RESUMO

Obesity presents a significant public health challenge, demanding effective dietary interventions. This study employed a high-fat diet-induced obesity mouse model to explore the impacts of inulin with different polymerization degrees on obesity management. Our analysis reveals that high-degree polymerization inulin (HDI) exhibited a significantly higher oil binding capacity and smaller particle size compared to low-degree polymerization inulin (LDI) (p < 0.05). HDI was more effective than LDI in mitigating body weight gain in high-diet induced obese mice, although neither LDI nor HDI affected blood sugar levels when compared to the high-fat diet control group (p < 0.05). Both HDI and LDI administrations reduced liver weight and enhanced brown adipose tissue thermogenesis compared to the high-fat diet induced control group (p < 0.05). Additionally, HDI suppressed hepatic lipogenesis, resulting in a further reduction in liver triglycerides compared to the high-fat diet-induced obese mice (p < 0.05). Notably, HDI improved gut health by enhancing intestinal morphology and modulating gut microbiota structure. HDI administration notably increased the relative abundance of cecal Akkermansia, a gut microbe associated with improved metabolic health, while LDI showed limited efficacy (p < 0.05 and p > 0.05, respectively). These findings underscore the importance of the structural properties of inulin in its potential to combat obesity and highlight the strategic use of inulin with varying polymerization degrees as a promising dietary approach for obesity management, particularly in its influence on gut microbiota composition and hepatic lipid metabolism regulation.

The effects of Lactobacillus plantarum in microencapsulation (LPM) on intestinal development in layer chicks were investigated in this study, as well as the colonization of L. plantarum in the gut. A total of 480 healthy Hy-Line Brown layer chicks at 0 d old were randomly divided into 4 groups (8 replicates each treatment), and the diets of these birds were supplemented with nothing (control), L. plantarum (0.02 g/kg feed; 109 CFU/kg feed), LPM (1.0 g/kg feed; 109 CFU/kg feed) and wall material of LPM (WM; 0.98 g/kg feed), respectively. Compared to control, LPM improved growth performance and intestinal development of layer chicks, evidenced by significantly increased body weight, average daily gain, average daily feed intake, villus height, villus height/crypt depth, as well as weight and length of the duodenum, jejunum and ileum (P < 0.05). These results could be attributed to the increased colonization of L. plantarum in the gut, which was verified by significant increases in lactic acid content, viable counts in chyme and mucosa (P < 0.05), as well as a visible rise in number of strains labeled with fluorescein isothiocyanate. Meanwhile, the relative abundances of Lactobacillus and Bifidobacterium significantly increased in response to microencapsulated L. plantarum supplementation (P < 0.05), accompanied by the significant up-regulation of colonization related genes (P < 0.05), encoding solute carrier family, monocarboxylate transporter, activin A receptor, succinate receptor and secretogranin II. To sum up, microencapsulated L. plantarum supplementation promoted intestinal development, which could be attributed to the enhancement of L. plantarum colonization in the intestine through the mutual assistance of Bifidobacterium and interactions with colonization related transmembrane proteins.

Disease-causing microorganisms such as Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) are among the primary contributors to morbidity and mortality of diarrhea in humans. Considering the challenges associated with antibiotic use, including antimicrobial resistance, this study aimed to develop a novel zinc-based agent for bacterial inactivation. To this end, zinc caproate (ZnCA) was synthesized using caproic acid (CA) and zinc oxide (ZnO) in anhydrous ethanol via the solvothermal method. Structural characterization techniques, including Fourier-transform infrared spectroscopy, single crystal X-ray diffraction analysis, and nuclear magnetic resonance spectroscopy, revealed the bidentate bridging coordination of zinc atoms with CA. The resulting two-dimensional ZnCA network was found to be composed of a distinct lamellar pattern, without any evident inter-layer interactions. Powder X-ray diffraction analysis, elemental analysis, and melting point analysis confirmed that ZnCA had an average particle size of 1.320 µm, a melting point of 147.2 °C, and a purity exceeding 98 %. Remarkably, ZnCA demonstrated potent antibacterial activity against E. coli and S. aureus, which exceeded the antibacterial efficacy of ZnO. ZnCA exerted its antibacterial effects by inhibiting biofilm formation, disrupting cell membrane integrity, increasing cell membrane permeability, and altering intracellular Ca2+-Mg2+-ATPase activity. These findings highlight the potential of ZnCA as a promising antibiotic substitute for the treatment of diarrhea in humans.

