BACKGROUND: Triticale is making its way on dairy farms as an alternative forage crop. This requires the availability of high-yielding triticale varieties with good digestibility. Triticale forage breeding mainly focussed on biomass yield, but efforts to improve digestibility are increasing. We previously investigated the interrelationships among different quality traits in soft dough triticale: starch, acid detergent fibre and in vitro digestibility of organic matter (IVOMD) and of neutral detergent fibre (IVNDFD) of the total plant, IVNDFD and Klason lignin of the stems, and ear proportion and stem length. Here we determine the genetic control of these traits, using a genome-wide association (GWAS) approach. A total of 33,231 DArTseq SNP markers assessed in a collection of 118 winter triticale genotypes, including 101 varieties and 17 breeding lines, were used. RESULTS: The GWAS identified a total of 53 significant marker-trait associations (MTAs). The highest number of significantly associated SNP markers (n = 10) was identified for total plant IVNDFD. A SNP marker on chromosome 1A (4211801_19_C/T; 474,437,796 bp) was found to be significantly associated with ear proportion, and plant and stem IVNDFD, with the largest phenotypic variation for ear proportion (R²p = 0.23). Based on MTAs, candidate genes were identified which were of particular relevance for variation in in vitro digestibility (IVD) because they are putatively involved in plasma membrane transport, cytoskeleton organisation, carbohydrate metabolic processes, protein phosphorylation, and sterol and cell wall biogenesis. Interestingly, a xyloglucan-related candidate gene on chromosome 2R, SECCE2Rv1G0126340, was located in close proximity of a SNP significantly associated with stem IVNDFD. Furthermore, quantitative trait loci previously reported in wheat co-localized with significantly associated SNP markers in triticale. CONCLUSIONS: A collection of 118 winter triticale genotypes combined with DArTseq SNP markers served as a source for identifying 53 MTAs and several candidate genes for forage IVD and related traits through a GWAS approach. Taken together, the results of this study demonstrate that the genetic diversity available in this collection can be further exploited for research and breeding purposes to improve the IVD of triticale forage.

Hydrolysable tannins (HT) show potential as silage additive for autumn herbage silages, high in (rumen degradable) protein, as they may reduce proteolysis. Additionally, they have abilities to form pH-reversible tannin-protein complexes, non-degradable in the rumen but degradable in the abomasum and intestines of ruminants. Therefore they can improve milk N efficiency and shift N excretions from urine to faeces, possibly mitigating the environmental impact of ruminants. In this study, two small bunker silos were filled with autumn grass. One was treated with 20 g/kg DM HT extract (TAN) (TannoSan-L), the other with 8 mg/kg DM inoculant containing lactic acid bacteria (INO) (Bonsilage Fit G). Secondly, micro-silos (2.75 L) were filled with four treatments; (1) grass without additive (CON) (n = 5); (2) TAN (n = 5); (3) INO (n = 5); and (4) TAN + INO (n = 5). The bunker silos were used in a cross-over feeding experiment with periods of 4 weeks involving 22 lactating Holstein cows (average ± SD: 183 ± 36.3 days in milk, 665 ± 71.0 kg body weight, and 33.8 ± 3.91 kg/day milk yield). The HT dose was insufficient to reduce proteolysis or alter chemical composition and nutritional value in the micro- and bunker silages. Including grass silage added with TAN (3.2 g HT/kg DM) in the diet, did not affect feed intake nor fat and protein corrected milk yield in comparison to feeding the grass silage added with INO in a similar diet. The TAN-fed cows had an increased faecal N excretion and decreased apparent total-tract N and organic matter digestibility, but no improvement in the cows' N utilization could be confirmed in milk and blood urea levels. Overall, feeding an autumn grass silage treated with 20 g/kg chestnut HT extract did not affect the performance of dairy cows in comparison to feeding an autumn grass silage treated with a lactic acid bacteria inoculant.

