RESUMO

PURPOSE: To evaluate the efficacy of optimized T1-Perfusion MRI protocol (protocol-2) with whole brain coverage and improved spatial resolution using Compressed-SENSE (CSENSE) to differentiate high-grade-glioma (HGG) and low-grade-glioma (LGG) and to compare it with the conventional protocol (protocol-1) with partial brain coverage used in our center. METHODS: This study included MRI data from 5 healthy volunteers, a phantom and 126 brain tumor patients. Current study had two parts: To analyze the effect of CSENSE on 3D-T1-weighted (W) fast-field-echo (FFE) images, T1-W, dual-PDT2-W turbo-spin-echo images and T1 maps, and to evaluate the performance of high resolution T1-Perfusion MRI protocol with whole brain coverage optimized using CSENSE. Coefficient-of-Variation (COV), Relative-Percentage-Error (RPE), Normalized-Mean-Squared-Error (NMSE) and qualitative scoring were used for the former study. Tracer-kinetic (Ktrans,ve,vp) and hemodynamic (rCBV,rCBF) parameters computed from both protocols were used to differentiate LGG and HGG. RESULTS: The image quality of all structural images was found to be of diagnostic quality till R = 4. NMSE in healthy T1-W-FFE images and COV in phantom images increased with-respect-to R and images provided optimum quality till R = 4. Structural images and maps exhibited artefacts from R = 6. All parameters in tumor tissue and hemodynamic parameters in healthy gray matter tissue computed from both protocols were not significantly different. Parameters computed from protocol-2 performed better in terms of glioma grading. For both protocols, rCBF performed least (AUC = 0.759 and 0.851) and combination of all parameters performed best (AUC = 0.890 and 0.964). CONCLUSION: CSENSE (R = 4) can be used to improve the resolution and brain coverage for T1-Perfusion analysis used to differentiate gliomas.

RESUMO

The development of synthetic small molecules capable of promoting neuronal fate in stem cells is a promising strategy to prevent the decline of hippocampal function caused by several neurological disorders. Within this context, isoxazole 9 (Isx-9) has been shown to strongly induce cell proliferation and neuronal differentiation in the subgranular zone (SGZ) of the hippocampal dentate gyrus (DG), while also improving hippocampal function in healthy mice. We have recently demonstrated that Isx-9 is able to restore normal neurogenesis levels after procedural stress. Here, we further discuss these findings highlighting the importance of including a naïve group in studies investigating the effects of either restraint stress or mild chronic unpredictable stress (CUS) on adult hippocampal neurogenesis.

Purpose To develop and evaluate magnetic resonance (MR) neurography of the brachial plexus with robust fat and blood suppression for increased conspicuity of nerves at 3.0 T in clinically feasible acquisition times. Materials and Methods This prospective study was HIPAA compliant, with institutional review board approval and written informed consent. A low-refocusing-flip-angle three-dimensional (3D) turbo spin-echo (TSE) sequence was modified to acquire both in-phase and out-of-phase echoes, required for chemical shift (Dixon) reconstruction, in the same repetition by using partial echoes combined with modified homodyne reconstruction with phase preservation. This multiecho TSE modified Dixon (mDixon) sequence was optimized by using simulations and phantom studies and in three healthy volunteers. The sequence was tested in five healthy volunteers and was evaluated in 10 patients who had been referred for brachial plexopathy at 3.0 T. The images were evaluated against the current standard of care, images acquired with a 3D TSE short inversion time inversion recovery (STIR) sequence, qualitatively by using the Wilcoxon signed-rank test and quantitatively by using the Friedman two-way analysis of variance, with P < .05 considered to indicate a statistically significant difference. Results Multiecho TSE-mDixon involving partial-echo and homodyne reconstruction with phase preservation achieved uniform fat suppression in half the imaging time compared with multiacquisition TSE-mDixon. Compared with 3D TSE STIR, fat suppression, venous suppression, and nerve visualization were significantly improved (P < .05), while arterial suppression was better but not significantly so (P = .06), with increased apparent signal-to-noise ratio in the dorsal nerve root ganglion and C6 nerve (P < .001) with the multiecho TSE-mDixon sequence. Conclusion The multiecho 3D TSE-mDixon sequence provides robust fat and blood suppression, resulting in increased conspicuity of the nerves, in clinically feasible imaging times and can be used for MR neurography of the brachial plexus at 3.0 T. © RSNA, 2016 Online supplemental material is available for this article.

Adult hippocampal neurogenesis can be modulated by various physiological and pathological conditions, including stress, affective disorders, and several neurological conditions. Given the proposed role of this form of structural plasticity in the functioning of the hippocampus (namely learning and memory and affective behaviors), it is believed that alterations in hippocampal neurogenesis might underlie some of the behavioral deficits associated with these psychiatric and neurological conditions. Thus, the search for compounds that can reverse these deficits with minimal side effects has become a recognized priority. In the present study we tested the pro-neurogenic effects of isoxazole 9 (Isx-9), a small synthetic molecule that has been recently identified through the screening of chemical libraries in stem cell-based assays. We found that administration of Isx-9 for 14days was able to potentiate cell proliferation and increase the number of immature neurons in the hippocampal DG of adult rats. In addition, Isx-9 treatment was able to completely reverse the marked reduction in these initial stages of the neurogenic process observed in vehicle-treated animals (which were submitted to repeated handling and exposure to daily intraperitoneal injections). Based on these results, we recommend that future neurogenesis studies that require repeated handling and manipulation of animals should include a naïve (non-manipulated) control to determine the baseline levels of hippocampal cell proliferation and neuronal differentiation. Overall, these findings demonstrate that Isx-9 is a promising synthetic compound for the mitigation of stress-induced deficits in adult hippocampal neurogenesis. Future studies are thus warranted to evaluate the pro-neurogenic properties of Isx-9 in animal models of affective and neurological disorders associated with impaired hippocampal structural plasticity.

