Loxosceles spp. (brown spiders) bites are responsible for the development of a syndrome consisting mainly of dermonecrotic lesions, and also systemic effects. Rabbits are one of the main experimental models used for better understanding the systemic and local effects of Loxosceles venom. The aim of this study is to evaluate the toxic and protective effects of rabbits immunized with Loxosceles spp. venom. Male New Zealand rabbits were allocated as a control group (CG; n = 5) that received adjuvant (Montanide) and phosphate-buffer saline (PBS), or as venom group (VG; n = 5) that received 21 µg of Loxosceles venom using Montanide as adjuvant. After five immunization cycles, a trial with 7 µg of Loxosceles intermedia (L. intermedia) venom was performed, and dermonecrotic lesions were measured. The rabbits were then euthanized, and their organs were collected for histopathology analysis. Rabbits that had undergone Loxosceles venom immunization protocol showed minor clinical disturbances during the experimental period. The used immunization protocol protected the rabbits against the toxic effect of the Loxosceles venom because they showed minor clinical disturbances during the experimental period.

Canine brucellosis is an infectious and zoonotic disease caused by Brucella canis, which has been reported worldwide, and is a major public health concern due to close contact between dogs and humans. In dogs, canine brucellosis manifests with abortion outbreaks, reproductive failure, enlargement of lymph nodes, and occasionally affects the osteoarticular system, although the occurrence of asymptomatic infections in dogs are not uncommon. In humans, the disease is associated with a febrile syndrome, commonly with non-specific symptoms including splenomegaly, fatigue, and weakness. Infection of dogs occurs mostly by the oronasal route when in contact with contaminated tissues such as aborted fetuses, semen, urine, and vaginal secretions. In humans, contact with contaminated fluids from infected dogs is an important source of infection, and it is an occupational risk for veterinarians, breeders, laboratory workers, among other professionals who deal with infected animals or biological samples. The diagnosis in dogs is largely based on serologic methods. However, serologic diagnosis of canine brucellosis remains very challenging due to the low accuracy of available tests. Molecular diagnostic methods have been increasingly used in the past few years. Treatment of infected dogs is associated with a high frequency of relapse, and should be employed only in selected cases. Currently there are no commercially available vaccines for prevention of canine brucellosis. Therefore, development of novel and improved diagnostic methods as well as the development of efficacious and safe vaccination protocols are needed for an effective control of canine brucellosis and its associated zoonotic risk.

Listeria monocytogenes is a cause of infectious food-borne disease in humans, characterized by neurological manifestations, abortion, and neonatal septicemia. It is intracellular bacterium, which limits the development of protective inactivated vacines. Adjuvants capable of stimulating cellular immune response are important tools for developing novel vaccines against intracellular bacteria. The aim of this study was to evaluate the vaccine potential of L. monocytogenes inactivated by gamma irradiation (KLM-γ) encapsulated in alginate microcapsules associated or not with chitosan against listeriosis in the murine model. At the fourth day after challenge there was a reduction in bacterial recovery in mice vaccinated with KLM-γ encapsulated with alginate or alginate-chitosan, with lower bacterial loads in the spleen (10 fold) and liver (100 fold) when compared to non-vaccinated mice. In vitro stimulation of splenocytes from mice vaccinated with alginate-chitosan-encapsulated KLM-γ resulted in lymphocyte proliferation, increase of proportion of memory CD4+ and CD8+ T cell and production of IL-10 and IFN-γ. Interestingly, the group vaccinated with alginate-chitosan-encapsulated KLM-γ had increased survival to lethal infection with lower L. monocytogenes-induced hepatic inflammation and necrosis. Therefore, KLM-γ encapsulation with alginate-chitosan proved to have potential for development of novel and safe inactivated vaccine formulations against listeriosis.

BACKGROUND/OBJECTIVES: Vaccination is the most important tool for controlling brucellosis, but currently there is no vaccine available for canine brucellosis, which is a zoonotic disease of worldwide distribution caused by Brucella canis. This study aimed to evaluate protection and immune response induced by Brucella ovis ΔabcBA (BoΔabcBA) encapsulated with alginate against the challenge with Brucella canis in mice and to assess the safety of this strain for dogs. METHODS: Intracellular growth of the vaccine strain BoΔabcBA was assessed in canine and ovine macrophages. Protection induced by BoΔabcBA against virulent Brucella canis was evaluated in the mouse model. Safety of the vaccine strain BoΔabcBA was assessed in experimentally inoculated dogs. RESULTS: Wild type B. ovis and B. canis had similar internalization and intracellular multiplication profiles in both canine and ovine macrophages. The BoΔabcBA strain had an attenuated phenotype in both canine and ovine macrophages. Immunization of BALB/c mice with alginate-encapsulated BoΔabcBA (108 CFU) induced lymphocyte proliferation, production of IL-10 and IFN-γ, and protected against experimental challenge with B. canis. Dogs immunized with alginate-encapsulated BoΔabcBA (109 CFU) seroconverted, and had no hematologic, biochemical or clinical changes. Furthermore, BoΔabcBA was not detected by isolation or PCR performed using blood, semen, urine samples or vaginal swabs at any time point over the course of this study. BoΔabcBA was isolated from lymph nodes near to the site of inoculation in two dogs at 22 weeks post immunization. CONCLUSION: Encapsulated BoΔabcBA protected mice against experimental B. canis infection, and it is safe for dogs. Therefore, B. ovis ΔabcBA has potential as a vaccine candidate for canine brucellosis prevention.