Cereal forages, such as triticale forage, progressively gain interest as alternative crop for maize. The main study objective was to investigate the variation in potential feeding value of triticale forage among maturity stage, growing season and genotype, using total plant and stem fractions. Therefore, near infrared spectroscopy (NIRS) was evaluated as fast screening tool. The prediction ability was good (ratio of prediction to deviation, RPD ≥3.0) for total plant residual moisture, starch, sugars and for stem crude ash (CAsh) and neutral detergent fibre (aNDFom); suitable for screening (2.0 ≤ RPD <3.0) for total plant CAsh, acid detergent fibre (ADFom), in vitro digestibility of organic matter (IVOMD), in vitro digestibility of neutral detergent fibre (IVNDFD) and for stem total lignin (TL) and IVNDFD; poor (1.5 ≤ RPD <2.0) for total plant crude protein, crude fat, aNDFom, lignin (sa) and for stem Klason lignin (KL); unreliable (RPD <1.5) for stem residual moisture and acid soluble lignin (ASL). The evolution in potential feeding value of 36 genotypes harvested at the medium and late milk to the early, soft and hard dough stage was followed. The most important changes occurred between the late milk and early dough stage, with little variation in quality after the soft dough stage. During 2 growing seasons, variation in feeding value of 120 genotypes harvested at the soft dough stage was demonstrated. Interestingly, variation in stem IVNDFD is almost twice as high as for the total plant (CV 12.4% versus 6.6%). Furthermore, Spearman correlations show no link between dry matter yield and digestibility of genotypes harvested at the soft dough stage. Based on linear regression models ADFom appears as main predictor of both plant IVOMD and plant IVNDFD. Stem IVNDFD is particularly determined by KL.

Lowering the CP level in piglet diets reduces the risk of postweaning diarrhea and N excretion to the environment. The question remains at what point CP becomes limiting. An experiment was designed with 2 standardized ileal digestible (SID) Lys levels (10 and 11 g) and 6 CP levels (140, 150, 160, 170, 180, 190 g/kg) in a 2 × 6 factorial design (with 6 pens of 6 animals each per treatment). Linear and quadratic (QP) mixed models of performance in function of CP were fitted to study the effect of SID Lys and CP and their interaction. To determine optima, QP models and broken line models with linear (BLL) or quadratic (BLQ) ascending portions were fitted through the data. It was hypothesized 1) that the response to a decreasing digestible CP level could be described with broken line models and 2) that the break point of these models is dependent on the dietary SID Lys level. Decreasing the CP level decreased ADG (P < 0.001). For G:F, the effect of decreasing CP level depended on the SID Lys level (P of the interaction = 0.028 in the linear model and P = 0.002 in the QP model). According to the BLL model, with 11 g SID Lys in the diet, G:F started to decline with CP levels < 176 g CP [SID Lys:CP = 0.062, SID Lys:apparent total tract digestible (ATTD) CP = 0.077], and with 10 g SID Lys, CP levels < 165 g/kg (SID Lys:CP = 0.061, SID Lys:ATTD CP = 0.075) depressed performance. Serum creatinine levels showed a linear decrease with increasing SID Lys:CP levels (P < 0.001). Across both SID Lys levels, when fitting a BLL model, minimal serum urea levels were reached at an SID Lys:CP ratio of 0.064. This seems to be the point where CP and not Lys limits muscle deposition. The small difference in break point between serum urea level and performance suggests that the composition of nonessential AA may also be at stake. The effect of decreasing CP level depends on SID Lys, and using a maximal SID Lys:CP ratio may be useful for optimizing the AA profile of dietary CP. When the SID Lys:CP ratio exceeds 0.064 (SID Lys:ATTD CP > 0.079), protein and not individual AA limits growth in most piglets between 4 and 9 wk of age.

Sixty non-pregnant, non-lactating double-muscled Belgian Blue (DMBB) cows were used to estimate the energy required to maintain body weight (BW). They were fed one of three energy levels for 112 or 140 days, corresponding to approximately 100%, 80% or 70% of their total energy requirements. The relationship between daily energy intake and BW and daily BW change was developed using regression analysis. Maintenance energy requirements were estimated from the regression equation by setting BW gain to zero. Metabolizable and net energy for maintenance amounted to 0.569 ± 0.001 and 0.332 ± 0.001 MJ per kg BW(0.75)/d, respectively. Maintenance energy requirements were not dependent on energy level (p > 0.10). Parity affected maintenance energy requirements (p < 0.001), although the small numerical differences between parities may hardly be nutritionally relevant. Maintenance energy requirements of DMBB beef cows were close to the mean energy requirements of other beef genotypes reported in the literature.

Effective assessment of sulphur (S) status in cattle is important for optimal health, yet remains difficult. Rumen fluid S concentrations are preferred, but difficult to sample under practical conditions. This study aimed to evaluate salivary S concentration as estimator of S status in cattle. Saliva and rumen fluid samples were collected from dairy cows (n = 16) as well as samples of different feedstuffs offered to the animals. The N and S concentrations were determined using the Dumas technique. The average dietary N and S content were calculated as well as N:S ratio of saliva, rumen fluid and diet. Salivary S concentrations could not predict rumen fluid or dietary S concentrations (p > 0.05). The log transformed salivary N:S ratio (x) could predict the rumen fluid N:S ratio (y) with a linear equation of y = 9.83 (±4.59) x + 0.39 (±4.56) (r = 0.497, p = 0.05), but left too much residual variation to serve as reliable predictor. Further research should investigate this relationship in the search for an S status estimator.

