RESUMO

The commercial nutrient media for investigation of the biologic properties of L. monocytogenes is developed. The efficiency of the use of developed media for determination of motility and lecithinase activity of Listeria in the establishment of the isolated culture to the pathogenic species is shown. These nutrient media are developed for visually accurate registration of the motility and lecithinase activity of Listeria when identifying the isolated cultures improving the diagnostics of listeriosis.

RESUMO

Tetrahydrofolate and its derivatives, commonly known as folates, are essential for almost all living organisms. Besides acting as one-carbon donors and acceptors in reactions producing various important biomolecules such as nucleic and amino acids, as well as pantothenate, they also supply one-carbon units for methylation reactions. Plants along with bacteria, yeast and fungi synthesize folates de novo and therefore constitute a very important dietary source of folates for animals. All the major steps of folate biosynthesis and metabolism have been identified but only few have been genetically characterized in a handful of model plant species. The possible differences in the folate pathway between various plant and algal species have never been explored. In this study we present a comprehensive comparative study of folate biosynthesis and metabolism of all major land plant lineages as well as green and red algae. The study identifies new features of plant folate metabolism that might open new directions to folate research in plants.

The purpose of study is to develop a traditional Endo's growth medium with the view of suppression of swarming of Proteus vulgaris and Proteus mirabilis. The object of study was a commercial dry Endo's growth medium. The inoculation of reference strains of enterobacteria and clinical sample (feces, urine, phlegm) was implemented according the normative documents. It is demonstrated that brining into Endo's growth medium tryptophan and sodium salts of bile acids in a particular ratio of their concentrations suppressed swarming of Proteus vulgaris and Proteus mirabilis. The obtained results demonstrated that application of enhanced Endo's growth medium significantly increases its differential and diagnostic characteristics.

Auxin plays a pivotal role in virtually every aspect of plant morphogenesis. It simultaneously orchestrates a diverse variety of processes such as cell wall biogenesis, transition through the cell cycle, or metabolism of a wide range of chemical substances. The coordination principles for such a complex orchestration are poorly understood at the systems level. Here, we perform an RNA-seq experiment to study the transcriptional response to auxin treatment  within gene groups of different biological processes, molecular functions, or cell components in a quantitative fold-change-specific manner. We find for Arabidopsis thaliana roots treated with auxin for 6 h that (i) there are functional groups within which genes respond to auxin with a surprisingly similar fold changes and that (ii) these fold changes vary from one group to another. These findings make it tempting to conjecture the existence of some transcriptional logic orchestrating the coordinated expression of genes within functional groups in a fold-change-specific manner. To obtain some initial insight about this coordinated expression, we performed a motif enrichment analysis and found cis-regulatory elements TBX1-3, SBX, REG, and TCP/site2 as the candidates conferring fold-change-specific responses to auxin in Arabidopsis thaliana.

RESUMO

The testing of specificity and diagnostic effectiveness was applied to new national chromogenic growth mediums. The algorithm was developed to isolate and express identify the clinically significant and sanitary demonstrative opportunistic enterobacteria using chromogenic growth mediums.

The nosocomial pneumonia is the most prevailing form of hospital-acquired infection. It is the leading cause of mortality among all forms of hospital-acquired infections. In the departments of resuscitation and intensive therapy nosocomial pneumonia consists more than 25% of all hospital-acquired infections. The analysis of lavages of various components of apparatuses of artificial pulmonary ventilation in the departments of resuscitation and intensive therapy of surgery hospital departments demonstrated that gram-negative micro-flora inoculated more often (72.0%). During last years, this type of micro-flora plays leading part in development of severe forms of nosocomial pneumonia. The gram-negative bacteria consisted up to 38/8% of all isolated cultures. The similar micro-flora was isolated and from tracheobronchial lavages and it confirms the possibility of infection spreading by anesthetic breathing equipment.

RESUMO

Cloning of the Arabidopsis thaliana genomic DNA fragment presumably corresponding to the promoter region of the ornithine-delta-aminotransferase (OAT) gene is reported. The reporter-gene construct, containing the Escherichia coli beta-glucouronidase gene under control of the OAT gene promoter was generated. The Nicotian tabacum SR1 transformants carrying this construct were obtained. It was demonstrated that in normal conditions, expression of the reporter gene was associated with the meristems and the zones of intensive shoot growth. Possible role of the OAT gene in nitrogen metabolism and shoot development is discussed.

Nosocomial infection is most commonly registered in the traumatology and surgery units, primarily in the intensive care unit (ICU) that is considered to be an increased risk unit owing to a large number of performed manipulations and the extensive empirical use of antibiotics. In ICU patients with nosocomial pneumonias at the Traumatology Center, nonfermentative gram-negative bacteria were isolated more frequently (51%), including P. aeruginosa (35.1%), from both patients' pathological material and environmental objects. All the cultures had typical properties; the majority of isolates belonged to serovar 02. P. aeruginosa strains were found to be polyresistant to test antibiotics.

AIM: Evaluation of bacterial contamination of six hospital environment of Perinatal Center of Makhachkala as part of epidemiologic surveillance for nosocomial infections. MATERIALS AND METHODS: One hundred twenty-eight air samples from different hospital units and 344 swabs from hospital equipment, instruments, and inventory were tested. Dry nutrient media manufactured by Scientific Manufacturing Organization "Pitatelnye Sredy" were used for isolation and identification of microorganisms. Species of microorganisms was determined on the basis of complex of tinctorial, morphological, biochemical, and serologic tests. RESULTS: Significant species diversity of opportunistic microorganisms was established. Cultures of Staphylococcus epidermidis (46; 18.5%) and Staphylococcus saprophyticus (44; 17.7%) were significantly more frequently isolated from swabs from environment. CONCLUSION: Microbiological monitoring of sanitary conditions of perinatal center assists sanitary-epidemiologic control for circulation of microorganisms--potential agents of nosocomial infections.

RESUMO

Studies to design a dry chromogenic nutrient medium for the diferentiation of Klebsiella were under way, by detecting the intracellular Klebsiella genus-specific enzyme of human potential pathogenicity--5-aminosalicylate decarboxylase. The composition of the proposed medium that ensured its high sensitivity and improved its differentiating properties as compared with the known traditional media was worked through. Klebsiella are isolated and identified on the proposed medium in one step, which substantially reduces diagnosis time and material costs and takes some burden from microbiologists.

RESUMO

The effectiveness of the bacteriological study of the blood depends in many aspects on the quality of nutrient media. The currently approved nutrient media for the isolation and yield of hemocultures have low isolation rates and require prolonged time for the isolation of causative agents. A two-phase nutrient medium for the yield and isolation of hemoculture has been developed. This medium has been found to have advantages over the control "double medium" in sensitivity, yield effect, growth rate and isolation rate with respect to the causative agents of bacteriemia and sepsis. The medium is standard and ready for use. The above-mentioned advantages of the medium increase the quality of the bacteriological diagnosis of bacteriemia and sepsis.
