This study aimed to evaluate the response of Toona ciliata seedlings to sublethal doses of glyphosate. The increasing use of glyphosate in agriculture concerns the scientific community, as the drift of this pollutant into aquatic systems or atmospheric currents can affect non-target species. Therefore, we need to understand how non-target species respond to small doses of this herbicide. T. ciliata seedlings (clone BV-1110) were exposed to sublethal doses of glyphosate (0, 9.6, 19.2, 38.4, 76.8 g ae ha-1). Anatomical, physiological, and photochemical analyses were performed 60 days after herbicide application, and growth assessments were carried out after 160 days of cultivation. We found that sublethal doses of glyphosate above 19.2 g ae ha-1 induced toxicity symptoms in Toona ciliata leaves. These symptoms were mild in some cases, such as chlorosis, but severe in other cases, such as tissue necrosis. We observed a positive relationship between increased plant height and photochemical yield with plant exposure to sub-doses 9.6 and 19.2 g ae ha-1. A sublethal dose of 38.4 g ae ha-1 improved the photosynthetic rate and carboxylation efficiency. Thus, we confirmed the hypothesis of a hormetic effect when T. ciliata was exposed to sub-doses of glyphosate equal to or lower than 38.4 g ae ha-1. However, the sublethal dose of 76.8 g ae ha-1 must be considered toxic, impacting photosynthetic activity and, consequently, the height of T. ciliata. The stem diameter of T. ciliata responded positively to increasing glyphosate doses. This occurs to compensate for the negative effect of glyphosate on water absorption. Further research will provide valuable information for harnessing the potential benefits of hormesis to improve the productivity of T. ciliata.

Background: The leaves of Serjania erecta Radlk (Sapindaceae) are renowned in ethnobotany for their medicinal properties and are significant as a medicinal resource for traditional Brazilian communities. As necrotic spots are common on these leaves, indicating interaction with phytopathogenic fungi, it was hypothesized that biotrophic fungal species colonize the leaf tissues of S. erecta. Methods: To test this hypothesis, we employed standard techniques in plant anatomy, which enabled us to investigate the interaction of fungal structures with plant tissues and describe the morphoanatomical and histochemical characteristics of the epidermis and limbus of S. erecta. Results: The anatomical analysis showed the existence of leaf teeth on the leaf tips. Additionally, hyphae, conidiospores, and spores of Bipolaris/Curvularia species were detected on the adaxial epidermis. Moreover, melanized microsclerotia were found in glandular areas of the leaf teeth and the phloem, providing evidence of biotrophic behavior. The hypothesis that biotrophic phytopathogenic fungi interact with S. erecta leaf tissues was confirmed, despite the presence of many bioactive compounds (such as flavonoids, alkaloids, and essential oils), as evidenced by histochemical analyses. The presence of tector, glandular, and scabiform trichomes on the leaf teeth and epidermis was also revealed. This study presents, for the first time, the synthesis of essential oils and alkaloids in the leaves of S. erecta. Additionally, it investigates previously unexplained aspects of the anatomy and histochemistry of the species, as well as its interaction with resident microorganisms. Therefore, it is recommended that future research focus on extracting and characterizing the oils and alkaloids of S. erecta, as well as exploring other aspects related to its microbiome and its relationship.

Many biological activities are described for the Clusiaceae family. Clusia fluminensis, a species from Brazilian flora, is mainly employed for ornamental purposes. This review aimed to depict the current knowledge of C. fluminensis from a bioprospecting standpoint. "Clusia fluminensis" search term was applied in Scopus, Web of Science, PubMed and Bireme databases according to PRISMA-ScR statement. Selected papers on Phytochemistry or Bioactivity followed hand searching procedures. Bioactivity preclinical studies considered in vitro or in vivo biological systems, treated with plant extracts or isolated compounds. The outcomes were compared with standard or no treatment control groups. Critical appraisal of individual trials considered completeness in the research fields. Our results showed that 81% of the selected papers presented high level of completeness, 69% revealed phytochemical parameters and 31% biological applications of plant extracts and isolated compounds. Polyisoprenylated benzophenones, terpenoids, sterols and phenolic compounds were identified. Antiviral, insecticidal and snake antivenom activities were reported. In conclusion, the phytochemical data reinforce the reported activities. Potential applications in personal care, nutritional supplementation and pharmaceutical, food, chemical or textile industries were also identified. Toxicological and phytochemical complementary studies may be required.

