As representative organic and inorganic additives, both biochar and smectite exhibit an excellent capacity to improve humification efficiency during composting. Nevertheless, the mechanisms underlying biochar/smectite-induced compost humification have still not been fully explored from the perspective of overall organic substances. In this study, three composting treatments were performed as follows: 10 % biochar-amended composting, 10 % smectite-amended composting and natural composting without any additive. UV-visible parameters and synchronous hetero two-dimensional correlation spectra showed that biochar accelerated dissolved organic matter (DOM) complications, unsaturation and aromatization. For example, biochar promoted the C2 and simple C3 peaks to convert into a sophisticated C3/360 peak. However, the effect of smectite was negligible in complicating the DOM structure. Both biochar and smectite displayed an invigorating role in promoting humic substance (HS) formation. The strengthened relations between bacterial richness and physicochemical indicators and HS fractions might contribute to the positive action of biochar/smectite on HS synthesis. Network analysis showed that both bacterial functional omnipotence and specialization in response to the addition of catalysts may contribute to compost humification. The chemical pathway involved in DOM humification was intensified by enhancing the role of pH in biochar composting and weakening the degradation of unsaturated aromatic compounds of DOM with smectite addition. These findings benefit the practical application of biochar/smectite in promoting composting efficiency.

RESUMO

Rheumatoid arthritis (RA) is an autoimmune disease characterized by severe joint damage and disability. However, the specific mechanism of RA has not been thoroughly clarified over the past decade. Nitric oxide (NO), a kind of gas messenger molecule with many molecular targets, is demonstrated to have significant roles in histopathology and homeostasis. Three nitric oxide synthases (NOS) are related to producing NO and regulating the generation of NO. Based on the latest studies, NOS/NO signaling pathways play a key role in the pathogenesis of RA. Overproduction of NO can induce the generation and release of inflammatory cytokines and act as free radical gas to accumulate and trigger oxidative stress, which can involve in the pathogenesis of RA. Therefore, targeting NOS and its upstream and downstream signaling pathways may be an effective approach to managing RA. This review clearly summarizes the NOS/NO signaling pathway, the pathological changes of RA, the involvement of NOS/NO in RA pathogenesis and the conventional and novel drugs based on NOS/NO signaling pathways that are still in clinical trials and have good therapeutic potential in recent years, with an aim to provide a theoretical basis for further exploration of the role of NOS/NO in the pathogenesis, prevention and treatment of RA.

The aim of this study was to assess the effectiveness of biochar and montmorillonite islands on heavy metal adsorptive immobilization and identify crucial driving factors and pathways during chicken manure composting. Compared to montmorillonite (6.74 and 89.25 mg/kg), biochar exhibited an obviously higher ability of Cu and Zn enrichment (41.79 and 167.77 mg/kg), might be attributed to its abundant active functional groups. Network analysis showed that compared to Cu, core bacteria positively and negatively related to Zn was obviously more and less in passivator islands, respectively, which might explain significantly higher Zn concentration. Structural Equation Model displayed that dissolved organic carbon (DOC), pH and bacteria were critical driving factors. Pretreatment of passivator packages, such as soaking in the solution being rich in DOC and inoculating specific microbial agents accumulating heavy metals via extracellular adsorption /intracellular interception would significantly improve the effectiveness of adsorptive passivation on heavy metals.

Acetylcholine, a neurotransmitter secreted by cholinergic neurons, is involved in signal transduction related to memory and learning ability. Alzheimer's disease (AD), a progressive and commonly diagnosed neurodegenerative disease, is characterized by memory and cognitive decline and behavioral disorders. The pathogenesis of AD is complex and remains unclear, being affected by various factors. The cholinergic hypothesis is the earliest theory about the pathogenesis of AD. Cholinergic atrophy and cognitive decline are accelerated in age-related neurodegenerative diseases such as AD. In addition, abnormal central cholinergic changes can also induce abnormal phosphorylation of ttau protein, nerve cell inflammation, cell apoptosis, and other pathological phenomena, but the exact mechanism of action is still unclear. Due to the complex and unclear pathogenesis, effective methods to prevent and treat AD are unavailable, and research to explore novel therapeutic drugs is various and active in the world. This review summaries the role of cholinergic signaling and the correlation between the cholinergic signaling pathway with other risk factors in AD and provides the latest research about the efficient therapeutic drugs and treatment of AD.

