BACKGROUND: Data on equine lumbar pathology hardly exist in breeds other than Thoroughbreds. OBJECTIVES: To describe pathological changes of the osseous lumbar vertebral column in Warmblood horses, Shetland ponies and Konik horses. STUDY DESIGN: Descriptive post-mortem study. METHODS: The lumbar vertebral columns of 34 Warmblood horses, 28 Shetland ponies, and 18 Konik horses were examined by computed tomography (CT). Osteoarthritis (OA) of articular processes (APJs), OA of intertransverse joints (ITJs), intervertebral disc (IVD) mineralisation, impingement of spinous (SPs) and transverse (TPs) processes and spondylosis were scored. Breed differences in prevalence and severity of pathologies were analysed by linear regression analysis and by calculating Spearman's rank correlation coefficients (rs). RESULTS: In Warmblood horses, the prevalence of OA of APJs, impingement of SPs and TPs was respectively 90%, 36%, and 35%, significantly higher than in the other breeds (p < 0.001). In Konik horses, IVD mineralisation (40%) and spondylosis (10%) were more frequent than in Warmbloods and Shetland ponies (p = 0.03). Severity score for OA of ITJs was highest in Shetland ponies (p < 0.001). For impingement of SPs, severity score was highest in Warmbloods (p = 0.03), and of TPs lowest in Shetland ponies (p = 0.003). For all parameters, except for spondylosis in Shetland ponies, there was a positive correlation between percentage of vertebrae affected and age, with IVD mineralisation scores increasing faster in Konik horses (p < 0.001). In all breeds, there was also a positive relation between scores of severity and age for OA of APJs and ITJs and for IVD mineralisation, with severity scores increasing faster in Shetland ponies (p = 0.04). Strong left/right correlations of the severity scores were seen for OA of the APJ, ITJ, impingement of TPs, and paramedian spondylosis (rs = 0.74-0.86, all p < 0.001). MAIN LIMITATIONS: Clinical histories were not available. CONCLUSIONS: There are distinct breed differences between prevalence and severity of osseous pathologies of the lumbar spine. Warmblood horses have higher scores for most pathologies with IVD mineralisation being more important in Konik horses and OA of ITJs in Shetland ponies.

BACKGROUND: Lameness assessment in horses is still predominantly performed using subjective methods. Visual assessment is known to have moderate to good intra-rater agreement but relatively poor inter-rater agreement. Little is known about inter- and intra-rater agreement on the evaluation of back motion, for which no objective measurement technique in a clinical setting is available thus far. OBJECTIVES: To describe inter- and intra-rater agreement of visual evaluation of equine back mobility. STUDY DESIGN: Rater reliability study using a fully crossed design in which all horses are rated by all observers. This data is compared with objective gait analysis. METHODS: Seventy equine professionals (veterinarians and physiotherapists) and veterinary students evaluated videos of 12 healthy horses at walk and trot on a hard, straight line. Nine parameters related to back mobility were scored: general mobility, thoracic, lumbar, lumbosacral flexion and extension and left and right thoracolumbar latero-flexion. All parameters were compared with simultaneously measured quantitative motion parameters. After 1 month, six randomly chosen horses were re-evaluated by 57 observers. RESULTS: For each parameter inter- and intra-rater agreements were calculated using intra-class correlation coefficients. For all parameters, inter-rater agreement was very poor (<0.2). The mean intra-rater agreement of all observers and for all parameters was poor (~0.4) but varied between 0.0 and 0.96 for individual observers. There was no correlation between the visual subjective scoring and objective gait analysis measurements. MAIN LIMITATIONS: Horses were scored from videos and by lack of any existing (semi-) quantitative system, a custom-made system had to be used. CONCLUSIONS: The poor inter- and intra-rater agreements of visual scoring of mobility of the equine back and the disagreement between subjective and objective gait analysis data, demonstrate the need for the development and introduction of objective, quantitative and repeatable techniques to assess equine back motion.