The soybean looper, Chrysodeixis includens, is a primary soybean pest that reduces crop productivity. This work examined control of C. includens populations with methanolic extract of Serjania erecta, a native Cerrado plant, while minimizing risks to pollinators, natural enemies and the environment. Serjania erecta specimens were collected, identified, and subjected to methanol extraction. Bioassays were performed using newly hatched and second-instar caterpillars and different extract concentrations on the diet surface to obtain IC50 values. Two replicates, containing 10 caterpillars, were established in triplicate. The IC50 values were 4.15 and 6.24 mg of extract mL-1 for first-instar and second-instar caterpillars, respectively. These growth inhibition results informed the extract concentrations assessed in subsequent development inhibition assays, in which the pupal weight was higher under the control than under the treatments. Extract treatments increased the duration of the larval, pupal and total development. The potential of different concentrations of S. erecta extract to inhibit the enzymes carboxylesterases was also evaluated. Carboxylesterases activity decreased by 41.96 and 43.43% at 7.8 and 15.6 µg mL-1 extract, respectively. At 31.3 µg mL-1 extract, enzymatic activity was not detected. Overall, S. erecta leaf methanolic extract showed inhibitory potential against carboxylesterases.

The objective was to evaluate the effect of the glyphosate on Hymenaea courbaril L. A randomized block design with five replications was implemented. Each experimental unit was composed of one plant in a 5 L container. The treatments were 0 "control"; 96; 240; 480; and 960 g ha-1 "corresponding to 10, 25, 50, and 100% of the commercial dose of glyphosate recommended for Caryocar brasiliense crop, respectively". The evaluations were performed at 24 h and 60 days after application. Visual and anatomical evaluations did not change regardless of the dose, while the histochemical evaluation showed an accumulation of starch grains in leaf tissues. There was an increase in the photosynthetic rate, in the electron transport rate, and in the effective quantum yield of photosystem II at 24 h after application. At 60 days after the application of the treatments, the photosynthetic rate showed a slight decrease and the transpiratory rate showed quadratic behavior. An increase in plant height was observed up to the dose of 480 g ha-1, a linear increase in stem diameter and a decrease in the number of leaves with increasing glyphosate doses. These results show that the cuticle protected the plant, and that the little absorbed glyphosate increased photosynthesis and transpiration to favor the plants. We can conclude that the H. courbaril species is able to survive after contact with glyphosate during the evaluated time, with no visual and/or anatomical damage, showing increases in growth and physiological characteristics for the tested doses.

