RESUMO

In Cabo Verde, several endemic species are used in traditional medicine. However, no scientific studies have been conducted on the quality, efficacy, and safety of most of these plants. This study focused on establishing the botanical and chemical identification parameters required for a quality monograph of Campylanthus glaber Benth. aerial parts, a medicinal plant of Cabo Verde traditionally used to treat fever and muscular pain. In addition, in vitro antioxidant and antihyperglycemic activity, cytotoxicity, and genotoxicity were assessed for this medicinal plant. Optical microscopy, LC/UV-DAD-ESI/MS, and colorimetric assays were used for botanical, chemical, and biological studies, respectively. Cytotoxicity was assessed by the MTT assay with HepG2 cells, and genotoxicity by the Ames test. Microscopically, the xeromorphic leaf of C. glaber presents a thick cuticle (13.6-25.5 µm), thick-walled epidermal cells, anomocytic-type stomata, glandular trichomes (stalk length = 49.4-120.8 µm), and idioblasts containing calcium oxalate microcrystals. The chemical screening of aqueous and hydroethanolic extracts of this medicinal plant revealed the presence of organic acids, iridoids, phenylethanoids, and flavonoids as the main classes of marker compounds, with malic acid, citric acid, and verbascoside being the main marker compounds identified. Both extracts showed similar LC/UV-DAD/ESI-MS qualitative profiles and DPPH radical scavenger activity (IC50 = 130.9 ± 1.4; 134.3 ± 3.1 µg/mL). The hydroethanolic extract inhibited both α-amylase and α-glucosidase enzymes in a dose-dependent manner. Both extracts showed no cytotoxicity (up to 1000 µg/mL) by the MTT assay and no genotoxic potential with or without metabolic activation up to 5 mg /plate. The results obtained are an important contribution to the monographic quality assessment of C. glaber aerial parts and suggest that this medicinal plant may be safe and potentially used as an herbal drug raw material for pharmaceutical purposes.

RESUMO

INTRODUCTION: Among the Organization for Economic Co-operation and Development members, Portugal has the highest reported consumption of anxiolytics, hypnotics, and sedatives, of which a large proportion are benzodiazepines or related drugs. These are known to cause tolerance and dependence. Other drugs with hypnotic effect, such as antidepressants, antihistamines, antipsychotics, or anticonvulsants have been identified by some reports as alternatives to benzodiazepines for the treatment of insomnia. In this regard, the aim of this study was to characterize the consumption of benzodiazepines, non-benzodiazepine anxiolytic, hypnotic or sedative effect drugs and other drugs with the potential to be used off-label to treat insomnia, and the results concerning benzodiazepine consumption related indicators in the primary health care setting in the Lisbon and Tagus Valley region. MATERIAL AND METHODS: From 2013 to 2020, a census, descriptive and retrospective study was conducted. The evolution of the variables total defined daily doses, defined daily doses per 1000 inhabitants per day (DHD) and relevant indicators were characterized. Data were extracted from the SIARS platform used in the Lisbon and Tagus Valley regional Health Administration. RESULTS: There was a decrease in the consumption of benzodiazepines (from 57.44 to 63.11 DHD) and an increase of non-benzodiazepines and of drugs with potential off-label use (from 6.56 to 8.56 DHD and from 14.70 to 25.95 DHD, respectively). Among non-benzodiazepines, zolpidem was the most consumed drug, also showing an increasing trend (from 4.86 to 6.96 DHD). For the group of drugs with off-label use potential, there was an increased consumption of trazodone (from 3.81 to 7.92 DHD), mirtazapine (from 3.52 to 6.48 DHD), pregabalin (from 3.15 to 4.87 DHD), quetiapine (from 2.68 to 4.59 DHD) and gabapentin (from 1.32 to 1.90 DHD), which was only the case (or, at least, more significantly) for the lower dose formulations. The median of results of the Primary Health Care setting indicator "proportion of elderly patients without prescription of sedatives, anxiolytics, and hypnotics", was 81.0 in 2015 and increased to 84.9 in 2020. For the indicator "proportion of patients without prolonged prescription of sedatives, anxiolytics, and hypnotics", the median was 93.6 in 2019 and 94.3 in 2020. CONCLUSION: There was, overall, a decreasing trend in the dispensing of benzodiazepines in the Lisbon and Tagus Valley Region. Even though this data suggests a change in the therapeutic pattern for insomnia, more robust studies are needed to confirm this observation.

