RESUMO

Flexible sensors are becoming the focus of research because they are very vital for intelligent products, real-time data monitoring, and recording. The flat silk cocoon (FSC), as a special form of cocoon, has all the advantages of silk, which is an excellent biomass carbon-based material and a good choice for preparing flexible sensors. In this work, a flexible piezoresistive sensor was successfully prepared by encapsulating carbonized flat silk cocoons (CFSCs) using an elastic matrix polydimethylsiloxane (PDMS). The sensing performance of the material is 0.01 kPa-1, and the monitoring range can reach 680.57 kPa. It is proved that the sensor can detect human motion and has excellent durability (>800 cycles). In addition, a sensor array for a keyboard based on CFSCs was explored. The sensor has a low production cost and a simple preparation process, and it is sustainable and environmentally friendly. Thus, it may have potential applications in wearable devices and human-computer interactions.

Multidrug resistance (MDR) is the main cause of clinical chemotherapy failure, and new strategies to overcome MDR are needed. We report multi-responsive silk fibroin nanoparticles (SFNs) co-loaded with the chemotherapeutic drug doxorubicin (DOX) and PX478 (a hypoxia-inducible factor inhibitor), which was functionalized with folic acid (FA). This combination could actively target tumor cells and respond to the release of PX478, inhibit the hypoxia-inducible factor (HIF) gene and its related downstream drug-resistant target genes. The FA-PX478-DOX-SFNs (F-P-D-S) combination showed accelerated drug release profiles in the media simulating the tumor microenvironment, which had acidic pH, high levels of reactive oxygen species and high levels of glutathione. Compared with PX478-DOX-SFNs (P-D-S) without targeted modification, the cellular uptake rate of F-P-D-S increased. In addition, F-P-D-S quickly achieved lysosomal escape, enabling DOX to rapidly enter the nucleus to kill the drug-resistant cells. A cytotoxicity test indicated that the IC50 of DOX against MCF-7/ADR cells was 1.0 µg/mL in F-P-D-S, which was 26 times lower than that of free DOX (25.6 µg/mL). F-P-D-S significantly down-regulated HIF-1α, MDR1, VEGF and GLUT-1 and P-gp protein to overcome multidrug resistance. This effective synergistic chemotherapy strategy for HIF inhibition has potential for use in the treatment of multidrug-resistant tumors.

RESUMO

The silkworm Bombyx mori is an important economic insect for producing silk, the "queen of fabrics". The currently available genomes limit the understanding of its genetic diversity and the discovery of valuable alleles for breeding. Here, we deeply re-sequence 1,078 silkworms and assemble long-read genomes for 545 representatives. We construct a high-resolution pan-genome dataset representing almost the entire genomic content in the silkworm. We find that the silkworm population harbors a high density of genomic variants and identify 7308 new genes, 4260 (22%) core genes, and 3,432,266 non-redundant structure variations (SVs). We reveal hundreds of genes and SVs that may contribute to the artificial selection (domestication and breeding) of silkworm. Further, we focus on four genes responsible, respectively, for two economic (silk yield and silk fineness) and two ecologically adaptive traits (egg diapause and aposematic coloration). Taken together, our population-scale genomic resources will promote functional genomics studies and breeding improvement for silkworm.

Bombyx mori silk fibroin (SF) is widely used in the field of biomaterials due to its excellent biocompatibility and mechanical properties. However, SF cannot be used directly in many applications and needs to be dissolved first. Lithium bromide (LiBr) is a traditional solvent which is usually used to dissolve SF. However, LiBr has several limitations, e.g., it is expensive, it is toxic to organisms, and it is environmentally unfriendly. Herein, we investigate the possibility of developing a ternary reagent system that is inexpensive, non-toxic to organisms, and environmentally friendly as an alternative for silk fibroin solubilization. The results confirm that regenerated silk fibroin (RSF) prepared using a ternary reagent has the same morphology and amino acid composition as that prepared using LiBr, but the RSF prepared using a ternary reagent still had a small amount of calcium residue even after long-term dialysis. Further research found that the residual calcium does not cause significant differences in the structure and biological performance of the RSF, such as its cytotoxicity, blood compatibility, and antibacterial properties. Therefore, we believe that ternary reagents are an ideal alternative solvent for dissolving SF.

The recycling and reuse of biomass waste for the preparation of carbon-based adsorbents is a sustainable development strategy that has a positive environmental impact. It is well known that a large amount of silk sericin (SS) is dissolved in the wastewater from the silk industry. Utilizing the SS instead of discharging it into the environment without further treatment would reduce environmental and ecological problems. However, effective enrichment of the SS from the aqueous solution is a challenge. Here, with the help of carboxymethyl chitosan (CMCS), which can form a gel structure under low voltage, an SS/CMCS hydrogel with SS as the major component was prepared via electrodeposition at a 3 V direct-current (DC) voltage for five minutes. Following a carbonization process, an SS-based adsorbent with good performance for the removal of methylene blue (MB) from an aqueous solution was prepared. Our results reveal that the SS/CMCS hydrogel maintains a porous architecture before and after carbonization. Such structure provides abundant adsorption sites facilitating the adsorption of MB molecules, with a maximum adsorptive capacity of 231.79 mg/g. In addition, it suggests that the adsorption is an exothermic process, has a good fit with the Langmuir model, and follows the intra-particle diffusion model. The presented work provides an economical and feasible path for the treatment of wastewater from dyeing and printing.