BACKGROUND: Polymorphisms in MTHFR gene influence risk and overall survival of patients with brain tumor. Global genomic DNA (gDNA) methylation profile from tumor tissues is replicated in peripheral leukocytes. This study aimed to draw a correlation between rs1801133 MTHFR variants, gDNA methylation and overall survival of patients with recurrent glioblastoma (rGBM) under perillyl alcohol (POH) treatment. METHODS: gDNA from whole blood was extracted using a commercially available kit (Axygen) and quantified by spectrophotometry. Global gDNA methylation was determined by ELISA and rs1801133 polymorphism by PCR-RFLP. Statistical analysis of gDNA methylation profile and rs1801133 variants included Mann-Whitney, Kruskal-Wallis, Spearman point-biserial correlation tests (SPSS and Graphpad Prism packages; significant results for effect size higher than 0.4). Prognostic value of gDNA methylation and rs1801133 variants considered survival profiles at 25 weeks of POH treatment, having the date of protocol adhesion as starting count and death as the final event. RESULTS: Most rGBM patients showed global gDNA hypomethylation (median = 31.7%) and a significant, moderate and negative correlation between TT genotype and gDNA hypomethylation (median = 13.35%; rho = - 0.520; p = 0.003) compared to CC variant (median = 32.10%), which was not observed for CT variant (median = 33.34%; rho = - 0.289; p = 0.06). gDNA hypermethylated phenotype (median = 131.90%) exhibited significant, moderate and negative correlations between TT genotype (median = 112.02%) and gDNA hypermethylation levels when compared to CC (median = 132.45%; rho = - 0,450; p = 0.04) or CT (median = 137.80%; rho = - 0.518; p = 0.023) variants. TT variant of rs1801133 significantly decreased gDNA methylation levels for both patient groups, when compared to CC (d values: hypomethylated = 1.189; hypermethylated = 0.979) or CT (d values: hypomethylated = 0.597; hypermethylated = 1.167) variants. Positive prognostic for rGBM patients may be assigned to gDNA hypermethylation for survivors above 25 weeks of treatment (median = 88 weeks); and TT variant of rs1801133 regardless POH treatment length. CONCLUSION: rGBM patients under POH-based therapy harboring hypermethylated phenotype and TT variant for rs1801133 had longer survival. Intranasal POH therapy mitigates detrimental effects of gDNA hypomethylation and improved survival of patients with rGBM harboring TT mutant variant for MTHFR rs1801133 polymorphism. Secretory structures were little studied in Gentianaceae. Glandular areas on the calyx dorsal region are commonly reported for Helieae species, the main tribe of Gentianaceae. So, the elucidation of nature of glandular areas is particularly relevant. Trichomes secreting mucilage, interpreted as colleters, are reported only for the sepals of Gentianinae species. We aimed to anatomically characterize and identify the nature of the calycinal secretory structures in Calolisianthus pedunculatus. Samples from floral buds, flowers, and fruits were collected, fixed, and processed following usual procedures for light and scanning electron microscopies. Histochemical tests were performed to determine the nature of the secretion. Glucose, fructose, and sucrose were measured with an ELISA reader. Colleters occur on the sepal ventral region and are composed of a multicellular secretory head and a stalk. These structures secrete polysaccharides and proteins, and the secretion is probably released through cuticle microchannels. Nectaries, on the other hand, occur on the sepal dorsal region. They are formed by 3-5 cells arranged in rosettes circling a central cell or pore. These structures also secrete polysaccharides (mainly fructose), lipids, and proteins. The identification of the secretory structures in the sepals of Calolisianthus pedunculatus highlights the importance of anatomical studies in this family. The interpretation of the glandular areas on the calyx of the Helieae species as nectaries has been proven, as well as the confirmation of colleters as common structures in the sepals of Gentianaceae. Besides the taxonomic and phylogenetic importance of nectars and colleters, we highlight the importance of the secretion for the protection of floral buds against dehydration.

OBJECTIVE: To examine the validity of the GT3X® ActiGraph accelerometer and the Google Fit® smartphone application in estimating stepping activity in people with chronic stroke. METHODS: Thirty-seven stroke survivors walked along a straight, 10 metre hallway over 5 min at their fastest speeds, wearing the GT3X® ActiGraph accelerometer and a smartphone on their paretic lower limb. The criterion-standard measurement made was the actual number of steps, counted by a trained examiner. RESULTS: The mean estimated steps measured by the GT3X® ActiGraph and Google Fit® respectively were 276.7 ±â€¯97.6 and 481.0 ±â€¯119.8; that calculated from the examiner's measurements was 472.0 ±â€¯93.9. Statistically significant associations were found between the actual steps and those estimated by the GT3X® ActiGraph (r = 0.56; p < 0.001) and Google Fit® (r = 0.89; p < 0.001). The Google Fit® application demonstrated the highest reliability coefficient (ICC[2,1] = 0.93; p < 0.001; p = 0.37), compared to that of the GT3X® ActiGraph (ICC[2,1] = 0.32; p < 0.001; p < 0.001). CONCLUSIONS: The GT3X® ActiGraph underestimated the data and may not be the most appropriate device to estimate the stepping activity of stroke patients. The findings support the validity of a smartphone application in estimating the stepping activity of individuals with stroke, when worn on the paretic side.

Resumo O objetivo deste estudo foi revisar a literatura sobre os efeitos do exercício físico nas estruturas e funções do sistema nervoso (SN) de idosos e reportar os exercícios e parâmetros frequentemente usados. Após ampla e sistemática busca em bases de dados eletrônicas, foram incluídos 22 ensaios clínicos aleatorizados e não aleatorizados que avaliaram os efeitos do exercício físico em variáveis neuroanatomofisiológica de idosos. A maioria (77,3%) investigou os efeitos no SN central e usou exercício aeróbico. O exercício físico apresentou benefícios nas estruturas e funções do SN de idosos, inclusive as funções cognitivas, e nas estruturas e funções musculares e cardiovasculares, na mobilidade e no equilíbrio. Ainda não há um consenso sobre quais parâmetros de treinamento são mais adequados.


