RESUMO

The migratory insect Mythimna separata is a major pest of grain crops in Asia. Unfortunately, the molecular mechanisms that control and regulate reproduction in this species remain unclear. In this study, transcriptome sequencing was utilized to identify genes associated with ovary development and oogenesis. Clean sequences totaling 117.71 Gb were assembled into 178,534 unigenes with a mean length of 647.37 bp and N50 length of 837 bp. Transcriptome analysis showed that 7921 unigenes were significantly expressed in ovaries with 4403 and 3518 unigenes up- and down-regulated, respectively. Enrichment analysis with the Kyoto Encyclopedia of Genes and Genomes database suggested that 729 differentially expressed genes were significantly enriched in the top 20 pathways (q-values <0.05). Twenty genes were associated with ovary development and oogenesis and included lipases, Nanos, small heat shock proteins (sHsps) and histones; these were further verified by qRT-PCR and may play essential roles in M. separata reproduction. Collectively, our findings reveal underlying mechanisms of M.separata reproduction and may lead to RNAi-based management strategies targeting reproductive physiology.

Background: Allergic rhinoconjunctivitis (ARC) and allergic rhinitis (AR) are prevalent allergic diseases. People are becoming increasingly aware of the impact of microbial disorders on host immunity and allergic diseases. Studies have demonstrated an association between allergic diseases and the microbiome, but much remains unknown. We assessed changes in the conjunctival microbiome and nasal microbiome in patients with ARC or AR. Methods: Conjunctival swabs and nasal swabs were collected from each participant for 16S rRNA amplicon sequencing. Bacterial communities were analyzed. Results: Forty patients with ARC, 20 patients suffering from AR, and 34 healthy controls (HCs) were recruited. This study found the abundance of conjunctival microbiome in patients with ARC or AR was significantly lower than that in HCs. The diversity of conjunctival microbiome in patients with AR was significantly lower than those in the other two groups. There is no significant difference in abundance of nasal microbiome between the three groups. The diversities of nasal microbiome in patients with ARC or AR were significantly lower than that in HCs. We found significant differences in microbiota compositions in patients with ARC or AR compared with those in HCs. However, no significant difference in microbiota compositions was found between patients with ARC and patients with AR. Microbiome functions in the ARC group and AR group were also altered compared with HCs. Conclusions: We revealed changes in the composition and function of the conjunctival microbiome and nasal microbiome of patients with ARC or AR, which suggests that there is a relationship between allergic conditions and the local microbiome.

The diamides, chlorantraniliprole (CHL) and cyantraniliprole (CYA), have been used as seed treatment agents against the fall armyworm (FAW), Spodoptera frugiperda in China. However, large-scale application of these two insecticides is prohibited because of their high cost. The neonicotinoid insecticides, clothianidin (CLO) and thiamethoxam (THI), are cheaper and widely used. In this study, we tested the efficacy of CHL + CLO and CYA + THI as seed treatment agents against FAW larvae both in laboratory and field conditions. Laboratory experiments showed that the two binary mixtures (both 240 g.a.i.100 kg-1 corn seeds) caused FAW mortality exceeded 84.00% at 14 days after seedling emergence (DAE). The mortality of the binary mixtures were similar to either CHL (300 g.a.i.100 kg-1corn seeds) or CYA (144 g a.i.100 kg-1corn seeds), but higher than CLO (120 g.a.i.100 Kg-1corn seeds) or THI (180 g a.i.100 kg-1corn seeds). Two independent field experiments showed that both binary mixtures resulted in above 68.00% control efficacy at 14 DAE, suggesting that these insecticidal combinations could effectively control FAW over a relative long period. In addition, both binary mixtures showed no negative effects on the growth and development of corn seedlings. The residues of binary mixtures in corn leave were also lower at 28 DAE as compared to residues in CHL or CYA alone. Most importantly, the costs of CHL + CLO were reduced up to 50% and CYA + THI up to 20% when compared to singly used chemical. Totally, our results indicated that CHL + CLO and CYA + THI had the same control efficacy as CHL or CYA alone, but with much lower cost.

Immune cells have an uncertain function during the progression of extranodal natural killer/T-cell lymphoma (ENKTL). The present study determined the distribution, phenotype, and clinical significance of B lymphocytes in ENKTL. Immunohistochemistry indicated high infiltration of CD20+ B lymphocytes in the tumour tissues of 40% of the patients, and that a high infiltration correlated with better overall survival. Moreover, B lymphocytes had an active mature phenotype in situ and suppressed the proliferation of ENKTL cells in vitro. These results suggest that tumour infiltration of CD20+ B lymphocytes may be a new prognostic indicator for patients with ENKTL.