European farms for broiler breeders often have raised slatted areas in front of the nests, but in other regions of the world no raised slatted areas are provided. This study aimed to investigate the effects of a raised slatted area on leg health, mating behaviour and floor laying behaviour. Ten groups of 33 broiler breeder hens and three males were housed in two pen types: with or without a raised slatted area in front of the nests. Each pen had one plastic and one wooden nest. Between 25 and 31 weeks of age, ten marked hens per pen were weighed and assessed weekly on foot pad dermatitis, hock burn and wounds. At the end of week 31, animals were euthanized and bone strength of the tibia and humerus of these individuals was assessed. At 24, 27 and 30 weeks of age, mating behaviour was observed for an hour per pen, noting both numbers of successful and unsuccessful copulations. The number of eggs laid in the nests and on the floor was recorded daily between 20 and 31 weeks of age. Foot pad dermatitis scores were affected by age, but not by pen type. Generally, there were only minor issues with foot pad dermatitis (scores <11 on a 0-100 scale), probably due to the young age of the hens. Body weight was not affected by pen type, while the prevalence of hock burns was too low to analyse and no difference in bone strength was found for the tibia and the humerus. Overall, mating behaviour was less frequent in pens with raised slats than in pens without raised slats (29 ±â€¯2 vs 35 ±â€¯3 times/h) and more frequent at 27 weeks of age than at 24 and 30 weeks of age (38 ±â€¯1 vs 31 ±â€¯4 and 27 ±â€¯2 times/h). The pens with raised slats had a lower percentage of floor eggs than pens without raised slats (11.2 ±â€¯0.4 vs 19.3 ±â€¯0.5%). The wooden nest was preferred over the plastic nest as on average 63% of the eggs were laid in the wooden nest. This study shows that providing raised slats decreases mating behaviour and percentage of floor eggs, although its effects on leg health remain inconclusive.

Animals in a negative affective state seem to be more sensitive to reward loss, i.e. an unexpected decrease in reward size. The aim of this study was to investigate whether early-life and current enriched vs. barren housing conditions affect the sensitivity to reward loss in pigs using a successive negative contrast test. Pigs (n = 64 from 32 pens) were housed in barren or enriched conditions from birth onwards, and at 7 weeks of age experienced either a switch in housing conditions (from barren to enriched or vice versa) or not. Allotting pigs to the different treatments was balanced for coping style (proactive vs. reactive). One pig per pen was trained to run for a large reward and one for a small reward. Reward loss was introduced for pigs receiving the large reward after 11 days (reward downshift), i.e. from then onwards, they received the small reward. Pigs housed in barren conditions throughout life generally had a lower probability and higher latency to get the reward than other pigs. Proactive pigs ran overall slower than reactive pigs. After the reward downshift, all pigs ran slower. Nevertheless, reward downshift increased the latency and reduced the probability to get to the reward, but only in pigs exposed to barren conditions in early life, which thus were more sensitive to reward loss than pigs from enriched early life housing. In conclusion, barren housed pigs seemed overall less motivated for the reward, and early life housing conditions had long-term effects on the sensitivity to reward loss.

REASONS FOR PERFORMING STUDY: Previous surveys have reported that mare and foal survival after correction of uterine torsion (UT) varies from 60 to 84% and from 30 to 54%, respectively. Furthermore, resolution via a standing flank laparotomy (SFL) has been associated with better foal, but not mare, survival. OBJECTIVES: To compare the success of SFL with other correction methods (e.g. midline or flank laparotomy under general anaesthesia; correction per vaginam). STUDY DESIGN: Retrospective analysis of clinical records. METHODS: Data on correction technique, stage of gestation, degree of rotation, survival and subsequent fertility for 189 mares treated for UT at 3 equine referral hospitals in The Netherlands during 1987-2007 were analysed. RESULTS: Mean stage of gestation at diagnosis was 283 days (range 153-369 days), with the majority of UTs (77.5%) occurring before Day 320 of gestation. After correction of UT, 90.5% of mares and 82.3% of foals survived to hospital discharge, between 3 and 39 days later, and to foaling. Multivariable logistic regression indicated that correction method and stage of gestation at UT affected survival of foals and mares. For foals, survival was 88.7% after SFL compared with 35.0% after other methods (P = 0.001). When UT occurred at <320 days, 90.6% of foals survived, compared with 56.1% at ≥320 days (P = 0.007). For mare survival, an interaction between stage of gestation and correction method was detected (P = 0.02), with higher survival after SFL (97.1%) than other methods (50.0%) at <320 days of gestation (P<0.01). When UT occurred at ≥320 days, mare survival did not differ between techniques (76.0 vs. 68.8%; P = 0.6). Of 123 mares that were bred again, 93.5% became pregnant; fertility did not differ between mares treated by SFL (93.9%) and other techniques (87.5%; P = 0.9). CONCLUSIONS: Standing flank laparotomy is the surgical technique of choice for resolving uncomplicated equine UT (i.e. with no coexisting gastrointestinal lesions) except when the stage of gestation exceeds 320 days.