The alleviating effects of Lactobacillus plantarum in microencapsulation (LPM) on lipopolysaccharide (LPS)-induced intestinal inflammatory injury were investigated in layer chicks. A total of 252 healthy Hy-Line Brown layer chicks were randomly divided into six groups. Birds were injected with saline or LPS except for the control, and the diets of birds subjected to LPS were supplemented with nothing, L. plantarum, LPM, and wall material of LPM, respectively. The viable counts of LPM reached 109 CFU/g, and the supplemental levels of L. plantarum, LPM, and WM were 0.02 g (109 CFU), 1.0 g, and 0.98 g, per kilogram feed, respectively. LPS administration caused intestinal damage in layer chicks, evidenced by increased proinflammatory factors accompanied by poor intestinal development and morphology (p < 0.05). LPM/LPS significantly increased body weight, small intestine weight and length, villus height, villus height/crypt depth, and mRNA relative expression of tight junction protein genes (p < 0.05) and performed better than free L. plantarum. These findings could be attributed to the significant increase in viable counts of L. plantarum in the small intestine (p < 0.05), as well as the enhanced levels of Actinobacteriota, Lactobacillaceae, and Lactobacillus in intestinal microbiota (p < 0.05). Such results could further significantly increase goblet and PCNA+ cell percentage (p < 0.05); the mRNA relative expressions of epithelial cell, fast-cycling stem cell, quiescent stem cell, endocrine cell, and Paneth cell; and goblet and proliferative cell marker genes, including E-cadherin, Lgr-5, Bmi-1, ChA, Lysozome, Mucin-2, and PCNA (p < 0.05). Furthermore, the mRNA relative expressions of key genes involved in epithelial cell proliferation, namely, c-Myc, Cyclin-1, Wnt-3, Lrp-5, and Olfm-4, exhibited significant upregulation compared with the LPS treatment, as well as the differentiating genes Notch-1 and Hes-1 (p < 0.05). To sum up, microencapsulated L. plantarum supplementation could alleviate intestinal injury in layer chicks induced by LPS by promoting the proliferation and differentiation of intestinal epithelial cells, which could be attributed to the increase in viable count of L. plantarum in the gut and optimization in intestinal microbial flora.

Ascorbic acid (AA) is an indispensable nutrient required to sustain optimal poultry health and performance, which is commonly excluded from the diet of broilers. To investigate the synthesis and distribution of AA during broiler growth and clarify its possible turnover, 144 1 d old healthy Arbor Acres broilers with a body weight of approximately 41 g were randomly assigned to eight groups of 18 broilers each. The kidney, liver, ileum, and spleen of one bird from each group were collected every week until 42 d to detect the synthesis capacity, tissue distribution, and transporter gene expression of AA. The results showed that kidney L-gulonolactone oxidase (GLO) activity responded quadratically (p < 0.001), with maximum activity observed at 7 to 21 d old. Hepatic total AA and dehydroascrobate (DHA) concentration increased linearly (p < 0.001) with age, as did splenic total AA (p < 0.001). In the ileum, mRNA expression of sodium vitamin C transporter 1/2 (SVCT1/2) decreased with the growing age of the broilers (p < 0.05). The expression of SVCT1 in the kidney was not influenced by the growing age of the broilers. The progressive buildup of AA in the liver and spleen of broilers as they age implies an amplified demand for this nutrient. The waning synthesis capacity over time, however, raises concerns regarding the possible inadequacy of AA in the latter growth phase of broilers. The addition of AA to the broilers' diet might have the potential to optimize their performance. However, the effectiveness of such dietary supplementation requires further investigation.