Abstract The objective of this study was to review the literature on the effects of physical exercise on the structures and functions of the nervous system (NS) of elderly people and to report frequently used exercises and its parameters. After extensive and systematic search in electronic databases, it was included 22 randomized and non-randomized clinical trials that evaluated the effects of physical exercise on neuroanatomophysiological variables of the elderly. The majority (77.3%) investigated the effects on central NS and used aerobic exercise. Physical exercise showed benefits in the structures and functions of the NS of elderly subjects, including cognitive functions, and in muscular and cardiovascular structures and functions, mobility and balance. There is still no consensus on which training parameters are most appropriate.


Resumen El objetivo de este artículo fue analizar la bibliografía sobre los efectos del ejercicio físico en las estructuras y funciones del sistema nervioso (SN) de las personas de edad avanzada e informar sobre los ejercicios y parámetros usados con frecuencia. Después de una extensa y sistemática búsqueda en bases de datos electrónicas, se incluyeron 22 ensayos clínicos de manera aleatoria y no aleatoria que evaluaban los efectos del ejercicio sobre las variables neuroanatomofisiológicas de personas de edad avanzada. La mayoría (77,3%) analizaba los efectos sobre el SN central y utilizaba el ejercicio aeróbico. El ejercicio mostraba beneficios en estructuras y funciones del SN de edad avanzada, incluyendo las funciones cognitivas, en estructuras y funciones musculares y cardiovasculares, y en la movilidad y el equilibrio. Todavía no existe un consenso sobre los parámetros de entrenamiento más adecuados.

OBJECTIVE: To examine the concurrent validity of the GT3X® ActiGraph accelerometer and Google Fit® smartphone application in estimating energy expenditure in people who had suffered a stroke, during fast overground walking. METHODS: Thirty community-dwelling stroke individuals walked on a 10-meter hallway over 5min at their fastest speeds, wearing a Cortex Metamax 3B® ergoespirometer, a GT3X® ActiGraph accelerometer, and a smartphone with the Google Fit® application. Pearson correlation coefficients were calculated to verify the associations between measures of energy expenditure, in kilocalories (kcal), estimated by both devices and those obtained with the Cortex Metamax 3B® ergoespirometer (gold-standard measure). RESULTS: Fair association was found between the energy expenditure values estimated from the combined formula of the ActiGraph GT3X® and those obtained with the gold-standard measure (r=0.37; p=0.04). No significant associations were found between the energy expenditure values estimated by the Google Fit® application and those provided by the gold-standard measure. CONCLUSIONS: The findings demonstrated that both the GT3X®ActiGraph accelerometer and the Google Fit® smartphone application do not provide valid measures of energy expenditure in chronic stroke individuals during fast overground walking.

Tumor infiltration into brain tissue usually remains undetected even by high-resolution imaging. Molecular markers are used to increase diagnostic accuracy, but with limited continuous monitoring application. We evaluated the potential of circulating cell-free DNA (cfDNA) as a molecular indicator of the response to therapy by the intranasal administration (ITN) of perillyl alcohol (POH) in brain tumors. The cohort included 130 healthy subjects arranged as control-paired groups and patients at terminal stages with glioblastoma (GBM, n = 122) or brain metastasis (BM, n = 55) from stage IV adenocarcinomas. Serum cfDNA was isolated and quantified by fluorimetry. Compared with the controls (40 ng/mL), patients with brain tumors before ITN-POH treatment had increased (p < 0.0001) cfDNA median levels: GBM (286 ng/mL) and BM (588 ng/mL). ITN-POH treatment was significantly correlated (rho = -0.225; p = 0.024) with survival of >6 months at a concentration of 599 ± 221 ng/mL and of.

This article aimed to determine the recruitment rate of chronic stroke survivors to cross-sectional studies and to determine their retention at the two days of assessments. Participants after six months of a unilateral stroke were screened for eligibility and invited to participate in two cross-sectional studies, by telephone. The number of people who were screened, eligible, and successfully recruited was recorded. Retention at the two days of assessments was also recorded. From a list of 654 individuals, 87 were ineligible. Of the 567 left, 216 had wrong contact numbers, 144 refused to participate, and 12 had died. A total of 165 subjects participated in both studies. Out of the 56 who agreed to attend to the second day of assessment, eight did not return. The results showed that individuals with chronic stroke had low rates of recruitment and retention.

