decreased the prostate cancer cell proliferation, migration, and invasion. RAB11A also induced the epithelial-mesenchymal transition. PF562271 suppressed the malignant characteristics of prostate cancer cells caused by RAB11A knockdown. Furthermore, the interference of RAB11A reduced the tumor growth and the protein levels of p-FAK/FAK and p-AKT/AKT in vivo. Conclusion: RAB11A promotes cell malignant progression and tumor formation in prostate cancer via activating FAK/AKT signaling pathway.

RESUMO

As a widely applied semiconductor nanomaterial, quantum dots (QDs) have drawn considerable interest. In this study, pumpkin and rice seedlings were hydroponically exposed to two core/shell CdSe/ZnS QDs coated with cysteamine (CdSe/ZnS-CA) and polyethylene glycol-carboxy (CdSe/ZnS-PEG-COOH) for 10 days to analyze their time-varying uptake, translocation, and transformation behaviors in plants. Both QDs were mainly adsorbed/absorbed by the roots in the particulate state, and more CdSe/ZnS-CA accumulated than CdSe/ZnS-PEG-COOH. For CdSe/ZnS-CA-treated plants, the Se and Cd concentrations (CSe and CCd) associated with the roots were 561 ± 75 and 580 ± 73 µg/g (dw) for rice and 474 ± 49 and 546 ± 53 µg/g (dw) for pumpkin, respectively, on day 10. For CdSe/ZnS-PEG-COOH-treated plants, the concentrations of Se and Cd associated with roots were 392 ± 56 and 453 ± 56 µg/g (dw) for rice and 363 ± 52 and 417 ± 52 µg/g (dw) for pumpkin, respectively. The surface charges and coatings significantly affected the accumulation of QDs at the beginning of exposure; however, the impaction decreased with time. The ratios between the Cd and Se concentrations (CCd/CSe) in the stems and leaves varied from those of the QD standards, indicating the transformation of the QDs in the exposure system. Se and Cd were more likely to translocate in CdSe/ZnS-PEG-COOH-treated plants than in CdSe/ZnS-CA-treated plants. The vertical translocation of Se was greater than that of Cd. Rice showed greater abilities of accumulation and translocation of Se and Cd from both QDs than pumpkin. These findings improve our understanding of the interference of QDs with plants and their environmental fate.

RESUMO

In this study, the facile carbothermal reduction method was enforced using urea as dopant to modify the structure and chemical composition of nanoscale zero-valent-iron/biochar hybrid thereby boosting its reduction performance. Through fine-tuning the N-doped amount, the optimal nZVI/N-doped BC was obtained, which exhibited more active sites (nZVI, persistent free radicals (PFRs), pyrrolic-N) and superior electrochemical conductivity. With these blessings, the electrons originating from galvanic cell reaction could zip along the highway within the hybrid. Taking nitrobenzene (NB) as the target pollutant, the quantitative analysis revealed that the NB reduction and adsorption removal efficiency were dramatically improved by 2.42 and 2.78 times, respectively. What's more, combining the in-situ experimental detection and theoretical calculations, unexpected NB reductive multipath with respect to PFRs and pyrrolic-N accelerating the Fe3+/Fe2+ cycle within the nZVI/N-doped BC system was decoded. The enhancement of Fe3+/Fe2+ cycle improved the electron utilization efficiency and maintained the reduction reactivity of the hybrid. This work raised awareness of the mechanisms regarding the reduction performance of nZVI/N-doped BC elevated by N-doped and the pollutant reductive pathway within the system, uncovered the dusty roles of PFRs and N-species during the reduction process.

