RESUMO

Oreocharisleveilleana Fedde was collected in Ta-pin in 1910 and published in 1911. The collected location was verified within western Luodian County, Guizhou Province, China. However, there have been no records of the species' collection for more than 100 years since then. After extensive investigations by our research team on the type locality and its surrounding areas, we found that it is widely distributed in western Luodian County and eastern Wangmo County, Guizhou Province, China. During further research on the original literature, type specimens and type locality of O.leveilleana, the taxonomic position of O.leveilleana, which was once treated as a synonym of O.auricula (S.Moore) C.B.Clarke, was found to have a taxonomic problem. Through morphological research combined with geographical distribution analysis, it has been determined that it should belong to the genus Petrocodon Hance and it is the same species as P.coccineus (C.Y.Wu ex H.W.Li) Yin Z.Wang. According to the regulations and suggestions of the 2018 "International Code of Nomenclature for Algae, Fungi, and Plants (Shenzhen Code)", we propose and confirm a new combination - Petrocodonleveilleanus (Fedde) X.X.Bai & F.Wen and treat P.coccineus as a synonym of the new combination. Due to its unique bright red flowers within Petrocodon, its original Chinese name has been retained.

Hemihydrate phosphogypsum, an industrial solid waste product of phosphoric acid production, is abundant and inexpensive. If the problem of poor water resistance is solved, this material could be substituted for cement and other traditional energy-consuming cementitious materials in the construction industry. This approach would confer important economic and environmental benefits while promoting the resource utilization of phosphogypsum (PG). In this study, hydrophobic and self-cleaning coatings of H-PDMS/PMHS/OTS hybrid nanosilica were prepared on a post-hydroxylated PG surface using sol-gel and impregnation methods. The water contact angle, Fourier-transform infrared spectroscopy, Three-dimensional surface morphology and roughness analysis, X-ray photoelectron spectroscopy, scanning electron microscopy, surface abrasion tests, and tape adhesion tests were used to evaluate the hydrophobicity of the coatings. The results demonstrated that the in situ reaction produced a hydrophobic siloxane/nanosilica hybrid network that bonded to the PG surface via hydrogen bonding, making the otherwise completely hydrophilic PG hydrophobic (PGH-3, contact angle (CA) = 144.1°). The PGH-3 sample exhibited excellent chemical stability, maintaining a contact angle greater than 135° under strongly acidic or alkaline conditions. The contact angle remained at 123.7° after 50 tape-bonding tests. After 100 wear cycles, the contact angle remained at 121.9°. This study presents an environmentally friendly method and a straightforward application procedure to impart hydrophobicity to solid waste PG. Its potential is thus demonstrated in the field of PG-based construction materials and the comprehensive utilization of solid waste.

China is currently experiencing a phase of high-quality development, and fostering the resilience of the urban economy is key to promoting this development. The growth of the digital economy is seen as critical to achieving this goal. Therefore, it is necessary to study the mechanism by which the digital economy affects urban economic resilience and the impact of carbon emissions. To this end, this paper empirically analyzes the mechanisms and impacts of the digital economy on urban economic resilience using panel data from 258 prefecture-level cities in China between 2004 and 2017. The study employs a two-way fixed effect model and a moderated mediation model. The results show that: (1) The development of the digital economy can significantly improve the resilience of the urban economy in different periods and different city sizes; (2) The development of the digital economy promotes the economic resilience of developed cities and eastern cities more significantly; (3) In the context of carbon emissions, the digital economy positively contributes to urban economic resilience through population quality and industrial structure but negatively contributes to urban economic resilience through above-scale enterprises; (4) Carbon emissions have a positive moderation effect on the historical path of the industrial structure, above-scale enterprises, and the front path of population quality in the mechanism of the role of the digital economy on the economic resilience of cities, and a negative moderation effect on the front path of above-scale enterprises. Based on these findings this paper proposes several suggestions, such as revolutionizing the digital development of cities, optimizing regional industrial collaboration, accelerating the training of digital talents, and preventing the disorderly expansion of capital.

The regulation of microsomal (e.g., FAD2) and plastidial (e.g., FAD6) oleate desaturases by cold, heat and salt stress were investigated. Gene expression levels and fatty acid compositions were determined in the roots, stems and leaves of safflower following stress treatments. A safflower plastidial oleate desaturase gene, CtFAD6, was cloned, and oleic acid desaturation was confirmed in Synechococcus sp. strain PCC7942. The results showed that temperature regulated oleate desaturation at the transcriptional level, and this regulation pattern was tissue-specific. CtFAD2-1, CtFAD2-2 and CtFAD6 were significantly induced under cold and heat stress in young leaves, and CtFAD2-2 and CtFAD6 were slightly induced in young stems. In contrast, CtFAD2-1, CtFAD2-11 and CtFAD2-10 were sensitive to salt stress in all safflower tissues (roots, stem and leaves). CtFAD6 was insensitive to salt and was slightly induced in leaves only.