Resumo Este artigo teve por objetivo determinar a taxa de recrutamento de indivíduos pós-Acidente Vascular Encefálico (AVE) em estudos transversais e determinar sua retenção nos dois dias de avaliação, tendo como referencial teórico a Classificação Internacional de Funcionalidade, Incapacidade e Saúde. Participantes após 6 meses a um AVE unilateral foram selecionados de acordo com a sua elegibilidade e convidados a participar em dois estudos transversais, por telefone. Foi obtido o número de pessoas contatadas, elegíveis e recrutadas com sucesso. A retenção nos dois dias de avaliação foi igualmente obtida. De uma lista de 654 indivíduos, 87 não eram elegíveis. Dos 567 restantes, 216 possuíam números telefônicos errados, 144 se recusaram a participar e 12 haviam ido a óbito. Um total de 165 indivíduos participaram dos dois estudos. Dos 56 indivíduos que concordaram em participar do segundo dia de avaliação, oito não retornaram. Os resultados demonstraram que indivíduos pós-AVE crônicos apresentam baixas taxas de recrutamento e retenção.

This study aimed to investigate the antitumor and immunomodulatory properties of the flavonoid apigenin (5,7,4'-trihydroxyflavone), which was extracted from Croton betulaster Mull, in glioma cell culture using the high-proliferative rat C6 glioma cell line as a model. Apigenin was found to have the ability to reduce the viability and proliferation of C6 cells in a time-dependent and dose-dependent manner, with an IC50 of 22.8 µmol/l, 40 times lower than that of temozolomide (1000 µmol/l), after 72 h of apigenin treatment. Even after C6 cells were treated with apigenin for 48 h, high proportions of C6 cells entered apoptosis (39.56%) and autophagy (22%) as shown by flow cytometry using annexin V/propidium iodide and acridine orange staining, respectively. In addition, the flavonoid apigenin induced cell accumulation in the G0/G1 phase of the cell cycle and inhibited glioma cell migration efficiently. Moreover, apigenin induced astroglial differentiation and morphological changes in C6 cells, characterized by increased expression of glial fibrillary acidic protein and decreased expression of nestin protein, a typical marker of neuronal precursors. The immunomodulating effects of apigenin were also characterized by a change in the inflammatory profile as evidenced by a significant decrease in interleukin-10 and tumor necrosis factor production and increased nitric oxide levels. Because apigenin can induce differentiation, apoptosis, and autophagy, can alter the profile of cytokines involved in regulating the immune response, and can reduce the survival, growth, proliferation, and migration of C6 cells, this flavonoid may be considered a potential antitumor drug for the adjuvant treatment of malignant gliomas.

Resumo Objetivo: Observar possíveis transições entre níveis de fragilidade, segundo o fenótipo de fragilidade em idosos de Belo Horizonte-MG, em um período de 24 meses. Método: Trata-se de um estudo exploratório longitudinal. Dos 601 indivíduos que compuseram a linha de base da Rede FIBRA de Belo Horizonte, selecionou-se 40 indivíduos de ambos os sexos, com mais de 65 anos, classificados como frágeis, residentes na comunidade, que possuíssem marcha independente ou com auxílio de dispositivos e que não fossem portadores de doenças que inviabilizassem a realização dos testes. Após 24 meses, os participantes foram reavaliados com o mesmo inquérito padronizado usado na linha de base do estudo. Resultados: A amostra inicial foi composta por 40 indivíduos, com média de idade de 78,03 (±6,46) anos. Após 24 meses, localizaram-se 22 indivíduos da amostra inicial. Foi realizada comparação da pontuação obtida para a classificação do fenótipo de fragilidade entre as duas amostras, considerando os mesmos 22 indivíduos. Esta comparação mostrou que 16 indivíduos apresentaram um menor número de itens positivos para o fenótipo de fragilidade na avaliação após 24 meses, três apresentaram pontuação positiva maior e três não apresentaram alteração na pontuação entre as avaliações (p=0,004). Conclusão: O presente estudo observou que a grande maioria dos indivíduos apresentou melhora na classificação do fenótipo de fragilidade.