RESUMO

One of the most crucial components of a student's language proficiency is basic language proficiency, which is also its fundamental component. The development of students' language skills is greatly aided by ELL (English Language and Literature). It can not only foster the growth of students' language thinking but also widen their perspectives and enhance their capacity for language comprehension. In this essay, the rules of English are examined from the multifaceted ELL viewpoint. This study extracts personality characteristic data from practical texts and incorporates it into a modelling process of students' knowledge changes based on DM- (data mining-) related technology and multidisciplinary expertise. According to the t-test results, ELL, which incorporates communicative competence, has started to take on a role in education and is helping students' language skills. The performance gains are 3.95, 4.21, and 3.66, when the number of computing nodes is increased to 10, 20, and 30 times that of the original nodes, and the linear expansion indices are 0.914, 0.928, and 0.93. This proves that the Hadoop+Hive-based data cleaning tool has excellent linear scalability.

RESUMO

Two municipal solid waste incineration fly ashes were selected for washing by deionized water and Na2CO3 solution for comparison. Results showed that the benefits of washing were two folds: (1) Washing was able to reduce the contents of Cl- and SO42- while increased the contents of CaO, SiO2, Al2O3 etc.; (2) Washing by Na2CO3 solution showed increased stability of heavy metals (Cr, Ni, Cd, Cu, Zn and Pb) and fly ash was safe for later reuse. Release of Cl- was high at more than 90% regardless of washing solution. SO42- and Ca2+ removal was highly dependent on the dissolution and precipitation equilibriums. Na2CO3 washing promoted the formation of CaCO3. Thus SO42- was washed off instead of precipitating as CaSO4 and retained in fly ash solid. SO42- removal was raised to more than 80% by Na2CO3 washing as compared with about 30% by deionized water. At the same time, Ca removal by Na2CO3 dropped to 1-2%. In addition, the basicity of fly ash was important as high basicity helped SO42- removal. Overall, washing by Na2CO3 appears to be a promising option for fly ash treatment.

RESUMO

In this work, a typical congener of short-chain chlorinated paraffins (SCCPs) with six chlorine atoms (CP-4, 1,2,5,6,9,10-C10H16Cl6, 250 ng/mL) was selected to elaborate the comprehensive environmental transformation of SCCPs in rice seedling exposure system. CP-4 was quickly absorbed, translocated, and phytovolatilized by seedlings with a small quality of CP-4 (5.81-36.5 ng) being detected in the gas phase. Only 21.4 ± 1.6% of an initial amount (10,000 ng) of CP-4 remained in the exposure system at the end of exposure. Among the transformed CP-4, some were attributed to the degradation of the rhizosphere microorganism (9.1 ± 5.8%), root exudates (2.2 ± 4.2%), and abiotic transformation (3.0 ± 2.8%) that were proved by several transformation products found in the root exudate exposure groups and unplanted controls, and a majority was phytotransformed by rice seedlings. Here, 61 products were determined through complex transformation pathways, including multihydroxylation, -HCl elimination, dechlorination, acetylation, sulfation, glycosylation, and amide acid conjugation. The acetylated and amide acid conjugates of CPs were first observed. Phase I and Phase II phytometabolic reactions of CPs were found intertwining. These findings demonstrate that multiactive transformation reactions contribute to the overlook of CPs accumulated in plants and are helpful for the environmental and health risk assessments of SCCPs in agricultural plants.

RESUMO

Early detection of lung cancer has a higher likelihood of curative treatment and thus improves survival rate. Low-dose computed tomography (LDCT) screening has been shown to be effective for high-risk individuals in several clinical trials, but has high false positive rates. To evaluate the risk of stage I lung cancer in the general population not limited to smokers, a retrospective study of 133 subjects was conducted in a medical center in Taiwan. Regularized regression was used to build the risk prediction model by using LDCT and health examinations. The proposed model selected seven variables related to nodule morphology, counts and location, and ten variables related to blood tests and medical history, achieving an area under the curve (AUC) value of 0.93. The higher the age, white blood cell count (WBC), blood urea nitrogen (BUN), diabetes, gout, chronic obstructive pulmonary disease (COPD), other cancers, and the presence of spiculation, ground-glass opacity (GGO), and part solid nodules, the higher the risk of lung cancer. Subjects with calcification, solid nodules, nodules in the middle lobes, more nodules, and diseases related to thyroid, liver, and digestive systems were at a lower risk. The selected variables did not indicate causation.

