PURPOSE: To determine whether inhibition of the F11 receptor/JAM-A (F11R) using F11R-specific antagonist peptide 4D results in inhibition of smooth muscle cell (SMC) proliferation and migration in vivo, known as neointimal hyperplasia (NIH), using a mouse focal carotid artery stenosis model (FCASM). MATERIALS AND METHODS: The mouse FCASM was chosen to test the hypothesis because the dominant cell type at the site of stenosis is SMC, similar to that in vascular access stenosis. Fourteen C57BL/6 mice underwent left carotid artery (LCA) partial ligation to induce stenosis, followed by daily injection of peptide 4D in 7 mice and saline in the remaining 7 mice, and these mice were observed for 21 days and then euthanized. Bilateral carotid arteries were excised for histologic analysis of the intima and media areas. RESULTS: The mean intimal area was significantly larger in control mice compared with peptide 4D-treated mice (0.031 mm2 [SD ± 0.024] vs 0.0082 mm2 [SD ± 0.0103]; P = .011). The mean intima-to-intima + media area ratio was significantly larger in control mice compared with peptide 4D-treated mice (0.27 [SD ± 0.13] vs 0.089 [SD ± 0.081]; P = .0079). NIH was not observed in the right carotid arteries in both groups. CONCLUSIONS: Peptide 4D, an F11R antagonist, significantly inhibited NIH in C57BL/6 mice in a FCASM.

The fruiting bodies of bracket fungi are a specific microhabitat colonized by various invertebrates of which mites (Acari) are rarely studied, and if they are, the study is usually faunistic. The aim of the research was to determine whether the diversification of mite assemblages (Mesostigmata, Oribatida) inhabiting the fruiting bodies of Fomitopsis pinicola (Sw.) P. Karst. (Polyporales) are connected with the character of the forests and/or the degree of decay (DD) of the fruiting bodies. The research was conducted at Bialowieza National Park (BNP), in forests close to natural ones and in Karkonosze National Park (KNP) which was affected by a large-scale forest dieback in the 1980s. Eighty fruiting bodies (40 at each study site) of F. pinicola belonging to four DD categories were collected. In total, 4,345 individuals of 120 mite species were recorded at BNP, and 13,912 individuals of 96 species were recorded at KNP. Analyses revealed that the sample dispersion at each study site was comparable, nevertheless the samples from each study site were clearly grouped into slightly overlapping sets which allow observation of the differences between them. In the less decayed fungi (DD 1 and 2) there were fewer mite species and individual mites than in the more decayed samples (DD 3 and 4). There were also significant differences between the fauna of the fungi in each particular DD: the fauna of DD 1 differed from all others, whereas the fauna of heavily decayed fungi (DD 3 and 4) was more comparable.

BACKGROUNDS AND AIMS: To address the problem of resistance to standard antiplatelet therapy in some patients, our team proposed a purinoceptor-dependent dual therapy. Its efficacy is also determined by the condition of the vascular endothelium which, by secreting numerous factors, is involved in hemostasis. Among them, thrombospondin-1 is important in the context of thrombotic events. Therefore we sought to determine if the novel dual purinoceptor-dependent concept is associated with TSP-1 changes in vascular endothelial cells. METHODS AND RESULTS: TSP-1 expression in human microvascular endothelial cells was determined at transcriptional and protein level. We performed real-time PCR, the Western blot analysis and ELISA test. We found that TSP-1 mRNA and protein expression levels significantly changed in response to P1R agonists treatment. Furthermore, we have observed that co-administration of selective A2AR agonists (UK-432,097 or MRE0094) with P2Y12R antagonists altered TSP-1 expression levels, and the direction of these changes was not synergistic. MRE0094 applied with ARC69931MX or R-138727 increased mRNA expression from 39 to 56 or 57%, respectively (*P < 0.05 vs. MRE0094; ***P < 0.001 vs. control). Also, in the case of the P2Y12R antagonists used together with UK-432,097, there was an increase from 53 to 71 and 70% (*P < 0.05 vs. UK-432,097; ***P < 0.001 vs. control). The observed trends in gene expression were reflected in the protein expression and the level of its secretion from HMEC-1. CONCLUSION: The article presents evidence which proves that the purinoceptor-dependent concept is associated with TSP-1 changes in endothelial cells (EC). Moreover, Human Microvascular Endothelial Cells treatment applied together with agonists (MRE0094 or UK-432,097) and P2Y12R antagonist did not result in any synergistic effect, implicating a possible crosstalk between G proteins in GPCRs dependent signaling. Therefore, we suggest that understanding of the specific mechanism underlying TSP-1 alterations in EC in the context of the dual purinoceptor-dependent approach is essential for antiplatelet therapies and should be the subject of future research.

