OBJECTIVE: This study aimed to evaluate the effectiveness of granulocyte colony-stimulating factor (G-CSF) for infertility and recurrent spontaneous abortion. METHODS: Existing research was searched in PubMed, Embase and Cochrane Library till Dec 2021. Randomized control trials (RCTs) that compared G-CSF administration with the control group in infertility women undergoing IVF were included. The primary outcomes included clinical pregnancy rate; the secondary outcomes included live birth rate, abortion ratebiochemical pregnancy rate, embryo implantation rate, as well as endometrial thickness. RESULT(S): 20 RCTs were included in this study. G-CSF increased the clinical pregnancy rate (RR = 1.85; 95% CI: 1.07, 3.18) and the endometrial thickness (MD = 2.25; 95% CI: 1.58,2.92;) in patients with thin endometrium undergoing IVF. G-CSF increased the biochemical pregnancy rate (RR = 2.12; 95% CI: 1.54, 2.93), the embryo implantation rate (RR = 2.51; 95% CI: 1.82, 3.47) and the clinical pregnancy rate (RR = 1.93; 95% CI: 1.63, 2.29) in patients with a history of repeated implantation failure undergoing IVF. No differences were found in pregnancy outcomes of general IVF patients. CONCLUSIONS: Granulocyte colony-stimulating factor is likely to be a potential option for infertility women undergoing IVF with thin endometrium or recurrent implantation failure . BACKGROUND: Heterotopic pregnancy (HP) is a rare condition in which both ectopic and intrauterine pregnancies occur. HP is uncommon after natural conception but has recently received more attention due to the widespread use of assisted reproductive techniques (ART) such as ovulation promotion therapy. CASE SUMMARY: Here, we describe a case of HP that occurred after ART with concurrent tubal and intrauterine singleton pregnancies. This was treated successfully with surgery to preserve the intrauterine pregnancy, resulting in the birth of a low-weight premature infant. This case report aims to increase awareness of the possibility of HP during routine first-trimester ultrasound examinations, especially in pregnancies resulting from ART and even if multiple intrauterine pregnancies are present. CONCLUSION: This case alerts us to the importance of comprehensive data collection during regular consultations. It is important for us to remind ourselves of the possibility of HP in all patients presenting after ART, especially in women with an established and stable intrauterine pregnancy that complain of constant abdominal discomfort and also in women with an unusually raised human chorionic gonadotropin level compared with simplex intrauterine pregnancy. This will allow symptomatic and timeous treatment of patients with better results.

In order to find a facile and practical method to synthesize amino cellulose in bulk with high regional selectivity and high degree of substitution, the reaction conditions to brominate cellulose and to reduce azido group were carefully studied and some interesting phenomena were observed. With the optimized method, 6-amino-6-deoxy cellulose could be easily prepared with very simple separation techniques. The degree of substitution of the amino group amounted to 0.97 which was determined by 1H NMR spectrum of 6-benzamido-6-deoxy cellulose. Moreover, the amino group was evidenced to be at the C6 of glucose unit by 1H-1H COSY NMR and 1H-13C HSQC NMR spectra. In addition, three cellulose 6-acetamido-6-deoxy-2,3-bis(phenylcarbamate)s were prepared from the 6-amino-6-deoxy cellulose prepared with the techniques optimized in the present study. The developed cellulose derivatives were used as chiral selectors with which chiral stationary phases (CSPs) were prepared. The CSPs exhibited enantioseparation power to some chiral compounds.

BACKGROUND: In plants, CONSTITUTIVELY PHOTOMORPHOGENIC 1 (COP1) is a key negative regulator in photoperiod response. However, the biological function of COP1-interacting protein 1 (CIP1) and the regulatory mechanism of the CIP1-COP1 interaction are not fully understood. RESULTS: Here, we identified the apple MdCIP1 gene based on the Arabidopsis AtCIP1 gene. Expression pattern analysis showed that MdCIP1 was constitutively expressed in various tissues of apple, and responded to stress and hormone signals at the transcriptional level. Ectopic expression of MdCIP1 complemented the phenotypes of the Arabidopsis cip1 mutant, and MdCIP1 inhibited anthocyanin biosynthesis in apple calli. In addition, the biochemical assay demonstrated that MdCIP1 could interact with MdCOP1 protein by their coiled-coil domain, and MdCIP1-OX/cop1-4 had a similar phenotype in photomorphogenesis with the cop1-4 mutant, suggesting that COP1 is epistatic to CIP1. Furthermore, the transient transformation assay indicated that MdCIP1 repressed anthocyanin biosynthesis in an MdCOP1-mediated pathway. CONCLUSION: Take together, this study finds that MdCIP1 acts as a repressor in regulating hypocotyl elongation and anthocyanin biosynthesis through MdCOP1 in apple.