RESUMO

Waste printed circuit boards (WPCBs) were co-pyrolyzed with iron oxides and iron salts. Solid, liquid, and gaseous products were collected and characterized. Co-pyrolysis with FeCl2, FeCl3, or FeSO4 was able to increase the yield of liquid product which was rich in phenol and its homologues. Also, the addition of co-pyrolysis reagents reduced the release of brominated organics to liquid as Br was either fixed as FeBr3 in solids or released as HBr. In particular, FeCl2 showed the best ability to reduce the release of Br-containing organics to liquid compared with FeCl3 and FeSO4. Solid residuals were rich in iron oxides, glass fibers, and charred organics with surface areas of 20.6-26.5 m2/g. CO2 together with a small amount of CH4 and H2 were detected in the gaseous products. Overall, co-pyrolysis could improve the quantity and quality of liquid oil which could be reused as chemical or energy sources. Pyrolysis of waste printed circuit board was promising as a method for recycling.

RESUMO

Polychloro-1,3-butadienes (polyCBDs) are attracting increasing concern due to their high toxicity. However, research on multiple polyCBDs in aquatic biota is still extremely limited. In this study, a sensitive method for simultaneous determination of nine polyCBD (Cl4-Cl6) congeners, including six tetrachlorobutadiene (TeCBD) isomers, two pentachlorobutadiene (PeCBD) isomers, and hexachlorobutadiene (HCBD), in shellfish was developed based on accelerated solvent extraction (ASE), solid-phase extraction (SPE) clean-up and gas chromatography-triple quadrupole mass spectrometry (GC-QqQ-MS/MS). Low method limits of detection (MDLs) in the range 0.03-0.21 ng/g dry weight for target analytes with satisfactory recoveries (47.7 %-70.6 %) were achieved. The valid method was then applied to analyze nine polyCBDs congeners in 42 shellfish and 11 fish samples collected from markets in eight coastal cities, China. Trace HCBD was detected in 14 samples, while TeCBDs and PeCBDs were under the MDLs in all the samples, indicating little contamination of these pollutants in the marketed shellfish and fish in China. Multiple polyCBDs especially TeCBDs and PeCBDs were firstly involved in the proposed method and investigation here, which lay the groundwork for future research on the environmental behavior and exposure risks of polyCBDs in aquatic biotas.

RESUMO

In plants, chloroplasts are the sites at which photosynthesis occurs, and an increased abundance of chloroplasts increases the nutritional quality of plants and the resultant color of fruits. However, the molecular mechanisms underlying chlorophyll synthesis and chloroplast development in tomato fruits remain unknown. In this study, we isolated a chlorophyll-deficient mutant, reduced chlorophyll mutant 1 (rcm1), by ethylmethanesulfonate mutagenesis; this mutant produced yellowish fruits with altered chloroplast development. MutMap revealed that Solyc08g005010 is the causal gene underlying the rcm1 mutant phenotype. A single-nucleotide base substitution in the second exon of SlRCM1 results in premature termination of its translated protein. SlRCM1 encodes a chloroplast-targeted metalloendopeptidase that is orthologous to the BCM1 protein of Arabidopsis and the stay-green G protein of soybean (Glycine max L. Merr.). Notably, the yellowish phenotype of the lutescent1 mutant can be restored with the allele of SlRCM1 from wild-type tomato. In contrast, knockout of SlRCM1 by the CRISPR/Cas9 system in Alisa Craig yielded yellowish fruits at the mature green stage, as was the case for lutescent1. Amino acid sequence alignment and functional complementation assays showed that SlRCM1 is indeed Lutescent1. These findings provide new insights into the regulation of chloroplast development in tomato fruits.