Cabo Verde's biodiversity is threatened by activities that meet human needs. To counteract this, an integration of scientific and indigenous knowledge is required, but no comprehensive list of the useful local plants is available. Thus, in this work, we assess (1) their diversity and phytogeography; (2) the role of geophysical, historical, and socio-economic factors on species distribution and uses; and (3) potentially relevant species for sustainable development. Data were obtained from flora, scientific publications, historical documents, herbarium specimens and field work. Many species were introduced since the 15th century to support settlement and commercial interests. We identified 518 useful taxa, of which 145 are native, 38 endemic and 44 endangered. The number of useful taxa is correlated with altitude and agricultural area, as well as with rural population indicators, but not with total population or socio-economic indicators such as gross domestic product. Native taxa are mostly used for fuelwood, forage and utilitarian purposes. Agrobiodiversity and traditional practices seem crucial to cope with recurrent droughts and ensure food security. Most of the introduced species do not present conservation problems, contrasting with the overuse of some native taxa. The safeguarding of native populations will ensure the sustainable exploitation of these resources and benefit the local economy.

RESUMO

Water treatment sludge was successfully thermally converted to obtain biochar as a stable material with resource potential. This research explored the application of sludge biochar as a supplementary cementitious material. The cement paste samples incorporating different amounts of sludge biochar were prepared, hardened, and analyzed for performance. The results show an improvement in hydration kinetics and mechanical properties of cement paste incorporating biochar, compared to raw sewage sludge. The mineralogical, thermal and microscopic analyses show evidence of pozzolanic activity of the biochar. The samples with 2% and 5% biochar showed higher heat release than the reference material. Specimens with 1%, 2% and 5% biochar showed a slightly higher compressive strength at 28 days compared to the reference material. Sludge conversion to biochar will incur an estimated cost of US$398.23/ton, which is likely to be offset by the substantial benefits from avoiding landfill and saving valuable cementitious materials. Therefore, this research has demonstrated that through conversion to biochar, water treatment sludge can be promoted as a sustainable and alternative cementitious material for cement with minimum environmental impacts, hence contributing to circular economy.

RESUMO

The effects of acetylcarvacrol on the reproductive performance of engorged female R. microplus were evaluated. Carvacrol was acetylated by a reaction with acetic anhydride, confirmed by the identification of its melting point and by infrared spectroscopy. Based on the median lethal concentration, females were exposed to sublethal concentrations (3.7, 4.6 and 5.0 µL/mL) of acetylcarvacrol by means of the adult immersion test. The following parameters were evaluated: female weight before oviposition, pre-oviposition period, egg mass weight, egg production index, incubation period, hatching rate, fecundity rate, percentage of reduction in oviposition, percentage of reduction in hatching and product efficacy. The greatest efficacy was observed in the group treated with the highest concentration (91.69%) due to the greater weight reduction in egg mass (34.91 ± 0.02 mg) and the lower hatching rate (7.23 ± 15.50%). Therefore, this compound is a promising alternative for the control of R. microplus infestations.

RESUMO

The aim of this study is to investigate on the compression performance of cellular Polylactic Acid (PLA) manufacturing while using Fused Deposition Modelling. Computer Aided Design (CAD) models of cellular structures are designed using the sequential addition of spherical voids with porosity content varying from 10% to 60%. The three-dimensional (3D) microstructures of cellular PLA are characterised using X-ray micro-tomography to retrieve the correlation between the process-induced defects and the cellular geometrical properties. Mechanical testing is performed under severe compression conditions allowing for the reduction in sample height up to 80%. Finite element computation that is based on real microstructures is used in order to evaluate the effect of defects on the compression performance. The results show a significant drop of the process-induced defects thanks to the use of small layer thickness. Both mechanical anisotropy and performance loss are reduced due to vanishing process-induced defects more significantly when the amount of intended porosities is large. The compression behaviour of 3D printed PLA cellular structures is then found to be only guided by the amount and distribution of the intended porosity.

RESUMO

OBJECTIVE: This study compared resting blood pressure (BP) using ambulatory BP monitoring (ABPM) responses in two groups of subjects trained in land exercise (LE) and aquatic exercise (AE), and assessed post-exercise hypotension (PEH) using ABPM, after land- and aquatic-based exercises. METHODS: ABPM (24 hours) was used to measure the baseline BP in elderly hypertensive women trained in LE and AE and the PEH induced by exercise. For this, 40 subjects were evaluated at rest and after a land- or aquatic-based exercise session (aerobic: 75% of reserve heart rate combined with resistance exercise). RESULTS: The daytime BP was lower for AE [systolic BP (SBP) 124 ± 1.0 mmHg, diastolic BP (DBP) 70 ± 1.5 mmHg] than for LE (SBP 134 ± 0.9 mmHg, DBP 76 ± 0.9 mmHg), but there were no differences at night-time. The aquatic exercise-induced PEH in the second hour was maintained at the 24th hour post-exercise. For land exercise-induced PEH, it was maintained at the 12th hour post-exercise. The SBP and DBP were lower at the 24th hour for AE than for LE. CONCLUSIONS: Elderly hypertensive people trained in AE had lower baseline BP during the daytime. SBP and DBP values were lower for individuals trained in AE, and their PEH was more rapid and longer lasting after AE.