Bracket fungi are seen mainly as the cause of economic losses in forestry, and their role as creators of biodiversity is relatively poorly understood. The aim of the study was defining the manner in which the degree of decay (DD) of the fruiting bodies determines the character of the invertebrate assemblages colonising them. The effect of this group of fungi on the modification of biodiversity of invertebrates (Aranae, Opiliones, Pseudoscorpionida, two groups of mites-Mesostigmata and Oribatida, and Collembola and Insecta) was investigated by analyzing 100 fruiting bodies of 10 species of bracket fungi divided into four DD classes. The material was collected at Bialowieza National Park, which is considered to be the largest area of natural forests in the North European Plain. 16 068 invertebrate individuals classified into 224 species were obtained. Oribatid mites (12 543 individuals) constituted the largest group of individuals, which were classified into 115 species with the most numerous Carabodes femoralis (8,811 individuals). Representatives of this group of mites have been reported previously in the publications on bracket fungi; however, the contributions of Oribatida and other groups of invertebrates were not broadly compared. Moreover, the species such as Hoploseius mariae and H. oblongus, which were predominantly found in fruiting bodies of bracket fungi, have also been discerned. The invertebrate fauna differs depending on DD of the samples: In the more decayed samples, a higher number of both individuals and species were recorded compared to the samples with lower DDs; however, this trend proved to be nonlinear. The DCA and cluster analysis revealed a similarity of the invertebrate assemblages from the 2 DD and 4 DD samples. They also indicated that the group 3 DD differed the most from all the other samples. The indicator species analysis identified species characteristic to individual DDs: For group 1 DD, it was, for example, Hoploseius oblongus; for 2 DD-Orchesella bifasciata; and for 3 DD-Chernes cimicoides, while for 4 DD-Dinychus perforatus.

In the recent years, the awareness of the role purinergic signaling plays as a therapeutic target has increased considerably. The purinoceptor allows the action of extracellular nucleotides (P2 receptors) and intermediary products of their metabolism, such as adenosine (P1 receptors), regulating pivotal processes occurring in the cardiovascular system. This study focuses on a dual purinoreceptor-dependent approach, based on the activation of adenosine P1 receptors with the simultaneous inhibition of P2Y12 receptors that can be used as novel platelet inhibitors in antithrombotic therapy. Endothelial cells are directly exposed to the drugs circulating in the bloodstream. That is why effects of our concept on human microvascular endothelial cells (HMEC-1) were examined in in vitro studies, such as enzyme-linked immunosorbent assay and scratch assays. In response to adenosine receptor agonists, levels of secreted vascular endothelial growth factor varied. Two of them, 5'-N-ethylcarboxamidoadenosine and MRE0094 remarkably increased vascular endothelial growth factor release. The elevated levels were reduced when used together with the P2Y12 receptor antagonist. Also, rates of wound closure in a scratch assay were significantly reduced in these cases. The results suggest that the proposed treatment does not impair endothelial cell condition. In addition, it is suggested as a collateral benefit, namely solving the problem of excessive activation of endothelial cells during antiplatelet therapy.

