RESUMO

We isolated and purified yak (Bos grunniens) endometrial epithelial cells and assayed different concentrations of estradiol (E2) and progesterone (P4) with respect to secretion of epidermal growth factor (EGF) and insulin-like growth factor-1 (IGF-1). Uterine epithelia were confirmed with Feulgen staining, karyotype analysis, and immunohistochemistry. Then, cells were treated with E2 and P4 and cultured in serum-free medium for 24 h. EGF and IGF-1 were measured with immunofluorescence, Real-time PCR (RT-PCR), Western blot (WB), and ELISA. When E2 and P4 were applied separately, the expression of EGF and IGF-1 did not change significantly, and EGF and IGF-1 gene and protein expression and epidermal growth factor receptor (EGFR) and insulin-like growth factor receptor (IGFR) gene expression were significantly increased when both hormones were combined. When the hormones were used singly, the optimal concentration of E2 was 10 ng/mL and of P4 was 100 ng/mL. When combined, the optimal E2 concentration was 10 ng/mL and P4 was 10 ng/mL. Thus, E2 and P4 can modulate expression of EGF and IGF-1 in endometrial epithelial cells at the morphological, gene, protein, and exocrine level, which is of great significance to improve the rate of yak embryo implantation and reproduction.

Microalgae cultivation in wastewater has gained significant attention as a cost-saving means for algae-based biofuel production. To evaluate the performance of Scenedesmus dimorphus cultivated in a 100-L continuously operated photobioreactor using domestic secondary effluent (DSE), algal growth, nutrients removal and energy evaluation were conducted in four scenarios. Prior to the application of continuous cultivation, S. dimorphus was grown in a batch operated 1.5-L bubble column photobioreactor to test the growth feasibility and lipids accumulation of S. dimorphus in DSE. The highest biomass achieved in DSE was 244 mg L(-1)with lipid content at 26.06%. Simultaneously, 98.72% of total phosphorus (TP) and 98.04% of total nitrogen (TN) in DSE were removed. Then, S. dimorphus were inoculated in the 100-L continuously operated photobioreactor using BG11, unsterilized DSE, N, P-added DSE and UV-sterilized DSE as the medium, respectively. Results showed that the highest biomass gained were 567, 174, 276 and 198 mg L(-1), respectively. TP removal rates in four scenarios were all above 90%. With adjustment to DSE, the overall TN removal rates increased up to 80%. Finally, energy evaluation demonstrated that although the case of BG11 as the medium provided the most energy production, the case using DSE with N and P supplementation was of the highest net energy rate, suggesting that microalgae cultivation for biodiesel production by DSE is of obvious potential and advantage over the synthesis medium like BG11.

BACKGROUND: Good pasture syndrome (GPS) has been paid much attention recently for the dangerous illness and high mortality. OBJECTIVE: To investigate the efficiency of plasma exchange (PE) to treat Goodpasture syndrome (GPS) in children associated with Turner's syndrome. METHOD: We report a case of a 15 year old female with GPS and Turner's syndrome. The patient has intermittent fever and cough for 45 days and oliguria for 6 days. Turner's syndrome was determined through blood karyotype analysis, and GPS was diagnosed because the patient was negative for antinuclear antibodies and antineutrophil cytoplasmic antibodies (ANCA), but positive for anti-glomerular basement membrane (anti-GBM) antibodies (200 RU/ml). PE was carried out in combination with immunosuppression therapy. RESULTS: The results show PE treatment can efficiently decrease the levels of anti-GBM antibodies. The antibody levels were >200 RU/ml and 184 RU/ml before and after the first PE treatment, respectively. The removal efficiency were 40%, 47%, 42%, 54%, 52% for the fifth, sixth, seventh, eighth and ninth PE procedures, respectively. CONCLUSION: The therapy with PE, hemodialysis, pulse methylprednisolone followed by oral prednisone and cyclophosphamide greatly contributed to improvement of this patient's condition, and resolved the patient's pulmonary haemorrhage. All these results demonstrate that PE contributed efficiently to the treatment for GPS in children.

The influences of water solvent on the structures and stabilities of the complex ion conformers formed by the coordination of alanine dipeptide (AD) and Na+ have been investigated using supramolecular and polarizable continuum solvation models at the level of B3LYP/6-311++G**, respectively; 12 monohydrated and 12 dihydrated structures of Na+-AD complex ion were obtained after full geometrical optimization. The results showed that H2O molecules easily bind with Na+ of Na+-AD complex ion, forming an ion-lone pair interaction with the Na-O bond length of 2.1-2.3 A. Besides, H2O molecules also can form hydrogen bonds OW-HW...O(1), OW-HW...O(2), N(1)-H(1)...OW or N(2)-H(2)...OW with O or N groups of the Na+-AD backbone. The most stable gaseous bidentate conformer C7AB of Na+-AD is still the most stable one in the solvent of water. However, the structure of the most unstable gaseous conformer alpha'B of Na+-AD collapses under the attack of H2O molecules and changes into C7AB conformation. Computations with IEFPCM solvation model of self-consistent reaction field theory give that aqueous C5A is more stable than C7eqB and that the stabilization energies of water solvent on monodentate conformers of Na+-AD complex ion (about 272-294 kJ/mol) are more than those on bidentate ones (about 243 kJ/mol).

The Mopan persimmon (Diospyros kaki L. cv. Mopan) is the major cultivar of astringent persimmon in northern China. This study investigates the radical scavenging activity against ABTS and DPPH radical, and the content of total and individual phenolics (catechin, epicatechin, epigallocatechin, chlorogenic acid, caffeic acid, and gallic acid) with apple, grape, and tomato as controls. The radical scavenging activities against ABTS and DPPH radicals of the Mopan persimmon are 23.575 and 22.597 microm trolox eq/g f.w., respectively. These findings suggest that the Mopan persimmon's antioxidant activity is significantly (P < 0.05) stronger than that of reference materials. The Mopan persimmon showed the highest content of total phenolics among the 4 materials tested. Significant correlations (R(2)= 0.993, P < 0.05, ABTS radical; R(2)= 0.980, P < 0.05, DPPH radical) are found between the total phenolics and the radical scavenging activities. The total content of these 6 kinds of phenolics (catechin, epicatechin, epigallocatechin, chlorogenic acid, caffeic acid, and gallic acid) is significantly correlated (R(2)= 0.831, P < 0.05, ABTS radical; R(2)= 0.745, P < 0.05, DPPH radical) with the individual radical scavenging activity of the 4 materials, although the total content of the 6 phenolics accounts for no more than 20% of the total phenolics in the Mopan persimmon. Gallic acid exhibits the strongest antioxidant activity in all 6 kinds of phenolics and its content is the largest in the Mopan persimmon, presumably being responsible for its much higher antioxidant activity as compared to apple, grape, and tomato.

The chemical constituents of the essential oils obtained from the leaves of Vitex negundo var. cannabifolia, V. negundo var. heterophylla, V. negundo and V. trifolia were analysed by GC-MS. Forty compounds including alpha-pinene, linalool, terpinyl acetate, beta-caryophyllene and caryophyllene oxide, etc. were identified. Their percentages in oils were given.
