RESUMO

Of intracranial tumors, primary central nervous system lymphoma (PCNSL) is rare in dogs. Herein, we describe our experience with two dogs (a 3-year-old intact female toy poodle and a 5-year-old spayed female toy poodle) that developed neurological symptoms. Magnetic resonance imaging (MRI) revealed intracranial disseminated lesions. Cerebrospinal fluid (CSF) examination revealed pleocytosis and B-cell monoclonal proliferation in both cases. PCNSL or secondary central nervous system lymphoma (SCNSL) was diagnosed on the basis of MRI findings and CSF examinations. Nimustine (ACNU) is a nitrosourea alkylating agent, a class of drugs that includes lomustine. Nimustine is mainly used to treat human intracranial neoplasia because of its high permeability across the blood-brain barrier. The dogs in this study were treated with combined chemotherapy comprising nimustine and prednisolone, which achieved complete or nearly complete remission of neurological symptoms and long-term survival (>2583 days and 1218 days), but with problematic adverse effects. We determined that the dose of nimustine for canine PCNSL or SCNSL with intravenous infusion was 25-30 mg/m2 every 3-4 weeks for a total of four times; however, the data were insufficient to determine the optimal regimen.

A 6-day-old male Japanese Black calf presented with a transverse fracture of the left calcaneus. In calcaneal fractures, traction of the gastrocnemius muscle causes substantial displacement of the proximal fracture fragment; therefore, external fixation alone is prone to failure of fusion or deformed fusion. Furthermore, internal fixation alone may result in refracture due to the high load on the implant. Therefore, internal and external fixation were used to treat the fracture. Bone fusion was observed on postoperative day 50; the wire was removed on postoperative day 90. Radiographic examination at 4 months postoperatively revealed that the bone had fused in normal alignment. Therefore, a good prognosis can be expected for calcaneal fractures treated with combined internal and external fixation.

Medical records of 16 Japanese Black calves with respiratory signs associated with perinatal rib fracture were analyzed, retrospectively, to speculate criteria of surgical intervention for the disease. For this analysis, the severity of respiratory condition was classified into three grades: Grade 1, with no wheezing, Grade 2, with wheezing after excitement but not at rest and Grade 3, with wheezing at rest. Grade 1 (n=3) received only conservative management. Seven of Grade 2 (n=8) and all of Grade 3 (n=5) had surgical management. Clinical outcome was good in all of Grade 1, and the surgical success rate was 83% in other groups. Our result suggests that surgical intervention should be considered for cases with more the severity of Grade 2.

Cryptorchidism is defined as the failure of the testis to descend into the scrotal position. Bulls with cryptorchidism have problems in both meat quality and husbandry management; thus, it is greatly important to accurately identify the retained testis and remove it during the early stage. Abdominal computed tomography (CT) was performed under general anesthesia in 34 bulls aged 3-9 months with cryptorchidism. All bulls underwent laparoscopic or incision approach for cryptorchidectomy, and 40 testes were dissected. The detection rates of retained testes were 64.5% in the abdominal cavity and 100% in the subcutaneous inguinal region, and the total detection rate was 72.5%. Furthermore, all cases in this study were suspected to have intra-abdominal cryptorchidism in primary care, but CT revealed that 22.5% of cases had cryptorchidism in the subcutaneous inguinal region. The CT value (mean ± standard deviation) of the retained testes was 20.96 ± 7.54 Hounsfield Unit, and the CT value and size of the retained testes showed a positive weak correlation with bovine age. Therefore, there is the demerit that general anesthesia and a huge device are necessary; nevertheless, CT is suggested to be useful in identifying the location of retained testes and selecting an appropriate surgical approach in bulls with cryptorchidism. Moreover, CT findings suggested that the maturation of the retained testes might depend not on the descending process but on age.

Osteochondrosis (OC) is not well recognized in cattle. Japanese Black (JB) cattle are valued for their beef and studies regarding OC are scarce. Hence, the aim of this study was to retrospectively discuss clinical and radiographical findings, diagnosis and treatment options for OC in JB cattle for successful outcomes. Medical records of 11 calves affected with OC were reviewed, retrospectively. Clinical and radiographic findings, treatment and outcomes and postmortem examination findings were analyzed. All calves had lameness. Stifle (4/14) and tarsal (4/14) were the most affected joints. Subchondral bone lucencies were observed in all joints (14/14) and accompanied with sclerosis (10/14). Radiographic grading was performed in calves. Calves were treated surgically (8/11) or conservatively (1/11). Surgically treated calves were auctioned (5/8). Postmortem examination was performed (2/11). Gross findings revealed ulcerative lesion at articular surface. Histopathologically, cartilage layer was defected at lesion. Inflammation (2/2) and granulation tissue (1/2) was also observed. In conclusion, OC in JB cattle can be diagnosed on the basis of radiography with history and clinical signs in clinical setting. Surgery should be performed in mild cases for successful outcomes. A modified radiographic grading criteria for clinical cases may hold potential in evaluating prognosis and outcomes.