Abstract Objective: To observe possible changes in frailty phenotype classification over time. Method: 40 individuals of both genders aged over 65 years, living in the metropolitan region of Belo Horizonte, Minas Gerais and identified in a base line Rede FIBRA assessment as frail, who could walk independently or with the aid of devices and who had no illnesses that would prevent tests being carried out, , were considered. A standardized survey previously established by Rede FIBRA was administered. Results: The initial sample was composed of 40 individuals with a mean age of 78.03 (±6.46) years. After 24 months, a follow up of the 22 individuals from the initial sample that could be found was performed. The frailty phenotype classification scores of the same 22 subjects from the two samples were compared. After comparing the groups, it was found that 16 individuals had a lower number of frailty phenotype positive items in the follow up, three had higher positive scores and there was no change in the score of three individuals between assessments (p=0.004). Conclusion: This study found that a large majority of individuals showed improvement in their frailty phenotype classification.

Introdução: A reabilitação física deveria ser realizada em intensidades capazes de promover estresse cardiorrespiratório adequado para minimizar o descondicionamento apresentado por indivíduos após Acidente Vascular Encefálico (AVE). Objetivo: Revisar a literatura acerca da intensidade das sessões de reabilitação física capazes de proporcionar estresse cardiorrespiratório em indivíduos pós-AVE. Métodos: Selecionaram-se estudos onde se avaliou o estresse cardiorrespiratório por meio da frequência cardíaca-alvo de indivíduos pós-AVE. A busca dos artigos foi realizada nas bases de dados Medline, SciELO, Embase e PEDro. Resultados: Foram encontrados três trabalhos transversais, e um longitudinal, com a amostra total de 78 hemiparéticos pós-AVE, totalizando 267 atendimentos. Em nenhuma das pesquisas a intensidade das sessões foi capaz de proporcionar estresse cardiorrespiratório adequado. A atividade com maior estresse cardiorrespiratório foi a marcha. Conclusões: A intensidade do exercício durante a reabilitação física não foi capaz de provocar efeitos cardiorrespiratórios adequados em indivíduos pós-AVE.


Introduction: Physical rehabilitation should have adequate intensities to provide cardiopulmonary stresses to minimize deconditioning presented by individuals post stroke. Objective: To review the literature regarding the intensity of the physical rehabilitation sessions, which were able to provide cardiorespiratory stresses in stroke subjects. Methods: This literature review included studies which evaluated cardiorespiratory stresses based upon the target heart rates of stroke subjects. A search of the literature was conducted in the Medline, SciELO, Embase, and PEDro databases. Results: Three cross-sectional and one longitudinal study were found, with the total sample of 78 stroke individuals; totalling 267 sessions. The results of all studies showed that the intensity of the sessions was not capable of providing adequate cardiopulmonary stresses. The activity which provided higher cardiorespiratory stresses was gait. Conclusion: Exercise intensity during physical rehabilitation is not able to provide adequate cardiorespiratory stresses in stroke individuals.

Laryngeal squamous cell carcinoma (LSCC) is the most common among several types of head and neck cancers. Current treatments have a poor effect on early and advanced cases, and further investigations for novel agents against LSCCs are desirable. In this study, we elucidate the cytotoxic enhancing effect of arsenic trioxide (As2O3) combined with L-buthionine sulfoximine (BSO) in LSCC. The effect of BSO with As2O3 or Cisplatin (CDDP) on cell viability was examined using 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT). The reactive oxygen species (ROS) levels, cell cycle, and apoptosis were measured by flow cytometry using 2',7'-dichlorodihydrofluorescein diacetate (DCFH-DA), propidium iodide (PI) and annexin V/PI. The acidic vacuolar organelles were visualized by fluorescence microscope and quantified using flow cytometry. Neither CDDP nor As2O3 when used alone reduced the cell viability. BSO was found to enhance only As2O3 sensitivity, leading to G2/M arrest and autophagy with no correlation of ROS induction. This result suggests that modulation of glutathione enhances autophagy, which interplays with apoptosis. In this study, we obtained initial preclinical evidence for the potential efficacy of these drugs in a combined therapy protocol.