Brucella ovis causes economic and reproductive losses in sheep herds. The goal of this study was to characterize infection with B. ovis field isolates in a murine model, and to evaluate protection induced by the candidate vaccine strain B. ovis ΔabcBA in mice challenged with these field isolates. B. ovis field strains were able to colonize and cause lesions in the liver and spleen of infected mice. After an initial screening, two strains were selected for further characterization (B. ovis 94 AV and B. ovis 266 L). Both strains had in vitro growth kinetics that was similar to that of the reference strain B. ovis ATCC 25840. Vaccination with B. ovis ΔabcBA encapsulated with 1% alginate was protective against the challenge with field strains, with the following protection indexes: 0.751, 1.736, and 2.746, for mice challenged with B. ovis ATCC25840, B. ovis 94 AV, and B. ovis 266 L, respectively. In conclusion, these results demonstrated that B. ovis field strains were capable of infecting and inducing lesions in experimentally infected mice. The attenuated vaccine strain B. ovis ΔabcBA induced protection in mice challenged with different B. ovis field isolates, resulting in higher protection indexes against more pathogenic strains.(AU)


Brucella ovis é responsável por perdas econômicas e reprodutivas em rebanhos ovinos. O objetivo deste trabalho foi caracterizar a infecção com as cepas isoladas de campo de B. ovis em modelo murino e avaliar a eficiência vacinal da mutante B. ovis ΔabcAB para proteção contra desafio com as cepas isoladas de campo. Foram utilizadas sete cepas isoladas de campo foram capazes de colonizar e provocar lesões no fígado e no baço de camundongos após sete dias pós-infecção. Após triagem, duas cepas foram selecionadas para a melhor caracterização (B. ovis 94 AV and B. ovis 266L). Ambas apresentaram crescimento em placa de cultivo semelhante ao da cepa de referência B. ovis ATCC 25840. A vacinação com a cepa de Brucella ovis ΔabcBA encapsulada com alginato a 1% foi capaz de proteger camundongos desafiados com as cepas isoladas de campo, com os seguintes índices de proteção: 0,751, 1,736 e 2,746, para camundongos desafiados com B. ovis ATCC 25840, B. ovis 94 AV e B. ovis 266 L, respectivamente. Estes resultados demonstraram que as cepas isoladas de campo de B. ovis são capazes de infectar e induzir lesão em camundongos experimentalmente infectados. O uso da cepa mutante atenuada B. ovis ΔabcBA para vacinação de fêmeas C57BL/6 desafiados com diferentes cepas de B. ovis induziu proteção nos camundongos desafiados com diferentes cepas de B. ovis. Deste modo, mostrando-se eficiente na proteção das cepas de campo de B. ovis.(AU)

Canine brucellosis is an infectious and contagious disease associated with reproductive losses in breeding kennels. As a zoonotic disease, it poses a risk to human health, especially for veterinarians and breeders who handle materials potentially contaminated with Brucella canis. However, canine brucellosis is a neglected and underestimated disease given the difficulties in establishing a definitive diagnosis. We evaluated the frequency of detection of B. canis in 5 breeding kennels by using various serologic methods and PCR. Circulation of B. canis in these kennels was confirmed by bacterial isolation. The frequency of positive serologic results varied from 6.3% by AGID to 16.5% by dot-ELISA. There was no positive serology for smooth Brucella. PCR testing was positive in 13.9% of samples. The only detection tests with reasonable agreement were PCR and 2ME-MAT. The diagnosis of canine brucellosis remains challenging. The use of a single laboratory method, or even the use of different laboratory methods, may not be sufficient to reach a definitive diagnosis.

ABSTRACT: Ruminants vary their walking trajectory according to the positioning of the trees in integrated systems, which can determine the concentration of dung deposition at certain locations. The aim of this study was to map the distribution of feces and the behavior of dairy heifers in sunny and shaded areas. This experiment was carried out in Mato Grosso, Brazil, where the main grazing conditioner is shade due to the presence of trees in the pastures. Shading levels used were: full sunlight (control), moderate shade (338 trees ha−1) and intensive shade (714 trees ha−1) in randomized complete blocks. The experimental period was divided into three evaluation periods based on rainfall distribution: rainy period = December 2012; transition period = March 2013 and dry period = June 2013. Animal behavior assessments and dung distribution mapping were performed. The full sunlight system displayed a higher concentration of feces patches at sites near the gate, cow drinkers and fences opposite the gate. Heifers picked shaded places for ruminating and idleness. The major concentration was reported in the central area and under trees, in shaded systems. There were places with greater feces concentrations, but when the pasture had trees, deposition did not only happen underneath trees but also in places under their influence. The distribution was more homogeneous when trees were present in large quantities. The shaded area available in pastures affects spatial distribution of dung, stimulating uniformity.


