Although there are many hormonal changes associated with reproduction, the effects of ovulation and early pregnancy on adrenocorticotropic hormone (ACTH) and insulin concentrations are poorly described. We hypothesise that both ovulation and early pregnancy will alter ACTH and insulin concentrations in healthy mares. Eighteen mares showing no clinical signs suggestive of, or laboratory findings consistent with, pituitary pars intermedia dysfunction PPID and insulin dysregulation (ID) are enrolled. ACTH, cortisol, insulin and glucose concentrations are measured over their peri-ovulatory period, as determined via ultrasounds and progesterone concentrations. The mares are grouped by age and gestation status, and a two-way repeated-measures ANOVA is used to determine the effects of age and early pregnancy, along with the peri-ovulatory period, on analyte concentrations. No significant effect of age, ovulation or early pregnancy is detected on the mares' cortisol, insulin or glucose concentrations; however, there is a significant effect of early pregnancy and ovulation on ACTH concentrations (p = 0.04 and p = 0.04 respectively). ACTH concentrations change around ovulation and with early pregnancy. Therefore, knowledge of a mare's reproductive status might be beneficial when interpreting ACTH concentrations.

Hendra virus (HeV) continues to pose a serious public health concern as spillover events occur sporadically. Terminally ill horses can exhibit a range of clinical signs including frothy nasal discharge, ataxia or forebrain signs. Early signs, if detected, can include depression, inappetence, colic or mild respiratory signs. All unvaccinated ill horses in areas where flying foxes exist, may potentially be infected with HeV, posing a significant risk to the veterinary community. Equivac® HeV vaccine has been fully registered in Australia since 2015 (and under an Australian Pesticides and Veterinary Medicines Authority special permit since 2012) for immunization of horses against HeV and is the most effective and direct solution to prevent disease transmission to horses and protect humans. No HeV vaccinated horse has tested positive for HeV infection. There is no registered vaccine to prevent, or therapeutics to treat, HeV infection in humans. Previous equine HeV outbreaks tended to cluster in winter overlapping with the foaling season (August to December), when veterinarians and horse owners have frequent close contact with horses and their bodily fluids, increasing the chance of zoonotic disease transmission. The most southerly case was detected in 2019 in the Upper Hunter region in New South Wales, which is Australia's Thoroughbred horse breeding capital. Future spillover events are predicted to move further south and inland in Queensland and New South Wales, aligning with the moving distribution of the main reservoir hosts. Here we (1) review HeV epidemiology and climate change predicted infection dynamics, (2) present a biosecurity protocol for veterinary clinics and hospitals to adopt, and (3) describe diagnostic tests currently available and those under development. Major knowledge and research gaps have been identified, including evaluation of vaccine efficacy in foals to assess current vaccination protocol recommendations.

Equine uterine development, including endometrial histogenesis, begins prenatally and is completed postnatally. Little is known about this process in the horse. Uterine tissue was acquired from 38 foals, ranging in developmental age from gestational day (GD) 300 to postnatal day (PND) 180, for assessment of endometrial histogenesis. Patterns of endometrial cell proliferation were evaluated by multispectral imaging of uterine tissue sections stained immunofluorescently for Ki-67. Labeling index (LI, % labeled cells) for Ki-67 was calculated for each endometrial cell compartment (luminal epithelium, glandular epithelium, stroma). Histologically, nascent endometrial glands were present in all pre- and postnatal uterine tissues. Overall, Ki-67 LI increased (P < 0.0001) from the pre-to postnatal periods, and was higher (P < 0.0001) in epithelium as compared to stroma. Postnatally, endometrial Ki-67 LI increased (P < 0.0001) from week 1 to week 24. Our findings confirm that, in contrast to neonatal patterns of uterine development described for domestic ungulates, equine endometrial histogenesis begins prenatally, marked by the appearance of uterine glands as early as GD 300. Epithelial proliferation associated with maturation of the equine endometrium is pronounced by postnatal week 24.

Mismating, or termination of pregnancy, is a commonly requested reproductive procedure for bitches and queens. Surgical treatment via ovariohysterectomy is the preferred choice when bitches or queens are not desired for future breeding purposes. Animals that are reproductively valuable can be treated with a variety of drugs to terminate the pregnancy. The choice of specific medical therapy is based on safety, efficacy, availability of the drug, and gestational age of the pregnancy. Currently, there is no US Food and Drug Administration-approved treatment of mismating in North America for dogs or cats.

Chlorhexidine gluconate solution is a potent antimicrobial and therefore could be used effectively for treatment of endometritis, but historically this substance has been implicated as irritating to mucous membranes, including the endometrium of the mare. The use of chlorhexidine hydrochloride suspension (Nolvasan Suspension, Zoetis, Florham Park, NJ, USA) was evaluated in the uterus of normal mares to determine if adverse effects on endometrial health were noted. Twelve healthy, adult light breed mares were included in this study. Procedures were approved by the Auburn University Institutional Animal Care and Use Committee. All mares were determined to be reproductively normal by evaluation of endometrial histopathology, cytology, and bacterial culture. Mares were randomly assigned to treatment or control groups (n = 6 per group). Each mare was treated during estrus with an intrauterine infusion of 1 g (28 mLs per tube; 35.7 mg/mL) of chlorhexidine hydrochloride suspension (treatment group) or an equal volume of lactated ringer's solution (control group) once daily for 3 consecutive days. Biopsy and cytology samples were taken 3, 7, and 14 days after completion of treatment. Cytology and biopsy samples were read by a board-certified pathologist (L.N.) blinded to treatments, and biopsy samples were graded using a standardized Kenney-Doig score. There was no difference with respect to biopsy grade, degree of endometrial fibrosis, or presence of cytologic inflammation comparing control and treatment groups (P = 0.55, 0.7, and 0.06, respectively), neither when accounting for sampling day. The suspension was visible within the uterine lumen when mares were examined with transrectal ultrasonography for up to 4 days after treatment. Treatment with chlorhexidine hydrochloride in this formulation and at this concentration does not appear to have a deleterious effect on short term endometrial health in mares.