This paper reports the covalent functionalization of graphene oxide (GO) by a presynthesized metal-organic framework NH2-MIL-101(Fe) via ultrasonication of the two components. The formation of Fe-O covalent bonding in the NH2-MIL-101(Fe)-GO nanohybrid is clearly evidenced, and the covalent bonding still remains after electrochemical reduction. The morphology and structure of the nanohybrid are characterized via scanning electron microscopy, transmission electron microscopy, UV-vis spectroscopy, Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, X-ray diffraction, and Raman spectroscopy. The electrode based on electrochemically reduced NH2-MIL-101(Fe)-GO shows ultrastable and high-sensitive performance in simultaneous electrochemical sensing of three purine metabolic derivatives (uric acid, xanthine, and hypoxanthine); in particular, no signal fading is seen even after running for 120 times. The covalent bonding within the nanohybrid is obviously the key to maintain such a stability.

We present the cluster-to-cluster transformations among three gold nanoclusters, [Au6(dppp)4]2+ (Au6), [Au8(dppp)4Cl2]2+ (Au8) and [Au11(dppp)5]3+ (Au11). The conversion process follows a rule that states that the transformation of a small cluster to a large cluster is achieved through an oxidation process with an oxidizing agent (H2O2) or with heating, while the conversion of a large cluster to a small one occurs through a reduction process with a reducing agent (NaBH4). All the reactions were monitored using UV-Vis spectroscopy and ESI-MS. This work may provide an alternative approach to the synthesis of novel gold nanoclusters and a further understanding of the structural transformation relationship of gold nanoclusters.

Visual deprivation during the critical period induces long-lasting changes in cortical circuitry by adaptively modifying neuro-transmission and synaptic connectivity at synapses. Spike timing-dependent plasticity (STDP) is considered a strong candidate for experience-dependent changes. However, the visual deprivation forms that affect timing-dependent long-term potentiation(LTP) and long-term depression(LTD) remain unclear. Here, we demonstrated the temporal window changes of tLTP and tLTD, elicited by coincidental pre- and post-synaptic firing, following different modes of 6-day visual deprivation. Markedly broader temporal windows were found in robust tLTP and tLTD in the V1M of the deprived visual cortex in mice after 6-day MD and DE. The underlying mechanism for the changes seen with visual deprivation in juvenile mice using 6 days of dark exposure or monocular lid suture involves an increased fraction of NR2b-containing NMDAR and the consequent prolongation of NMDAR-mediated response duration. Moreover, a decrease in NR2A protein expression at the synapse is attributable to the reduction of the NR2A/2B ratio in the deprived cortex.

Permutational isomers of trigonal bipyramidal [W2RhIr2(CO)9(η(5)-C5H5)2(η(5)-C5HMe4)] result from competitive capping of either a W2Ir or a WIr2 face of the tetrahedral cluster [W2Ir2(CO)10(η(5)-C5 H5)2] from its reaction with [Rh(CO)2(η(5)-C5HMe4)]. The permutational isomers slowly interconvert in solution by a cluster metal vertex exchange that is proposed to proceed by Rh-Ir and Rh-W bond cleavage and reformation, and via the intermediacy of an edge-bridged tetrahedral transition state. The permutational isomers display differing chemical and physical properties: replacement of CO by PPh3 occurs at one permutational isomer only, while the isomers display distinct optical power limiting behavior.