OBJECTIVES: The objectives of this study were to: estimate the prevalence of extended-spectrum ß-lactamase (ESBL)- and AmpC ß-lactamase-producing Escherichia coli carriage among broiler farmers, their family members and employees; identify and quantify risk factors for carriage, with an emphasis on contact with live broilers; and compare isolates from humans and broilers within farms with respect to molecular characteristics to gain insight into transmission routes. METHODS: A cross-sectional prevalence study was conducted on 50 randomly selected Dutch broiler farms. Cloacal swabs were taken from 20 randomly chosen broilers. Faecal swabs were returned by 141 individuals living and/or working on 47 farms. ESBL/AmpC-producing E. coli were isolated and, for selected isolates, phylogenetic groups, plasmids and sequence types were determined. Questionnaires were used for risk factor analysis. RESULTS: All sampled farms were positive, with 96.4% positive pooled broiler samples. The human prevalence was 19.1%, with 14.3% and 27.1% among individuals having a low and a high degree of contact with live broilers, respectively. Five pairs of human-broiler isolates had identical genes, plasmid families and E. coli sequence types, showing clonal transmission. Furthermore, similar ESBL/AmpC genes on the same plasmid families in different E. coli sequence types in humans and broilers hinted at horizontal gene transfer. CONCLUSIONS: The prevalence among people on broiler farms was higher than in previous studies involving patients and the general population. Furthermore, an increased risk of carriage was shown among individuals having a high degree of contact with live broilers. The (relative) contribution of transmission routes that might play a role in the dissemination of ESBL/AmpC-encoding resistance genes to humans on broiler farms should be pursued in future studies.

Fish-borne zoonotic trematodes (FZTs) can cause major human health problems. The aim of this study was to quantify the transmission of parapleurolophocercous cercariae to common carp (Cyprinus carpio) and to study the effect of the density of cercariae and the density of fish on transmission with respect to the volume of water and surface area of the bottom. Fish were kept individually either as controls (n= 91) or were exposed to 250 cercariae in tubes with a volume of 25, 50, 100, 250 or 500 ml water (n= 190) with a surface area of 4, 12, 21, 30 or 49 cm2 (n= 195). The dose to which the fish were exposed was kept constant. Infection occurred in 94-100% of fish, with a mean of 15-18 metacercariae per fish and the proportion of FZTs established at 0.06-0.07 metacercariae per cercariae per fish. Neither the prevalence of infection with FZTs nor the number of metacercariae per fish nor the proportion of FZTs established were significantly associated with differences in the density of cercariae or the density of fish per ml water or per cm2 surface area. Thus, it was concluded that the transmission of cercariae to fish is independent of density.

Prevalence of, and risk factors for, carriage of extended-spectrum ß-lactamase (ESBL) -producing Enterobacteriaceae were determined for 1025 Dutch adults in municipalities with either high or low broiler densities. Overall prevalence of ESBL carriage was 5.1%. The hypothesis that individuals in areas with high broiler densities are at greater risk for ESBL carriage was rejected, as the risk was lower (OR = 0.45; p 0.009) for these individuals. Owning a horse increased the risk (OR = 4.69; p ≤0.0001), but horse owners often owned multiple species of companion animals. Routes of transmission from animals to humans in the community, and the role of poultry in this process, remain to be elucidated.

This study aimed to determine the prevalence and risk factors for methicillin-resistant Staphylococcus aureus (MRSA) on 50 Dutch broiler farms. Of 145 persons living and/or working on these farms, eight tested positive for MRSA (5.5%). Investigation of 250 pooled throat samples of broilers and 755 dust samples resulted in four farms where MRSA-positive samples were present (8.0%). All isolates belonged to the CC398 complex. Living and/or working on a MRSA-positive farm was a risk for MRSA carriage; 66.7% of people on positive farms were MRSA positive vs. 1.5% on negative farms (P<0.0001). Due to the low number of positive farms and persons, and high similarity in farm management, it was impossible to draw statistically valid conclusions on other risk factors. For broiler farming, both farm and human MRSA prevalence seem much lower than for pig or veal farming. However, MRSA carriage in people living and/or working on broiler farms is higher compared to the general human population in The Netherlands (5.5% vs. <0.1%). As broiler husbandry systems are not unique to The Netherlands, this might imply that people in contact with live broilers are at risk for MRSA carriage worldwide.