In the present work, a sustained-release film composed of silk fibroin (SF), curcumin (Cur), glutaraldehyde (GA), and glycerol (Gly) was prepared successfully for wound dressings. Features relevant to wound dressings of SF/Gly/GA/Cur film were assessed. Physical and chemical properties of the fabricated materials were also characterized. The results showed that the prepared SF/Gly/GA/Cur film demonstrated a good sustained-release performance, flexibility, and gas permeability. In addition, it was found that the prepared SF/Gly/GA/Cur film possessed the capability to effectively inhibit the growth of bacteria and prevent bacterial penetration with a suitable water vapor transmission rate. Furthermore, the prepared composite film was non-cytotoxic, which makes it an ideal material for wound dressings.

Natural Bombyx mori silk fibroin (SF) fibers were modified with 2-methacryloyloxyethyl isocyanate (MOI) first for the introduction of vinyl groups. Then, 1H,1H,2H,2H-perfluorodecanethiol was grafted onto the SF fibers via thiol-ene click chemistry using ultraviolet light. The formations of MOI-modified and PFDT-grafted SF fibers were analyzed using Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy, respectively. The morphology of samples was also revealed by a scanning electron microscope. In addition, differential scanning calorimetry results demonstrated that SF fibers did not show significant change in thermal behavior, regardless of the chemical modification. To confirm the cytotoxicity of the prepared SF fibers, MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] assay was performed, and no toxicity was observed with PFDT-grafted SF fibers. The results also showed that PFDT-grafted SF fibers exhibited good antifouling properties when Chlorella vulgaris (C. vulgaris) was selected as a model for algal cells adhesion experiment.

BACKGROUND: Recruitment of endogenous stem cells has been considered an alternative to cell injection/implantation in articular cartilage repair. PURPOSE: (1) To develop a cartilage tissue-engineering scaffold with clinically available biomaterials and functionalize the scaffold with an aptamer (Apt19s) that specifically recognizes pluripotent stem cells. (2) To determine whether this scaffold could recruit joint-resident mesenchymal stem cells (MSCs) when implanted into an osteochondral defect in a rabbit model and to examine the effects of cartilage regeneration. STUDY DESIGN: Controlled laboratory study. METHODS: The reinforced scaffold was fabricated by embedding a silk fibroin sponge into silk fibroin/hyaluronic acid-tyramine hydrogel and characterized in vitro. A cylindrical osteochondral defect (3.2 mm wide × 4 mm deep) was created in the trochlear grooves of rabbit knees. The rabbits were randomly assigned into 3 groups: Apt19s-functionalized scaffold group, scaffold-only group, and control group. Animals were sacrificed at 6 and 12 weeks after transplantation. Repaired tissues were evaluated via gross examination, histologic examination, and immunohistochemistry. RESULTS: In vitro, this aptamer-functionalized scaffold could recruit bone marrow-derived MSCs and support cell adhesion. In vivo, the aptamer-functionalized scaffold enhanced cell homing in comparison with the aptamer-free scaffold. The aptamer-functionalized scaffold group also exhibited superior cartilage restoration when compared with the scaffold-only group and the control group. CONCLUSION: The Apt19s-functionalized scaffold exhibited the ability to recruit MSCs both in vitro and in vivo and achieved a better outcome of cartilage repair than the scaffold only or control in an osteochondral defect model. CLINICAL RELEVANCE: The findings demonstrate a promising strategy of using aptamer-functionalized bioscaffolds for restoration of chondral/osteochondral defects via aptamer-introduced homing of MSCs.

Regenerated Silk biomaterials are usually pre-formed from silk fibroin solutions. However, the dissolution of silk fibroin in proper solvents by a simple and low cost way is still a challenge. Here, we employed a CaCl2-methanol solvent system with a very low CaCl2 concentration of 6wt% to dissolve silk fibroin. During the dissolution process, the evaporation of methanol cause the changing of solvation sheath of ions in the solvent. The remaining solvent with the incomplete solvation sheath is absorbed by the silk fiber and interacts with fibroin chains to complete the solvation sheath, which accounts for the dissolution of silk fibroin. Silk fibroin dissolution stops as all the solvation sheaths are complete. The final CaCl2 concentration is ca. 26% and silk fibroin is completely dissolved with a yield of about 90%. Silk fibroin is dissolved into multi-scale nanofibrils solution which is potential for producing regenerated silk fibroin materials for functional applications.