Polysaccharide decolorization has a major effect on polysaccharide function. In the present study, the decolorization of Rehmannia glutinosa polysaccharides (RGP) is optimized using two methods-the AB-8 macroporous resin (RGP-1) method and the H2O2 (RGP-2) method. The optimal decolorization parameters for the AB-8 macroporous resin method were as follows: temperature, 50 °C; macroporous resin addition, 8.4%; decolorization duration, 64 min; and pH, 5. Under these conditions, the overall score was 65.29 ± 3.4%. The optimal decolorization conditions for the H2O2 method were as follows: temperature, 51 °C; H2O2 addition, 9.5%; decolorization duration, 2 h; and pH, 8.6. Under these conditions, the overall score was 79.29 ± 4.8%. Two pure polysaccharides (RGP-1-A and RGP-2-A) were isolated from RGP-1 and RGP-2. Subsequently, their antioxidant and anti-inflammatory effects and mechanisms were evaluated. RGP treatment activated the Nrf2/Keap1 pathway and significantly increased the activity of antioxidant enzymes (p < 0.05). It also inhibited the expression of pro-inflammatory factors and suppressed the TLR4/NF-κB pathway (p < 0.05). RGP-1-A had a significantly better protective effect than RGP-2-A, likely owing to the sulfate and uronic groups it contains. Together, the findings indicate that RGP can act as a natural agent for the prevention of oxidation and inflammation-related diseases.

Polysaccharide decolorization is a key determinant of polysaccharide structure. In this study, two purified Rehmannia glutinosa polysaccharides, RGP-1-A and RGP-2-A, were obtained after decolorization using the AB-8 macroporous resin and H2O2, respectively. RGP-1-A (molecular weight (Mw) = 18,964 Da) and RGP-2-A (Mw = 3305 Da) were acidic and neutral heteropolysaccharides, respectively, and were both polycrystalline in structure. FTIR analysis revealed that RGP-1-A was a sulfate polysaccharide, while RGP-2-A had no sulfate group. Experiments on IPEC-1 cells showed that RGPs alleviated oxidative stress by regulating the Nrf2/Keap1 pathway. These findings were confirmed by the upregulation of Nrf2, NQO1, and HO-1; the subsequent increase in the levels of antioxidant indicators (SOD, LDH, CAT, and MDA); and the restoration of mitochondrial membrane potential. Overall, the antioxidant capacity of RGP-1-A was significantly higher than that of RGP-2-A. These results suggest that RGPs may be a potential natural antioxidant and could be developed into functional foods.

We investigated the effect of photoperiod on ileal morphology, barrier function, short-chain fatty acid (SCFA) contents, microbial flora, melatonin expression, and synthesis in laying ducks. After adaption, a total of 180 Jinding laying ducks (252 days old) were randomly divided into three treatments, receiving 12L (hours of light):12D (hours of darkness), 16L:8D, or 20L:4D. Each treatment had six replicates with 10 birds each. The formal experiment lasted 58 days. Compared with 12L:12D, the significantly higher values of villus height and goblet cell percentage (GCP) were observed in 16L:8D treatment, accompanied with the higher mRNA relative expression of zonula occludens-1, zonula occludens-2, zonula occludens-3, claudin-1, occludin, and mucin 2 (P < 0.05). Besides, significantly higher values of acetate and propionate, butyrate and total SCFA concentrations were simultaneously observed in ileal chyme of 16L:8D treatment (P < 0.05). For the ileal microbial community, the results of principal coordinate analysis (PCoA) visually presented that three photoperiod groups were mainly scattered into three clusters, indicating that the microbiota composition in different photoperiod treatments were quite dissimilar. Lower values of Shannon indicators were observed in the 20L:4D treatment (P < 0.05), meaning that the microbiota α-diversity decreased in the 20-h photoperiod. The relative abundance of Actinobacteria, Fusobacteria, and Proteobacteria at phylum level and Fusobacterium, Clostridium_sensu_stricto_1, and Pectobacterium at genus level kept an appropriate balance in the 16L:8D photoperiod. Melatonin level in serum decreased with the increasing photoperiods at 6:00 and 12:00, which was consistent with melatonin receptor expressions in the hypothalamus and ileal tissue. Meanwhile, the adenosine 3',5'-cyclic phosphate (cAMP) contents were significantly downregulated in the pineal gland (P < 0.05), in response to the increase in photoperiod. In conclusion, an appropriate photoperiod could improve ileal morphology, barrier function, SCFA profile, and microbial flora, which may be attributed to the appropriate regulation of the circadian rhythm through melatonin as well as its receptor expression, and 16 h could be an adequate photoperiod for laying ducks.