A multianalyte method was developed to identify and quantitate 26 mycotoxins simultaneously in maize silage by means of ultrahigh-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). The extraction and cleanup procedure consists of two extraction steps followed by purification on a Waters Oasis HLB column. The method developed was validated with the requirements of Commission Decision 2002/657/EC taken into account. The limit of detection and quantitation ranges were 5-348 and 11-695 ng/g, respectively. Apparent recovery varied between 61 and 116%, whereas repeatability and reproducibility were within the ranges of 3-45 and 5-49%, respectively. The method developed was successfully applied for maize silage samples taken at the cutting surface and 1 m behind that surface. Mainly Fusarium toxins (beauvericin, deoxynivalenol, enniatins, fumonisins, fusaric acid, and zearalenone) were detected, but postharvest toxins such as mycophenolic acid and roquefortine C were identified as well.

To reduce N pollution by dairy cows, two levels of rumen degradable protein balance (RDPB) and the use of protected protein sources were studied in two feeding trials. Trial 1 investigated if the effect of lowering the RDPB, at a constant DPI (digestible protein in the intestine) supply, could be compensated by using protected protein sources (formaldehyde treated (= formolated) soybean meal (FSBM) or a combination of FSBM and formolated rapeseed meal at a ratio of 60:40). Trial 2 studied if FSBM is as effective as soybean meal (SBM) at a low RDPB level. Each trial was set up as a Latin square design, with three treatments and three periods, involving 18 lactating Holstein cows. Reducing RDPB from about 150 g/d to below -40 g/d resulted in small negative effects on the milk production, independent of the protein source (protected or not). However, N excretion per kg fat and protein corrected milk (FPCM) and N efficiency were both improved. At a low RDPB level, the performance of FSBM (35.0 kg FPCM) was no better than after feeding a combination of FSBM and formolated rapeseed meal (34.7 kg FPCM). The use of FSBM (32.8 kg FPCM and 34.2% N efficiency) was as effective as SBM (32.5 kg FPCM and 34.2% N efficiency). As such, SBM use can approximately be halved by using FSBM, possibly in combination with locally produced rapeseed meal.

Feeding measures with a potential to improve N efficiency in dairy cattle husbandry were studied at two levels of undegradable protein balance (OEB). In each of the two experiments, two simultaneous Latin squares were conducted, each with three treatments and three lactating Holstein cows. Decreasing the OEB of the diet improved N efficiency and resulted in lower N excretion per kg milk. To avoid a negative effect of the decreased OEB on the production results, spreading the concentrate intake (as TMR or in five meals) seemed to be most promising, although only few significant effects were found. The use of protected protein sources or the addition of clinoptilolite were not successful in improving N-efficiency. The rather well balanced mixed basal diet and the large variation between animals seem to have hampered the assessment of such significant influences.

Weaning at a different daily concentrate intake was investigated during a 140-d experimental period, using 54 male and 68 female newborn Belgian Blue double-muscled animals. They were divided into three comparable groups and received milk at 10% of their birth weight up to weaning. Concentrate was levelled off at a maximum daily intake of 3 kg, while grass hay was freely available. Weaning occurred at a daily concentrate intake level (CL) of 0.5, 0.75 and 1.0 kg, respectively. Weaning at an increased CL prolonged the milk-feeding period by 13.1 and 14.6 days, and resulted in a higher pre- and post-weaning growth rate (p < 0.05). Daily gain during the entire experimental period averaged 0.84, 0.85 and 0.88 kg for the respective groups (p = 0.065). Daily concentrate intake was not different among groups, with only a small effect of CL on intake around weaning. Early weaning resulted in a significant reduction of hay intake (p = 0.032). Total daily net energy intake increased slightly with a higher CL at weaning, so that energy conversion was slightly improved, amounting to 17.7, 17.6 and 17.4 MJ/kg gain, respectively. Energy balance during the first week after weaning was negative for CL 0.5 kg (-22%), while it was close to 0 for CL 0.75 kg (-2%) or positive for CL 1.0 kg. Most effects of CL at weaning were similar for males and females, but male calves tended to have a higher intake and a faster growth rate than females. It can be concluded that weaning should be delayed until Belgian Blue double-muscled calves consume at least 0.75 kg per day or more for reasons of welfare, although performance was hardly improved by weaning at a daily concentrate intake of more than 0.5 kg per day.