Breathing patterns can be considered a vital sign providing health information. Infrared thermography is used to evaluate breathing patterns because it is non-invasive. Our study used not only sequence temperature data but also RGB images to gain breathing patterns in cattle. Mask R-CNN was used to detect the ROI (region of interest, nose) in the cattle RGB images. Mask segmentation from the ROI detection was applied to the corresponding temperature data. Finally, to visualize the breathing pattern, we calculated the temperature values in the ROI by averaging all temperature values in the ROI. The results in this study show 76% accuracy with Mask R-CNN in detecting cattle noses. With respect to the temperature calculation methods, the averaging method showed the most appropriate breathing pattern compared to other methods (maximum temperature in the ROI and integrating all temperature values in the ROI). Finally, we compared the breathing pattern from the averaging method and that from the thermal image observation and found them to be highly correlated (R2 = 0.91). This method is not labor-intensive, can handle big data, and is accurate. In addition, we expect that the characteristics of the method might enable the analysis of temperature data from various angles.

This is a histopathologic and endocrinologic study of 6 calves diagnosed with cryptorchidism. Cases 1-3 were diagnosed as resembling testicular regression syndrome. In cases 1 and 2, the extracted tissue was a small, firm, gray-white mass, and there was lack of obvious testicular tissue in case 3. Histopathologically, the excised tissue in cases 1-3 was a fibrotic testicular remnant with inflammation, mineralization, hemosiderin-laden macrophages or lipofuscin-laden macrophages, and lack of germ cells and interstitial endocrine cells. These findings were compared with cases 4-6, which were diagnosed as testicular hypoplasia due to cryptorchidism. These cases had small but otherwise grossly unremarkable intra-abdominal testicular tissue and histologically had a few germ cells and sustentacular cells with arrested spermatogenesis and an increase in interstitial endocrine cells. Cases 1-3 had more severe degenerative changes compared with cases 4-6. In case 2, the average diameter of the seminiferous tubules was much smaller than in cases 4-6, and there were few tubule cross sections. Anti-Müllerian hormone (214 pg/ml) was detected in the plasma of case 2. Based on the macroscopic and histopathologic findings as well as endocrinologic profiles, the testicular degeneration in cases 1-3 was considered similar to that of testicular regression syndrome. In this condition, it is thought that a normally developing intra-abdominal testis undergoes degeneration due to heat or a vascular disorder such as torsion.

Two-month-old (Case 1) and one-year-old (Case 2) Japanese Black cattle presented with bilateral rostral mandibular open fractures. At presentation, the proximal edges of each fracture were discolored and had a severe stench, indicating necrosis caused by infection. In both cases, a bilateral rostral mandibulectomy over the symphysis was performed. Although the tongues of both animals prolapsed post-surgery, they had no significant problems with eating or drinking. Case 1 showed excellent growth during fattening, and Case 2 successfully became pregnant and gave birth. Rostral mandibulectomy can be an effective surgical option for the treatment of cattle with difficulty in internal or external fixation due to unfavorable necrotic cranial mandibular open fractures.

A 3.5-year-old female Chihuahua was presented with complaint of neck pain, intermittent cough and dysphagia. Physical examination and diagnostic imaging of neck region revealed a solid and highly vascularized mass involving the retropharyngeal region. Histologically, the mass showed an atypical zellballen pattern which comprised of high density of type I chief cells with high nuclear cytoplasmic ratio and separated by delicate fibrovascular stroma. Immunoreactivity for neuroendocrine markers was diffusely positive in cytoplasm of tumor cells. Disseminated tumor emboli in external jugular vein were detected 6 months after initial surgery. An electron microscopic study revealed numerous electron-dense intracytoplasmic neurosecretory granules. Based on these findings, carotid body carcinoma was diagnosed.

Runt-related transcription factor 2 (Runx2) is essential for osteogenesis. This study aimes at identification of the genomic region differentially methylated in DNA for regulation of Runx2 expression. In the proximal promoter of mouse Runx2, DNA methylation was frequent at the region further than 3 kb relative to the transcription start site, in contrast to lower methylation status of the closer locus within 2 kb from the transcription start site. At the intermediate part, we identified a novel differentially methylated region in the Runx2 promoter region (Runx2-DMR): from -2.7 to -2.2 kb relative to the start site of Runx2 transcription in mice. In this region, the DNA methylation rate correlated negatively with Runx2 expression among mouse organs as well as among primary cultures of bone marrow from different dogs. Induction of mouse and dog mesenchymal-like cells into osteoblastic differentiation decreased the methylation rate of Runx2-DMR. Thus, in this study, we identified a novel genomic region in which DNA methylation status is related to Runx2 expression and detected demethylation of Runx2-DMR during osteoblastic differentiation in mouse and dog.