The aim of this study was to examine the dynamics of parasite specific antibody development in Trichinella spiralis and Toxoplasma gondii co-infections in pigs and to compare these with antibody dynamics in T. spiralis and T. gondii single infections. In this experiment, fifty-four pigs were divided into five inoculated groups of ten animals, and one control group of four animals. Two groups were inoculated with a single dose of either T. gondii tissue cysts or T. spiralis muscle larvae, one group was inoculated simultaneously with both parasites and two groups were successively inoculated at an interval of four weeks. Specific IgG responses to the parasites were measured by ELISA. T. gondii burden was determined by MC-PCR carried out on heart muscle and T. spiralis burden by artificial digestion of diaphragm samples. Specific IgG responses to T. gondii and T. spiralis in single and simultaneously inoculated animals showed a respective T. gondii and T. spiralis inoculation effect but no significant interaction of these parasites to the development of specific antibodies with the serum dilutions used. Moreover, our data showed that the specific IgG response levels in groups of animals successively or simultaneously co-infected were independent of a respective previous or simultaneous infection with the other parasite. Additionally, no differences in parasite burden were found within groups inoculated with T. gondii and within groups inoculated with T. spiralis. Conclusively, for the infection doses tested in this experiment, the dynamics of specific antibody development does not differ between single and simultaneous or successive infection with T. gondii and T. spiralis. However, lower parasitic doses and other ratios of doses, like low-low, low-high and high-low of T. gondii and T. spiralis in co-infection, in combination with other time intervals between successive infections may have different outcomes and should therefore be studied in further detail.

In 2005, methicillin-resistant Staphylococcus aureus was found in pig herds and in humans in contact with pigs. To determine the prevalence of, this now-called livestock-associated (LA) MRSA among pig herds in The Netherlands and to identify and quantify risk factors, an observational study of 202 pig herds was performed between 2007 and 2008. Five environmental wipes and 60 nasal swabs from each herd were collected, and microbiological analysis was performed on single environmental samples and pooled nasal samples. A herd was considered MRSA-positive if ≥1 sample tested positive. The prevalence of MRSA-positive herds was 67% in breeding herds and 71% in finishing herds. Multivariable logistic regression analysis was then performed on data from 171 breeding herds. The number of MRSA-positive herds increased from ∼30% at the start to ∼75% at the end of the study, most likely due to transmission between herds. The prevalence of MRSA increased with herd size, as ∼40% of smaller herds (<250 sows) were MRSA-positive compared to >80% of larger herds (>500 sows). Other risk factors (e.g. antimicrobial use, purchase of gilts and hygiene measures) were not significantly associated with MRSA, though associated with herd size. Herd size appeared to be a compilation of several factors, which made larger herds more often MRSA positive.

This study evaluated the effect of feeding level and protein content in feed in first- and second-parity sows during the first month of gestation on sow BW recovery, farrowing rate, and litter size during the first month of gestation. From d 3 to 32 after the first insemination, sows were fed either 2.5 kg/d of a standard gestation diet (control, n = 49), 3.25 kg/d (+30%) of a standard gestation diet (plus feed, n = 47), or 2.5 kg/d of a gestation diet with 30% greater ileal digestible AA (plus protein, n = 49). Feed intake during the experimental period was 29% greater for sows in the plus feed group compared with those in the control and plus protein groups (93 vs. 72 kg, P < 0.05). Sows in the plus feed group gained 10 kg more BW during the experimental period compared with those in the control and plus protein groups (24.2 ± 1.2 vs. 15.5 ± 1.2 and 16.9 ± 1.2 kg, respectively, P < 0.001). Backfat gain and loin muscle depth gain were not affected by treatment (P = 0.56 and P = 0.37, respectively). Farrowing rate was smaller, although not significantly, for sows in the plus feed group compared with those in the control and plus protein groups (76.6% vs. 89.8 and 89.8%, respectively, P = 0.16). Litter size, however, was larger for sows in the plus feed group (15.2 ± 0.5 total born) compared with those in the control and plus protein groups (13.2 ± 0.4 and 13.6 ± 0.4 total born, respectively, P = 0.006). Piglet birth weight was not different among treatments (P = 0.65). For both first- and second-parity sows, the plus feed treatment showed similar effects on BW gain, farrowing rate, and litter size. In conclusion, an increased feed intake (+30%) during the first month of gestation improved sow BW recovery and increased litter size, but did not significantly affect farrowing rate in the subsequent parity. Feeding a 30% greater level of ileal digestible AA during the same period did not improve sow recovery or reproductive performance in the subsequent parity.