This study was carried out at Ny-Ålesund on Spitsbergen in Svalbard (High Arctic). Eight study sites were established along a transect from the fjord to the snout of the glacier. The sites differed from each other by the type of vegetation cover and soil characteristics. Soil samples were collected and placed in Tullgren funnels. Extracted arthropods were represented by two groups of mites (Mesostigmata and Oribatida) and springtails (Collembola). The pioneer species that occurred first after retreat of the glacier were representatives of the Collembola (Agrenia bidenticulata and Hypogastrura concolor). Later, other springtails appeared including Folsomia alpha, Folsomia quadrioculata, Hypogastrura concolor, Isotoma anglicana, Sminthurinus concolor and the first species of oribatid mites; Camisia foveolata and Tectocepheus velatus velatus. Arthropod communities recorded along the transect were characterized by large variations in both species composition and abundance of individuals. The greater the distance from the glacier snout, the greater the species richness (2 to 22 species). The mean number of species per sample was the lowest at site 8 (1 ± 0.71) (the closest to the glacier) and greatest at site 1 (14 ± 1.41) (furthest from the glacier). The Simpson's diversity index (D) was distinctly greater at sites 1 (4.61 ± 0.06) and 3 (3.94 ± 0.11) than at other sites, especially site 8 (1.07 ± 0.06). Densities were least in the samples closest to the glacier (30 to 101 individuals; density 3000-10,100 individuals/m2). At the other locations, abundance was highly variable (905 to 7432 individuals; density 90,500-743,200 individuals/m2). The mean abundances were greatest at sites 2 and 3. The great variations in total abundances observed were often due to the presence or absence of one or more dominant species exhibiting extreme abundance variability between sites. The microarthropod community of the High Arctic is composed of heterogeneous circumpolar species, yet on a landscape scale is extremely dependent on local environmental conditions which may be subject to rapid change.

Secretion of PDI from platelets and endothelial cells is an important step of all thrombotic events. In the absence of extracellular PDI thrombus formation and fibrin generation may be impaired. Thrombin-mediated PDI secretion is regulated by the stimulation of P2Y12 receptors. This paper provides evidences that P2Y12 antagonists or AR agonists may modulate release of PDI molecules from platelets and with less efficiency from endothelial cells. Moreover P2Y12 antagonization or AR agonization modulates platelet-endothelial interaction. We prove that combinations of P2Y12 antagonists and AR agonists inhibit platelet-dependent adhesion of cancer cells to endothelium and attenuate cancer cell invasiveness, but longer exposition to AR agonists may stimulate migration of invasive breast cancer cells through endothelium thus leading to increased metastasis.

BACKGROUND: Though vitamin D deficiency is a global problem with wide spectrum of severe public health consequences, inadequate vitamin D status still remains one of the most common and untreated medical conditions. Thyroid diseases, including hypothyroidism, also represent the most frequent endocrinopathies in general population. OBJECTIVES: To determine the vitamin D status in hypothyroid patients and to ascertain the status of thyroid hormone replacement. METHODS: The 25(OH)D concentrations (ECLIA) in 71 hypothyroid patients recruited in the Outpatient Clinic of Endocrinology or Department of Clinical Endocrinology were assessed. The examined group was composed of 59 subjects diagnosed with primary hypothyroidism of different etiology and 12 patients with secondary hypothyroidism. The control group included 16 healthy individuals. RESULTS: Mean serum 25(OH)D concentration in healthy volunteers was significantly lower than in hypothyroid subjects (13.09±1.63 vs. 19.92±1.37 ng/mL). Patients with a history of thyroidectomy presented with significantly higher mean 25(OH)D concentration than controls (23.25±2.75 vs. 13.09±1.63 ng/mL). Mean serum 25(OH)D concentration in effectively treated hypothyroidism was significantly higher than in controls (21.90±1.47 vs. 13.09±1.63 ng/mL) or undertreated hypothyroidism (21.90±1.47 vs. 13.52±3.39 ng/mL). Hypothyroid patients aged under 60 years presented with significantly lower mean 25(OH)D concentration than elders (16.46±1.54 vs. 24.39±1.18 ng/mL). The major 25(OH)D deficient (≤10 ng/mL) or deficient (≤20 ng/mL) hypothyroid patients were significantly younger than those with 25(OH)D concentrations exceeding 10 ng/mL or 20 ng/mL respectively. CONCLUSIONS: These findings confirm the necessity for vitamin D status improvement in the general population and more effective healthcare of hypothyroid patients.