Nitrate is essential for plant growth and development. When nitrate availability is low, plants produce more lateral roots (LRs) to seek nitrate from the soil. In this study, by DNA electrophoretic mobility shift and luciferase assays, it was showed that NIN-like protein 7 (NLP7) transcription factor activated expression of TAR2 by directly binding to its promoter. Finally, through genetic analysis, it was speculated that NLP7 regulated LR development through TAR2. In conclusion, NLP7 binds to the TAR2 promoter and activates TAR2 expression, thereby promoting nitrate-dependent LR development.

COP1, an important RING ubiquitin ligase E3, is a molecular switch for light regulation in plant development. As an interacting protein of COP1, CIP8 contains a RING-H2 domain, but its biological function is unclear. Here, the apple MdCIP8 was identified based on its homology with AtCIP8 in Arabidopsis. MdCIP8 was constitutively expressed at different levels in various apple tissues, and the expression level of MdCIP8 was not affected by light and dark conditions. MdCIP8 reversed the short hypocotyl phenotype of the cip8 mutant under light conditions. Furthermore, the yeast two-hybrid experiment showed that MdCIP8 interacted with the RING domain of MdCOP1 through its RING-H2 domain. MdCIP8-OX/cop1-4 exhibited the phenotype of the cop1-4 mutant, indicating that CIP8 acts upstream of COP1. In addition, an apple transient injection experiment showed that MdCIP8 inhibited anthocyanin accumulation in an MdCOP1-dependent pathway. Overall, our findings reveal that CIP8 plays an inhibitory role in the light-regulation responses of plants.

Endometriosis is a common debilitating gynecologic disease. Almost 10% of reproductive-age women are affected by this disease; they commonly suffer pelvic pain and/or infertility. Early diagnosis of this multifactorial disease remains difficult because its etiology is not clear and the early symptoms are nonspecific. In addition, many reproductive-age women are unwilling to undergo invasive laparoscopic surgery because of the possibility of decreasing fertility. Thus, identifying biomarkers for the early diagnosis of endometriosis a key focus of current research. Long noncoding RNAs (lncRNAs) are a class of noncoding transcripts that have length of > 200 nucleotides and lack protein-coding ability but still influence gene expression in various ways. With advances in genome-wide analysis, researchers have determined that lncRNAs play an important role in many human diseases, particularly tumors. Moreover, the role of lncRNAs in the pathogenesis of endometriosis has been continually recognized. In this review, we discuss the status of current research on dysregulated lncRNAs and their roles in the pathogenesis of endometriosis. We aim to stimulate new investigations toward the identification of lncRNAs as biomarkers for the early diagnosis and therapy of this long-term gynecological disease.

Sulfate-reducing prokaryotes (SRP) and sulfur-oxidizing prokaryotes (SOP) play vital roles in the sulfur cycle. The SRP community was used to represent a microbial community with high richness and diversity. The 454 pyrosequencing, Illumina high-throughput sequencing, and traditional clone library methods that target the dissimilatory sulfite reductase ß subunit gene (dsrB), which encodes a key enzyme in the sulfate reduction pathway, were used to compare the differences in SRP community characteristics. Comparative analyses suggested that Illumina high-throughput sequencing was a more appropriate method for SRP (high richness and diversity) community studies. The SOP soxB gene (~750 bp) was used as a representative of the long-sequence segment. The 454 pyrosequencing and Illumina high-throughput sequencing methods were used to compare the differences in SOP community characteristics. The results revealed that 454 pyrosequencing did not reflect its advantage of a longer read length; whereas, the Illumina high-throughput sequencing with more numerous and shorter sequence reads was more suitable when the soxB gene was used to investigate the community composition and diversity of SOP.