Since the first report on methicillin resistant Staphylococcus aureus (MRSA) CC398 in pigs, several countries have determined the prevalence of MRSA-positive pig herds using different sampling and laboratory techniques. The objective of the study was to compare three sampling methods for MRSA-classification of herds. Therefore, nasal swabs of pigs and environmental wipes were collected from 147 herds with breeding pigs. Per herd, laboratory examination was done on 10 pools of 6 nasal swabs (NASAL), 5 single environmental wipes (ENVSINGLE) and one pool of 5 environmental wipes (ENVPOOL). Large differences in apparent prevalence of MRSA-positive herds between methods were found: 19.1% for ENVPOOL, 53.1% for ENVSINGLE, and 70.8% for NASAL. Pairwise comparisons of methods resulted in relative sensitivities of 26.9% (ENVPOOL vs. NASAL), 34.6% (ENVPOOL vs. ENVSINGLE), and 72.1% (ENVSINGLE vs. NASAL) with relative specificities of respectively 100%, 98.6% and 93.0%. Cohen's kappa was respectively 0.18, 0.32 and 0.55, thus varying between very poor and moderate agreement. Examination of environmental wipes is an easy and non-invasive method to classify herds for MRSA. The number of environmental wipes needed depends on e.g. required detection limits and within-herd prevalence. In low prevalent herds (e.g. herds with <3 positive pools of nasal swabs), 25 single environmental wipes are required to be 90% sure that MRSA is detected at a detection limit similar to analyzing 10 pools of nasal swabs. Individual analysis of environmental wipes is highly recommended, as pooling 5 environmental samples resulted in a substantial reduction of the apparent prevalence.

In 2005, a distinct clone of methicillin resistant Staphylococcus aureus (MRSA CC398) was found in pigs and people in contact with pigs. The structure of the pig production chain in high technology pig husbandry enables pathogens to spread during animal trading, with an increasing prevalence in herds further down the chain. The objective of this study was to quantify the effect of the MRSA status of the supplying herd on the MRSA status of the receiving herd in order to gain more insight into the role of animal trading as a transmission route for MRSA CC398. Nasal samples (60-80 pigs per herd) were collected from 38 herds; in 20 herds, environmental samples were collected as well. Ten MRSA-positive herds (based on the results of nasal swabs of 10 individual pigs per herd) from a prior study were included in the data analysis. Herds were classified as MRSA positive if at least one sample tested positive. The 48 herds were part of 14 complete (40 herds) and 4 incomplete (8 herds) pig production chains. Fifty-six percent of the herds were classified as MRSA positive. MRSA-positive herds were observed at the start (breeding herds), middle (farrowing herds) and the end (finishing herds) of the pig production chain. All of the herds in 8 chains tested MRSA positive;, all of the herds in 5 chains tested MRSA negative and in the remaining 5 chains, MRSA-positive and MRSA-negative herds were detected. Seven spa types were found, which were all previously confirmed to belong to CC398. All of the isolates were susceptible to mupirocin, linezolid, rifampicin, fusidic acid and cotrimoxazole. Resistance against tetracycline, erythromycin and clindamycin was found in 100, 74 and 76% of the isolates, respectively. Seventy-nine percent of herds with a MRSA-positive supplier of pigs were MRSA positive, whereas 23% of herds with a MRSA-negative supplier were MRSA positive (OR=10.8; 95% CI: 1.5-110.1; P=0.011). The presence of entirely MRSA-positive and MRSA-negative chains and the strong association between the MRSA status of herds and their suppliers illustrates a large risk associated with purchasing pigs from MRSA-positive herds; a top-down strategy for future control programs is, therefore, a basic requirement. However, 23% of herds with a MRSA-negative supplier were MRSA positive and furthermore, 46% of the herds at the top of the pig production chain without a supplier tested MRSA positive. This underlined the need for the identification of additional risk factors for MRSA.