Presented herein is a newly designed strategy that rapidly introduces ethyl difluoroacetate radicals through a dialkylzincs induced radical 1,6-conjugate addition pathway. Besides achieving high yields and excellent functional group compatibility, this protocol allowed the incorporation of a gem-difluoromethylene motif to be accomplished within minutes.

RESUMO

A novel diethylzinc-mediated radical 1,2-addition of perfluoroalkyl iodides to unactivated alkenes and alkynes is presented, which demonstrates a novel way to generate an ethyl difluoroacetate radical. This method is highly efficient and gives full conversions of the substrates, high yields of the products, and negligible byproducts and requires no column chromatography purifications. The mild conditions enable this protocol to exhibit excellent functional group compatibility.

RESUMO

A Cu-catalyzed enantio- and diastereoselective diarylmethylation of 1,3-dicarbonyl compounds is described. It is a successful example of constructing all-carbon quaternary stereocenters from 3 °C-H nucleophiles, a challenging topic in synthetic chemistry. In the present work, two contiguous stereocenters are constructed with high levels of stereoselectivity and atom economy. The broad scope of 1,3-dicarbonyl nucleophiles and the tolerance of a wide range of functional groups make this protocol of great importance in the synthesis of chiral diarylmethane compounds.

Epstein-Barr virus (EBV) establishes lifelong latent infection in humans and is associated with several lymphoid and epithelial cancers. In nasopharyngeal carcinoma (NPC), EBV expresses few viral proteins but elevated levels of Bam-HI A rightward transcripts (BARTs) RNA, which includes viral microRNAs and long non-coding RNAs (lncRNAs). BART lncRNAs localize within the nucleus of EBV-infected cells and knockdown of BART lncRNAs significantly affects the expression of genes associated with cell adhesion, oxidoreductase activity, inflammation, and immunity. Notably, downregulation of IKAROS family zinc finger 3 (IKZF3/Aiolos), which plays a role in lymphocyte development and cell attachment, occurred in NPC C666-1 cells following BART lncRNA-knockdown. Since Aiolos expression is normally restricted to lymphoid cells and rarely observed in epithelial cells, induction of Aiolos by BART lncRNA was confirmed by expressing the major BART lncRNA isoform, RPMS1, in EBV-positive and -negative cells. BART lncRNA associated with the CBP/p300 complex and RNA polymerase II (Pol II) in the nucleus, suggesting that BART lncRNAs may mediate epigenetic regulation of gene expression through interaction with the chromatin remodeling machinery. This contention is further supported by evidence that BART lncRNA appears to stall Pol II at the promoter region and may regulate IFNB1 and CXCL8 expression by inhibiting transcription by Pol II in NPC. We hypothesize that EBV BART lncRNA expression modulates host gene expression and maintains EBV latency by interfering with histone methylation and acetylation processes. Aberrant expression of affected host genes mediated by BART lncRNA may lead to immune evasion, progression, and metastasis of NPC.

The genus Streptomyces is a widespread genus within the phylum Actinobacteria and has been isolated from various environments worldwide. However, little is known about whether biogeography affects distributional pattern of Streptomyces in salty environments. Such information is essential for understanding the ecology of Streptomyces. Here we analyzed four house-keeping genes (16S rRNA, rpoB, recA and atpD) and salty-tolerance related genes (ectA-ectD) of 38 Streptomyces strains isolated from saline environments in Yunnan and Xinjiang Provinces of western China. The obtained Streptomyces strains were classified into three operational taxonomic units, each comprising habitat-specific geno- and ecotype STs. In combination with expressional variations of salty-tolerance related genes, the statistical analyses showed that spatial distance and environmental factors substantially influenced Streptomyces distribution in saline environments: the former had stronger influence at large spatial scales (>700 km), whereas the latter was influential at large (>700 km) and small spatial scales (<700 km). Plus, the quantitative analyses of salty-tolerence related genes (ectA-D) indicated that Streptomyces strains from salt lakes have higher expression of ectA-D genes and could accumulate larger quantities of ectoine and hydroxyectoine than strains from salt mines, which could help them resist to salinity in the hypersaline environments.

The genus Nocardiopsis is a widespread group within the phylum Actinobacteria and has been isolated from various salty environments worldwide. However, little is known about whether biogeography affects Nocardiopsis distribution in various hypersaline environments. Such information is essential for understanding the ecology of Nocardiopsis. Here we analyzed 16S rRNA, gyrB, rpoB and sodA genes of 78 Nocardiopsis strains isolated from hypersaline environments in Yunnan and Xinjiang Provinces of western China. The obtained Nocardiopsis strains were classified into five operational taxonomic units, each comprising location-specific phylo- and genotypes. Statistical analyses showed that spatial distance and environmental factors substantially influenced Nocardiopsis distribution in hypersaline environments: the former had stronger influence at large spatial scales, whereas the latter was more influential at small spatial scales.

