RESUMO

Here, a molecularly imprinted electrochemical sensor (MIECS) was designed and fabricated for specifically monitoring norfloxacin (NFX), an entirely synthetic antibiotic. In which, Cu2+ dopped covalent organic framework (COF) was used to connect NFX imprinting layer and glassy carbon electrode through covalence. Under optimized conditions, the linear range is as wide as 5 orders of magnitude, and the detection limit is 5.94 × 10-3 µM (estimated based on S/N = 3). Average recoveries are among 92.4%-99.0% with relative standard deviations ≤ 4.05% (n = 3) in (spiked) whole, low-fat, and skimmed milk, validated by independent HPLC assays. The excellent performance can be ascribed to the significant recognition and enriching ability of the imprinting layer, improved conductivity of Cu2+ dopped covalent organic framework, and high stability of covalence between layers. We hope the work will act as a model of MIECSs for rapidly and selectively detecting trace drug residue in complex real samples.

RESUMO

Anomalous solid-like liquids at the solid-liquid interface have been recently reported. The mechanistic factors contributing to these anomalous liquids and whether they can stably exist at high vacuum are interesting, yet unexplored, questions. In this paper, thin slices of silica tubes soaked in hexadecane were observed under a transmission electron microscope at room temperature. The H-spectrum of hexadecane in the microtubules was measured by nuclear magnetic resonance. On the interior surface of these silica tubes, 0.2-30 µm in inside diameter (ID), a layer (12-400 nm) of a type of non-volatile hexadecane was found with thickness inversely correlated with the tube ID. A sample of this anomalous hexadecane in microtubules 0.4 µm in ID was found to be formable by an ion beam. Compared with the nuclear magnetic resonance H-spectroscopy of conventional hexadecane, the characteristic peaks of this abnormal hexadecane were shifted to the high field with a broader characteristic peak, nuclear magnetic resonance hydrogen spectroscopy spectral features typical of that of solids. The surface density of these abnormal hexadecanes was found to be positively correlated with the silanol groups found on the interior silica microtubular surface. This positive correlation indicates that the high-density aggregation of silanol is an essential factor for forming the abnormal hexadecane reported in this paper.

RESUMO

Lycopene is a useful phytochemical that holds great commercial value. In our study the lycopene production pathway in E. coli originating from the precursor isopentenyl diphosphate (IPP) of the non-mevalonate pathway was reconstructed. This engineered strain of E. coli accumulated lycopene intracellularly under aerobic conditions. As a next step, the production of lycopene was enhanced through metabolic engineering methodologies. Various competing pathways at the pyruvate and acetyl-CoA nodes were inactivated to divert more carbon flux to IPP and subsequently to lycopene. It was found that the ackA-pta, nuo mutant produced a higher amount of lycopene compared to the parent strain. To further enhance lycopene production, a novel mevalonate pathway, in addition to the already existing non-mevalonate pathway, was engineered. This pathway utilizes acetyl-CoA as precursor, condensing it to form acetoacetyl-CoA and subsequently leading to formation of IPP. Upon the introduction of this new pathway, lycopene production increased by over 2-fold compared to the ackA-pta, nuo mutant strain.