An impaired reproductive performance in second parity compared to first parity sows, decreases reproductive efficiency and, perhaps, longevity of sows. This study aims to quantify the effect of live weight development and reproduction in first parity on reproductive performance of second parity sows, i.e. pregnancy rate as well as litter size. Measures of sow development (live weight at first insemination, farrowing and weaning) and reproduction (total number of piglets born, weaning to insemination interval, lactation period, number piglets weaned) were recorded on two experimental farms. Logistic regression analysis was done for the binary outcome 'non-pregnancy from first insemination after first weaning' (yes/no). General linear regression analysis was used for litter size from 1st insemination in second parity. Repeat breeders were omitted from the analysis on litter size in second parity, since a prolonged period between weaning and conception can positively influence litter size. Farms differed significantly in measures of sow live weight development and therefore data were analyzed per farm. Compared with gilts from farm A, gilts from farm B were older and heavier at: first insemination (275±0.9 days and 145±0.8kg for farm B vs. 230±0.6 days and 124±0.5kg for farm A), first farrowing (resp. 189±1.1 vs. 181±0.9kg) and first weaning (resp. 165±1.1 vs. 156±0.9kg). Weight loss during pregnancy was similar for both farms (resp. 24.9±0.7 and 23.7±1.0kg). Gilts from farm A, however, gained more weight in the period between first insemination and first weaning compared with gilts from farm B (resp. 36.1±0.8 and 20.9±1.3kg). Non-pregnancy in second parity was 11% for farm A and 15% for farm B. Litter sizes in first and second parity were, respectively, 10.7±0.1 and 11.6±0.2 for farm A and 11.8±0.1 and 11.6±0.1 for farm B. Variables associated with non-pregnancy and litter size in second parity differed between farms. On farm A, mainly sow live weight development was associated with non-pregnancy and litter size in second parity, whilst on farm B variables like total number born in 1st parity and sow line, were associated with non-pregnancy and litter size in second parity. On both farms, higher weight gain from first insemination to first weaning was associated with a decrease in non-pregnancy (odds ratio 0.7 per 10kg for farm A and 0.8 per 10kg for farm B) and on farm A with higher litter size in second parity (ß=0.42 per 10kg weight gain). Results show that sow live weight development affects reproductive performance in second parity, especially on farm A where gilts are relatively light or young at first insemination. Management of these animals should aim to optimize development at first insemination and to increase growth between first insemination and first weaning in order to optimize production in second parity.

The objective of this study was to evaluate Fourier transform infrared (FTIR) spectrometry to measure milk ketone bodies to detect hyperketonemic cows and compare this method with milk fat to protein ratio to detect hyperketonemia. Plasma and milk samples were obtained weekly from calving to wk 9 postpartum from 69 high-producing dairy cows. The reference test for hyperketonemia was defined as plasma concentration of beta-hydroxybutyrate (BHBA) >or=1,200 micromol/L. The weekly prevalence of hyperketonemia during the first 9 wk of lactation was, on average, 7.1%. Both BHBA and acetone in milk, determined by FTIR, had a higher sensitivity (80%) to detect hyperketonemia compared with milk fat to protein ratio (66%). Specificity was similar for the 3 diagnostic tests (71, 70, and 71%). In conclusion, FTIR predictions of BHBA or acetone in milk can detect cows with hyperketonemia in early lactation with a higher accuracy compared with the use of milk fat to protein ratio. Because of the high proportion of false-positive tests, there are concerns about the practical applicability of FTIR predictions of acetone, BHBA, and fat to protein ratio in milk to detect hyperketonemic cows.

The course and clinical appearance of an Eimeria species infection in chicken flocks depend on the response of an individual bird to infection and on population-dynamics of the infection in the flock. Differences in ingested numbers of oocysts may affect oocyst load in the flock and the subsequent infectious dose for not yet infected birds. To study the link between numbers of oocysts excreted by infected birds and transmission of Eimeria acervulina, experiments were carried out with 42 pairs of broiler chickens using inoculation doses with 5, 50, 500 or 50,000 sporulated oocysts. In each pair one bird was inoculated and the other bird was contact-exposed. All contact birds became infected, which occurred on average within 34h after exposure to an inoculated bird. Although a higher inoculation dose resulted in higher oocyst excretion in inoculated and contact-infected birds, only small non-significant differences in transmission rates between groups were found.