2.3-month-old (Case 1), one-month-old (Case 2) and 6-month-old (Case 3), Japanese Black calves presented with mild to severe wheezing. All calves had histories of dystocia at birth with breech presentation. Physical examination, thoracic radiography, endoscopy or computed tomography indicated wheezing associated with tracheal collapse and stenosis caused by perinatal rib fractures. Partial resection of the fractured first and second ribs was performed on all calves. The respiration in Cases 1 and 2 immediately improved after the surgery, while Case 3 required two weeks to improve. Cases 1 and 3 grew up healthy and were sold at auction, but Case 2 had a recurrence of wheezing at three months post-discharge and showed growth retarding. Partial costectomy may be an effective solution for control of respiration, however, further cases are required to discuss the criteria for surgical management and to obtain favorable postoperative prognosis in calves with tracheal collapse and stenosis caused by perinatal rib fractures.

The clinical utility of plasma natriuretic peptide concentrations in dogs with right-sided congestive heart failure (CHF) remains unclear. We investigated whether plasma levels of atrial natriuretic peptide (ANP) and N-terminal pro B-type natriuretic peptide (NT-proBNP) are useful for assessing the congestive signs of right-sided heart failure in dogs. This retrospective study enrolled 16 healthy dogs and 51 untreated dogs with presence (n=28) or absence (n=23) of right-sided CHF. Medical records of physical examinations, thoracic radiography and echocardiography were reviewed. The plasma concentration of canine ANP was measured with a chemiluminescent enzyme immunoassay. Plasma NT-proBNP concentrations were determined using an enzyme immunoassay. Plasma ANP and NT-proBNP concentrations in dogs with right-sided CHF were significantly higher than in healthy controls and those without right-sided CHF. The plasma NT-proBNP concentration >3,003 pmol/l used to identify right-sided CHF had a sensitivity of 88.5% and specificity of 90.3%. An area under the ROC curve (AUC) was 0.93. The AUC for NT-proBNP was significantly higher than the AUCs for the cardiothoracic ratio, vertebral heart score, ratio of right ventricular end-diastolic internal diameter to body surface area, tricuspid late diastolic flow and ratio of the velocities of tricuspid early to late diastolic flow. These results suggest that plasma ANP and NT-proBNP concentrations increase markedly in dogs with right-sided CHF. Particularly, NT-proBNP is simple and helpful biomarkers to assess the right-sided CHF.

The performance of horses undergoing regular intense exercise is adversely affected by oxidative stress. Thus, it is important to increase antioxidant production in horses in order to reduce oxidative stress. Ozonated autohemotherapy (OAHT) reportedly promotes antioxidant production. This study aimed to evaluate the effects of OAHT on antioxidant capacity. Ten Thoroughbred horses were used in this study. After the OAHT, we collected serum samples and measured biological antioxidant potential (BAP). We found that BAP began to increase after the OAHT and was significantly higher in the OAHT group than at 3 (P<0.01) and 7 days (P<0.05) after OAHT than in the control group at 3 and 7 days after starting collection of blood samples. Therefore, it was shown that OAHT improved the antioxidant capacity of the horses.

The present study evaluated the effects of single-dose filgrastim on hematology in 16 healthy horses after long-distance transportation. Horses were assigned to receive filgrastim (0.23 µg/kg, SC, once; G-CSF group; n=8) or saline (0.9% NaCl) solution (0.3 ml, SC, once; control group; n=8) ≤ 1 hr before transportation. Horses were transported 2,530 km using commercial vans over the course of approximately 44 hr. Clinical examinations and hematologic analyses were performed on all horses before and after transportation. Because the post-transportation white blood cell counts and bacillary neutrophil to segmented neutrophil ratio were significantly higher in the G-CSF group, filgrastim may have promoted the mobilization of neutrophils from marrow. Filgrastim deserves a further study for efficacy in preventing horse shipping fever.

Laparoscopic cryptorchidectomy without insufflation was applied in 10 standing bulls aged 3 to 15 months. Nine bulls were preoperatively pointed out intra-abdominal testes by computed tomography. Preoperative fasting for a minimum of 24 hr provided laparoscopic visualization of intra-abdominal area from the kidney to the inguinal region. Surgical procedure was interrupted by intra-abdominal fat and testis size. It took 0.6 to 1.5 hr in 4 animals weighing 98 to 139 kg, 0.8 to 2.8 hr in 4 animals weighing 170 to 187 kg, and 3 and 4 hr in 2 animals weighing 244 and 300 kg to complete the cryptorchidectomy. In conclusion, standing gasless laparoscopic cryptorchidectomy seems to be most suitable for bulls weighing from 100 to 180 kg.