RESUMO

The phenolic profile and antioxidant activity of Cape Verde's Artemisia gorgonum Webb and Hyptis pectinata (L.) Poit. infusions before and after in vitro simulation of the gastrointestinal digestion were determined. The LC-UV/DAD fingerprinting analysis allowed the identification of 3-O-caffeoylquinic acid, chlorogenic acid, 3,5 dicaffeoylquinic acid, 4,5-dicaffeoylquinic acid and other caffeoylquinic acids derivatives on A. gorgonum infusion, and of caffeoylquinic acid and quercetin derivatives on H. pectinata infusion. Despite some decrease in the chromatographic area of several peaks, no relevant qualitative alterations on the chromatographic profile were observed between the digested and undigested herbal infusions. Results obtained showed a decrease on the antioxidant capacity of both tested herbal infusions after the in vitro digestion. This decrease was more pronounced for H. pectinata than for A. gorgonum and was also more pronounced regarding the radical scavenging capacity than regarding the reducing capacity. After complete digestion the superoxide anion and the DPPH-radical scavenging capacities decreased ≈ 43 and 75% for H. pectinata and ≈ 31 and 70% for A. gorgonum. Despite the observed differences before and after simulated gastrointestinal digestion, both infusions still had antioxidant activity at the end of this process. Thus, the antioxidant potential of A. gorgonum and H. pectinata infusions from Cape Verde, prepared as traditionally used, seems to be kept in some extend throughout the digestive system.

RESUMO

In many applications in metallography and analysis, many regions need to be considered and not only the current region. In cases where there are analyses with multiple images, the specialist should also evaluate neighboring areas. For example, in metallurgy, welding technology is derived from conventional testing and metallographic analysis. In welding, these tests allow us to know the features of the metal, especially in the Heat-Affected Zone (HAZ); the region most likely for natural metallurgical problems to occur in welding. The expanse of the Heat-Affected Zone exceeds the size of the area observed through a microscope and typically requires multiple images to be mounted on a larger picture surface to allow for the study of the entire heat affected zone. This image stitching process is performed manually and is subject to all the inherent flaws of the human being due to results of fatigue and distraction. The analyzing of grain growth is also necessary in the examination of multiple regions, although not necessarily neighboring regions, but this analysis would be a useful tool to aid a specialist. In areas such as microscopic metallography, which study metallurgical products with the aid of a microscope, the assembly of mosaics is done manually, which consumes a lot of time and is also subject to failures due to human limitations. The mosaic technique is used in the construct of environment or scenes with corresponding characteristics between themselves. Through several small images, and with corresponding characteristics between themselves, a new model is generated in a larger size. This article proposes the use of Digital Image Processing for the automatization of the construction of these mosaics in metallographic images. The use of this proposed method is meant to significantly reduce the time required to build the mosaic and reduce the possibility of failures in assembling the final image; therefore increasing efficiency in obtaining results and expediting the decision making process. Two different methods are proposed: One using the transformed Scale Invariant Feature Transform (SIFT), and the second using features extractor Speeded Up Robust Features (SURF). Although slower, the SIFT method is more stable and has a better performance than the SURF method and can be applied to real applications. The best results were obtained using SIFT with Peak Signal-to-Noise Ratio = 61.38, Mean squared error = 0.048 and mean-structural-similarity = 0.999, and processing time of 4.91 seconds for mosaic building. The methodology proposed shows be more promissory in aiding specialists during analysis of metallographic images.

Despite of sewage sludge agricultural recycling is one of the most adequate forms for sludge final disposal, plants grown in sludge-amended areas are under the risk of heavy metal contamination. This study was aimed at studying the distribution of Cd, Cu, Zn, Mn, and Fe in corn plants grown on soil treated with the sewage sludge from Companhia Pernambucana de Saneamento (COMPESA), Pernambuco state, Brazil. The sludge was applied to an Ultisol at six doses (0, 7.7, 15.4, 29.7, 45.1, and 60.5t ha-1). Heavy metal concentrations were determined in various plant parts (stem, leaves, stalk, straw involving the cob, and grains). The results showed that addition of sludge increased the Zn uptake by plants, with Zn accumulating mainly in the straw involving the cob and leaves. This increase in Zn absorption decreased Cu concentration in plants grown in sludge-treated soils. Except for grains, Fe concentrations were increased in all the plant parts due to sludge addition. On the other hand, most Mn was mainly accumulated in stalk and stem of the corn plants. Sewage sludge application on soil cultivated with corn did provoke neither phytotoxicity nor grains contamination with the heavy metals studied.