RESUMO: Os ruminantes variam a trajetória de sua caminhada de acordo com o posicionamento das árvores em sistemas integrados. A deposição de fezes pode estar mais ou menos concentrada em determinados locais, de acordo com a disposição desses condicionadores de pastejo que definem o caminhamento animal. O objetivo deste experimento foi mapear a localização e a distribuição de fezes de bezerras leiteiras de acordo com seu comportamento afetado pela disponibilidade de áreas ensolaradas e sombreadas. Este experimento foi conduzido em Mato Grosso, no Brasil. O principal condicionador de pastejo foi o sombreamento promovido pela presença de árvores nas pastagens. Foram impostos níveis de sombreamento: pleno sol (controle), sombreamento moderado (338 árvores ha− 1) e sombreamento intenso (714 árvores ha− 1) em blocos completos casualizados. O período experimental foi dividido em períodos de avaliação com base na distribuição das chuvas: Período Chuvoso = dezembro de 2012; Período de Transição = Março de 2013 e Período Seco = Junho de 2013. Foram realizadas avaliações de comportamento animal e mapeamento da distribuição das placas de fezes na área. Para atividades de ruminação e lazer, as novilhas preferiram os locais mais sombreados. O sistema pleno sol promoveu maior concentração de placas de fezes em locais próximos a porteiras e bebedouros, no lado oposto ao da porteira. Nos sistemas sombreados, a maior concentração foi na área central e sob sombra. Havia locais com maior concentração de fezes, mas quando havia árvore na pastagem, a deposição não acontecia apenas abaixo da árvore, mas também nos locais sob sua influência. A distribuição foi mais homogênea quando as árvores estavam em maiores quantidades. A área de sombra disponível no pasto afeta a distribuição espacial do esterco estimulando a uniformidade.

Nas últimas décadas o mercado brasileiro de carne ovina aumentou substancialmente, e o estado de Mato Grosso apresentou a maior taxa de crescimento do Brasil, apesar da falta de informações sobre os sistemas de produção nesta região. Este estudo teve como objetivo caracterizar os sistemas de produção de ovinos da região meio-norte do Mato Grosso, utilizando um questionário com perguntas qualitativas e quantitativas. Dentre as 40 propriedades avaliadas foram predominantes (82,5%; 33/40) rebanhos com menos de 100 animais, com exploração semi-intensiva, voltada para a produção de carne. A ovinocultura foi citada como atividade econômica secundária em 92,5% (37/40) dos rebanhos e, em 60,0% (24/40) das propriedades, os animais eram mantidos apenas para consumo familiar. O manejo sanitário deficiente resultou na ocorrência de diversas doenças, sendo os casos de miíases os mais citados (65,0%; 26/40). A ovinocultura possui grande potencial para o desenvolvimento em Mato Grosso. No entanto, a atividade ainda é negligenciada em muitos aspectos, incluindo a saúde animal.


In recent decades, the Brazilian market for sheep meat has increased substantially, and the State of Mato Grosso had the highest growth rate in Brazil. However, information about ovine production systems in this region is lacking. This study aimed to characterize productive systems of sheep in the medium northern region of Mato Grosso, by application of qualitative and quantitative questionnaires. Among 40 properties evaluated, herds there were predominately with fewer than 100 animals (82.5%, 33/40), with semi-intensive exploration focused on meat production. Livestock sheep was a secondary economic activity in 92.5% (37/40) of farms, and sheep were kept only for family consumption in 60.0% (24/40). Poor health management resulted in the occurrence of various diseases, and myiasis was the most frequently cited disease (65.0%, 26/40). Sheep farming has a great potential for development in Mato Grosso. However, the activity is neglected in many ways, including animal health.

The intensification of the livestock production system has gained prominence over the last decades. In addition to the reduction of grazing areas and increased productivity per hectare, the intercropping involving forest tree species and ruminants has been established as a sustainable production model, generating income and valuation of natural capital. Besides the social, economic, and environmental aspects, the animal welfare is a noteworthy factor. The objective of this study was to evaluate the microclimatic conditions in an open-pasture and in silvopastoral systems, considering the Temperature Humidity Index (THI) and alterations in animal behavior. Three different pasture arrangements were analyzed in this study: total absence of trees in an open-pasture (ArrA), presence of peripheral trees (Eucalyptus spp.) along the border fences (ArrB), and an intensive wooded area aggregated with pasture (ArrC). A herd of 24 crossbreed heifers (3/4 and 7/8 Holstein-Girolando breed) was evaluated. Behavior data were collected every 15 min starting at 08 h00 with readings ending at 16 h00. THI was used to evaluate the environmental comfort. The THI found in the system with open-pasture and in the two systems with silvopastoral arrangement reached critical levels. The two arrangements with eucalyptus rows were not capable of eliminating heat stress in the conditions found in the north region of Mato Grosso State although better conditions were obtained under the tree canopy. The differences between the microclimatic variables for the three arrangements modified the behavior of the animals regarding their location and activity, except for water consumption.
