Aptamers, as single-stranded DNA or RNA fragments, have been widely applied as the bio-recognition element for fabrication of flexible and reliable aptasensors to be used in food safety control, clinical therapy and diagnosis and environment monitoring fields. With increasingly fierce antibiotics resistance appearing as a worldwide problem, a highly efficient method is urgently needed to detect antibiotics residues in animal-sourced food. Herein, a simply operated aptasensor based on quantitative real-time PCR (qRT-PCR) was fabricated to realise the simultaneous detection of two antibiotics (i.e. chloramphenicol and kanamycin). The limit of detection (LOD) of 6.13 ng/mL for chloramphenicol and of 19.17 ng/mL for kanamycin of this dual-aptasensor were achieved. Actually, such LOD values were not as good as that of an aptasensor individually established for each antibiotic. The circular dichroism analysis suggested that in the dual-aptasensor, adjacent aptamers might disturb each other's binding with their respective target. Although certain detection sensitivity was lost, the dual-aptasensor could still fulfil the detection requirements, and more importantly, it would improve the detection efficiency. Finally, this dual-aptasensor was applied for detecting chloramphenicol and kanamycin in real spiked food samples, and results indicated good recovery rates. These results demonstrated this developed dual-aptasensor to be a promising highly efficient method with low cost for simultaneous detection of chloramphenicol and kanamycin residues in animal-sourced food samples.

Tinnitus may cause anxiety, depression, insomnia, which impair the quality of life of millions worldwide. However, the mechanism of tinnitus remains to be understood, it has been previously hypothesized that the activation of N-methyl-D-aspartate (NMDA) receptor is involved in the tinnitus processes and blockade of the NMDA receptor is regarded as a therapeutic strategy for tinnitus treatment even if the rescue treatment is still proved invalid in some cases. To demonstrate the therapeutic effect of the NMDA receptor blocker on tinnitus, we examined here the spontaneous firing rate (SFR) and the neurochemical dynamics in the auditory cortex (AC) of rats after sodium salicylate (SS) injection, which is a widely used model for tinnitus research. We also recorded their responses to MK-801 treatment. Electrophysiological studies showed that MK-801 significantly suppresses SFR in AC of rats with SS-induced tinnitus. In addition, by using a technique that combining in vivo microdialysis with an online electrochemical system (OECS) and a high-performance liquid chromatography (HPLC), we found that the levels of both glutamate and ascorbate in AC dramatically increased after SS injection and that MK-801 administration attenuated those response. Further studies found that MK-801 given at a time point of 30 min pre- or post-injection of SS were more effective than that given at a time point of 60 min post-SS injection, indicating that the time point of MK-801 intervention has a critical impact on the therapeutic effect. These findings suggest that MK-801 plays a neuroprotective role against hyperactivity during tinnitus induced by SS and that the therapeutic effect depends on the time point of MK-801 intervention, which would advance the studies on understanding of the therapeutic potential of NMDA receptor antagonist in tinnitus therapy.

BACKGROUND: Tinnitus is a common disorder that causes significant morbidity; however, the neurophysiological mechanism is not yet fully understood. A relationship between tinnitus and limbic system has been reported. As a significant component of the limbic system, the hippocampus plays an important role in various pathological processes, such as emotional disturbance, decreased learning ability, and deterioration of memory. This study was aimed to explore the role of the hippocampus in the generation of tinnitus by electrophysiological technology. METHODS: A tinnitus model was established in rats through intraperitoneal injection of salicylate (SA). Subsequently, the spontaneous firing rate (SFR) of neurons in the hippocampal CA1 area was recorded with in vivo multichannel recording technology to assess changes in excitability induced by SA. To investigate the effect of excitability changes of hippocampus on the auditory pathway, the hippocampus was electrically stimulated and neural excitability in the auditory cortex (AC) was monitored. RESULTS: Totally 65 neurons in the hippocampal CA1 area were recorded, 45 from the SA group (n = 5), and 20 from the saline group (n = 5). Two hours after treatment, mean SFR of neurons in the hippocampal CA1 area had significantly increased from 3.06 ± 0.36 Hz to 9.18 ± 1.30 Hz in the SA group (t = -4.521, P < 0.05), while no significant difference was observed in the saline group (2.66 ± 0.36 Hz vs. 2.16 ± 0.36 Hz, t = 0.902, P > 0.05). In the AC, 79.3% (157/198) of recorded neurons showed responses to electrical stimulation of the hippocampal CA1 area. Presumed pyramidal neurons were excited, while intermediate neurons were inhibited after electrical stimulation of the hippocampus. CONCLUSIONS: The study shows that the hippocampus is excited in SA-induced tinnitus, and stimulation of hippocampus could modulate neuronal excitability of the AC. The hippocampus is involved in tinnitus and may also have a regulatory effect on the neural center.

Four methicillin-resistant coagulase-negative staphylococci (MRCoNS), one Staphylococcus haemolyticus and three Staphylococcus cohnii, from infections of humans collected via the Ministry of Health National Antimicrobial Resistance Surveillance Net (Mohnarin) program in China were identified as linezolid-resistant. These four isolates were negative for the 23S rRNA mutations, but positive for the gene cfr. Mutations in the gene for the ribosomal protein L3, which resulted in the amino acid exchanges Gly152Asp and Tyr158Phe, were identified in S. haemolyticus 09D279 and S. cohnii NDM113, respectively. In each isolate, the cfr gene was located on a plasmid of ca. 35.4 kb, as shown by S1 nuclease pulsed-field gel electrophoresis and Southern blotting experiments. This plasmid was indistinguishable from the previously described plasmid pSS-02 by its size, restriction pattern, and a sequenced 14-kb cfr-carrying segment. Plasmid pSS-02 was originally identified in staphylococci isolated from pigs. This is the first time that a cfr-carrying plasmid has been detected in MRCoNS obtained from intensive care patients in China. Based on the similarities to the cfr-carrying plasmid pSS-02 from porcine coagulase-negative staphylococci, a transmission of this cfr-carrying plasmid between staphylococci from pigs and humans appears to be likely.

A platinum plate coated with single-walled carbon nanotubes (SWCNTs@Pt) was prepared by means of electrophoretic deposition. Using the SWCNTs@Pt plate, an electrosorption-enhanced solid-phase microextraction (EE-SPME) technique was proposed for the extraction of trace anions in water, described as follows: a positive potential was applied to the SWCNTs@Pt plate to extract F(-), Cl(-), Br(-), NO(3)(-) and SO(4)(2-) from water using electrosorption, and then a negative potential was applied to the plate placed in ultra-pure water for the desorption of the absorbed anions, and finally the desorbed anions were analyzed using ion chromatography (IC). The EE-SPME parameters, including extraction potential and time as well as desorption potential and time, were investigated. An analytical method based on the above procedures, i.e., EE-SPME-IC, was established and used for the analysis of trace anions in water. The results showed that the application of potential on the SWCNTs@Pt plate significantly enhanced the ion extraction efficiency, and an enrichment factor of 15-38 was achieved. The SWCNTs@Pt plate could be used more than 50 times without significant decay. The linear range, the limit of detection (S/N=3), the limit of quantification (S/N=10) and repeatability (n=7) of our EE-SPME-IC method were 1.0-150.0 µg/L, 0.06-0.26 µg/L, 0.19-0.85 µg/L and 2.1-8.0%, respectively. The proposed method was successfully applied for the analysis of trace anions in deionized water, and acceptable recoveries between 65.3 and 121.1% were obtained for the spiked deionized water samples.
