RESUMO

Background: Prior studies have explored the association between perceived parental rejection-warmth and adolescents' rumination, but it is unclear whether the interaction between perceived parental rejection and warmth can predict adolescents' rumination in a Chinese context and whether this interaction varies by children's gender during the post-COVID-19 era. Objective: This study aimed to address these issues in Chinese early adolescents from a family system perspective. Methods: A total of 910 adolescents (M age = 13.63, 48.50% female) from two middle schools in Chongqing and Changsha, China participated in the survey, answering measures for demographics, perceived parental rejection-warmth, and rumination. Results: The results show that adolescents' rumination was positively related to perceived paternal rejection (r = 0.326, p <.001) and maternal rejection (r = 0.343, p <.001), and negatively related to perceived paternal warmth (r = -.184, p <.001) and maternal warmth (r = -0.125, p <.001). Moreover, perceived maternal warmth significantly moderated the link between perceived paternal rejection and adolescents' rumination (boot effect = -0.066, 95CI% = [-0.124, -0.010]), but this moderating effect is only presented in boys not in girls (boot effect = -0.063, 95CI% = [-0.015, 0.140]). However, perceived paternal warmth moderated the link between perceived maternal rejection and rumination in adolescents (boot effect = -0.052, 95CI% = [-0.103, -0.001]), and this interaction varied by adolescents' gender (boot effect = 0.103, 95CI% = [0.029, 0.177]). Conclusions: Perceived Parental rejection and parental warmth co-exist in the Chinese family system, and they exert an interactive effect on adolescents' rumination depending on their gender. It implies that both parents should be more accepting, caring, considerate, and warm toward their daughters, and it is also in line with the tradition and status quo of parenting in Chinese families. These findings have implications for Chinese parental co-parenting practices in families with adolescents and adolescence mental health counseling work.

RESUMO

In the present work, an inducible assembly of di-block polymer compounds approach was employed for the synthesis of mesoscopic gyrus-like In2O3 by using lab-made high-molecular-weight amphiphilic di-block copolymer poly(ethylene oxide)-b-polystyrene (PEO-b-PS) as a revulsive, with indium chloride as an indium source and THF/ethanol as the solvent. The obtained mesoscopic gyrus-like In2O3 indium oxide materials exhibit a large surface area and a highly crystalline In2O3 nanostructure framework, and the gyrus distance is about 40 nm, which can facilitate the diffusion and transport of acetone vapor molecules. By using this material as a chemoresistance sensor, the obtained gyrus-like indium oxides were used as sensing materials, showing an excellent performance to acetone at a low operating temperature (150 °C) due to their high porosity and unique crystalline framework. The limit of detection of the thick-film sensor based on indium oxides is appropriate for diabetes exhaled breath acetone concentration detection. Moreover, the thick-film sensor shows a very fast response-recovery dynamics upon contacting acetone vapor due to its abundant open folds mesoscopic structure, and also to the large surface area of the nanocrystalline gyrus-like In2O3.

RESUMO

Posttranslational modifications of antibody products affect their stability, charge distribution, and drug activity and are thus a critical quality attribute. The comprehensive mapping of antibody modifications and different charge isomers (CIs) is of utmost importance, but is challenging. We intended to quantitatively characterize the posttranslational modification status of CIs of antibody drugs and explore the impact of posttranslational modifications on charge heterogeneity. The CIs of antibodies were fractionated by strong cation exchange chromatography and verified by capillary isoelectric focusing-whole column imaging detection, followed by stepwise structural characterization at three levels. First, the differences between CIs were explored at the intact protein level using a top-down mass spectrometry approach; this showed differences in glycoforms and deamidation status. Second, at the peptide level, common modifications of oxidation, deamidation, and glycosylation were identified. Peptide mapping showed nonuniform deamidation and glycoform distribution among CIs. In total, 10 N-glycoforms were detected by peptide mapping. Finally, an in-depth analysis of glycan variants of CIs was performed through the detection of enriched glycopeptides. Qualitative and quantitative analyses demonstrated the dynamics of 24 N-glycoforms. The results revealed that sialic acid modification is a critical factor accounting for charge heterogeneity, which is otherwise missed in peptide mapping and intact molecular weight analyses. This study demonstrated the importance of the comprehensive analyses of antibody CIs and provides a reference method for the quality control of biopharmaceutical analysis.

RESUMO

Novel 3D biogenic C-doped Bi2MoO6/In2O3-ZnO Z-scheme heterojunctions were synthesized for the first time, using cotton fiber as template. The as-prepared samples showed excellent adsorption and photodegradation performance toward the hazardous antibiotic doxycycline under simulated sunlight irradiation. The morphology, phase composition and in situ carbon doping could be precisely controlled by adjusting processing parameters. The carbon doping in Bi2MoO6/In2O3-ZnO was derived from the cotton template, and the carbon content could be varied in the range 0.9-4.4 wt.% via controlling the heat treatment temperature. The sample with Bi2MoO6/In2O3-ZnO molar ratio of 1:2 and carbon content of 1.1 wt.% exhibited the highest photocatalytic activity toward doxycycline degradation, which was 3.6 and 4.3 times higher than those of pure Bi2MoO6 and ZnInAl-CLDH (calcined layered double hydroxides), respectively. It is believed that the Z-scheme heterojunction with C-doping, the 3D hierarchically micro-meso-macro porous structure, as well as the high adsorption capacity, contributed significantly to the enhanced photocatalytic activity.

RESUMO

Human GLP-1 (glucagon-like peptide-1) can produce a remarkable improvement in glycemic control in patients with type 2 diabetes. However, its clinical benefits are limited by its short half-life, which is less than 2 min because of its small size and rapid enzymatic inactivation by dipeptidyl peptidase IV. We engineered GLP-1-IgG2σ-Fc, a 68-kDa fusion protein linking a variant human GLP-1 (A8G/G26E/R36G) to a human IgG2σ constant heavy-chain. A stably transfected Chinese hamster ovary cell line was obtained using electroporation. Western blotting showed that the expressed protein was immunoreactive to both GLP-1 and IgG antibodies. GLP-1-IgG2σ-Fc stimulated insulin secretion from INS-1 cells in a dose- and glucose-dependent manner and increased insulin mRNA expression. The half-life of GLP-1-IgG2σ-Fc in cynomolgus monkeys was approximately 57.1 ± 4.5 h. In the KKAy mouse model of diabetes, one intraperitoneal injection of GLP-1-IgG2σ-Fc (1 mg/kg) reduced blood glucose levels for 5 days. A 4-week repeat-administration study identified sustained effects on blood glucose levels. Oral glucose tolerance tests conducted at the beginning and end of this 4-week period showed that GLP-1-IgG2σ-Fc produced a stable glucose lowering effect. In addition, KKAy mice treated with GLP-1-IgG2σ-Fc showed statistically significant weight loss from day 23. In conclusion, these properties of GLP-1-IgG2σ-Fc demonstrated that it represented a potential long-acting GLP-1 receptor agonist for the treatment of type 2 diabetes.
