RESUMO

The study of circulating complexes fibronectin-fibrin in 21 patients with immunocomplex affections revealed their high levels which occurred in inverse relationships with fibronectin concentrations in the blood, but in direct correlation with indicators of inflammation (fibrinogen, cialic acid). It is suggested that the complexes are involved in the development of fibrosis at the sites of chronic inflammation in immune disorders.

RESUMO

Blood levels of fibrin degradation products and free haemoglobin were studied in 108 inpatients with acute transmural myocardial infarction in the period of rehabilitation. At the same time contents of cadmium, vanadium, lead and nickel in blood cells and plasma were determined spectrographically. It was established that heavy metals take part in activation of blood clotting and haemolysis which decrease equidistance and accelerates sedimentation of erythrocytes.

Postmyocardial infarction patients on hospital rehabilitation in case of no complications were examined for time course changes in the levels of fibrinogen degradation products (FDP), free hemoglobin (fHg) and fibronectin (Fn) using enzyme immunoassay. As shown by FDP and fHg values, the patients had DIC syndrome with its maximum on observation days 6-12 and 22-24. Fn fall occurred on the disease day 2-4 preceding maximum hemostasis activation up to 6-12th day. Less marked Fn reduction on the disease day 15-17 predicted a rise in hemocoagulation on day 22-24. It is recommended to determine FDP, fHg and Fn levels in blood of postmyocardial infarction patients to detect DIC syndrome and prevent hemostasis activation.

RESUMO

The content of ceruloplasmin, fibronectin and selenium was evaluated in the blood plasma of acute myocardial infarction patients within the first three days after onset if the disease. Results showed a reduction of fibronectin during the first hours after onset of the disease. The content of ceruloplasmin was increased, selenium reduced during the entire period of examination indicating pronounced disturbances of lipid peroxidation in the acute period of myocardial infarction as well as potentiation of antioxidant processes.

RESUMO

The content of soluble immunoreactive fibronectin in wound exudate in 104 patients after appendectomy, laparotomy and thermal injuries was studied both in non-complicated and purulent postoperative wounds. It was shown that in the case of non-complicated wound healing fibronectin reached maximum level at the 2nd and 3rd postoperative days. At the following days the fibronectin content decreased and returned at the 4th-5th days to the first day level. In the purulent wound the amount of fibronectin in the exudate remains low in the hydratation phase rising significantly in the phase of dehydration. The described changes correlate well with the clinical signs of wound healing. The data suggest that the fibronectin content in wound exudate can be used in evaluation of wound healing, prediction of possible complications as well as in control of treatment efficiency.

RESUMO

The theoretical and methodological aspects of fibronectin isolation and quantitative determination in biological fluids are analysed on the basis of the communications available in the literature and the authors' own data. The procedure of applying the analyzed methods, potential complexity and errors in interpreting the data obtained by various methods are briefly outlined. Possible fields of their application, such as basic and clinical investigations, biotechnological production, etc. are also discussed.

RESUMO

Examination of the effect of calcium antagonists on the functional activity of monocytes revealed the possibility of using these cells as an object for testing antiatherosclerotic agents. The functional activity of monocytes was evaluated by their capacity of producing thromboxane B2 and by the cellular content of cyclic AMP. It is suggested that calcium antagonists verapamil and niphedipin, in particular, are potential antiatherosclerotic agents.

RESUMO

Comprehensive morphologic studies (including light microscopy, morphometry, autoradiography) of experimental rabbits have revealed a stimulating effect of fibronectin instillations on healing of central penetrating wounds of the cornea. A polypotent mechanism of the stimulating effect of fibronectin instillations during all the stages of the reparative process in the corneal tissue was proved.

RESUMO

A new type of substances is described, related to T. pallidum cultural strain structure and interacting with human and animal blood serum proteins, named T-PBC. T-PBC were found to react with albumin, immunoglobulins G, M, and A, fibronectin, CIg component of human blood serum complement, goat and bovine blood serum immunoglobulins. When injected to animals, T-PBC induced production of specific antibodies identifying 4 components in T-PBC, 2 of these components being resistant to heating at 100 degrees C for 5 min. All these components could be stained with protein stains.