The trigonal bipyramidal clusters M2Ir3(µ-CO)3(CO)6(η(5)-C5H5)2(η(5)-C5Me4R) (M = Mo, R = Me 1a, R = H; M = W, R = Me, H) reacted with isocyanides to give ligand substitution products M2Ir3(µ-CO)3(CO)5(CNR')(η(5)-C5H5)2(η(5)-C5Me4R) (M = Mo, R = Me, R' = C6H3Me2-2,6 3a; M = Mo, R = Me, R' = (t)Bu 3b), in which core geometry and metal atom locations are maintained, whereas reactions with PPh3 afforded M2Ir3(µ-CO)4(CO)4(PPh3)(η(5)-C5H5)2(η(5)-C5Me4R) (M = Mo, R = Me 4a, H 4c; M = W, R = Me 4b, H), with retention of core geometry but with effective site-exchange of the precursors' apical Mo/W with an equatorial Ir. Similar treatment of trigonal bipyramidal MIr4(µ-CO)3(CO)7(η(5)-C5H5)(η(5)-C5Me5) (M = Mo 2a, W 2b) with PPh3 afforded the mono-substitution products MIr4(µ-CO)3(CO)6(PPh3)(η(5)-C5H5)(η(5)-C5Me5) (M = Mo 5a; M = W 5b), and further reaction of the molybdenum example 5a with excess PPh3 afforded the bis-substituted cluster MoIr4(µ3-CO)2(µ-CO)2(CO)4(PPh3)2(η(5)-C5H5)(η(5)-C5Me5) (6). Reaction of 1a with diphenylacetylene proceeded with alkyne coordination and C≡C cleavage, affording Mo2Ir3(µ4­Î·(2)-PhC2Ph)(µ3-CPh)2(CO)4(η(5)-C5H5)2(η(5)-C5Me5) (7a) together with an isomer. Reactions of 2a and 2b with PhC≡CR afforded MIr4(µ3­Î·(2)-PhC2R)(µ3-CO)2(CO)6(η(5)-C5H5)(η(5)-C5Me5) (M = Mo, R = Ph 8a; M = W, R = Ph 8b, H; M = W, R = C6H4(C2Ph)-3 9a, C6H4(C2Ph)-4), while addition of 0.5 equivalents of the diynes 1,3-C6H4(C2Ph)2 and 1,4-C6H4(C2Ph)2 to WIr4(µ-CO)3(CO)7(η(5)-C5H5)(η(5)-C5Me5) gave the linked clusters [WIr4(CO)8(η(5)-C5H5)(η(5)-C5Me5)]2(µ6­Î·(4)-PhC2C6H4(C2Ph)-X) (X = 3, 4). The structures of 3a, 4a­4c, 5b, 6, 7a, 8a, 8b and 9a were determined by single-crystal X-ray diffraction studies, establishing the core isomerization of 4, the site selectivity for ligand substitution in 3­6, the alkyne C≡C dismutation in 7, and the site of alkyne coordination in 7­9. For clusters 3­6, ease of oxidation increases on increasing donor strength of ligand, increasing extent of ligand substitution, replacing Mo by W, and decreasing core Ir content, the Ir-rich clusters 5 and 6 being the most reversible. For clusters 7­9, ease of oxidation diminishes on replacing Mo by W, increasing the Ir content, and proceeding from mono-yne to diyne, although the latter two changes are small. In situ UV-vis-near-IR spectroelectrochemical studies of the (electrochemically reversible) reduction process of 8b were undertaken, the spectra becoming increasingly broad and featureless following reduction. The incorporation of isocyanides, phosphines, or alkyne residues in these pentanuclear clusters all result in an increased ease of oxidation and decreased ease of reduction, and thereby tune the electron richness of the clusters.

Metal cluster core expansion at tetrahedral group 6-group 9 mixed-metal clusters MIr3(µ-CO)3(CO)8(η(5)-L) (M = W, Mo, L = C5H5; M = Mo, L = C5Me5) with the iridium capping reagents Ir(CO)2(η(5)-L') (L' = C5Me5, C5Me4H) in refluxing toluene afforded the trigonal-bipyramidal clusters MIr4(µ-CO)3(CO)7(η(5)-C5H5)(η(5)-L') (M = Mo, L' = C5Me5, 1a; M = W, L' = C5Me5, 1b; M = Mo, L' = C5Me4H, 1c; M = W, L' = C5Me4H, 1d) and MoIr4(µ3-H)(µ-CO)2(µ-η(1):η(5)-CH2C5Me4)(CO)7(η(5)-C5Me5) (2). Related reactions with M2Ir2(µ-CO)3(CO)7(η(5)-L)2 (M = W, Mo, L = C5H5; M = Mo, L = C5Me5) afforded M2Ir3(µ-CO)3(CO)6(η(5)-C5H5)2(η(5)-L') (M = Mo, L' = C5Me5, 3a; M = W, L' = C5Me5, 3b; M = Mo, L' = C5Me4H, 3c; M = W, L' = C5Me4H, 3d), W2Ir3(µ-CO)4(CO)5(η(5)-C5H5)2(η(5)-C5Me4H) (4), and Mo2Ir3(µ-CO)3(CO)6(η(5)-C5Me5)3 (5). Single-crystal X-ray diffraction studies of 1a-1d, 2, 3a-3d, and 4 confirmed their molecular structures, including the µ-η(1):η(5)-CH2C5Me4 ligand at hydrido cluster 2, derived from a C-H bond activation of one of the methyl groups. Density functional theory (DFT) studies were employed to suggest the structure of 5. The redox behavior of the new clusters was examined through cyclic voltammetry; all clusters exhibit oxidation and reduction processes (with respect to the resting state), with the oxidation processes being the more reversible, and increasingly so on decreasing Ir content of the clusters, replacing W by Mo, and increasing alkylation of the cyclopentadienyl ligands. In situ IR and UV-vis-near-IR spectroelectrochemical studies of the reversible oxidation processes in 1a and 3a were undertaken, with the spectra of the former suggesting progression to an all-terminal CO geometry concomitant with the first oxidation and a significant structural change upon the second oxidation step. DFT studies of 1a revealed that its crystallographically-confirmed Mo-equatorial core geometry is essentially isoenergetic with a possible Mo-apical isomer, and identified several bridging CO structures for the charged states.

{[Cu(6)(TTTMB)(8)(OH)(4)(H(2)O)(6)]·8(NO(3))·34.5H(2)O}(n) can reversibly transform to {[Cu(6)(TTTMB)(8)I(3)]·9I·26H(2)O}(n) upon a single crystal to single crystal process. The transformation is accompanied by the formation of weak Cu(2+)I(-) interactions and changes in the coordination numbers, as well as modulation in their properties.
