Little is known about the toxin profiles, toxinotypes and variations of toxin Clostridioides difficile C (tcdC) in Iranian C. difficile isolates. A total of 818 stool specimens were obtained from outpatients (n = 45) and hospitalized patients (n = 773) in Tehran, Iran, from 2011 to 2017. The 44 C. difficile isolates were subjected to PCR of toxin C. difficile A (tcdA), toxin C. difficile B (tcdB), tcdA 3'-end deletion, toxinotyping and sequencing of the tcdC gene. Thirty-eight isolates (86.36%) were identified as tcdA and tcdB positive, and the remaining six isolates (13.63%) were nontoxigenic. All tcdA- and tcdB-positive isolates yielded an amplicon of 2535 bp by PCR for the tcdA 3' end. Fourteen (36.84%), seventeen (44.73%) and seven (18.43%) isolates belonged to wild-type, toxin C. difficile C subclone3 (tcdC-sc3) and tcdC-A genotype of tcdC, respectively. Thirty-one isolates (81.57%) belonged to toxinotype 0, and seven isolates (18.42%) were classified as toxinotype V. This study provides evidence for the circulation of historical and hypervirulent isolates in the healthcare and community settings. Furthermore, it was also demonstrated that the tcdC-A genotype and toxinotype V are not uncommon among Iranian C. difficile isolates.

BACKGROUND: Crystalline silica is a commonly used mineral in various industries and construction activities, and it is so important introducing potential biomarkers to identify early indicators of biological effects in its high-risk occupational exposures. AIM: The present study was aimed to assess the blood and urinary neopterin as an early biomarker of exposure in the workers of an insulator manufacturing plant who are exposed to crystalline silica. SUBJECTS AND METHODS: This analytical descriptive study was done among two groups of exposed workers (n = 55) and unexposed office workers (n = 38) of an insulator manufacturing plant. Statistical software R was used to determine sample size and select the participants by random sampling among nonsmoker workers. Sampling of airborne silica in breathing zone of participants was done based on the National Institute for Occupational Safety and Health method 7601. The urinary and blood samples were collected and prepared for analysis by high-performance liquid chromatography to determine the level of urinary and serum neopterin. All of the statistical analyses were carried out using SPSS 22. RESULTS: The airborne silica concentration was significantly different between two exposed and unexposed groups (P < 0.001, 0.27 [0.11] vs. 0.0028 [0.0006] mg/m3, respectively). The urinary neopterin in exposed group is significantly higher than the unexposed one (P < 0.001, 97.67 [30.24] vs. 55.52 [2.18] µmol/mol creatinine, respectively). Neopterin level of serum in exposed group is higher than the unexposed group, and there is a significant difference between them (P < 0.001, 6.90 [2.70] vs. 2.20 [1.20] nmol/l, respectively). The positive significant correlations were found between silica exposure concentration with urinary and serum neopterin (P < 0.001, r = 0.36 and 0.59, respectively). CONCLUSIONS: Considering the sensitively and easily measurement of neopterin in biological fluid and also the statistically significant positive relationships which were found between the airborne silica concentration and neopterin levels in the present study, the serum and urinary neopterin levels can be considered the potential biomarkers of silica exposure for doing further comprehensive studies in this area.

BACKGROUND: Welding can produce dangerous fumes containing various metals especially carcinogenic ones. Occupational exposure to welding fumes is associated with lung cancer. Therefore, welders in Gas Transmission Pipelines are known as a high-risk group. This study was designed to determinate the amounts of metals Cr, Ni, and Cd in breathing zone and urine of welders and to assess the possibility of introducing urinary metals as a biomarker due to occupational exposure. METHODS: In this cross sectional study, 94 individuals from Gas Transmission Pipelines welders, Iran, Borujen in 2011 were selected and classified into 3 groups including Welders, Back Welders and Assistances. The sampling procedures were performed according to NIOSH 7300 for total chromium, nickel, and cadmium and NIOSH 7600 for Cr+6. For all participants urine samples were collected during the entire work shift and metals in urine were determined according to NIOSH 8310. RESULTS: Back Welders and Assistances groups had maximum and minimum exposure to total fume and its elements, respectively. In addition, results showed that there are significant differences (P<0.05) between Welders and Back Welders with Assistances group in exposure with total fume and elements except Ni. Urinary concentrations of three metals including Cr, Cd and Ni among all welders were about 4.5, 12 and 14-fold greater than those detected in controls, respectively. Weak correlations were found between airborne and urinary metals concentrations (R2: Cr=0.45, Cd=0.298, Ni=0.362). CONCLUSION: Urinary metals concentrations could not be considerate as a biomarker for welders' exposure assessment.

