RESUMO

Osteoclasts are pivotal in regulating bone metabolism, with immune cells significantly influencing both physiological and pathological processes by modulating osteoclast functions. This is particularly evident in conditions of inflammatory bone resorption, such as rheumatoid arthritis and periodontitis. This review summarizes and comprehensively analyzes the research progress on the regulation of osteoclast formation by immune cells, aiming to unveil the underlying mechanisms and pathways through which diseases, such as rheumatoid arthritis and periodontitis, impact bone metabolism.

Leaf angle is determined by lamina joint inclination and is an important agronomic trait that determines plant architecture, photosynthetic efficiency, and crop yield. Cytokinins (CKs) are phytohormones involved in shaping rice (Oryza sativa L.) architecture, but their role in leaf angle remains unknown. Here, we report that CK accumulation mediated by rice CK OXIDASE/DEHYDROGENASE3 (OsCKX3) controls lamina joint development and negatively regulates leaf angle. Phenotypic analysis showed that rice osckx3 mutants had smaller leaf angles, while the overexpression lines (OsCKX3-OE) had larger leaf angles. Histological sections indicated that the leaf inclination changes in the osckx3 and OsCKX3-OE lines resulted from asymmetric proliferation of the cells and vascular bundles in the lamina joint. Reverse transcription quantitative PCR, promoter-fused ß-glucuronidase expression, and subcellular localization assays indicated that OsCKX3 was highly expressed in the lamina joint, and OsCKX3-GFP fusion protein localized to the endoplasmic reticulum. The enzyme assays using recombinant protein OsCKX3 revealed that OsCKX3 prefers trans-zeatin (tZ) and isopentenyladenine (iP). Consistently, tZ and iP levels increased in the osckx3 mutants but decreased in the OsCKX3 overexpression lines. Interestingly, agronomic trait analysis of the rice grown in the paddy field indicated that osckx3 displayed a smaller leaf angle and enhanced primary branch number, grain size, 1,000-grain weight, and flag leaf size. Collectively, our results revealed that enhancing CK levels in the lamina joint by disrupting OsCKX3 negatively regulates leaf angle, highlighting that the CK pathway can be engineered to reduce leaf angle in rice and possibly in other cereals.

Promotion of crop yield by compost application is generally thought to be ascribed to a better supply of macro and micronutrients, however the importance of compost DOM on plant growth has not been well demonstrated. In this study, composting of chicken manure, spent mushroom and sawdust was conducted under aerobic or anaerobic condition to determine the effects of compost DOM on plant growth. It was found that dissolved organic matter (DOM) first increased and then decreased in compost, and DOM of anaerobic compost was slightly higher than that of aerobic compost. When compost extract was applied to maize, among N, P, K and DOM content, it was DOM content that was most significantly and strongly related to plant biomass (r = 0.843, p<0.001). Compost DOM was also strongly related to soil properties, the improvement of which can also promote plant growth. Compost application confirmed that higher compost DOM results in greater plant biomass. In order to facilitate compost DOM accumulation, we designed a novel composting process which combined aerobic and anaerobic treatments, and the resulting compost (A-Ana compost) with the highest amount of DOM displayed the best performance in promotion of plant growth. A-Ana compost was able to increase maize biomass by 32.71% and 12.40% compared with only anaerobic or aerobic compost, respectively. Therefore, DOM is a critical factor determining compost quality and it is feasible to manipulate composting oxygen supply condition to increase compost DOM, which will lead to increased plant yield.

Excessive cadmium (Cd) in rice grain has become a major global public health problem. Here, the effect of foliar glycerol application on Cd accumulation in brown rice was examined. Various spraying concentrations of glycerol between 0.4mM and 50mM were investigated and the results showed that 0.8 mM was the best application concentration for decreasing Cd content in brown rice. After different application period experiment, filling stage was considered as the optimal spraying time. 0.4mM-5mM glycerol application one time at the filling stage could significantly reduce Cd concentration in brown rice by 28.5%-60.4%. Cd transport factors (the ratio of brown rice and flag leaf/node) were decreased by 48.5% and 27.3%, respectively, with glycerol application. Glycerol application also significantly increased Cd concentration in soluble fraction in flag leaf while reduced inorganic Cd and water-soluble Cd in both flag leaf and stem. Our results showed foliar spraying glycerol inhibited Cd transport to brown rice through Cd compartmentalisation in the vacuole and transformation of cadmium chemical form. This study may provide a new method to effectively alleviate the problem of excessive Cd in rice.