BACKGROUND: Testicular tumours are common in elderly male dogs, and Sertoli cell tumours (SCTs) are among the most common. An increase in blood estradiol concentration is often seen in canine SCTs, but such measurements do not necessarily correlate with the clinical signs. CASE REPORT: A 6-year-old male Pembroke Welsh corgi was referred for nonpruritic alopecia. Clinical examination revealed cryptorchidism of the right testicle, and blood tests showed an increased estradiol concentration. The cryptorchid testis was removed by laparotomy, and SCT was diagnosed histologically. An enzyme-linked immunosorbent assay kit designed to measure human anti-Müllerian hormone (AMH) revealed a very high preoperative serum AMH concentration, which decreased after surgery. The serum AMH concentrations of two intact healthy control male dogs were lower than that of the dog with the SCT before treatment but higher than thoseof two healthy castrated male dogs. CONCLUSIONS AND CLINICAL IMPORTANCE: Canine serum AMH concentrations, as measured by a human AMH enzyme-linked immunosorbent assay, may be useful as a marker for canine SCT.

The effect of sugar supplementation with 1 g/kg BW twice a week for eight weeks on rumen protozoa was determined in ten retarded growth calves. Rumen juice was sampled by abdominal paracentesis during the experiment. Papillae development of rumens excised by experimental laparotomy was macro- and micromorphologically determined before and after sugar supplementation in a selected calf. The numbers of Entodinium, Isotricha, Dasytricha and Epidinium protozoa increased by 3 to 12 folds after 1-3 wk of supplementation and subsequently decreased. The heights of the rumen papillae after sugar supplementation showed marked development compared with before supplementation (Post vs. Pre: 4.44 ± 0.43 vs. 1.36 ± 0.24 mm). Sugar supplementation accommodates the rumen protozoa profile and stimulates papillae development in retarded growth calves.

The growth effect of sugar supplementation was determined in 49 retarded growth calves. Calves were supplemented with sugar at 1 g/kg BW 2 times weekly for 8 weeks. Glucose tolerance tests prior to the experiment showed no difference between the retarded growth calves and normal growth controls. After sugar supplementation, the calves were classified into 4 groups characterized by high (H) or low (L) periodic changes in daily weight gain (DG) with a breakpoint of 0.8 kg/d in three periods, birth to sugar supplementation (Birth-Pre), the 8 weeks during supplementation (Pre-Post) and after feeding to delivery to market (Post-Market). The periodic DG showed a marked increase after supplementation in Pre-Post and Post-Market compared with before supplementation during Birth-Pre in 2 groups (0.93 and 1.11 vs. 0.51 kg/day for L-H-H [n=19], 0.66 and 1.19 vs. 0.42 kg/day for L-L-H [n=24]), but no difference was observed in L-H-L (n=3) and L-L-L (n=3). Peripheral blood was collected on the day before supplementation (Pre), 8 weeks after supplementation (Post) and eight weeks after cease of supplementation. The blood concentrations of both insulin-like growth factor-1 (IGF-1) and glucose showed significant increases in L-H-H and L-L-H, but decreases in non-esterified fatty acid were observed in L-H-H and L-L-L on day Post compared with day Pre, respectively (p<0.05). At delivery to market, the sugar-supplemented calves had body weights similar to the market average. The growth effect of sugar supplementation could be stimulated through rumen papillae development induce by sucrose, the main component of table sugar.

Canine distemper caused by canine distemper virus (CDV) is a contagious, incurable, often fatal, multisystemic viral disease that affects the respiratory gastrointestinal and central nervous system. Strains Vn86 and Vn99 of CDV were isolated, we believe for the first time, in Vietnam from two 4-month-old autopsied dogs pathologically showing non-suppurative encephalitis with pneumonia, lymphoid depletion and severe gastroenteritis. These strains caused syncytium cytopathic effect in Vero cells and Vero cells expressing canine signaling lymphocyte activation molecules. The titers of cell-associated viruses of both strains were higher than for released viruses. Molecular analysis showed that both new isolates of CDV joined to the group of classic type that is far from the Asia 1 and Asia 2 groups. These results indicated that first isolation and characterization of canine distemper virus in Vietnam with the immunohistochemical examination of the dog.

RESUMO

A 5-year-old male mixed-bred boar-hunting dog with a Plott hound background weighing 23 kg was brought to the Veterinary Teaching Hospital, University of Miyazaki, in October 2002. The dog was diagnosed with active infection with the lung fluke Paragonimus westermani by serological testing and also by detection of parasite eggs in his feces. Subsequent examination of four other dogs working with this dog as a boar-hunting team revealed that all five dogs were infected with P. westermani.