Specific IgM antibodies were assayed in blood sera of 230 patients with various forms of syphilis and of 561 controls. Enzyme immunoassay (EIA) sensitivity has made up 95.8 percent for primary syphilis, 93.8 percent for secondary new, 78.3 percent for secondary recurrent, 35.4 percent for early latent, 37.5 percent for late latent, and 80 percent for unknown syphilis. False-positive reactions were recorded in 5.5 percent of cases and were as a rule seen in the sera from patients with diffuse diseases of connective tissue. IgM EIA has proved highly effective in testing the sera from patients with primary seronegative syphilis and from subjects who had sexual intercourse with patients suffering from contagious forms of syphilis.

It has been found out that tripeptide Arg-Gly-Asp being under natural conditions a fibronectin fragment, responsible for adhesion, is capable in vitro to stimulate ingestion ability of the rat blood polymorphonuclear leucocytes and monocytes. Maximum increase of phagocytes index, phagocytes number and phagocyte per cent, which has ingested more than 10 latex particles, has been discovered after leuko-suspension incubation with tripeptide in initial concentrations 10(-3) mM and 10(-2) mM. Tripeptide can be regarded as a perspective phagocyte function regulator.

The content of the fibronectin, an extracellular glycoprotein in the drainage outflow system of human eyes was determined by the indirect immunoperoxidase staining technique. The degree of fibronectin accumulation in ocular tissues was evaluated by quantitative morphometric analysis. It was shown that the fibronectin level was elevated in ageing. Increased deposit of fibronectin in trabecular tissues, mainly, in the inner wall of Schlemm's canal and juxta-canalicular zone was demonstrated along with ageing. Comparison of fibronectin accumulation in glaucoma and ageing support the idea that ageing is a risk factor of glaucoma.

RESUMO

The content of blood nickel and myoglobin was studied in 25 patients with macrofocal and transmural myocardial infarction during the acute stage. It was established that acute myocardial infarction results in an increase of the content of nickel and myoglobin in the blood of these patients. The diagnostic importance of these examinations is discussed.

The examination of 147 surgical patients with cardiac diseases revealed that the onset of an operative intervention (skin incision) was accompanied with a substantial rise in endogenous insulin antibody titers in the blood of the operated patients, which is likely to trigger glucose metabolic disturbances. Insulin, 0.8-1.0 U/(kg/hour), and glucose, 0.4-0.5 g/(kg/hour), therapy (IGT) initiated prior to the skin incision and continued up to the application of forceps to the aorta was found to prevent the increase in insulin antibody titers both during the surgery and within 16-18 hours of the nearest postoperative period. IGT was demonstrated to improve myocardial contractility of patients in the preperfusion period and to provide an additional protection of the heart during its temporary anoxia. Insulin synthetic processes were activated in the nearest postoperative period in patients receiving IGT, unlike patients from the control group, the latter showing signs of diminished functional activity of B cells in the pancreas along with a moderate enhancement in the titer of antibodies to endogenous insulin. It was concluded that intraoperative IGT is a pathogenetically substantiated therapeutic modality in the complex of anesthetic supply during open heart surgeries.

We have previously found low levels of C1 and C4 INH in the sera of chronic lymphocytic leukaemia (CLL) patients. Hypocomplementaemia was supposed to be the consequence of a permanent activation of the classical pathway. We have compared the levels of C1 INH-C1rC1s and C1q-FN complexes in the sera of 95 CLL patients and 100 healthy controls, because these complexes are known to be formed in the early stage of classical pathway activation. A significant increase in the level of both types of complexes was found in sera of CLL patients as compared to the controls. These findings support the assumption that the classical complement pathway is activated in the patients with CLL.

A study is presented of indices of unspecific resistance of the body in 63 patients with erysipelas during the acute period of the disease and during recovery. In the acute period of the disease all patients revealed depending on the degree of endogenous intoxication: reduction of the content of fibronectin in the blood, significant reduction of the resistance of erythrocytes--increase of the per cent of peroxidation hemolysis, increase of the content of malonic dialdehyde--an index of disorders of lipid peroxidation, increased content middlemolecular peptides, circulating immune complexes and products of fibrin-fibrinogen degradation. The authors substantiate the expediency of using sodium thiosulfate possessing detoxicating, antiinflammatory and antiallergic antioxidant properties and the capacity to bind circulating immune complexes.