Biomass and lipid accumulation of heterotrophic microalgae Chlorella protothecoides by supplying mixed waste substrate of brewer fermentation and crude glycerol were investigated. The biomass concentrations of the old and the new C. protothecoides strains on day 6 reached 14.07 and 12.73 g/L, respectively, which were comparable to those in basal medium with supplement of glucose and yeast extract (BM-GY) (14.47 g/L for old strains and 11.43 g/L for new strains) (P>0.05). Approximately 81.5% of total organic carbon and 65.1% of total nitrogen in the mixed waste were effectively removed. The accumulated lipid productivities of the old and the new C. protothecoides strains in BM-GY were 2.07 and 1.61 g/L/day, respectively, whereas in the mixed waste, lipid productivities could reach 2.12 and 1.81 g/L/day, respectively. Our result highlights a new approach of mixing carbon-rich and nitrogen-rich wastes as economical and practical alternative substrates for biofuel production.

Trains of spin echoes are normally modulated by homonuclear scalar couplings. It has long been known that echo modulations are quenched when the pulse-repetition rates are much larger than the offsets of the coupling partners, because the spin systems behave as if they consisted of magnetically equivalent spins when the offsets are suppressed. This type of quenching of the echo modulations can occur when the radio-frequency (RF) pulses are ideal, that is, when they are perfectly homogeneous, properly calibrated to induce rotations through an angle, pi, and have an RF amplitude, omega(1)=-gammaB(1), that is strong compared to the largest offset, Omega(S)=omega(0S)-omega(RF), with respect to the carrier frequency. Recently, it was discovered that echo modulations can also be quenched when the RF pulses are nonideal, that is, when they are too weak to bring about an ideal rotation of the magnetization of the coupling partners, so that the effective fields associated with the RF pulses are tilted in the rotating frame. This phenomenon typically occurs when the pulse-repetition rates are much slower than the offset of the coupling partner. Under such conditions, it turns out, however, that for certain offsets, when the phase, Phi(S) (which arises from a free precession of the magnetization of the coupling partner, S, in the pulse interval, 2tau, and the pulse length, tau(pi)), approaches a multiple of 2pi, the echo modulations are restored. However, the frequencies of these echo modulations are not simply determined by the homonuclear scalar coupling, J(IS). The Fourier transforms of the echo trains (the so-called "J spectra") reveal surprising multiplet patterns, and the amplitudes of the echo modulations depend on the offsets of the coupling partners. Herein, we present a unified theory, based on an average-Hamiltonian approach, to describe these effects for two-spin systems. Experimental evidence of echo modulations in a system of two spins is presented. Experiments with three and more spins, backed up by extensive numerical simulations, will be presented elsewhere.

Singlet-states |S=(|alphabeta> - |betaalpha>)/sq.rt.2 can be excited in pairs of coupled spins I and S, first by preparing either a non-vanishing zero-quantum coherence I(+)S(-) or a state of longitudinal two-spin order I(z)S(z) and then by applying a coherent radio-frequency (RF) irradiation with a carrier frequency omega(rf) = (Omega(I) + Omega(S))/2 that lies half-way between the chemical shifts of the two spins involved. The life-times T(S) can be much longer than the spin-lattice relaxation time T(1) of longitudinal magnetization, but singlet-states are ultimately relaxed, not only by dipolar interactions between the active spins or with the external spins, but also as a result of a non-vanishing offset Deltaomega = omega(rf) - (Omega(I) + Omega(S))/2 or an insufficient amplitude of the RF irradiation that fails to fulfill the condition omega(1) >> DeltaOmega = (Omega(I) - Omega(S)). In this work, the effect of off-resonance irradiation is explored and an approximate formula for the effective relaxation rate of the singlet population is provided on the basis of perturbation theory. The qualitative features of the dependence of the relaxation rate of the singlet population on the offset Deltaomega and on the difference DeltaOmega of the chemical shifts of the two spins are illustrated by comparison with numerical simulations.

Spin locking of the nuclear magnetization of a spin with S=1 such as deuterium in the presence of a radio-frequency field under magic angle spinning (MAS) is described in terms of adiabatic modulations of the energy levels. In a brief initial period, part of the initial density operator nutates about the Hamiltonian and is dephased. The remaining spin-locked state undergoes persistent oscillatory transfer processes between various coherences with a periodicity given by the rotation of the sample. While all crystallites in the powder undergo such periodic transfer processes, the phases of the oscillations depend on the angle gamma of the crystallites. The angle gamma is the azimuthal angle defining the orientation of the unique axis of the quadrupolar interaction tensor in a rotor-fixed frame. The theory is extended to describe cross-polarization between spins S=1 and I=12 under MAS. There are four distinct Hartmann-Hahn matching conditions that correspond to four zero-quantum matching conditions, all of which are shifted and broadened compared to their spin S=12 counterparts. These matching conditions are further split into a family of sideband conditions separated by the spinning frequency. The theory allows the calculation of both shifts and broadening factors of the matching conditions, as verified by simulations and experiments.
