We have applied our amplitude-modulated flow analysis concept to extend the dynamic range to saturated analytical signals. Sample solution, the flow rate FS of which is periodically varied with a triangular control signal Vc, is merged with a reagent solution delivered at a constant flow rate FR. While the total flow rate FT is kept constant, a diluent with FT-(FS + FR) flow rate is aspirated from the third channel. The analytical signal Vd obtained downstream is processed by fast Fourier transform to obtain the amplitudes of the wave components in Vd. When the sample concentration CS is low, Vd shows a triangular profile like Vc; the sum of the amplitudes ΣA is proportional to CS. When CS is high, Vd shows a trapezoidal profile because of the Vd saturation. A linear calibration curve can also be obtained for such saturated signals by plotting CSΣA against CS. The proof of the concept is validated by applying it to spectrophotometric determinations of a dye (Methylene Blue) and colored complexes of Fe2+ - phenanthroline and Fe3+ - Tiron.

World health trends are focusing on a balanced food and beverage intake for healthy life. Refined deep-sea water (RDSW), obtained from deep-sea water collected offshore in Muroto (Japan), is mineral-rich drinking water. We previously reported that drinking RDSW improves human gut health. Here, we analyzed the effect of drinking RDSW on the gut ecosystem to understand this effect. This was a randomized double-blind controlled trial. Ninety-eight healthy adults were divided into two groups: RDSW or mineral water (control). The participants consumed 1 L of either water type daily for 12 weeks. A self-administered questionnaire and stool and urine samples were collected through the intervention. The following were determined: fecal biomarkers of secretory immunoglobulin A (sIgA), five putrefactive products, and nine short-chain-fatty-acids (SCFAs) as the primary outcomes; and three urinary isoflavones and the questionnaire as secondary outcomes. In post-intervention in the RDSW group, we found increased concentrations of five SCFAs and decreased concentrations of phenol and sIgA (p < 0.05). The multiple logistic analysis demonstrated that RDSW significantly affected two biomarkers (acetic and 3-methylbutanoic acids) of the five SCFAs mentioned above (p < 0.05). Similarly, the concentrations of urinary isoflavones tended to increase in post-intervention in the RDSW group. Constipation was significantly alleviated in the RDSW group (94%) compared with the control group (60%). Drinking RDSW improves the intestinal environment, increasing fecal SCFAs and urinary isoflavones, which leads to broad beneficial effects in human.

An automated sequential injection (SI) spectrophotometric system has been developed for evaluation of tyramine oxidase (TOD) inhibitory activity. The method is based on the inhibition of TOD that catalyzes the oxidation of tyramine substrate to produce aldehyde and hydrogen peroxide (H2O2). The produced H2O2 reacts with vanillic acid and 4-aminoantipyrine (4-AA) in the presence of peroxidase (POD) to form a quinoneimine dye, the absorbance of which is measured of absorbance at wavelength of 490 nm. The decrease of the quinoneimine dye is related to an increase of TOD inhibitory activity. Under the optimum conditions: 1.0 mM tyramine, 8 U mL(-1) TOD, 1.0 mM vanillic acid, 1.0 mM 4-AA and delay time of 10 s, some flavonoid compounds were examined for the TOD inhibitory activity expressed as IC50 value. It was found that flavonols (quercetin and myricetin) and flavans (epicatechin gallate (ECG) and epigallocatechin (EGC)) showed higher TOD inhibitory activity than flavones and flavanones. The results of IC50 values obtained from the proposed method and a batch-wise method were not significantly different from each other. Moreover, the SI system enabled automation of the analysis, leading to more convenient, more sensitive and faster analysis than the batch-wise method. A precise timing of the system also improves precision and accuracy of the assay, especially when the measurement of absorbance at non-steady state condition is involved.

A sequential injection (SI) spectrophotometric method with absorbance detection at 475 nm has been developed for evaluating activity of some compounds on an inhibition of the mushroom tyrosinase. The method involved a reaction of 3,4-dihydroxyphenylalanine (L-DOPA) and mushroom tyrosinase to form the o-dopaquinone. The decrease of the o-dopaquinone was related to an increase of tyrosinase-inhibitory activity. Under the optimum conditions (concentration and volume of L-DOPA and mushroom tyrosinase of 2.0 mM, 60 µL and 142 U mL(-1), 15 µL, respectively), some antioxidant compounds were examined for the tyrosinase-inhibitory activity. A batch enzymatic assay of tyrosinase-inhibitory activity was applied as the reference method for comparison. The results of IC(50) values obtained from the proposed method and the batch method were correlated well, with r(2) of 0.969. The SIA provides higher precision and degrees of automation, consumes smaller amounts of chemicals and it is simpler and faster than the batch method.

RESUMO

A new type of a reactor column, a crushed BaSO(4) reactor column used for the flow injection spectrophotometric determination of sulfate ion using the exchange reaction of sulfate ion and barium-dimethylsulfonazo III is proposed. The column is very simple and economical. It can be continuously used for 8h before washing with water for repeated usage of at least 1 month. The procedure is sensitive. Application to various water samples was demonstrated.