We investigated the effect of photoperiod on eggshell quality, bone quality characteristics and bone metabolism related enzymes and factors in laying ducks. After adaption, 300 Jinding laying ducks (252-day-old) were randomly divided into 5 treatments, receiving 12L (hours of light):12D (hours of darkness), 14L:10D, 16L:8D, 18L:6D or 20L:4D, respectively. Each group had 6 replicates of 10 birds each. The feeding experiment lasted 8 wk. Compared with 12L:12D, the higher values of eggshell breaking strength occurred in ≥18 h photoperiods at the end of 6 wk, and in ≥16 h photoperiods at the end of 4 wk, with the common highest values in 18 h photoperiod (P ˂ 0.05). Besides, 18L:6D had higher values of ultimate load Fu and cortical cross-sectional area A in tibia, femur, and ulna (P ˂ 0.05), compared with 12L:12D. The higher values of proximal bone mineral content (BMC; tibia), distal BMC (ulna), total Ca (tibia), and cortical volumetric bone mineral density (vBMC; tibia and ulna) were observed in 16L:8D and 18L:6D treatments (P ˂ 0.05). Meanwhile, 18 h photoperiod group had the higher proximal BMC (femur) and total Ca in ulna (P ˂ 0.05). In serum, compared with 12L:12D group, the higher ALP activity occurred in ≥16 h photoperiods (0:00 and 18:00), with the highest values in 18L:6D treatment (P ˂ 0.05); the higher values of TGF-ß (6:00) and OC (6:00 and 18:00) were simultaneously observed in 18 h photoperiod (P ˂ 0.05). Moreover, values of trACP activity, TNF-α and IL-6 contents decreased in ≥18 h photoperiods at 0:00 (P ˂ 0.05), compared with 12L:12D group. To sum up, an appropriate photoperiod could improve eggshell quality, bone strength and mineral content through increasing osteogenesis during the light time and decreasing resorption activity during the dark, and 18 h is an adequate photoperiod for the eggshell and bone quality of laying ducks.

We investigated the effect of photoperiod on performance, ovarian morphology, reproductive hormones levels, and their receptors mRNA expressions in laying ducks. After adaption, 300 252-day-old Jinding laying ducks were randomly allocated to 5 groups, receiving 12L:12D, 14L:10D, 16L:8D, 18L:6D, or 20L:4D, respectively. Each treatment had 6 replicates of 10 birds each. The feeding trial lasted 8 wk. Egg production, egg mass, and ADFI increased linearly and quadratically with increasing photoperiods (P < 0.05), and the higher values of them occurred in photoperiods ≥ 16 h, compared with 12L:12D (P > 0.05). Initial and bare stroma weight increased quadratically, while total large white follicle (LWF) number and weight increased linearly and quadratically, with increasing photoperiods (P < 0.05). The higher values of them occurred in 16L:8D and 18L:6D treatments as well as the higher total LWF weight also occurred in 20L:4D, compared with 12L:12D (P > 0.05). Besides, 16.93 and 16.93 h were the optimal photoperiods for bare stroma (follicles ≥ 2 mm in diameter removed) weight and total LWF weight, respectively, calculated from reliable regression equations (R2 ≥ 0.5071). Compared with 12L:12D, the higher levels of estradiol, progesterone, follicle-stimulating hormone (FSH) as well as the higher expressions of estrogen, luteinizing hormone (LH) and progesterone receptors were observed in ≥16 h photoperiods (P < 0.05), while the higher LH level and FSH receptor expression only occurred in 16L:8D and 18L:6D (P < 0.05). In the hypothalamus, higher mRNA expression of gonadotropin-releasing hormone occurred in 16L:8D and 18L:6D groups (P < 0.05). Meanwhile, gonadotropin-inhibitory hormone and prolactin increased in 20-hour photoperiod (P < 0.05), and the latter may be due to theup-regulation of vasoactive intestinal peptide expression (P < 0.05). To sum up, an appropriate photoperiod could improve the performance and reproductive organ and ovarian follicles development through reproductive hormones and their receptors, and 16.56 to 10.93 h is an adequate photoperiod for laying ducks.