Polycystic ovary syndrome (PCOS) is one of the most common endocrine disorders in reproductive-aged women. However, the exact pathophysiology of PCOS remains largely unclear. We performed deep sequencing to investigate the mRNA and long noncoding RNA (lncRNA) expression profiles in the ovarian tissues of letrozole-induced PCOS rat model and control rats. A total of 2147 mRNAs and 158 lncRNAs were differentially expressed between the PCOS models and control. Gene ontology analysis indicated that differentially expressed mRNAs were associated with biological adhesion, reproduction, and metabolic process. Pathway analysis results indicated that these aberrantly expressed mRNAs were related to several specific signaling pathways, including insulin resistance, steroid hormone biosynthesis, PPAR signaling pathway, cell adhesion molecules, autoimmune thyroid disease, and AMPK signaling pathway. The relative expression levels of mRNAs and lncRNAs were validated through qRT-PCR. LncRNA-miRNA-mRNA network was constructed to explore ceRNAs involved in the PCOS model and were also verified by qRTPCR experiment. These findings may provide insight into the pathogenesis of PCOS and clues to find key diagnostic and therapeutic roles of lncRNA in PCOS.

Ovarian cancer (OC) is the leading cause of death among all gynecological malignancies in the world and its underlying mechanism is still unclear. Compared with research on microRNAs, research on long non-coding RNAs (lncRNAs) is still in its infancy. Studies in recent years have demonstrated that lncRNAs exhibit multiple biological functions in various stages of OC development. In this review, we conclude that lncRNAs are closely involved in the pathogenesis of OC. The expression of lncRNAs indicates the early diagnosis, prognosis, and response to chemotherapy of OC. An attractive approach to treatment of OC is lncRNA small interfering RNA or acting as a plasmid targeting the expression of toxic genes, which is a novel step toward a major breakthrough in the treatment of human OC. E2-regulated lncRNA and its polymorphism, methylation, are also involved in OC. Further research efforts are needed before fully identifying, characterizing, and elucidating the actual functions of lncRNAs in OC at the molecular level and putting them into clinical practice.

Marine nitrogen cycle is of great significance to maintain the marine ecological balance. Anaerobic ammonia oxidation (ANAMMOX) process driven by anaerobic ammonia-oxidizing bacteria is an important way of promoting the marine nitrogen removal. In this study, quantitative PCR (qPCR), combining with environmental factors analysis, were used to study the distribution of ANAMMOX bacteria in the sediments from Yangtze River Estuary and its adjacent waters in summer, 2011. The results showed that the spatial abundance distribution of ANAMMOX bacteria was closely related to environmental factors. Total organic carbon, salinity, and concentration of ammonium played vital roles in regulating ANAMMOX bacteria abundances in this area. The abundance distribution of ANAMMOX bacteria was significantly positively correlated with total organic carbon content (P<0.01,r=0.69), reducing along with the increasing concentration of nitrite and DO, while the influence from salinity was complicated.

Nitrogen cycle is a key process in material circulation of marine ecosystem, which plays an important role in maintaining ecological balance. The ammonia oxidation process promoted by aerobic ammonia-oxidizing microorganism (AOM) is a rate-limiting step of nitrification. Real-time quantitative polymerase chain reaction (qPCR ), along with the determination of potential nitrification rates (PNR) was carried out in this study to understand the distribution of AOM in sediments of adjacent waters of Rushan Bay in August, 2014. The results indicated that the abundance of total ammonia-oxidizing bacteria (AOB) was always greater than that of total ammonia-oxidizing archaea (AOA) in the three sampling stations; the ratio of active AOB to total AOB was less than 1%, while no active AOA was detected in this study; the PNR significantly decreased after adding ampicillin which could inhibit the activity of AOB (P < 0.05). It was speculated that AOB might play a more important role in the ammonia oxidation in sediments of adjacent waters of Rushan Bay in August, 2014. Dissolved oxygen concentrations, temperature and ammonium concentrations played a significant role in distribution of AOM in sediments of adjacent waters of Rushan Bay.

RESUMO

Globozoospermia is a rare and serious teratozoospermia, which is one of the important causes contributing to human male infertility. The assisted reproductive technique remains the only means for such patients to produce offspring. However, the pathogenesis of globozoospermia is not yet clear. In recent years, related studies have shown that some genes are connected with the onset of globozoospermia. This paper outlines the progress in the studies of pathogenicity genes, aiming to contribute to the molecular diagnosis and mechanism investigation of the disease.

Recurrent spontaneous abortion (RSA) has various complicated causes, and more and more researches are focused on its etiology. Genetic factors are the most common risk factors of RSA; immune factors, infection factors, male factors and female factors play an important role; environmental pollution and some other unknown factors may also be conspirators. This article presents an overview on the possible risk factors of RSA.