In recent years, there has been a significant increase in molecularly imprinted solid-phase extraction (MISPE) technique for the purification and clean-up of environmental samples. In this study, solid-phase extraction using the imprinted polymer has been optimized with the experimental design approach for a triazine herbicide, named ametryn with regard to the critical factors such as sample pH, sample concentration, sample flow-rate, sample volume, elution solvent, washing solvent and sorbent mass. These factors were evaluated statistically and also validated with spiked drinking water samples and showed a good reproducibility over six consecutive days as well as six within-day experiments. Also, in order to the evaluate efficiency of the optimized MISPE protocols, enrichment capacity, reusability and cross-reactivity of cartridges have been studied. Finally, a selective MISPE was successfully demonstrated for ametryn with a recovery of above 90% for spiked drinking water samples. It was concluded that the central composite design could prove beneficial for aiding the MIP and MISPE development.

Among the solid-phase extraction (SPE) techniques, a novel system for a triazine herbicide named ametryn, has been developed based on a molecular imprinted polymer (MIP) phase. Through this method, the synthesis of the complementary to ametryn MIP was accomplished and the factors influencing its efficiency have been optimized. Through the optimization process, the type and the amounts of functional monomer and solvents, template amount, cross-linker, initiator as well as the polymerization temperature were considered to be evaluated. Based on the obtained results, the optimum conditions for the efficient polymerized sorbent, considering the recovery efficiency were solvent: acetonitrile, 6.41 mL; monomer: methacrylic acid, 5.41 mmol; template: 1.204 mmol; cross-linker: 27.070 mmol; initiator: 2.03 mmol; temperature: 40.86 degrees C. The optimum molar ratio among the template, monomer and cross-linker for ametryn was 1:4.49:22.48. The reversed-phase HPLC-UV was used for the ametryn determination, using an isocratic solvent delivery system (acetonitrile: H(2)O, 60:40), flow-rate of 0.8 mL min(-1) and a UV wavelength of 220 nm. In line with the obtained results, using central composite design (CCD) can increase the precision and accuracy of synthesis and optimization of MIP to ametryn and possibly other similar analogues.

Chromium is an important constituent widely used in different industrial processes for production of various synthetic materials. For evaluation of workers' exposure to trace toxic metal of Cr (III), environmental and biological monitoring are essential processes, in which, preparation of samples is one of the most time-consuming and error-prone aspects prior to analysis. The use of solid-phase extraction (SPE) has grown and is a fertile technique of sample preparation as it provides better results than those produced by liquid-liquid extraction (LLE). SPE using mini columns filled with XAD-4 resin was optimized regarding to sample pH, ligand concentration, loading flow rate, elution solvent, sample volume, elution volume, amount of resins, and sample matrix interferences. Chromium was retained on solid sorbent and was eluted with 2 M HNO(3) followed by simple determination of analytes by using flame atomic absorption spectrometry. Obtained recoveries of metal ion were more than 92%. The optimized procedure was also validated with three different pools of spiked urine samples and showed a good reproducibility over six consecutive days as well as six within-day experiments. Through this study, suitable results were obtained for relative standard deviation, therefore, it is concluded that, this optimized method can be considered to be successful in simplifying sample preparation for trace residue analysis of Cr in different matrices for evaluation of occupational and environmental exposures. To evaluate occupational exposure to chromium, 16 urine samples were taken, prepared, and analyzed based on optimized procedure.

This study was conducted to assess various environmental exposure measurements (total dust, ammonia and endotoxin) of poultry workers at the province of Isfahan, Iran. The results show that the workers who worked in enclosed systems of parent stock barns have the highest exposure to total and respirable dust: 21.3 +/- 3.2 and 4.6 +/- 0.9 mg/m3, respectively. In comparison with different ages of chicken, the highest concentration of total and respirable dust were 5.4 +/- 0.7 and 3.3 +/- 0.7 mg/m3 in the 45th day. In the above mentioned situation, the results of endotoxin concentrations were 20.6 +/- 1.1, 23.6 +/- 2.2, 21.3 +/- 1.2 and 26.8 +/- 1.8 ng/m3, respectively. Ammonia concentrations had the highest rate in enclosed systems of laying hens in winter and the 45th day of chicken age, measuring 33.2 +/- 5.2 and 20.2 +/- 3.0 mg/m3, respectively.