A Gram-positive, catalase- and oxidase-positive, strictly aerobic, endospore-forming rod bacterium, designated K3514(T), was isolated from the leaves of Nicotiana tabacum. The strain was able to grow at temperatures of 8-40°C, pH 5.0-10.0 and NaCl concentrations of 0-7%. The predominant quinones (>30%) of this strain were MK-7(H2) and MK-7. Phylogenetic analysis of 16S rRNA gene sequence showed that strain K3514(T) was affiliated to the genus Lysinibacillus, with its closest relatives being Lysinibacillus mangiferihumi (98.3% sequence similarity), Lysinibacillus sphaericus (97.9% sequence similarity), Lysinibacillus fusiformis (97.4% sequence similarity), and Lysinibacillus xylanilyticus (97.3% sequence similarity). However, low levels of DNA-DNA relatedness values suggested that the isolate was distinct from the other closest Lysinibacillus species. Additionally, based on analysis of morphological, physiological, and biochemical characteristics, the isolate could be differentiated from the closest known relatives. Therefore, based on polyphasic taxonomic data, the novel isolate likely represents a novel species, for which the name Lysinibacillus tabacifolii sp. nov. and the type strain K3514(T) (=KCTC 33042(T) =CCTCC AB 2012050(T)) are proposed.

OBJECTIVE: In order to explore the diversity of cultured halophilic archaeon from hypersaline environments in Lop Nur region and their potential application. METHODS: Total 13 soil samples were collected from Lop Nur regions. Halophilic archaea strains were isolated and identified by 16S rRNA gene sequence analysis. In addition, 17 strains were selected based on different branches in pylogenetic tree, and their salt concentration tolerance and amylase, protease, esterase activities were further detected by conventional methods. RESULTS: The 16S rRNA gene sequences of 56 selected strains were determined, and the phylogenetic analysis was carried out. These strains were classified into 10 known genera and 5 new potential genera, and the Shannon index was 1.820. The range of salt concentration tolerance of most strains was 10% - 35% (optimum at 20% - 25%). Amylase positive rate was 70.6%, protease positive rate was 35.3% and esterase positive rate was 82.4%. CONCLUSION: Diverse halophilic archaeon were discovered in Lop Nur regions. The isolation methods that we used were successful for isolating halophilic archaeon from these areas, which provided the technical basis to future explore the resources of halophilic archaeon in Lop Nur regions.

OBJECTIVE: To investigate the relationship between microbial community and the blight diseases in rhizosphere of carnation for biological control. METHODS: Bacterial strains isolated from the rhizosphere of healthy and blight carnation plants in greenhouse were replicated by morphology and 16S rRNA gene similarity, and investigated by using phylogenetic analysis based on 16S rRNA gene sequence alignments. RESULTS: Isolates belonged to 4 phyla of bacteria, 65 isolates from the samples of healthy carnation plants belonged to 9 genera and Bacillus, Streptomyces, Mortierella were the dominant bacteria. However, 33 isolates from the samples of blight carnation plants belonged to 12 genera and Stenotrophomonas, Sphingobacterium, Pseudomonas, Chryseobacterium, Amycolatopsis and Fusarium were only isolated from the sample of blight carnation plants. At least 13 isolates should represent potential novel species based on lower similarities of 16 S rRNA gene (90%-98% ). CONCLUSION: The result showed that either the percentage of the fungus in the total strains or the abundance of Bacillus groups in the total strains can be the referential targets to evaluate whether the carnation soil would be healthy for the carnational growth or lead to fusarium wilt diseases, accurately forecasting potential risks of the disease.

Fleeting activation of NMDA receptors (NMDARs) induces long-term modification of synaptic connections and refinement of neuronal circuits, which may underlie learning and memory and contribute to pathogenesis of a diversity of neurological diseases, including epilepsy. Here, we found that NR2A and NR2B subunit-containing NMDARs were coupled to distinct intracellular signaling, resulting in differential BDNF expression and extracellular signal-regulated kinase 1/2 (ERK1/2) activation. Selective activation of NR2A-containing NMDARs increased BDNF gene expression. Activation of NR2B-containing NMDARs led to ERK1/2 phosphorylation. Furthermore, selectively blocking NR2A-containing NMDARs impaired epileptogenesis and the development of mossy fiber sprouting in the kindling and pilocarpine rat models of limbic epilepsy, whereas inhibiting NR2B-containing NMDARs had no effects in epileptogenesis and mossy fiber sprouting. Interestingly, blocking either NR2A- or NR2B-containing NMDARs decreased status epilepticus-induced neuronal cell death. The specific requirement of NR2A and its downstream signaling for epileptogenesis implicates attractive new targets for the development of drugs that prevent epilepsy in patients with brain injury.