To determine methicillin-resistant Staphylococcus aureus (MRSA) carriage in poultry and slaughterhouse personnel, 40 Dutch broiler flocks, in six slaughterhouses and 466 personnel were sampled. Of the employees, 26 were positive (5.6%), indicating a higher risk of exposure when compared to the general Dutch population (0.1%). This risk was significantly higher for personnel having contact with live animals (5.2%) - especially hanging broilers on the slaughterline (20.0%) - than for all other personnel (1.9%). Conventional electric stunning conferred a significantly higher risk of MRSA carriage for employees than CO2 stunning (9.7% vs. 2.0%). A total of 405 broilers were sampled upon their arrival at the slaughterhouse, of which 6.9% were positive. These broilers originated from 40 Dutch slaughter flocks of which 35.0% were positive. MRSA contamination in the different compartments of slaughterhouses increased during the production day, from 8% to 35%. Of the 119 MRSA isolates, predominantly livestock-associated MRSA ST398 was found, although 27.7% belonged to ST9 (spa type t1430). There is an increased risk of MRSA carriage in personnel working at broiler slaughterhouses, particularly those having contact with live animals.

Coccidiosis is an economically important disease in chickens, caused by infection with Eimeria species parasites. Diagnosis of coccidiosis is frequently based on oocyst enumeration in pooled faecal samples or litter. In studies on infection dynamics and for monitoring in the field, samples from individual chickens may be more appropriate as these support the determination of infection status of individual birds and more accurately reflect oocyst output at time of sampling. Faecal samples from individual birds can be collected, but the counting procedure limits the number of samples that can be processed and unequivocal microscopic differentiation between Eimeria species is very difficult. A test that overcomes these drawbacks would improve efficiency and quality of the diagnosis. The aim of this study was to compare two methods for Eimeria oocyst quantification in samples from individual birds. A real-time PCR that quantifies oocysts in cloacal swabs (qPCR) and oocyst counts in single droppings were compared to the standard procedure of oocyst counts in bulked 24h faeces. Faecal samples were collected daily from 30 broiler chickens, inoculated with different doses of Eimeria acervulina. The three techniques produced comparable oocyst counts for all inoculation doses. Single dropping counts are applicable for small sample sizes and when a single Eimeria species is used. For larger sample sizes qPCR is preferable as it can be carried out on samples that have been frozen for storage. Furthermore, qPCR can identify and quantify different Eimeria species, which makes it a valuable diagnostic tool for field or experimental work.

Egg storage longer than 7 d is associated with a delay in hatch time and a decline in hatchability and chick quality. Prestorage incubation is suggested as a method to reduce the negative effects of prolonged storage times by altering the developmental stage of the embryo, but earlier research has shown that prestorage incubation can both be detrimental and beneficial for hatchability. The reason for these ambiguous results is not clear and the effect of prestorage incubation on chick quality is not studied extensively. The objective of this study was to investigate changes in developmental stage of embryos during prestorage incubation and the effect of prestorage incubation on hatchability and chick quality. Two experiments were conducted. In experiment I, eggs were stored for 3, 5, 8, or 12 d. In experiment II, eggs were stored for 5 or 11 d. Half of the eggs was stored immediately at 16 to 18 degrees C and the other half was exposed to prestorage incubation for 6 h in experiment I and for 4.5 h in experiment II. According to the classification table of Eyal-Giladi and Kochav (EG), embryonic development was advanced by prestorage incubation from developmental stage EG11.67 to developmental stage EG13.26 in experiment I (P = 0.02) and from developmental stage EG9.22 to developmental stage EG12.63 in experiment II (P < 0.0001). In experiment I, prestorage incubation reduced hatchability of set eggs from 59.3 to 51.5% when storage time was 12 d but did not reduce hatchability when storage time was 3, 5, or 8 d (interaction P = 0.02). Prestorage incubation increased chick length (P = 0.004). In experiment II, prestorage incubation increased hatchability of fertile eggs from 80.6 to 85.9% when storage time was 11 d but did not increase hatchability when storage time was 5 d (interaction P = 0.0009). Prestorage incubation increased percentage of second grade chicks (P = 0.0007). It seems that storage time, embryonic development at egg collection, and prestorage incubation duration determine the effect of prestorage incubation on hatchability and chick quality.