A new series of derivatives characterized by the presence of the 3,4,5-trimethoxylbenzamide substituted benzofurans were synthesized and evaluated for antiproliferative activity against four cancer cell lines and one normal human cell line. Among them, derivative 6g with greatest cytotoxicity significantly inhibited the growth of MDA-MB-231, HCT-116, HT-29 and HeLa cell lines with IC50 values of 3.01, 5.20, 9.13, and 11.09 µM, respectively. Importantly, 6g possessed excellent selectivity over non-tumoral cell lines HEK-293 (IC50 > 30 µM). Moreover, mechanistic studies revealed that 6g induced HeLa cells arrested in G2/M phase in a concentration-dependent manner, and inhibited polymerization of tubulin via a consistent way with CA-4. In general, these observations suggest that 6g is a promising anti-cancer lead and is worth further investigation to generate potential antitumor agents.

In this research, pH-sensitive semi-interpenetrating polymer network hydrogels based on sodium carboxymethyl cellulose and poly(methacrylic acid) were synthesized using free radical polymerization and semi-interpenetrating polymer network approach for oral administration of insulin. The chemical structure and thermal stability of the hydrogels were characterized using Fourier transform infrared spectroscopy, X-ray diffraction, and thermogravimetric analysis measurements. The interior morphology was observed by scanning electron microscopy and the inner structure exhibited a porous honeycomb-like shape. The investigations on the swelling properties of hydrogels revealed their ability to response to pH value change. The in vitro release behavior of insulin was pH dependent and the release of insulin was much lower at pH 1.2 compared to pH 6.8. In vitro cytotoxicity assay indicated that the hydrogels were noncytotoxic to HeLa cells. A sustained reduction in blood glucose level was observed after oral administration of insulin-loaded hydrogel to diabetic rats at 75 IU/kg. These results indicated that the hydrogel would be a promising vehicle for oral insulin delivery systems.

RESUMO

Considering the low accuracy of prediction in the positive samples and poor overall classification effects caused by unbalanced sample data of MicroRNA (miRNA) target, we proposes a support vector machine (SVM)-integration of under-sampling and weight (IUSM) algorithm in this paper, an under-sampling based on the ensemble learning algorithm. The algorithm adopts SVM as learning algorithm and AdaBoost as integration framework, and embeds clustering-based under-sampling into the iterative process, aiming at reducing the degree of unbalanced distribution of positive and negative samples. Meanwhile, in the process of adaptive weight adjustment of the samples, the SVM-IUSM algorithm eliminates the abnormal ones in negative samples with robust sample weights smoothing mechanism so as to avoid over-learning. Finally, the prediction of miRNA target integrated classifier is achieved with the combination of multiple weak classifiers through the voting mechanism. The experiment revealed that the SVM-IUSW, compared with other algorithms on unbalanced dataset collection, could not only improve the accuracy of positive targets and the overall effect of classification, but also enhance the generalization ability of miRNA target classifier.

OBJECTIVE: To apply needle-knife to treat lumber disc herniation (LDH) and surface electromyography were used to analyze biomechanical characteristic of patient's lumber muscle to make a comprehensive evaluation on its efficacy. METHODS: Thirty patients who met the inclusive criteria were selected and treated with needle-knife, once a week for 2 weeks. Visual analogue scale (VAS), ASLR and JOA score before and after treatment were observed. Surface electromyography was applied to test the surface electromyography signals. AEMG, MFs and MPF were calculated before and after the treatment. RESULTS: After treatment, VAS was significantly reduced, ASLR, JOA, AEMG and MPF were obviously increased, and the absolute value of MFs was lowed (all P < 0.01). CONCLUSION: The needle-knife could significantly relieve lumbar muscle strength, muscle tone and muscle fatigue, improve in the imbalance of lumbar extensor muscle group, leading to the recovery of biomechanical characteristic, and the clinical efficacy is superior.