Objetivo: Avaliar o perfil epidemiológico dos pacientes que sofreram traumatismos alvéolo-dentários em dentes permanentes, identificando também o tipo de trauma, os dentes mais atingidos e as causas mais frequentes. Métodos: A amostra foi constituída por todos os prontuários de pacientes atendidos em cinco anos, no Departamento de Cirurgia da Faculdade de Odontologia de Pelotas, totalizando 308 casos e 793 dentes traumatizados. Os dados obtidos foram tabulados em uma planilha e analisados estatisticamente. Resultados: A faixa etária prevalente foi dos 13 aos 19 anos (32,7%); 74,6% dos traumatismos ocorreram entre pessoas do sexo masculino; a causa mais comum foram os acidentes ciclísticos (23,3%); os tipos de trauma mais frequentes foram a avulsão (17,2%) e a fratura coronária nãocomplicada(17,2%) e os dentes mais acometidos foram os incisivos centrais superiores (66,7%).Conclusão: Foi possível concluir que, nesta pesquisa, a distribuição dos traumatismos foi muito similar a maioria dos estudos publicados, no entanto,em relação à variável causa constatou-se algumas características loco-regionais, reforçando a importância de estudos epidemiológicos para o desenvolvimento de condutas preventivas e terapêuticas específicas para os serviços de saúde.


Objetive: The present study aimed to evaluate the epidemiological profile of patients with traumatic dental injuries in permanent teeth, as well as to identify the type of trauma, the teeth most affected and the most frequent causes of dentoalveolar trauma. Methods: The sample comprised all of the dental records of patients treated at the Oral and Maxillofacial Surgery Department at the Faculty of Dentistryat the Federal University of Pelotas, in the Brazilian state of Rio Grande do Sul, over a five-year period, totaling 308 cases and 793 teeth with traumatic injury. The data were evaluated statistically. Results: The results obtained demonstrated that the most prevalent age group was 13-19 (32.7%); males were affected in 74.6% of injuries; the most common etiology was cycling accidents (23.3%); the most frequent types of trauma were avulsion (17.2%) and non-complicated crown fracture (17.2%); and the most affected teeth were the maxillary central incisors (66.7%). It was concluded that the distribution of trauma injuries, in this paper, is very similar to that presented in most of the published studies. However, with regard to the etiology variable,some local/regional characteristics were found, reinforcing the importance of epidemiological studies for the development of preventive and therapeutic methods specific to the health service.

Gliomas, the most frequent primitive central nervous system tumors, have been suggested to originate from astrocytes or from neural progenitors/stem cells. However, the precise identity of the cells at the origin of gliomas remains a matter of debate because no pre-neoplastic state has been yet identified. Transforming growth factor (TGF)-alpha, an epidermal growth factor family member, is frequently overexpressed in the early stages of glioma progression. We previously demonstrated that prolonged exposure of astrocytes to TGF-alpha is sufficient to trigger their reversion to a neural progenitor-like state. To determine whether TGF-alpha dedifferentiating effects are associated with cancerous transforming effects, we grafted intracerebrally dedifferentiated astrocytes. We show that these cells had the same cytogenomic profile as astrocytes, survived in vivo, and did not give birth to tumors. When astrocytes dedifferentiated with TGF-alpha were submitted to oncogenic stress using gamma irradiation, they acquired cancerous properties: they were immortalized, showed cytogenomic abnormalities, and formed high-grade glioma-like tumors after brain grafting. In contrast, irradiation did not modify the lifespan of astrocytes cultivated in serum-free medium. Addition of TGF-alpha after irradiation did not promote their transformation but decreased their lifespan. These results demonstrate that reversion of mature astrocytes to an embryonic state without genomic manipulation is sufficient to sensitize them to oncogenic stress.

TGFalpha and its receptor EGFR participate in the development of a wide range of tumors including gliomas, the main adult primary brain tumors. TGFalpha soluble form results from the cleavage by the metalloprotease TACE/ADAM17 of the extracellular part of its transmembrane precursor, pro-TGFalpha. To gain insights into the mechanisms underlying TGFalpha bioavailability, a yeast two-hybrid screen was performed to identify proteins interacting with pro-TGFalpha intracellular domain (ICD). DLG1/SAP97 (Discs Large Gene 1 or Synapse Associated Protein 97) was found to interact with both pro-TGFalpha and TACE ICDs through distinct PDZ domains. An in vivo pro-TGFalpha-DLG1-TACE complex was detected in U251 glioma cells and in gliomas-derived tumor initiating cells. Interaction between DLG1 and TACE diminished in response to stimulations promoting pro-TGFalpha shedding. Manipulation of DLG1 levels revealed dual actions of DLG1 on pro-TGFalpha shedding, favoring approximation of pro-TGFalpha and TACE, while limiting TACE full shedding activity. These results show that DLG1 participates in the control of TGFalpha bioavailability through its dynamic interaction with the growth factor precursor and TACE.