RESUMO

A large amount of sequencing data is produced in microbial community ecology studies using the high-throughput sequencing technique, especially amplicon-sequencing-based community data. After conducting the initial bioinformatic analysis of amplicon sequencing data, performing the subsequent statistics and data mining based on the operational taxonomic unit and taxonomic assignment tables is still complicated and time-consuming. To address this problem, we present an integrated R package-'microeco' as an analysis pipeline for treating microbial community and environmental data. This package was developed based on the R6 class system and combines a series of commonly used and advanced approaches in microbial community ecology research. The package includes classes for data preprocessing, taxa abundance plotting, venn diagram, alpha diversity analysis, beta diversity analysis, differential abundance test and indicator taxon analysis, environmental data analysis, null model analysis, network analysis and functional analysis. Each class is designed to provide a set of approaches that can be easily accessible to users. Compared with other R packages in the microbial ecology field, the microeco package is fast, flexible and modularized to use and provides powerful and convenient tools for researchers. The microeco package can be installed from CRAN (The Comprehensive R Archive Network) or github (https://github.com/ChiLiubio/microeco).

We evaluated the effect of photoperiod on ovarian morphology, reproductive hormone secretion, and hormone receptor mRNA expression in layer ducks during the pullet phase. A total of 480 71-d-old Jinding layer ducks were randomly divided into 5 groups that received 6L (hours of light):18D (hours of darkness), 8L:16D, 10L:14D, 12L:12D, or 14L:10D, respectively. Each group had 6 replicates with 16 birds each. The photoperiod feeding trial lasted 80 d until 150 d of age. The age at first egg (AFE), the total number, and weight of eggs increased linearly with increasing photoperiods (P < 0.05); lower values of AFE occurred with photoperiods ≥8 h, whereas a higher total number and weight of eggs occurred with photoperiods ≥10 h, compared with 6L:18D (P > 0.05). Oviduct weight, ovary percentage, and initial and bare stroma (weight and percentage) increased quadratically with increasing photoperiods (P < 0.05), and 10.24, 10.01, and 10.10 h were the optimal photoperiods for oviduct weight, bare stroma (follicles ≥2 mm in diameter removed) weight, and bare stroma percentage, respectively, as calculated from reliable regression equations (R2 ≥ 0.5791). Compared with 6L:18D, 10L:14D had a higher total large white follicle weight, small yellow follicle number, and weight (P < 0.05). In addition, higher serum levels of follicle-stimulating hormone, luteinizing hormone, and progesterone were observed with ≥10-h photoperiods (P < 0.05), as were levels of hormone receptor mRNA expression in ovarian follicles (P < 0.05), with the highest values for both measures at 10L:14D. In the hypothalamus, mRNA expression of gonadotropin-releasing hormone increased in ≥8-h photoperiods, with the highest value at 10L:14D. In contrast, gonadotropin-inhibitory hormone increased in photoperiods ≥12 h (P < 0.05). In conclusion, an appropriate photoperiod led to early sexual maturity and improved the development of reproductive organs and ovarian follicles through effects on reproductive hormones and their receptors; 10 to 10.24 h is an adequate photoperiod for layer ducks during the pullet phase.

This study aimed to evaluate the effect of photoperiod on growth performance and bone development in layer ducks during the pullet phase. A total of 480 11-wk-old Jinding layer ducks were randomly divided into 5 groups, receiving 6L (light): 18D (dark), 8L: 16D, 10L: 14D, 12L: 12D, 14L: 10D, respectively. Each group had 6 replicates of 16 birds each. The feeding trial lasted 80 d until 150 d of age. The results showed that 8L: 16D increased body weight (BW) and average daily gain (ADG; P < 0.05) compared with 6L:18D, whereas average daily feed intake, feed conversion ratio, and mortality rate had no difference among all treatments (P > 0.05). The midpoint perimeter (tibia and femur) and work to fracture (Wu; tibia) increased quadratically (P < 0.05), in response to photoperiod increment. Compared with 6L:18D, 8L:16D had higher tibia midpoint perimeter, ultimate load (Fu), and cortical cross-sectional area (P < 0.05), accompanied with higher content of total Ca, distal bone mineral content (BMC), and cortical volumetric BMC (vBMC; P < 0.05). A linear increase was observed in trabecular volumetric bone mineral density (vBMD) and vBMC (tibia and femur), elastic energy (femur), work to fracture Wu and young modulus of elasticity E (tibia) in response to photoperiod increase (P < 0.05), and treatment with 14L:10D had higher ultimate load Fu and total Ca of tibia (P < 0.05). Besides, estradiol and testosterone levels (serum), estrogen receptor gene mRNA expression (bone tissue), alkaline phosphatase and tartrate-resistant acid phosphatase activities, transforming growth factor-ß, osteocalcin and tumor necrosis factor-α contents increased in ≥12 h photoperiods (P < 0.05). In summary, appropriate photoperiod increased BW, ADG, cortical bone generation, and bone mineralization, whereas only medullary bone formation increased with photoperiods. These results suggest that 8L:16D could be an adequate photoperiod for layer ducks' growth and bone development during the pullet phase.

RESUMO

The objective of this study was to evaluate the effects of dietary supplementation with Moringa oleifera leaf (MOL) on performance, carcass characteristics, meat quality, and oxidative stability of breast muscle in broilers. A total of 720 1-d-old male Arbor Acres birds were randomly divided into 6 dietary groups, which were fed a basal diet supplemented with 0, 1, 2, 5, 10, and 15% MOL, respectively. Each group had 6 replicates of 20 birds each. The feeding trial lasted for 42 d. The results showed dietary MOL supplementation linearly and quadratically decreased body weight and average daily gain (P < 0.01), and increased feed conversion ratio (P < 0.001). Abdominal fat decreased linearly and quadratically in response to the supplementation of MOL in diets, both on d 21 and 42 (P < 0.001). In breast muscle, dietary supplementation with MOL quadratically increased the contents of C18:2, C18:3n-3, C20:4, polyunsaturated fatty acids (PUFA), n-3 PUFA, n-6 PUFA (P < 0.01), and decreased thrombogenic index (TI; P = 0.019). Dietary inclusion of MOL improved meat color, evidenced by quadratically reduced b* (yellowness) values (45 min postmortem, P = 0.001; 24 h postmortem, P = 0.018) and increased a* (redness) values (24 h postmortem, P < 0.001). Besides, diets supplemented with MOL quadratically decreased malondialdehyde (MDA) levels in breast muscle during storage (P < 0.001). Plasma total anti-oxidative capacity, total superoxide dismutase, glutathione peroxidase activities increased quadratically (P < 0.01), whereas MDA decreased quadratically (P < 0.001), in response to dietary MOL supplementation. In summary, MOL could be used as a feed ingredient for broilers to improve PUFA contents, oxidative stability, color of breast muscle, and abdominal fat without adverse effects on growth performance, with an inclusion of 1.56% in the diets.

The objective of this study was to determine the preliminary characteristics and protective effects of alfalfa polysaccharides (APS) on hepatocytes in vitro. The crude APS was purified by DEAE-cellulose and Sephadex G-100 chromatography, resulting in the four purified fractions: APS-1, APS-2, APS-3 and APS-4. The results indicated that APS-3 had higher carbohydrate and uronic acid contents and that APS-4 had a more complicated monosaccharide composition compared to the other purified fractions. The average molecular weights of APS-1, APS-2, APS-3 and APS-4 were 48,536, 6,221, 66,559 and 13,076 Da, respectively. Furthermore, APS (crude and its purified fractions) restored the activities of antioxidant enzymes and increased the total antioxidant capacity of hepatocytes subjected to H2O2-induced oxidative stress. Furthermore, APS treatment counteracted the increases in lactic dehydrogenase and malonaldehyde in the culture supernatant. These results clearly demonstrate that APS possesses a protective effect against oxidative injury in hepatocytes.
